ABSTRACT

Traditional pig breeding has a long cycle and high cost, and there is an urgent need to use new technologies to revitalize the pig breeding industry. The recently emerged CRISPR/Cas9 genome editing technique shows great potential in pig genetic improvement, and has since become a research hotspot. Base editor is a new base editing technology developed based on the CRISPR/Cas9 system, which can achieve targeted mutation of a single base. CRISPR/Cas9 technology is easy to operate and simple to design, but it can lead to DNA double strand breaks, unstable gene structures, and random insertion and deletion of genes, which greatly restricts the application of this technique. Different from CRISPR/Cas9 technique, the single base editing technique does not produce double strand breaks. Therefore, it has higher accuracy and safety for genome editing, and is expected to advance the pig genetic breeding applications. This review summarized the working principle and shortcomings of CRISPR/Cas9 technique, the development and advantages of single base editing, the principles and application characteristics of different base editors and their applications in pig genetic improvement, with the aim to facilitate genome editing-assisted genetic breeding of pig.

ABSTRACT

Abiotic stresses, predominately drought, heat, salinity, cold, and waterlogging, adversely affect cereal crops. They limit barley production worldwide and cause huge economic losses. In barley, functional genes under various stresses have been identified over the years and genetic improvement to stress tolerance has taken a new turn with the introduction of modern gene-editing platforms. In particular, clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated protein 9 (Cas9) is a robust and versatile tool for precise mutation creation and trait improvement. In this review, we highlight the stress-affected regions and the corresponding economic losses among the main barley producers. We collate about 150 key genes associated with stress tolerance and combine them into a single physical map for potential breeding practices. We also overview the applications of precise base editing, prime editing, and multiplexing technologies for targeted trait modification, and discuss current challenges including high-throughput mutant genotyping and genotype dependency in genetic transformation to promote commercial breeding. The listed genes counteract key stresses such as drought, salinity, and nutrient deficiency, and the potential application of the respective gene-editing technologies will provide insight into barley improvement for climate resilience.

The objective of the present paper was to investigate the inheritance of seedlessness in the 'Brazilian seedless' sugar apple and INO gene conservation in Annona squamosa and Annona cherimola x Annona squamosa genotypes by assessing their homology with the INO database genes. The F1 generation was obtained by crossing the mutant 'Brazilian seedless' (male genitor) (P1) with the wild-type A. squamosa with seeds (M1 and M2, female genitors). The INO gene was studied in mutant and wild-type A. squamosa (P1, M1, M2 and M3) and in the Gefner atemoya (A. cherimola x A. squamosa) (M4) cultivar. The DNA was extracted from young leaves, and four sets of specific primers flanking the INO gene were amplified. The seedless characteristic was identified as stenospermatic in the fruits of parental P1, suggesting monogenic inheritance with complete dominance. High sequence similarity of the INO gene amplifications in the sugar apple accessions (M1, M2, M3) and the atemoya cultivar Gefner (M4) reinforces the hypothesis of their conservation.


Resumo A herança da característica de fruto sem sementes de Annona squamosa ainda não foi esclarecida. Técnicas moleculares podem auxiliar em programas de melhoramento, principalmente na seleção assistida do gene de interesse. O gene INO pode estar relacionado ao desenvolvimento da semente dessas frutas. O objetivo foi investigar a herança da ausência de sementes em Annona squamosa e a conservação do gene INO nos genótipos Annona squamosa e Annona cherimola x Annona squamosa avaliando sua homologia com banco de dados de genes INO. A geração F1 foi obtida pelo cruzamento do mutante 'Brazilian seedless' (genitor masculino) (P1) com o tipo selvagem com sementes (A. squamosa) (M1 e M2, genitores femininos). O gene INO foi estudado em A. squamosa, mutante e selvagem (P1, M1, M2 e M3) e na cultivar Gefner atemoya (A. cherimola x A. squamosa) (M4). O DNA foi extraído de folhas jovens, e quatro conjuntos de primers específicos flanqueando o gene INO foram amplificados. A característica sem sementes foi identificada como estenospermática nos frutos do parental P1, sugerindo herança monogênica com dominância completa. A alta similaridade de sequência das amplificações do gene INO nos acessos de pinha (M1, M2, M3) e na cultivar de atemóia Gefner (M4) reforça a hipótese de sua conservação.

RESUMEN El comportamiento meiótico es de gran importancia para el desarrollo de programas de mejoramiento genético, especialmente en especies como la vainilla (Vanilla planifolia Jacks.), que se ven severamente afectadas por diversos factores bióticos y abióticos derivados de su estrecha base genética. Por ello, el presente trabajo se planteó con el objetivo de evaluar el comportamiento meiótico del morfotipo comercial "Mansa" del municipio de Papantla de Olarte, Veracruz, México. Para ello, se colectaron botones florales de diferentes tamaños y se fijaron en Carnoy, utilizando la técnica de la calabaza, seguido de una tinción con acetocarmín al 2% (p/v) para observar claramente todas las etapas del proceso meiótico. Se observó un comportamiento meiótico normal y un número de cromosomas haploides (n = 8). Estos resultados pueden ser útiles para el desarrollo de programas de mejora de este cultivo.


ABSTRACT Meiotic behavior is of great importance for the development of genetic improvement programs, especially in species such as vanilla (Vanilla planifolia Jacks.), which are severely affected by various biotic and abiotic factors derived from their narrow genetic base. Therefore, the present work was proposed with the objective of evaluating the meiotic behavior of the commercial morphotype "Mansa" from the municipality of "Papantla de Olarte" in Veracruz, Mexico. For this purpose, flower buds of different sizes were collected and fixed in Carnoy using the squash technique, followed by staining with 2% (w/v) acetocarmine to clearly observe all the stages of the meiotic process. Normal meiotic behavior and a haploid chromosome number (n = 8) were observed. These results may be useful for the development of breeding programs for this crop.

ABSTRACT

Background: Litter size at birth (LSB) is one of the most important economic traits in sheep and could be used in genetic improvement schemes for meat production. LSB is inherently a categorical trait and should be analysed with threshold models. Objective: Bayesian threshold models were used to analyze sheep LSB to estimate genetic parameters. Methods: Data was based on 7,901 LSB records from 14,968 dams and 682 sires collected from 1986 to 2012 at Makouie Sheep Breeding Station in Iran. Means of posterior distributions (MPDs) of LSB's genetic parameters were estimated, and the best-fitted models were selected using the deviance information criterion. Results: In the repeated measurement analysis, the estimated direct and maternal heritabilities, and permanent environmental effect (±SE), according to the best-fitted model (model 5), were 0.01 (0.010), 0.02 (0.014), and 0.01 (0.011), respectively. In the univariate analysis, the best estimates of direct and maternal heritabilities were 0.12 (0.064) and 0.08 (0.045), respectively. An increasing trend for direct and maternal heritabilities was observed in parity 2 (0.15 (0.082) and 0.25 (0.083), respectively). In the bivariate analysis, the best estimates of direct and maternal heritabilities for LSB were 0.03 (0.027) and 0.22 (0.041), respectively. The direct and maternal genetic correlations among parities were 0.25 (0.054) and 0.12 (0.021), respectively. Conclusions: The results showed a considerable influence of environmental factors on LSB in each parity of sheep; also, statistically different genetic parameters (p<0.05) were obtained from one parity to another, indicating the different and large influences of genetic and environmental factors for each parity.


Antecedentes: El tamaño de la camada al nacer (LSB) es inherentemente un rasgo categórico y debe analizarse con modelos de umbral. El LSB es uno de los rasgos de producción de carne más importantes en las ovejas y podría usarse en esquemas de mejora genética para la producción de carne. Objetivo: Se utilizaron modelos de umbral bayesiano para analizar el tamaño de la camada de ovejas al nacer (LSB) y estimar parámetros genéticos. Métodos: Los datos se basaron en 7.901 registros de LSB de 14.968 ovejas y 682 carneros recolectados de 1986 a 2012 en la estación de cría de ovejas Makouie en Irán. Se estimaron las medias de distribuciones posteriores (MPD) de los parámetros genéticos de LSB y se seleccionaron los modelos mejor ajustados utilizando el criterio de información de desviación. Resultados: En los análisis de medición repetida, la heredabilidad materna y directa estimada y el efecto ambiental permanente (±SE), según el modelo mejor ajustado (modelo 5), fueron 0,01 (0,010), 0,02 (0,014) y 0,01 (0,011), respectivamente. En el análisis univariado, las mejores estimaciones de heredabilidad directa y materna fueron 0,12 (0,064) y 0,08 (0,045), respectivamente. Se observó una tendencia creciente de heredabilidades directas y maternas en la paridad 2 (0,15 (0,082) y 0,25 (0,083), respectivamente). En el análisis bivariado, las mejores estimaciones de heredabilidad directa y materna para LSB fueron 0,03 (0,027) y 0,22 (0,041), respectivamente. Las correlaciones genéticas directas y maternas entre partos fueron 0,25 (0,054) y 0,12 (0,021), respectivamente. Conclusiones: Los resultados mostraron una influencia considerable de los factores ambientales sobre el LSB en cada parto de las ovejas; además, se obtuvieron parámetros genéticos estadísticamente diferentes (p<0.05) de un parto a otro, indicando las diferentes y grandes influencias de factores genéticos y ambientales para cada parto en ovejas. Los resultados de este estudio se pueden precisar aún más utilizando datos de SNP de todo el genoma sobre diferentes partes para manejar una amplia gama de problemas relacionados con la interacción del entorno genético del rasgo LSB.

Selection can affect growth, changing performance and asymptotic values. However, there is little information about the growth of families in fish breeding programs. The aim of this study was to evaluate the performance and growth of families of Nile tilapia AquaAmérica. Twenty AquaAmérica families cultivated in a net cage (13.5 m3) for 181 days were evaluated. The nonlinear Gompertz regression model was fitted to the data by the weighted least squares method, taking the inverse of the variance of weight in different families and at different ages as the weighting variable. The model was adjusted to describe the growth in weight and morphometric characteristics. Two families showed highest (P<0.05) weights at both 133 days (family AA10: 743.2 g; family AA16: 741.2 g) and 181 days (family AA10: 1,422.1 g; family AA16: 1,393.4 g) of the experiment. In both experimental periods, the males showed a heavier weight, with the greatest contrast between the sexes occurring at 181 days. The analysis of the three most contrasting families (AA1, AA9 and AA14) showed that the asymptotic value for weight was higher (P<0.05) in family AA9 (3,926.3 g) than in family AA14 (3,251.6 g), but specific growth rate and age at the inflection point did not differ significantly between families. In conclusion, two of the 20 families were superior; males exhibited a greater growth, mainly in the period of 181 days; and the growth curve differed between the families, especially for asymptotic weight.


A seleção pode impactar a forma de crescimento, mudando o desempenho e os valores assintóticos. No entanto, existem poucas informações sobre o crescimento das famílias em programas de criação de peixes. O objetivo deste estudo foi examinar o desempenho e as curvas de crescimento de famílias de tilápia-do-Nilo AquaAmérica. Foram avaliadas 20 famílias AquaAmérica cultivadas em tanques-rede (13,5 m3) por 181 dias. O modelo de regressão não linear de Gompertz foi ajustado aos dados pelo método dos mínimos quadrados ponderados, tomando o inverso da variância do peso nas diferentes famílias e nas diferentes idades como variável de ponderação. O modelo foi ajustado para descrever o crescimento em peso e características morfométricas. Duas famílias apresentaram pesos maiores (P <0,05) em 133 dias (família AA10: 743,2 g; família AA16: 741,2 g) e 181 dias (família AA10: 1422,1 g; família AA16: 1393,4 g) de experimento em relação a outras famílias. The nonlinear Gompertz regression model was fitted to the data by the weighted least squares method, taking the inverse of the variance of weight in different families and at different ages as the weighting variable. The model was adjusted to describe the growth in weight and morphometric characteristics. Two families showed highest (P 0.05) weights at both 133 days (family AA10: 743.2 g; family AA16: 741.2 g) and 181 days (family AA10: 1,422.1 g; family AA16: 1,393.4 g) of the experiment. In both experimental periods, the males showed a heavier weight, with the greatest contrast between the sexes occurring at 181 days. The analysis of the three most contrasting families (AA1, AA9 and AA14) showed that the asymptotic value for weight was higher (P 0.05) in family AA9 (3,926.3 g) than in family AA14 (3,251.6 g), but specific growth rate and age at the inflection point did not differ significantly between families. In conclusion, two of the 20 families were superior; males exhibited a greater growth, mainly in the period of 181 days; and the growth curve differed between the families, especially for asymptotic weight.


Resumo A seleção pode impactar a forma de crescimento, mudando o desempenho e os valores assintóticos. No entanto, existem poucas informações sobre o crescimento das famílias em programas de criação de peixes. O objetivo deste estudo foi examinar o desempenho e as curvas de crescimento de famílias de tilápia-do-Nilo AquaAmérica. Foram avaliadas 20 famílias AquaAmérica cultivadas em tanques-rede (13,5 m3) por 181 dias. O modelo de regressão não linear de Gompertz foi ajustado aos dados pelo método dos mínimos quadrados ponderados, tomando o inverso da variância do peso nas diferentes famílias e nas diferentes idades como variável de ponderação. O modelo foi ajustado para descrever o crescimento em peso e características morfométricas. Duas famílias apresentaram pesos maiores (P 0,05) em 133 dias (família AA10: 743,2 g; família AA16: 741,2 g) e 181 dias (família AA10: 1422,1 g; família AA16: 1393,4 g) de experimento em relação a outras famílias. Em ambos os períodos experimentais, os machos apresentaram maior peso, com maior contraste entre os sexos ocorrendo aos 181 dias. A análise das três famílias mais contrastantes (AA1, AA9 e AA14) mostrou que o valor assintótico para o peso foi maior (P 0,05) na família AA9 (3926,3 g) do que na família AA14 (3251,6 g), mas a taxa de crescimento específica e a idade no ponto de inflexão não diferiu significativamente entre as famílias. Em conclusão, duas das 20 famílias eram muito superiores; machos exibiram um maior crescimento, principalmente no período de 181 dias; e a curva de crescimento diferiu entre as famílias, principalmente quanto ao peso assintótico.

RESUMEN El objetivo del estudio fue evaluar los parámetros genéticos de las características biométricas del fruto y semillas de Caesalpinia ebano. La investigación se realizó en Montería (Colombia), se seleccionaron diez árboles, y de cada uno diez frutos tomados al azar. Se realizaron análisis de varianza y estimación de parámetros genéticos para nueve características: peso fresco del fruto, largo del fruto, ancho del fruto, grosor del fruto, número de semillas/fruto, peso de semillas por fruto, peso de una semilla, volumen de una semilla y densidad de una semilla. Se detectaron diferencias estadísticas (p < 0,01) entre árboles, para todas las características, excepto para el ancho del fruto y peso de una semilla; denotando la existencia de variabilidad genética entre los árboles para peso del fruto, largo del fruto, grosor del fruto, número de semillas y densidad de una semilla. El peso y largo del fruto presentaron la heredabilidad media familiar y heredabilidad en el sentido estricto con los valores más altos (h2 Fam > 82 % y h2 E > 30 %). La ganancia genética esperada fue mayor del 10 % en cinco de las nueve características estudiadas, y mayor del 20 % para número de semillas y peso de las semillas (27,31 y 27,87 %, respectivamente). Los caracteres peso y largo del fruto se podrían transmitir a las generaciones sucesivas, con mayor posibilidad de éxito, mediante la selección fenotípica de árboles.


ABSTRACT The objective of the study was to evaluate the genetic parameters of the biometric characteristics of the fruit and seeds of C. ebano. The investigation was carried out in Montería, Córdoba (Colombia); ten trees were selected and from each, ten fruits taken at random. Variance analysis and estimation of genetic parameters for nine traits were carried out: fresh fruit weight, fruit length, fruit width, fruit thickness, number of seeds/fruit, weight of seeds by fruit, weight of one seed, volume of one seed and density of one seed. Statistical differences (p < 0.01) were detected between trees, for all traits except for width of the fruit and weight of a seed; denoting the existence of genetic variability between trees for fruit weight, fruit length, fruit thickness, number of seeds and density of a seed. The weight and length of the fruit presented the highest family heritability and narrow sense heritability (h2 Fam > 82 % and h2 E> 30 %). The expected genetic gain was greater than 10 % in five of the nine studied traits, and greater than 20 % for number of seeds and weight of seeds (27.31 and 27.87 %, respectively). The traits fruit weight and length could be transmitted to successive generations, with greater chance of success, by phenotypic selection of trees.

ABSTRACT

The objective of this study was to estimate genetic parameters and genetic trends of different conformation and management traits regularly measured within the context of the National Dairy Gir Breeding Program (PNMGL). The estimation of genetic and residual variances for each trait was performed using average information restricted maximum likelihood (AI-REML) procedure in AIREMLF90 program software. The population was divided into three subpopulations constituted by measured females (with phenotype records), all females, and males. Linear regressions were applied for each trait, considering two periods of birth (1st period: 1938-1996; 2nd period: 1997-2012). The estimated heritability of conformation and management traits varied from 0.01 to 0.53, denoting a perspective of genetic improvement through selection and corrective matings for purebred Dairy Gir populations. The average genetic changes in conformation and management traits were, in general, variable and inexpressive, showing that the selection of Dairy Gir may have had been directed essentially to increase milk yield. The analysis of the two periods of birth indicated that some linear traits present progress (although inexpressive) in the 2nd period (more recent period).(AU)


O objetivo deste estudo foi estimar os parâmetros genéticos e as tendências genéticas para diferentes características de conformação e manejo de animais puros da raça Gir Leiteiro, pertencentes ao Programa Nacional de Melhoramento do Gir Leiteiro (PNMGL). A estimativa das variâncias genéticas e residuais para cada característica foi realizada usando-se o procedimento de máxima verossimilhança restrita (AI-REML), por meio do programa AIREMLF90. A população foi dividida em três subpopulações, constituídas por fêmeas mensuradas (com registros de fenótipo), todas fêmeas e machos. As regressões lineares para cada característica foram ainda divididas em dois períodos de anos de nascimento (1º período: 1938 a 1996; 2º período: 1997 a 2012). As herdabilidades estimadas variaram de 0,01 a 0,53 para as características de conformação e manejo, possibilitando a perspectiva de melhoramento mediante seleção e acasalamentos corretivos na população pura da raça Gir Leiteiro. As mudanças genéticas nas características conformação e manejo foram, em geral, variáveis e inexpressivas, sugerindo que a seleção no Gir Leiteiro possa ter sido direcionada essencialmente para maior produção de leite. Ao serem observados os dois períodos distintos de anos de nascimento, infere-se que algumas características lineares apresentaram progresso (embora inexpressivo) no 2º período analisado.(AU)

ABSTRACT

The aim of this research was to evaluate the genetic divergence between 164 genotypes of Manihot esculenta from the South-Central mesoregion of the State of Mato Grosso. The genotypes are from projects conducted by the Laboratory of Genetic Resources & Biotechnology of the University of the State of Mato Grosso, Cáceres-Mato Grosso (UNEMAT), and the Brazilian Public Agricultural Research Corporation - Agrosilvopastoral (EMBRAPA). The agronomic descriptors evaluated were plant height, height of first branching, branching levels, weight of the aerial part of the plant, total weight of the plant, number of roots per plant, average weight of roots per plant, yield of commercial roots, yield of non-commercial roots, number of rotten roots per plant and harvest index. For the analysis of genetic divergence, multivariate analysis based on the standardized Euclidean mean distance was employed, later performing the Hierarchical UPGMA and Tocher Optimization agglomerative methods. The degree of preservation of the genetic distances in the dendrogram was verified using the Cophenetic Correlation Coefficient. The Singh criterion was used to quantify the relative contribution of characteristics to genetic divergence. The genotypes presented genetic dissimilarity for the evaluated characteristics and based on the results of the dissimilarity matrix and groupings, it is recommended the crossings between the genotypes allocated in group II with the genotype allocated in group V, for the development of segregated populations with high genetic variability.

ABSTRACT

Despite the importance of passion fruit for the Brazilian fruit market, there are still many agronomic and fruit quality problems to be solved, in order to increase this crop performance. The objective of this study was to evaluate the quality of twelve genotypes of wild, sweet and yellow passion fruit, aiming to identify promising materials considering fruit quality, in Federal District, Brazil. An experiment was carried out at the Água Limpa Farm of the Universidade de Brasília (UnB) from 2016 to 2018, in a randomized block design, with 12 treatments, 4 replicates and 6 plants/plot. At the harvesting time, six fruits per plot were randomly collected for the following physicochemical analysis: fruit mass, pulp mass with and without seeds, length/longitudinal diameter, width/transverse diameter, length/width ratio, husk thickness, predominant color of the pulp (L*, C*, h*), number of seeds, seed size, total soluble solids content, total titratable acidity, total soluble solids/total titratable acidity ratio and pH. High heritability values ​​and relation of genetic/environment variation coefficients ratio were observed for most of the characteristics evaluated. The genotypes of yellow passion fruit MAR20#21 P2 x FB 200 P1 R2 and MAR20#19 ROXO R4 x ECRAM P3 R3 showed the best characteristics of fruit mass and pulp mass with seed. All the genotypes studied showed values ​​of total soluble solids above 11ºBrix. Positive and significant correlation was observed between fruit mass and length/width ratio, indicating that oblong fruits have higher fruit mass.

ABSTRACT

The evaluation of coffee quality in Brazil for commercialization is conducted mainly through sensory analysis, also known as the "cup test", in which professional tasters evaluate and score various attributes. The adoption of chemical methods could complement the sensory classification of beverages, if correlations between these chemical and sensory analyses exist, making classification less subjective. This work aimed to identify the relationships between the chemical and sensorial traits of coffee-beverage quality and to evaluate the use of these traits as criteria for the selection of Bourbon cultivars. Twenty coffee genotypes from the first three harvests across five municipalities of the state of Minas Gerais, Brazil were evaluated. The genotypic values, predicted for each genotype, were used to determine the index based on the sum of ranks from Mulamba and Mock. The genetic correlations among the evaluated traits were also estimated. The presented evaluations were not able to efficiently detect genetic and phenotypic relationships between the chemical and sensorial characteristics of drink quality, but as selection criteria for generation advancement in the beverage quality, it is possible to use these characteristics. Bourbon Amarelo LCJ 9-IAC, Bourbon Amarelo-Procafé, Bourbon Amarelo-Boa Vista, Bourbon Vermelho-São João Batista, and Bourbon Amarelo-Samambaia were the genotypes with the most promising cup quality in the studied regions. Through the selection of these five genotypes, the selection gain was 1.65% for sensory score for beverage quality, when the interaction among the studied environments was removed. The heritability was 92% for improving this trait.

Abscisic acid (ABA) is associated with bud dormancy, leaf abscission, and germplasm growth inhibition in in vitro conservation. We evaluated the effects of ABA in four wild Manihot accessions and one cassava accession (M. esculenta Crantz) to refine in vitro conservation methods for these species. The experiment was performed at the Laboratory for Tissue Culture from Embrapa, Cruz das Almas, Bahia State, Brazil. The statistical design was completely random in a 5 × 5 factorial scheme [(5 ABA dosages (0, 0.25, 0.50, 0.75, and 1 mg L-1) and 5 Manihot species (M. pseudoglaziovii, M. tristis, M. flabellifolia, M. chlorosticta, and M. esculenta)], with 15 replicates. Mini-cuttings of 1 cm were used, each inoculated in 10 mL of modified Murashige and Skoog medium, solidified with Phytagel® (2.4 g L-1) containing the respective ABA dosages. Tubes containing these mini-cuttings were placed in a germplasm conservation room with an irradiance of 30 µmol m-2 s-1, temperature of 22 ± 1°C, and photoperiod of 12 hours. Plant height (cm), the number of living and senescent leaves, shoots, and mini-cuttings (1 cm), and fresh and dry weights of shoots and roots (mg) were evaluated after 150 days. Growth reduction was prominent in M. pseudoglaziovii, M. tristis, and M. flabellifolia during the in vitro conservation period. ABSTRACT Climate change will have an impact on the Colombian agricultural sector, by 2050 increases in temperature and distribution of erratic rainfall are expected. Passion fruit cultivation does not tolerate water deficit, it reduces flower induction, generates fruit drop and defoliation. To tackle this problem, somaclonal variants (VS) of passion fruit were selected in-vitro, seeking tolerance to water deficit. Four phases were developed: I) callogenesis, II) direct and indirect organogenesis, II) Induction and evaluation of the water deficit with Polyethylene glycol 6000 (PEG 6000) and IV) in vitro selection of VS by morphometric measurements, chlorophyll and total sugars contents. Differences in callogenesis were found with different concentrations of 2,4-D, the concentration of 2 mg L-1 presented better results producing calluses in less time and in greater quantity (8 days, 90% of the leaf area). In indirect and direct organogenesis the medium MS + ANA + BAP (0.3: 0.6), showed significant statistical differences with respect to other means, for the variables root length (15.14 cm), stem (16.72 cm) and leaves ( 14.51 cm) and root thickness (0.76 cm) stem (1.25) and leaf width (6.75). The influence of PEG 6000 showed significant differences, the treatment with 30 g L-1 showed the smallest leaf width, the greatest width was found in 25 g L1. Statistical differences were found in chlorophyll levels and total sugar contents, the highest contents were recorded in the VS 25VS1, showing the possibility of obtaining seedlings tolerant to the water deficit of passion fruit by inducing somaclonal variation.


RESUMEN El cambio climático tendrá impactos en el sector agropecuario colombiano, para el 2050 se prevén aumentos de temperatura y distribución de lluvias erráticas. El cultivo de maracuyá no tolera el déficit hídrico, este disminuye la inducción floral, genera caída de frutos y defoliación. Para abordar esta problemática se seleccionaron in-vitro variantes somaclonales (VS) de maracuyá, buscando tolerancia al déficit hídrico. Se desarrollaron cuatro fases: I) callogénesis, II) organogénesis directa e indirecta, III) Inducción y evaluación del déficit hídrico con Polientilenglicol 6000 (PEG 6000) y IV) selección in vitro de VS por mediciones morfométricas, contenidos de clorofila y azúcares totales. Se hallaron diferencias en callogénesis con diferentes concentraciones de 2,4-D, la concentración de 2 mg-L-1 presentó mejores resultados produciendo callos en menor tiempo y en mayor cantidad (8 días, 90% del área foliar); en organogénesis indirecta y directa el medio MS + ANA + BAP (0.3:0.6), mostró diferencias estadísticas significativas respecto a otros medios, para las variables longitud de raíz (15.14 cm), tallo (16.72 cm) y hojas (14.51 cm) y grosor de raíz (0.76 cm) tallo (1.25) y ancho de hojas (6.75). La influencia de PEG 6000 mostró diferencias significativas, el tratamiento con 30 g-L-1 mostró menor ancho de hojas, el mayor ancho se encontró en 25 g-L-1. Se hallaron diferencias estadísticas en niveles de clorofila y contenidos de azúcares totales, los mayores contenidos se registraron en el VS 25VS1, mostrando la posibilidad de obtener plántulas tolerantes al déficit hídrico de maracuyá mediante la inducción de variación somaclonal.

BACKGROUND: Fragrance is one of the most important quality traits in rice, and the phenotype is attributed to the loss-of-function betaine aldehyde dehydrogenase (BADH2) gene. At least 12 allelic variations of BADH2 have been identified, and some of these have been applied to rice fragrance breeding using traditional molecular markers and Sanger sequencing techniques. However, these traditional methods have several limitations, such as being very expensive, imprecise, inefficient, and having security issues. Thus, a new molecular marker technology must be developed to improve rice fragrance breeding. RESULTS: In this study, more than 95% of the cultivated fragrant rice varieties belonged to a 7-bp deletion in exon 2 (badh2-E2) or an 8-bp deletion and 3-bp variation in exon 7 (badh2-E7). Both allelic variations resulted in the loss of function of the badh2 gene. We developed two novel SNP molecular markers, SNP_badh2-E2 and SNP_badh2- E7, related to the alleles. Their genotype and phenotype were highly cosegregated in the natural variation of rice accessions, with 160 of the 164 fragrant rice varieties detected with the two markers. These markers cosegregated with the fragrance phenotype in the F2 population. CONCLUSIONS: Two functional SNP molecular markers of badh2-E2 and badh2-E7 allelic variations were developed. These functional SNP molecular markers can be used for genotype and genetic improvement of rice fragrance through marker-assisted selection and will significantly improve the efficiency of fragrant rice breeding and promote commercial molecular breeding of rice in the future.

ABSTRACT: Logistic regression analysis is a technique that may aid genetic breeding programs in the selection of clones, especially in the early stages where experimental accuracy is low. This research aimed to identify the most important agronomic traits for energy cane clonal selection, and to verify the efficiency of the logistic model in predicting the genotypes to be selected. Evaluations were carried out on 220 clones in the first ratoon. The data were subjected to binary logistic regression analysis. Stalk number per meter was the most important trait in the selection of energy cane clones. In addition, plants with lower grade for smut incidence had a greater chance of being selected. The predictive capacities of the qualitative and quantitative models were 94% and 88%, respectively. The use of a qualitative model proved to be effective at predicting the number of energy cane genotypes to be selected and could be used as a selection strategy.


RESUMO: A análise de regressão logística é uma técnica que pode auxiliar programas de melhoramento genético na seleção de clones, especialmente nos estágios iniciais, em que a precisão experimental é baixa. Este trabalho teve como objetivo identificar os principais caracteres agronômicos para a seleção clonal de cana energia e verificar a eficiência do modelo logístico na predição dos genótipos a serem selecionados. As avaliações foram realizadas em 220 clones na primeira soca. Os dados foram submetidos à análise de regressão logística binária. O número de colmos por metro foi um caractere de maior importância na seleção de clones de cana energia. Além disso, os clones com nota menor para incidência de carvão tiveram maior chance de serem selecionados. As capacidades preditivas dos modelos qualitativos e quantitativos foram de 94% e 88%, respectivamente. O uso de um modelo qualitativo mostrou-se eficaz na previsão do número de genótipos de cana energia a serem selecionados e pode ser usado como estratégia de seleção.

ABSTRACT

The objective of this study was to evaluate the ecophysiological behavior of 'Tahiti' acid lime/rootstock grafted on Sunki mandarin hybrids during the prefloration stage of the plants, in the field, under saline stress. The progenies originating from crossings of the 'Sunki' mandarin with 'Rangpur' lime and Poncirus trioliata, as rootstocks, combined with the 'Tahiti' acid lime as a scion. The experiment was carried out under field conditions, at the Federal University of Campina Grande (UFCG), Pombal PB fields. In drainage lysimeter of 150 dm3, studying 10 scion/rootstock combinations, irrigated with two water salinity levels (CEa) (S1 = 0.3 and S2 = 3.0 dS m-1) making a 10 x 2 factorial scheme. The experimental design was a randomized block design, with 3 replicates and each plot consisting of a useful plant, totalizing 60 parcels. The scion/rootstock plants were transplanted at one year of age from the rootstocks sowing, being the beginning of the applications of the saline stress to the 15 days after the transplant lasting until the period of flowering, that is, up to 195 days after transplanting, During this period the plants were analyzed for growth and chlorophyll fluorescence a. The combinations less sensitive to salinity were composed of the 'Tahiti' grafted with TSKFL x (LCR x TR) - 018, TSKFL x TRBK - 011 and TSKFL x TRBK - 030, it being possible to use saline water with up to 3.0 dS m-1 in irrigation. Salinity did not damage the biochemical phase of the scion/rootstock combinations, only affecting the initial fluorescence of the lighted tissue.


Objetivou-se estudar o crescimento e a fluorescência da clorofila a em limeira ácida 'Tahiti' enxertada em híbridos de tangerineira 'Sunki' sob salinidade da água durante a fase de préfloração das plantas. Assim, com base em um delineamento experimental de blocos ao acaso, foram avaliadas dez combinações copa/porta-enxerto, referentes à referida limeira ácida em combinação com dez porta-enxertos (nove híbridos e o limoeiro 'Cravo Santa Cruz' como testemunha), irrigadas com dois níveis de salinidade da água (CEa) (S1=0,3 e S2=3,0 dS m-1), perfazendo um esquema fatorial 10 x 2, que foi repetido em três blocos e com parcela constituída por uma planta útil, totalizando 60 parcelas. As mudas enxertadas foram transplantadas aos 365 dias após a semeadura dos porta-enxertos, em lisímetros de 150 dm3, que foram preenchidos com um Neossolo Flúvico da região, sendo o início das aplicações do estresse salino aos 15 dias após o transplante (DAT), perdurando até o período de floração, ou seja, até os 195 DAT, avaliando o crescimento e a fluorescência da clorofila a. As combinações com maior crescimento quando irrigadas com água de condutividade de 0,3dS-1 foram aquelas com os porta-enxertos TSKC x (LCR x TR) ­ 017, TSKC x (LCR x TR) ­ 012, TSKFL x (LCR x TR) ­ 018, TSKFL x TRBK ­ 011 e TSKFL x TRBK ­ 028. A salinidade não ocasionou dano à fase bioquímica das combinações copa/porta-enxerto, apenas afetado a fluorescência inicial do tecido iluminado. É possível usar água salina de até 3,0 -1 no cultivo de citros, caso se empreguem combinações menos sensíveis, como aquelas tendo como porta-enxertos os híbridos TSKC x (LCR x TR) ­ 017, TSKFL x (LCR x TR) ­ 018, TSKFL x TRBK ­ 011 e TSKFL x TRBK ­ 030.
