ABSTRACT

Metallomics is a frontier interdisciplinary subject at its vigorous development stage. Its goal is to systematically study the content, distribution, chemical species, structural characteristics and functions of metal elements in biological system. It is also a comprehensive discipline to study the existing state and function of free or complex metal elements in life. Metallomics is an ideal tool to study the biological behavior of inorganic elements, which can be used to solve many problems in the research of mineral Chinese medicine(MCM). It provides a strong theoretical basis and technical support for the research of MCM. Its theory and methods provide re-ference and enlightenment for the in-depth study of MCM, and also provide new ideas and open up new ways for the research of MCM. The application of metallomics theory and methods in the research of MCM is of great significance to reveal the material basis and mec-hanism of MCM, promote the process of basic research on MCM, fully exploit and utilize medicinal mineral resources and carry forward the traditional MCM treasure in China. In this paper, we introduced the concept, academic development, research content and research methods of metallomics, and discussed the application prospects of metallomics in the analysis of inorganic element composition characteristics and quality control, material basis and mechanism of MCM, so as to provide reference for further researches on MCM.

This paper introduces the application and financing of programs of efficacy material base of traditional Chinese medicine funded by the National Natural Science Foundation of China(NSFC), the Youth Science Fund and the Regional Science Fund from 2016 to 2019, and conducts analysis and summary in terms of research objects and analysis methods, with the aim to provide reference for applicants for programs of efficacy material base of traditional Chinese medicine.

ABSTRACT

The four natures are the basic properties of Chinese materia medica (CMM). At present, it is one of the research hotspots to reveal the scientific connotation of CMM property theory. Clarifying the formation process and influencing factors of the medicinal properties of each single herb in a long historical period is the basic condition for the induction and summary of common characteristics of TCM property theory. Through the textual research of ancient books and documents, combined with the research results of modern Chinese medicine science, this paper combs the influence factors in the formation of the medicinal properties of Lonicera japonica, and reveals its scientific connotation. That is to say, the formation of the medicinal properties of each single herb of TCM is the result of its efficacy material base, the efficacy reflected in the process of clinical treatment and the tendency of clinical medication in previous dynasties. Based on the above research, the cause hypothesis of single herb property is put forward for the first time. The hypothesis provides a reference for the theoretical study of medicine property of TCM.

ABSTRACT

The function of Traditional Chinese Medicine (TCM) represents the overall effect of the formula. The effect is the expression of numerous biological effects of chemical components of TCM in certain pathological conditions of the body function adjustment, and these chemicals are the material basis of the TCM function. Limited by many conditions, few studies have been done on the material base of TCM function, and most of them have been supplemented by the substance basis of TCM efficacy. This leads to the disconnection between the theory of traditional Chinese medicine and modern research, and it is difficult to scientifically explain the functional indications of Chinese medicine. This paper discusses and summarizes the ideas and methods of modern research and evaluation of the material base of TCM function.Under the guidance of TCM theory and clinical practice, we set up an index system for evaluating the TCM function, and further explained the material basis and action mechanism of TCM formula. Through the organic combination of multidisciplinary and multi-technology, it provides a reference for the establishment of a comprehensive research model of the functional material basis of TCM formula.

ABSTRACT

Chinese materia medica (CMM) is the product of clinical practice of traditional Chinese medicine. Due to the complexity of the chemical composition of CMM and human body, the functional material base and its mechanism researches were facing a dilemma. In this paper, the chemical composition, antitumor material base, and related mechanism of Pinellia Rhizoma and Magnolia Officinalis Cortex were summarized and analyzed, and the existing problems were found. In order to better elucidate the functional material base and mechanism of the antitumor effects of Pinellia Rhizoma-Mangnolia Officinalis Cortex couplet medicines and reveal the scientific connotation of couplet medicines compatibility, the research thoughts of metabonomics combined with network pharmacology were proposed. With a view to providing theoretical basis and suggestion for the further research on the compound compatibility law.

ABSTRACT

As the basic form of Chinese materia medica (CMM) prescriptions, herb pair established a bridge linking single herb and CMM formula. Studying the compatibility mechanism of herb pair has important significance for revealing the compatibility mechanism of CMM formula. At present, many researchers studied the changes of the material basis, pharmacokinetics, and pharmacological effects after herb pair combined as the breakthrough point to explore compatibility mechanism of the herbal pair. In this paper, the research status were summarized and put forward prospects of the compatibility mechanism of the herbal pair and the foundation for modern research were expected to lay on compatibility mechanism and development of new drug based on the herb pair.

ABSTRACT

Cell membrane chromatography (CMC) was first proposed by Professor He in 1996. As one of bio-affinity chromatography technique, CMC was a simple and convenient technology in the study of interactions of active components in traditional Chinese medicines (TCMs) with membrane receptors in vitro, and screen active components from complicated TCMs. Recently, the CMC technology was developed rapidly, and widely applied in the discovery of lead compounds from nature product. This review article is focused on the application of cell membrane chromatography in the identification of active components in traditional Chinese medicine, together with the recent development of CMC methodology. Combining with our previous works in the analysis of the composition of complex substances, biochromatography and TLC bioautography for quality evaluation of TCM, we proposed a new holistic quality evaluation strategy of TCM related with bioactivity, which could be summarized as the integration of screen (screening of quality control marker by CMC), macroscopic characterization (characterizing the chemical material basis by multi-dimension and multi data fingerprinting) and microscopic description (multi-component quantification). The proposed strategy would provide a new idea for the holistic quality evaluation of TCM in the composition and concentration of bioactive components as quality evaluation indicators.

ABSTRACT

Prostaglandin (PG) E2 is an active substance in pathological and physiological mechanisms, such as inflammation and pain. The in vitro high-throughput assay for screening the inhibitors of reducing PEG2 production is a useful method for finding out antiphlogistic and analgesic candidates. The assay was based on LPS-induced PGE2 production model using a homogeneous time-resolved fluorescence(HTRF) PGE2 testing kit combined with liquid handling automation and detection instruments. The critical steps, including the cell density optimization and IC50 values determination of a positive compound, were taken to verify the stability and sensibility of the assay. Low intra-plate, inter-plate and day-to-day variability were observed in this 384-well, high-throughput format assay. Totally 5 121 samples were selected from the company's traditional Chinese medicine(TCM) material base library and used to screen PGE2 inhibitors. In this model, the cell plating density was 2 000 cells for each well; the average IC₅₀ value for positive compounds was (7.3±0.1) μmol; the Z' factor for test plates was more than 0.5 and averaged at 0.7. Among the 5 121 samples, 228 components exhibited a PGE2 production prohibition rate of more than 50%, and 23 components exhibited more than 80%. This model reached the expected standards in data stability and accuracy, indicating the reliability and authenticity of the screening results. The automated screening system was introduced to make the model fast and efficient, with a average daily screening amount exceeding 14 000 data points and provide a new model for discovering new anti-inflammatory and analgesic drug and quickly screening effective constituents of TCM in the early stage.

ABSTRACT

The compatibility of Salviae Miltiorrhizae Radix et Rhizoma (SMRR) and Carthamii Flos (CF) is a common pair-herb used in the modern traditional Chinese medicine prescriptions. As a function of promoting blood circulation and removing blood stasis, both SMRR and CF have been clinically used for hundreds of years and enduring popularity until today. At present, there have been many listed Chinese medicine prescriptions which were compatible with SMRR and CF. In this paper, we reviewed the research status of the formulae preparations, pair-herb compatibility, and component compatibility of SMRR and CF. We clarified the research advances in the chemistry, pharmacology, effective material base, and pharmacokinetics before and after the compatibility of SMRR and CF. Further, we indicated the scientificity and necessity of its compatibility and provided the references for the further research and modern formulation development of SMRR and CF.
