ABSTRACT

Objective: To investigate the chemical constituents of Gracilaria lemaneiformis. Methods: Compounds were isolated by normal phase silica gel and Sephadex LH-20 gel column chromatography. Their structures were elucidated by analyses of spectroscopic data. Results: Nine compounds were isolated from the ethanol extract of G. lemaneiformis, their structures were elucidated as palmitic acid (1), 1, 3-dipalmitin (2), 1-monopalmitin (3), uracil (4), adenosine (5), 2α-oxa-2-oxo-5α-hydroxy-3,4-dinorcholestane (6), 1-O-β-D-galactosyl-(6→1)-α-D-galactosyl-2, 3-O-dihexadecanoyl- glycerol (7), 1-O-α-D-glucose-2, 3-O-dihexadecanoyl-glycerol (8), and β-sitosterol (9). Conclusion: All of these compounds are obtained from this plant for the first time.

ABSTRACT

In this paper, in vitro anti-influenza virus activities of sulfated polysaccharide fractions from Gracilaria lemaneiformis were investigated. Cytotoxicities and antiviral activities of Gracilaria lemaneiformis polysaccharides (PGL), Gracilaria lemaneiformis polysaccharide fraction-1 (GL-1), Gracilaria lemaneiformis polysaccharide fraction-2 (GL-2) and Gracilaria lemaneiformis polysaccharide fraction-3 (GL-3) were studied by the Methyl thiazolyl tetrazolium (MTT) method, and the inhibitory effect against Human influenza virus H1-364 induced cytopathic effect (CPE) on MDCK cells were observed by the CPE method. In addition, the antiviral mechanism of PGL was explored by Plaque forming unit (PFU), MTT and CPE methods. The results showed: i) Cytotoxicities were not significantly revealed, and H1-364 induced CPE was also reduced treated with sulfated polysaccharide fractions from Gracilaria lemaneiformis; ii) Antiviral activities were associated with the mass percentage content of sulfate groups in polysaccharide fractions, which was about 13%, in polysaccharides (PGL and GL-2) both of which exhibited higher antiviral activity; iii) A potential antiviral mechanism to explain these observations is that viral adsorption and replication on host cells were inhibited by sulfated polysaccharides from Gracilaria lemaneiformis. In conclusion, Anti-influenza virus activities of sulfated polysaccharide fractions from Gracilaria lemaneiformis were revealed, and the antiviral activities were associated with content of sulfate groups in polysaccharide fractions.

ABSTRACT

The inhibitory effect of the crude phycoerythrin(PE) on the frequency of PCE micronucleus induced by CP and its antitumor effect on mice bearing S 180 sarcoma were studied. The result showed that the PE could decrease significantly the frequency of PCE micronucleus with a fair dose-effect relationship, and showed obvious antitumor effect on S 180 bearing mice with an inhibitory rate 44%.

ABSTRACT

Objective To investigate the composition and cytotoxic activity of the organic acids extracted from a red alga Gracilaria lemaneiformis.Methods Growth inhibition rate of the extract on HeLa cell was determined by MTT assay.The composition and contents of the organic acids were analysed by GC-MS after methylied.Results The organic acids from Gracilaria lemaneiformis showed potent cytotoxic activity against HeLa cell,IC_(50)=6.5?g? mL~(-1).Thirteen compounds were characterized from methylied organic acids.The major constituent was palmitic acid methyl ester (61.34%).Their relative acids were mainly C_(16)or C_(18)fatty acids and phthalates.Of them,palmitic acid,stearic acid,palmitoleic acid,oleic acid and linoleic acid have been reported to show antitumor activity.Conclusion The fatty acids mentioned above maybe a part of antitumor components of Gracilaria lemaneiformis.

ABSTRACT

Objective To investigate the antitumor effects of phycoerythrin from Gracilaria lemaneiftheormis.Methods The coarse phycoerythrin(CPE) was ig administered to sarcoma180 mice,then the antitumor effect of CPE in vivo was evaluated by the inhibition rate,thymus and spleen indexes.The growth inhibitory effect of phycoerythrin(PE) on cell lines HeLa,EC109,and vascular endothelial in vitro was examined with MTT assay.Influences on HeLa cell cycle and apoptosis of PE were observed by flow cytometry and annexin v-fitc/PI fluorescence staining.Results CPE exhibited inhibitory effect against sarcoma180 growth in mice at the dosage of 100~300 mg?kg-1with a fair dose-effect relationship,and the inhibitory rate was 49.23% at the dosage of 300 mg? kg-1.CPE could also obviously increase the weight of thymus;PE could obviously inhibit the growth of HeLa cell in a dose-effect relationship,the inhibitory rate could reach more than 70%.It could prevent the transforming of HeLa cell cycle from G2/M to S phase to cause apoptosis.But PE couldn't inhibit the growth of EC109 cell,while some growth stimulating effects of PE on vascular endothelial cell were observed.Conclusion The PE from Gracilaria lemaneiftheormis could observably inhibit sarcoma180 growth in vivo.Effects of PE on different cells in vitro were differ,but showed potent inhibitory effect to HeLa cell.

ABSTRACT

Objective To investigate the effects of antitumor and antioxidation of polysaccharide from Gracilaria Lemaneiformis(PGL) in sarcoma180 bearing mice.Methods The PGL extracted with hot water was administered to Sarcoma180 mice,then the superoxide dismutase(SOD) activity in red blood cell and lipoperoxide(LPO) content of liver homogenate of Sarcoma180mice were determined to estimate the antioxidant effect of PGL. Results PGL could significantly inhibit the growth of Sarcoma180 in mice.At the dose of 150mg?kg-1,ig,the inhibition rate was 63.56%.PGL obviously decreased LPO content of liver homogenate in Sarcoma180 mice.Conclusion The polysaccharide from Gracilaria Lemaneiformis could increase the antioxidant effect of Sarcoma180 mice,and it may be one of the mechanisms to realize its antitumor activivy.
