ABSTRACT

Combined with the related research progress of the application of chemical space in the field of Traditional Chinese Medicine (TCM), the common database resources and calculation tools of chemical space research are summarized and analyzed. The compatibility law of chemical space in TCM prescriptions, the screening of effective ingredients of TCMs, and TCM compounds are summarized. It was found that the current studies mainly focus on the efficacy and multi-target studies of TCMs and TCM compound, among which there are more studies on osteoarthritis and rheumatoid arthritis. The existing studies have clarified the efficacy and targets of TCMs or TCM compound for diseases, but there is still a lack of a database on the main components of all TCMs and the efficacy and main targets of TCM compound. Therefore, there needs a new database for the main efficacy molecules and targets of TCMs.

ABSTRACT

Objective:To establish an UHPLC-PDA method for determinating the content of 7 iridoid glycosides and 4 flavonoids constitutents simultaneously in Xiaoer-Ganyan granules. Methods:To take Agilent Ecilipse C18 (2.1 mm × 100 mm, 1.6 μm), and the colunm temperature was at 30 ℃. The mobile phase consists of acetonitrile (A) and 0.05% phosphoric acid (B) in gradient elution at a flow rate of 0.3 ml/min. The detection wavelength was set at 240 and 278 nm.Results:The linear ranges of swertiamain, gentiopicrin, sweroside, shanzhiside methy ester, gardenoside, genipin-1-β-D-gentiobioside, geniposide, baicalin, wogonoside, baicalein and wogonin were 19.16-306.56 μg ( r=0.999 4), 3.34-53.28 μg ( r=0.999 1), 5.30-84.64 μg ( r=0.999 5), 0.80-12.80 μg ( r=0.999 4), 0.46-7.20 μg ( r=0.999 2), 2.78-44.48 μg ( r=0.999 6), 6.02-96.16 μg ( r=0.999 9), 33.22-531.36 μg ( r=0.999 9), 3.92-62.72 μg ( r=0.999 2), 2.38-37.92 μg ( r=0.999 7), 1.32-20.96 μg ( r=0.999 9), respectively. The average recoveries ( n=9) varied from 94.62%-107.53% with RSDs no more than 3.0%. Conclusion:Method validation suggested that the developed method was suitable for simultaneous determination of 11 major constitutents in Xiaoer-Ganyan granules, thus providing reference for the improvement of quality standard of the drug.

ABSTRACT

As a traditional Chinese medicine, Ziziphi Spinosae Semen (ZSS) has the functions of tonifying liver, tranquilizing heart, astringent sweat and producing body fluid, which is used to treat neurasthenia, insomnia, dreaminess, debility, night sweat and so on. With the rapid and constant development of ZSS resource industry and its medicinal value, a large number of by-products and waste generated in the production and processing process, resulting in serious environmental problems. In general, the utilization rate of ZSS resources was still not high. Based on this, the chemical components and potential resources of ZSS were systematically combed from the perspective of the medicinal parts and bioactive components in this paper, and the authors had summarized that the widely application of ZSS and its by-products (fruit, leaf, root, etc.) in the fields of food, medicine, functional food and other areas was discovered and summarized as feed, feed additives, activated carbon, organic fertilizer, etc. In addition, this paper systematically summarized the current environmental protection problems of the industry, and put forward suggestions for improvement, aiming at reducing environmental pollution and improving the utilization efficiency of resources, so as to provide reference and basis for the comprehensive utilization of ZSS and its by-products, and promote the green, economical and double-effect development of the industry.

ABSTRACT

Objective:To identify the chemical constituents of Platycladi Cacumen<italic> </italic>before and after being carbonized. Method:Chemical constituents in 3 batches of Platycladi Cacumen and its carbonized products<italic> </italic>were identified and compared by ultra performance liquid chromatography-quadrupole-time of flight-mass spectrometry (UHPLC-QTOF-MS/MS). Chromatographic separation was performed on an ACQUITY UPLC BEH C<sub>18</sub> column (2.1 mm×100 mm, 1.7 μm) with 0.1% formic acid aqueous solution (A)- acetonitrile (B) as mobile phase for gradient elution (0-3.5 min, 5%-15%B; 3.5-6 min, 15%-30%B; 6-6.5 min, 30%B; 6.5-12 min, 30%-70%B; 12-12.5 min, 70%B; 12.5-18 min, 70%-100%B; 18-22 min, 100%B). The flow rate was 0.4 mL·min<sup>-1</sup> and the injection volume was 5 μL. Mass spectrometry was performed by an electrospray ionization, and the primary and secondary mass spectrometry data were collected with the full scan mode of positive and negative ions, the peaks containing MS/MS data were identified by self-established secondary mass spectrometry database and corresponding fragmentation law matching method. Result:A total of 77 and 76 substances with the same change trend were identified under positive and negative ion modes. After being<italic> </italic>carbonized, the disappeared components of Platycladi Cacumen were mainly amino acids, ketone aldehydes and other volatile components. Among newly produced components, there were 6 kinds of flavonoid aglycones (rhamnetin, 6,7,3'-trihydroxyflavone, 3,6,3'-trihydroxyflavone, 4'-hydroxy-2'-methyl-3,4,5-trimethoxychalcone, herbacetin and 3',5'-dimethoxy-3,5,7,4'-tetrahydroxyflavone), 3 kinds of coumarins (7-hydroxycoumarin, 7,8-dihydroxycoumarin and 8-acetyl-7-hydroxycoum-arin) and 3 kinds of benzoic acids (3-methylcatechol, pyrocatechol and chromone-3-carboxylic acid). There were a total of 40 flavonoids (quercitrin, quercetin, kaempferol, etc.) among these identified chemical constituents. Conclusion:There are significant quantitative and qualitative changes in the chemical compositions of Platycladi Cacumen after being carbonized. The flavonoids, the identified main active ingredients, can provide data reference for further study on the material basis of efficacy changes of Platycladi Cacumen<italic> </italic>before and after being carbonized.

ABSTRACT

Objective:To quickly analyze and identify the components in raw and wine-processed products of <italic>Polygonatum cyrtonema</italic> (PC) dried rhizomes by ultra-high performance liquid chromatography-quadrupole-time-of-flight mass spectrometry (UPLC-Q-TOF-MS), and then find out the differential components before and after processing. Method:The ACQUITY UPLC BEH C<sub>18</sub> column (2.1 mm×100 mm, 1.8 μm) was used with 0.1% formic acid aqueous solution-acetonitrile as the mobile phase for gradient elution at a flow rate of 0.25 mL·min<sup>-1</sup>. Electrospray ionization was selected for collection and detection in positive and negative ion modes, and the data were analyzed by PeakView 1.2.0.3. According to the retention time, accurate relative molecular weight and fragmentation ion information provided by MS, and combined with the reference substance and literature, the components were identified. After normalized treatment, the MS data of each sample were analyzed with principal component analysis (PCA) and orthogonal partial least squares-discriminant analysis (OPLS-DA), and then the differential components before and after processing were screened according to the principle that variable importance in the projection (VIP) value was >1. Result:A total of 38 components were identified from raw and wine-processed products of PC dried rhizomes, including 15 steroidal saponins, 6 alkaloids, 3 flavonoids, 2 amino acids, 2 organic acids and 10 others. The results of PCA and OPLS-DA showed that there were significant differences in the contents of components in PC dried rhizomes before and after processing, and 16 differential components such as kingianoside Z, disporopsin and linoleic acid were screened. Conclusion:UPLC-Q-TOF-MS technique can accurately and comprehensively identify the components in PC dried rhizomes, these components are mainly steroidal saponins, flavonoids and alkaloids. It takes a great difference in the contents of components before and after processing, and transformation of the same category components is the main reason for the differences of raw and wine-processed products, which will provide reference for the researches on material basis and processing chemistry of PC dried rhizomes.

ABSTRACT

Paridis Rhizoma(PR) is prepared from the dried rhizome of Paris polyphylla var. yunnanensis(PPY) or P. polyphylla var. chinensis(PPC) in Liliaceae family. The rapid development of PPY or PPC planting industry resulted from resource shortage has caused the waste of a large number of non-medicinal resources. To clarify the chemical compositions in rhizomes, fibrous roots, stems, leaves, seeds and pericarps of PPC, and explore the comprehensive application value and development prospect of these parts, the qualitative and quantitative analyses on the different parts of PPC were carried out by ultra-high performance liquid chromatography tandem quadrupole time-of-flight mass spectrometry(UPLC-Q-TOF-MS/MS) and high performance liquid chromatography(HPLC). A total of 136 compounds were identified, including 112 steroidal saponins, 6 flavonoids, 11 nitrogen-containing compounds and 7 phytosterols. Rhizomes, fibrous roots, and seeds mainly contained protopennogenyl glycosides and pennogenyl glycosides; leaves and stems mainly contained protodiosgenyl glycosides and diosgenyl glycosides; pericarps mainly contained pennogenyl glycosides, followed by diosgenyl glycosides. The total level of four saponins was the highest in fibrous roots and rhizomes, followed by those in the pericarps and arillate seeds, and the lowest in the stems and exarillate seeds. This study can provide data support for the comprehensive development and rational application of non-medicinal parts of PPC.

ABSTRACT

Drynariae Rhizoma is warm in nature and bitter in taste, mainly acting on liver and kidney systems. It is a common Chinese herbal medicine for the treatment of fracture and bone injury. The chemical compositions of Drynariae Rhizoma mainly include flavonoids, triterpenoids, phenylpropanoids and lignans. At present, modern pharmacological and clinical studies have shown that Drynariae Rhizoma has the effects of anti osteoporosis, promoting fracture healing, kidney protection, anti-inflammatory, promoting tooth growth, preventing and treating aminoglycoside ototoxicity and lowering blood lipid. In addition, the toxicity evaluation experiment of Drynariae Rhizoma has also shown that it has no obvious toxic and side effects. Naringin is a kind of dihydroflavone in Drynariae Rhizoma. Many studies have shown that naringin and other total flavonoids play an important role in anti-osteoporosis, promoting fracture healing, anti-inflammation, promoting tooth growth and lowering blood lipid. In this study, the research progresses on chemical consti-tuents and pharmacological activities of Drynariae Rhizoma in recent years were reviewed, and some mechanisms of action were summarized, to provide references for the further research and development of Drynariae Rhizoma.

ABSTRACT

Rhei Radix et Rhizoma(RRR) is a commonly used traditional Chinese medicine, with extensive pharmacological effects and clinical applications. This paper summarized processing history evolution of RRR and its effect on chemical compositions and pharmacological effects, and provided feasible insights for further studies on the chemical compositions and pharmacological effects of RRR before and after processing. Relevant information demonstrated that RRR has a long history of processing and various methods. At pre-sent, Chinese Pharmacopoeia mainly records four processing methods: cleaning(raw RRR), wine processing(RRR stir-fried with wine), steaming processing(RRR wine steaming), fried charcoal(RRR charring). RRR has a good effect in clearing heat effect, hemostatic effect and blood promoting effect, and its main chemical components are anthraquinone/anthrones, stilbene, phenylbutanone, chromogens, flavonoids and tannin compounds. This paper reviewed the history evolution of RRR and its effect on chemical composition and pharmacological changes, and put forward further study ideas, with the aim to provide a basic reference for processing mechanism, effective material basis and clinical application of RRR.

ABSTRACT

The chemical constituents of Rehmanniae Radix Preparatawere prepared according to the traditional method of "jiu zheng jiu shai" and investigated using multiple chromatographic methods. Six alkaloids were isolated and their structures were elucidated from spectral data and physicochemical properties, as follows: rehmanniae alkaloid A (4-{[(5-O-á-D-galactopyranosyloxy)methyl]-1H-pyrrole-2-carbaldehyde-1-yl}butyric acidmethyl ester) (1), baimantuoluoamide B (2), capparisine C (3), harman-3-carboxylic acid (4), (2S)-1-[2-(furan-2-yl)-2-oxoethyl]-5-oxopyrrolidine-2-carboxylate (5), and 1-[2-(furan-2-yl)-2-oxoethyl]pyrrolidin-2-one (6). Among them, compound 1 is a new alkaloid. Compounds 2-6 were newly isolated from Rehmannia glutinosa Libosch.The effect of compounds 1-6 on NRK-52e cell injury induced by LPS was investigated. The results show that compounds 1-3 exhibit protective effects against LPS-induced damage to NRK-52e cells.

ABSTRACT

Objective:To identify the quality differential markers of different processed products of Glycyrrhiza uralensis dry roots and rhizomes. Method:Ultra-high performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF-MSE) was used to collect high-precision mass-charge ratio and ion response strength information of the components in G. uralensis dry roots and rhizomes before and after processing by negative ion mode. The data set collected after pretreatment was analyzed with principal component analysis (PCA) and orthogonal partial least squares-discriminant analysis (OPLS-DA) to quickly search the differential components in different processed products of G. uralensis dry roots and rhizomes. Differential components were identified according to the relative molecular weight, fragment ion, mass spectrum database and related literature information, then the migration of components before and after processing was studied. Result:A total of 10 quality differential markers were searched from raw products, roasted products and honey-roasted products of G. uralensis dry roots and rhizomes, mainly derivatives of liquiritin and glycyrrhizic acid. Among them, the contents of 6''-O-acetylliquiritin apioside, 6''-O-acetylliquiritin apioside isomer, 6''-O-acetylliquiritin, formononetin and 11-deoxo-18β-glycyrrhetic acid were the highest in the raw products, the contents of 6''-O-acetylisoliquiritin apioside, 6''-O-acetylisoliquiritin, isoliquiritin and glycyrrhetic acid 3-O-glucuronide were the highest in the roasted products, the content of liquiritin was the lowest in the honey-roasted products. Conclusion:There are some chemical differences among the three products. This study can provide material basis for the quality control and pharmacodynamic research of processed products of G. uralensis dry roots and rhizomes.

ABSTRACT

Objective: To compare the chemical compositions of Scutellariae Radix (SR) before and after wine-frying, and provide a reference for establishing a comprehensive quality evaluation method for SR decoction pieces. Methods: Characteristic chromatogram of SR and wine-processed Scutellariae Radix (wpSR) were established using HPLC, the control characteristic chromatogram maps were generated, the common peaks were calibrated and the similarity was evaluated. Chemical compositions of SR and wpSR were identified by UPLC-Q-TOF/MS. The content profiles of 11 flavonoids (baicalin, baicalein, wogonoside, wogonin, oroxylin A, oroxyloside, scutellarin, apigenin, hispidulin, luteoloside and chrysin) of SR and wpSR was determined by UPLC-TQMS, and PCA was used to analysis the content of 11 flavonoids. Results: The characteristic chromatogram of SR and wpSR were established, and nine common peaks were calibrated. The similarity of two pieces were all greater than 0.947. Through the analysis of the multistage tandem mass spectrometry, retention time matching combined with the software of database search and literatures, 50 compositions were found and 44 compositions were identified in two pieces. The quantitative results showed that the content of flavonoid glycosides such as baicalin and wogonoside decreased slightly, while the content of flavonoid aglycones such as baicalein and wogonin increased slightly. After multivariate statistical analysis, two pieces were divided into two types. The differences of the content of baicalin, wogonoside and oroxyloside may be the main factors causing the change of chemical compositions of wine-frying of SR. Conclusion: Chemical profiles did not change after wine-frying of SR, but the content profiles of some compositions were changed. The established method can provide reproducible, efficient, as well as accurate analysis for Chinese medicinal materials. And it should be an eligible tool for the quality evaluation of SR.

In recent years, scholars have gradually deepened their research on various aspects of Cimicifugae Rhizoma, and its new chemical components and pharmacological effects have been discovered. Based on the review of the literature, the summary of its chemical composition and pharmacological effects, according to the definition of Q-marker, from the aspects of the source route, drug efficacy, traditional drug properties, and the significant components in different compatibility, the Q-marker of Cimicifugae Rhizoma is predicted, in order to provide a scientific basis for the quality evaluation of Cimicifugae Rhizoma.

On the basis of the studies, and according to the definition of quality marker(Q-marker), we predict its Q-markers from the aspects of the traditional efficacy, traditional medicinal properties, new clinical use, measurable composition, composition changes before and after processing, the plasma composition, and so on, which provides a scientific basis for the quality evaluation of Phytolaccae Radix.

Objective::To identify the active constituents of Paidu Qingzhi tablets by UPLC-Q-TOF-MS technique, and to reveal its potential targets and molecular mechanisms by network pharmacology. Method::Chromatography separation was achieved on an ACQUITY UPLC HSS T3 column (2.1 mm×100 mm, 1.8 μm) with mobile phase consisted of 0.1% formic acid aqueous solution and acetonitrile for gradient elution (0-17 min, 95%-5%A; 17-17.01 min, 5%-95%A; 17.01-20 min, 95%A), the flow rate was 0.3 mL·min-1, the column temperature was 40 ℃. Data acquisition was carried out in electrospray ionization (ESI) under the negative ion mode, the scanning range was 50-1 200.Ingredients in Paidu Qingzhi tablets were identified according to reference substance, relative molecular weight, mass spectrometric cleavage rule and literature information. Multiple databases were used to retrieve the targets of these identified ingredients and related diseases treated by the tablets, and metabolic pathways. A visual network of " herbs-compounds-targets-pathways-diseases" was constructed by Cytoscape 3.7.1. Result::A total of 33 active compounds in methanol extract of Paidu Qingzhi tablets were identified by UPLC-Q-TOF-MS, including 27 compounds from Rhei Radix et Rhizoma, 4 compounds from Panacis Quinquefolii Radix, 2 compounds from Ophiopogonis Radix. Totally 18 direct targets and 58 indirect targets corresponding to 31 ingredients were obtained, 7 metabolic pathways including steroid hormone biosynthesis pathway, arachidonic acid metabolic pathway, insulin signaling pathway were strongly correlated with the treatment of hyperlipidemia, acne and simple obesity by Paidu Qingzhi tablets. Conclusion::Ingredients of Paidu Qingzhi tablets are revealed by UPLC-Q-TOF-MS, based on these identified ingredients, targets and related metabolic pathways are visualized by network pharmacology. The current research can provide theoretical basis for quality control and clinical application of Paidu Qingzhi tablets.

Bupleuri Radix is a traditional Chinese medicine commonly used in clinical practice, which has the effects of relieving fever, relieving liver and depression, and promoting Yangqi. At present, the varieties of Bupleuri Radix are relatively chaotic, and the processing in the production areas is relatively extensive. In the processing of Bupleuri Radix, the processed products contained in the 2015 edition of Chinese Pharmacopoeia are raw products and vinegar-processed products. Berberine (BBR) is a natural isoquinoline alkaloid extracted from coptis chinensis.Berberine has several pharmacological activities,including disease-resistant microorganisms,anti-inflammatory,anti-tumor,heart protection,hypoglycemia,regulation of lipid metabolism and immunomodulatory properties.However,BBR's effects on neurodegenerative diseases remained relatively unexplored until its ability to stunt AD progression was characterized.Substantial studies suggest that berberine may be beneficial to AD by limiting the pathogenesis of extracellular amyloid plaques and intracellular neurofibrillary tangles.The studies in recent years have also found that berberine can additionally treat AD by affecting neurotransmitter,anti-oxidative stress,metabolism and other multi-target pathways.This article discusses the treatment pathway of berberine on AD,and summarizes the inhibition mechanism of berberine on AD.

The combined technique of Liquid chromatography-mass spectrometry (LC/MS) is regarded as one of the most powerful analytical techniques for the analysis of traditional Chinese medicine (TCM) since it combines the high efficiency of LC separation with the confident structural identification of mass spectrometric detection.Studieson pharmacodynamic material basisof TCM are key to TCM modern research.This paper briefly reviewed its application in the study of chemical components,serum pharmacochemistry and metabonomics of TCM,as well as the study of the fingerprints of herbal drugs,cell membrane chromatography and affinity ultrafiltration mass pectrometry,which offered reference for further application of LC/MS in traditional Chinese medicine.
