ABSTRACT

BACKGROUND:In recent years,it has been found that some traditional Chinese medicine monomers can alleviate oxidative stress and apoptosis of the skin flap, promote vascular regeneration of the skin flap and prevent skin flap necrosis by activating autophagy. OBJECTIVE:To review the research progress of traditional Chinese medicine monomer regulating autophagy in preventing flap necrosis. METHODS:The Chinese and English key words were"traditional Chinese medicine(TCM),autophagy, skin flaps".The first author searched the relevant articles published in CNKI and PubMed databases from January 2010 to October 2022.A total of 196 articles were retrieved in the preliminary screening and then screened according to the inclusion and exclusion criteria. The quality assessment was conducted by reading the literature titles and abstracts.Finally,55 articles were summarized. RESULTS AND CONCLUSION:(1)The regulation of autophagy is mediated by AMPK/mTOR,PI3K/AKT and other signaling pathways.Activation of autophagy can alleviate the oxidative stress and apoptosis of the flap, promote the regeneration of blood vessels in the flap, and prevent flap necrosis.(2)Terpenoids(Betulinic acid,Andrographolide,Notoginseng Triterpenes,Catalpa),phenolic compounds(Resveratrol,Curcumin,Gastrodin),phenolic acids(Salvianolic acid B)and steroid compounds(Pseudoginsenoside F11)in traditional Chinese medicine monomers can alleviate oxidative stress and apoptosis of skin flap by regulating related signaling pathways to activate autophagy, promote skin flap angiogenesis and promote skin flap survival.(3)Studying the research progress of traditional Chinese medicine monomer to prevent flap necrosis by regulating autophagy can provide a reference and theoretical basis for traditional Chinese medicine to prevent flap necrosis and promote flap healing in the clinic.

ABSTRACT

BACKGROUND:Sarcopenia is a progressive, generalized skeletal muscle disease that is closely related to the occurrence of osteoarthritis, fractures, limb disability and death in the elderly.Establishing animal models of sarcopenia is essential to understand the pathophysiology of sarcopenia and to identify effective treatment strategies. OBJECTIVE:To review the evaluation criteria of mouse models of sarcopenia and the modeling methods of mouse models of sarcopenia, and to analyze and compare the advantages and disadvantages of various modeling methods,in order to provide reference for the research and diagnosis and treatment of sarcopenia. METHODS:"Sarcopenia, skeletal muscle aging, mouse model, animal model"in Chinese and English were used as Chinese and English search terms, respectively. The search formula was"(sarcopenia OR skeletal muscle aging)AND(mouse model OR animal model)."CNKI,WanFang and PubMed were searched for related articles published from January 2010 and October 2022.A total of 59 articles were finally included for analysis. RESULTS AND CONCLUSION:There is a faster modeling time in SAMP8 mice and the type of muscle atrophy is consistent with that of patients with sarcopenia. Therefore,it is an ideal model.Although the surgical method can successfully induce muscle atrophy,it requires precise surgical operation, which is difficult and time-consuming.Hindlimb suspension modeling in mice is similar to that of the elderly and can be regarded as an effective model of senile sarcopenia.Although reagent injection molding is simple to perform, both the dose and number of days of administration of reagents are not clear and need to be further investigated.Transgenic mouse models are less commonly used and their model stability needs further study. The search for a mouse model with low cost, short time consumption and high simulation of human sarcopenia is still a future research direction.

ABSTRACT

BACKGROUND:Osteoarthritis is a chronic degenerative disease of skeletal muscle, and the incidence of osteoarthritis is increasing yearly, but its pathogenesis is not clear. The commonly used drugs for osteoarthritis include non-steroidal anti-inflammatory drugs and glucocorticoid.Adverse drug reactions to these drugs can reduce patient compliance and ultimately affect the efficacy of treatment.Active ingredients in Chinese medicine are closely scrutinized for their safety and other characteristics. OBJECTIVE:Through reviewing the research progress in the effect of active ingredients of traditional Chinese medicine on osteoarthritis-related signaling pathways,to provide ideas and theoretical basis for the research, development, and application of new drugs in the prevention and treatment of osteoarthritis. METHODS:CNKI,WanFang,VIP,PubMed,and GeenMedical were searched for relevant literature in the last 10 years. The key words were"OA,osteoarthritis, traditional Chinese medicine, active ingredients of traditional Chinese medicine, chondrocyte, inflammation, signal pathways, mechanism"in Chinese and English. We excluded the irrelevant repetitive and older literature, and finally included 63 articles for further review. RESULTS AND CONCLUSION:The active ingredients of traditional Chinese medicine regulate the levels of inflammatory factors, promote the proliferation and differentiation of chondrocytes, regulate the apoptosis of chondrocytes, and delay the degeneration of chondrocytes through Wnt/β-catenin,NK-ΚB,P38 MAPK,PI3K/AKT,JAK2/STAT3 and other signaling pathways, thus slowing the progression of osteoarthritis.

ABSTRACT

BACKGROUND:Studies have found that chondrocyte ferroptosis can be used as an important pathogenesis to induce the occurrence and development of osteoarthritis. With the continuous development of Chinese medicine, some scholars have found that some traditional Chinese medicine monomers and traditional Chinese medicine compounds can inhibit chondrocyte ferroptosis through various mechanisms of action, and finally play a role in the treatment of osteoarthritis. OBJECTIVE:To explore the relationship between ferroptosis and osteoarthritis and the mechanism by which traditional Chinese medicine deals with ferroptosis in the treatment of osteoarthritis,in order to provide new ideas for the treatment and prevention of osteoarthritis. METHODS:With"osteoarthritis, ferroptosis, chondrocytes, lipid metabolism, traditional Chinese medicine,ROS,GPX 4"as the English and Chinese search terms,CNKI,PubMed,WanFang, and VIP databases were searched for relevant articles regarding the relationship between ferroptosis and osteoarthritis and its traditional Chinese medicine intervention until 2022.Then,a systematic analysis was conducted. RESULTS AND CONCLUSION:Intracellular iron overload will cause lipid peroxidation, resulting in the accumulation of reactive oxygen species and eventually the occurrence of ferroptosis, while excessive iron will accelerate the apoptosis of chondrocytes and affect chondrocyte homeostasis, thereby inducing the development of osteoarthritis. When amino acid metabolism is abnormal,an imbalance in glutamate and cystine exchange causes the accumulation of glutamate outside the cells, and high extracellular concentration of glutamate will cause the depletion of glutathione, indirectly inhibiting the function of ferroptosis-related factors, and contributing to ferroptosis. When cellular lipid metabolism is abnormal, unsaturated fatty acids,an important component of the cell membrane, lose their diallyl hydrogen atoms, leading to the destruction of cells to induce ferroptosis, while lipid metabolism is closely involved in the progression of osteoarthritis.Traditional Chinese medicine can play a role in the prevention and treatment of osteoarthritis by inducing the occurrence of ferroptosis. The research on iron death is still immature, and further research on its action mechanism is expected to open up new ways for the clinical treatment of osteoarthritis.

ABSTRACT

BACKGROUND:The pathogenesis of osteoporosis is complex, and its essence is the weakening of bone formation and the enhancement of bone absorption caused by various reasons, resulting in the imbalance of bone metabolism. In recent years,N6-methyladenosine has been found(N6-methyladenosine,m6A)methylation can prevent and treat osteoporosis by regulating bone metabolism. OBJECTIVE:Taking the regulation of bone metabolism by m6A methylation as an entry point,to systematically sort out and summarize the research progress of m6A methylation in osteoporosis,so as to provide certain theoretical reference bases for the search of new therapeutic targets for osteoporosis. METHODS:CNKI,WanFang,VIP,PubMed,MEDLINE,Nature, and Cochrane databases were retrieved for relevant literature published from database inception to 2023.The keywords were"osteoporosis,m6A methylation, bone metabolism, bone marrow mesenchymal stem cells, osteoblasts, osteoclasts"in Chinese and English.Duplicates and obsolete non-referenced documents were excluded, and a total of 73 standard papers were included for further review. RESULTS AND CONCLUSION:m6A methylation can affect the activity and differentiation of bone marrow mesenchymal stem cells, osteoblasts, and osteoclasts through various pathways to regulate bone metabolism and prevent osteoporosis. The regulatory process of m6A methylation is extremely complex, and its related proteins play different roles in different cells.Even in the same kind of cells, the same type of proteins may have radically different roles, regulating different physiological and pathological processes.

ABSTRACT

Breast cancer is the most prevalent malignant tumor among women globally, posing a serious threat to women's health. With the establishment of staging and typing principles for breast cancer diagnosis and treatment, and the development and application of novel antitumor drugs, the survival and quality of life of breast cancer patients have been continuously improving. In China, the large base of breast cancer patients possesses unique incidence characteristics, necessitating ongoing exploration of more appropriate treatment strategies;the volume and level of clinical research are also continuously advancing. In 2023,significant clinical research results were reported for different subtypes of breast cancer. In surgical treatment, clinical trials on targeted axillary lymph node dissection and the establishment of a predictive model BRCA-CRisk for contralateral breast cancer risk provide more evidence for de-escalation in surgical treatment. In the area of human epidermal growth factor receptor 2(HER2)-positive breast cancer, pyrotinib has shown significant efficacy in advanced breast cancer treatment. In triple-negative breast cancer, precision subtype treatment and immunotherapy continue to improve patient survival. For hormone receptor-positive breast cancer, significant research results were obtained in exempting low-risk patients from chemotherapy and exploring alternative options after resistance to endocrine therapy. In the aspect of BRCA mutations,BGB-290-201 further confirmed the therapeutic efficacy and safety of poly(ADP-ribose)polymerase(PARP)inhibitors for the Chinese population. For advanced HER2-negative patients carrying germline BRCA(gBRCA)1/2 mutations, pamiparib will be an ideal treatment choice. This article reviews the important clinical research in the field of breast cancer in China in 2023,summarizes key results, and aims to provide reference ideas for future clinical research.

ABSTRACT

Mesenchymal stem cells(MSCs)are self-regenerating,rapidly proliferating pluripotent stem cells that depend primarily on their derived pro-angiogenic,inflammatory regulatory, and tro-phic factors to exert beneficial effects that attenu-ate deleterious inflammatory responses, reduce vascular damage, and promote tissue repair and re-generation.Obstructive sleep apnea hypoventila-tion syndrome(OSAHS)is a chronic disorder marked by oropharyngeal collapse during sleep,re-sulting in transient reduced airflow, large fluctua-tions in intrathoracic pressure, and intermittent hy-poxia and hypercapnia.OSAHS subsequently cyto-kine-mediated inflammatory cascades, oxidative stress, and ischemia, recruit MSCs from inflamed and damaged tissues through MSCs-derived of anti-inflammatory and pro-angiogenic factor activity,re-duce hypoxia, suppress inflammation, promote re-generation,and prevent fibrosis in OSAHS-injured tissues. In this paper,we will describe the patho-genesis of inflammation, oxidative stress, fibrosis and ischemia from the perspective of OSAHS,high-lighting the current research progress on MSCs-de-pendent regulation of OSAHS-related pathology.
