ABSTRACT
Aim To observe the effects of active com-ponent of Radix Isatidis ( ACRI ) on anti-anoxia and anti-fatigue in mice and investigate its possible mecha-nism. Methods Based on the weights, the mice were randomly divided into 5 groups: blank control group, ACRI 25, 50 and 100 mg·kg-1 groups, positive drug ( American ginseng liquid) control group 3 mL·kg-1 . Drugs were administered to the mice for about 14 con-secutive days, and during the experiment general situa-tions of mice were observed. The experiment of bearing hypoxia at normal pressure and the experiment of swim-ming while weight-bearing were conducted to study the effect of ACRI on anti-anoxia and anti-fatigue in mice. Then the superoxide dismutase ( SOD ) activities, the content of maleic dialdehyde ( MDA ) of mice serum and liver and blood urea nitrogen, blood lactic acid, liver glycogen were detected, in order to investigate its mechanism. Results ACRI decreased the growth rate of body weight in mice significantly, obviously pro-longed the survival time of anoxic mice at normal pres-sure and the swimming time of loaded mice, enhanced the SOD activities of mice blood and liver, decreased the MDA content of mice blood and liver, increased the content of liver glycogen, and decreased the blood urea nitrogen and blood lactic acid in mice after swim-ming. Conclusion ACRI has the anti-anoxia and anti-fatigue functions.

Objective To explore effects of ephedrine on epinephrine (EPI) and estradiol( E2)concentration in blood, histological structures and activity of superoxide dismutase (SOD) and maleic dialdehyde (MDA) content in the ovary of female mice.Methods Pregnant female mice in the experimental group were treated with ephedrine (2g/L,4g/L, 6g/L), and the mice in the control group were treated with saline by intraperitoneal injection .Using ELISA method to detect concentration changes of EPI and E 2 in plasma, the changes in the enzymatic activity of SOD and MDA content were analyzed by colorimetry .The ovarian weight was measured , histological structures in ovary were observed by microscopy and the number of ovarian follicles were counted .Results EPI concentration in plasma of female mice increased , and the E2 concentration decreased dose-dependently in the experimental group .The activities of SOD were lower , and MDA content significantly increased compared to the control .The ovary weight of experimental groups was lower than the control group.The numbers of primary follicle , secondary follicle and atretic follicle increased and corpus luteum decreased in the experimental groups .There were pyknotic ovarian tissues , including oocyte ateophy , degradation granulose cells , and karyopyknotic granulose lutein cells in the experimental groups .Conclusion Ephedrine has a great effect on the ovarian histological structure and ovarian function of female mice , which may be correlated with the low activities of SOD .

Objective To compare the changes in nitric oxide (NO), total ability of anti-oxidize antioxidase, superoxide dismutase (SOD), glutathion peroxidase (GSH-PX), catalase (CAT) and maleic dialdehyde(MDA) in rats after single and accumulative 60Co γ-irradiation . Methods 48 rats were randomly divided into two groups of single and accumulative irradiation. Each group was irradiated by 60Co gamma-rays. The total irradiation doses were 0.4, 0.8, 1.6, 3.2 and 6.4 Gy, respectively in each group. The NO, total ability of anti-oxidize antioxidaze, SOD, GSH-PX, CAT and MDA in serum were measured at day 1 after last irradiation. Results Compared with the single irradiation group, the NO (52.6-117.9 μmol/ml), total ability of anti-oxidize antioxidaze (3.3-26.2 U/ml), the antioxidase activity of the SOD (26.3-167.5 U/ml), GSH-PX (740.8-2462.4 U/ml), CAT (3.3-29.4 U/ml) and the content of MDA(29.3-155.1 nmol/ml) of mt serum in accumulative irradiation group were increased after irradiation, which was related with the accumulative irradiation dose. For instance, total ability of anti-oxidize antioxidase (26.2 U/ml), antioxidase activity of the SOD (167.5 U/ml) and CAT (29.4 U/ml) in 0.4 Gy group of accumulative irradiation were significandy increased when compared with those of control group. However, the content of MDA in accumulative irradiation group was obviously higher than that in single radiation group when the irradiation doses delivered over 3.2 Gy, which might be correlated with higher antioxidase activity. Conclusions Low dose of accumulative gamma-rays irradiation can induce the stimulative effect of antioxidase activity. However, higher dose of accumulative gamma-rays irradiation can damage the activity of antioxidase.

Objective To explore the joint toxic effect of formaldehyde and benzene on testicles of the mice. Methods 78 Kunming male mice were divided into groups, in the formaldehyde treatment groups were exposed at dosage of 0.2 mg/kg,2 mg/kg,20 mg/kg, in the benzene treatment groups were exposed at dosage of 100.0 mg/kg,200.0 mg/kg,400.0 mg/kg and the in formaldehyde and benzene combined treatment groups were exposed at dosage of 0.1+50.0 mg/kg,1+100.0 mg/kg,10+200.0 mg/kg respectively. The treatments were conducted by peritoneal injection for five consecutive days,once a day,then killed on 6th day. The activity of superoxide dismutase(SOD),the contents of maleic dialdehyde(MDA),the contents of copper(Cu),zinc(Zn) in testicles of the mice were determined. [Objective] To observe the curative effects and the influence of activity of Superoxide dismutase(SOD) and contents of Maleic dialdehyde(MDA) of Qingchangyukui Gel in Rats with Experimental Ulcerative Colitis. [Method]Ulcerative Colitis model was established in Wistar rats by injecting trinitro-benzene-sulfonic acid (TNBS)into anus, and then treated with Qingchangyukui Gel and salisy-lazosulfa-pyridine(SASP) respectively. Isolating the colon and measuring the SOD and MDA after treating for two weeks. [Results]Colon mucous membrane in the model group had serious erosion, ulcer, damaging of glandular organ, high inflammation grade; activity of SOD in model group was significantly lower than in normal group and contents of MDA increased. The other groups, especially in Qingchangyukui Gel group, had different levels of inflammation repairing, ulcer healing, increasing of SOD and reducing of MDA. [Conclusion]TNBS can cause Colon mucous membrane inflammation and ulcer, reducing of SOD and increasing of MDA , which are the typical pathological change of Ulcerative Colitis; the Qingchangyukui Gel has the effects of increasing the activity of SOD and reducing contents of MDA, therefore it can effectively treat the UC rat.

Objective To prove insulin resistance-induced by free fatty acid(FFA)with hyperinsulinemic-euglycemic clamp and assess the prevention from this resistance by N-acetylcysteine(NAC).Methods Twenty-four female Wistar rats were randomly divided into 4 groups,each of which received infusion of IH(20% intralipid +20 U/mL heparin),IH+NAC,NAC,and saline respectively for 7 h.Two-hour hyperinsulinemic-euglycemic clamp was made after 5h infusion.During the infusion and clamp the rats were awake.Blood samples were taken for measuring insulin,FFA and maleic dialdehyde(MDA)at 0 min,280 min,290 min,300 min,340 min,380 min,400 min,410 min and 420 min.The insulin sensitive index was calculated.Results Plasma FFA and MDA levels in IH group were significantly elevated compared with those in control,IH+NAC and NAC groups,P

Objective To observe the elimination of oxygen free radicals in rat myocardium mitochondria by diazoxide(DE) during different duration of hypothermic preservation.Methods The Langendorff model of isolated rat heart was used.After stored in 4℃ Celsior solution or Celsior solution containing 30 ?mol/L of DE for different duration(0,3,6,9 or 12h),the activity of the superoxide dismutase(SOD) and the content of maleic dialdehyde(MDA) in heart mitochondrial were measured.The myocardial mitochondria ultrastructure was also evaluated.Results(1) After hypothermic preservation,the activity of SOD was decreased,the content of MDA was increased.The ultrastructure of myocardium mitochondria was also destrored in preserved rat hearts in a time-dependent manner.(2) Compared with the corresponding Celsior solution hypothermic preservation groups,DE significantly improved the above indexes(SOD,MDA and myocardial mitochondria ultrastructure).Conclusion Diazoxide can protect rat myocardial mitochondria from long-term hypothermic preservation injury by direct elimination of oxygen free radials and enhancement of the activity of SOD.

Objective: To study the effects of pharmaco-serum from rabbits administrated intragastrically with Fuzheng Jiedu decoction(FJD) on injured human bronchial epithelial(16HBE) cells exposed to nickel sulfate(NiSO4).Methods: Cultured 16HBE cells were treated with both NiSO4 and different doses of FJD pharmaco-serum in vitro.The detections of cell activities and viabilities were carried out.Meanwhile,activities of glutathione peroxidase(GSH-Px) and superoxide dismutase(SOD) as well as maleic dialdehyde(MDA) contents were measured.Results: FJD pharmaco-serum could decrease mortalities and increase viabilities of 16HBE cells exposed to NiSO4.In cells co-treated with NiSO4 and FJD pharmaco-serum,the content of MDA was decreased,while GSH-Px and SOD activities were increased at the same time.High dose of FJD pharmaco-serum had the most dramatic effect.Conclusion: This study suggested that FJD could antagonize NiSO4-induced toxicity,which may be involved in its antioxidant function.

Objective To study the oxidative stress state of mice before and after air simulated diving. Methods Forty-eight mice were divided into 6 groups randomly. Every group was exposed to corresponding pressure for 60 minutes, twice a day for consecutive 3 days. Blood was obtained at the corresponding time to observe the changes of maleic dialdehyde, glutathione, glutathione-s-transferase, glutathione peroxidase, superoxide dismutase and catalase. Results Compared with the control, the activity of glutathione-s-transferase, glutathione peroxidase, superoxide dismutase, catalase and the level of glutathione were markedly inhibited and maleic dialdehyde increased in HO group and HA1 group (P
