ABSTRACT

O objetivo foi avaliar a microbiota dos filés de surubim (Pseudoplatystoma spp.) submetidos à defumação com alecrim. Foram coletados fragmentos dos filés para cada etapa de avaliação (in natura, salmoura e defumação em 2, 4, e 6 horas). Da salga para a defumação houve uma redução, no qual, os microrganismos aeróbios mesófilos diminuíram de 1,4x105UFC/g para 1,8x104 UFC/g nos filés sem alecrim e 1,4x104 UFC/g nos filés com alecrim e na contagem de microrganismos aeróbios psicrotróficos foi observada redução de 1,9x105UFC/g para 7,8x104 UFC/g nos filés sem alecrim e 2,6x104 UFC/g nos filés com alecrim. Portanto, os filés adicionados de alecrim apresentaram queda constante e mais brusca na contagem bacteriana e resultou em uma boa redução da carga bacteriana.

ABSTRACT

O presente trabalho teve como objetivo avaliar e quantificar S. aureus e presença ou ausência de salmonela em peixes frescos nativos provenientes de viveiros escavados localizados na Região Norte. Foram analisados 4 lotes de pintado e 4 lotes de tambaqui, com 4 amostras (peixes) em cada lote, onde o lote representava uma despesca em tanque escavado. As análises foram realizadas com kits da 3M mundialmente utilizados e recomendados pela Association of Official Analytical Chemists (AOAC). Os resultados demonstraram presença de salmonella em todos os lotes de tambaqui e em três lotes de pintado. Quanto à contagem de S. aureus, apenas uma amostra (peixe) apresentou acima do estabelecido, da espécie pintado.

ABSTRACT

The Gram-negative intestinal microbiota of Hypostomus auroguttatus and Pimelodus maculatus, a detritivorous and an omnivorous fish species, respectively, were compared between fishes from the reservoir and the stretch of the river below the dam of the Funil hydroelectric plant, Rio de Janeiro, Brazil. Four selective culture media were used under aerobic and two under anaerobic conditions. The omnivorous species had microbiota with higher population levels compared to the detritivorous species. The number of morphotypes and population levels of total bacteria, vibrio and Bacteroides tended to be higher in summer and autumn in the reservoir, and not different in the river. The number of morphotypes of enterobacteria and total bacteria were higher in the lotic environment compared with the lentic one. The bacteria Aeromonas hydrophila and Plesiomonas shigelloides and the obligate anaerobic Fusobacterium mortiferum were the most frequently identified microorganisms in the intestine of both H. auroguttatus and P. maculatus. Both season and habitat influenced the Gram-negative intestinal microbiota of H. auroguttatus and P. maculatus. Environmental factors influenced the Gram-negative intestinal microbiota of both species with possible impact on the interrelationship between the fishes and their digestive ecosystem, although the gut microbiota composition of fishes may result from host-specific selective pressures within the gut.

ABSTRACT

273 bacterial strains were isolated from 20 Chinese longsnout catfish samples. The biochemical characteristics of all strains conformed to the species description of Aeromonas veronii bv. veronii on the basis of Vitek GNI+ card. Furthermore, 16S rDNA, gyrB and rpoD sequences of the representative strain PY50 were sequenced and showed high similarity with A. veronii bv. veronii in Genbank. Antibiotic-resistance of the representative strain PY50 was assessed by the Kirby-Bauer disk diffusion method, and the results showed it was susceptible and moderately susceptible to 13 and 4 of the 21 antimicrobial agents tested. Extracellular products of strain PY50 contained gelatinase, lecithinase, elastase, most of lipase and lipopolysaccharide. Virulence of strain PY50 and extracellular products to Chinese longsnout catfish were also tested, and LD50 were about 3.47~10(4) CFU per fish and 11.22 ƒÊg per fish in intraperitoneal injection respectively. This is the first report that A. veronii bv. veronii was the pathogenic agent of ulcerative syndrome in Chinese longsnout catfish.

ABSTRACT

Bacterial infection among the most common cultured mudfish Clarias gariepinus in Africa, has become a cause of concern, because it constitutes the largest economic loss in fish farms. In order to provide useful biological data of the pathogens for good management practices, samples were collected monthly between January 2008 and December 2009 in three monoculture nursery ponds, located in three different positions: upriver (A, grassland), mid-river (B, mixed forest and grassland) and downriver (C, rainforest) along 200km length of Cross River floodplains, Nigeria. A total of 720 fingerlings between 15.1 and 20.7g were analyzed to determine the degree of infection. The bacterial pathogens were taken from their external surfaces, and were isolated and identified by standard methods. The caudal fins of fingerlings from pond A had the highest bacterial load (5.8x10³cfu/g), while the least counts (1.2x103cfu/g) were identified on the head of fish from pond C, with Flexibacter columnaris as the major etiological agent. Pseudomonas fluorescens, Aeromonas hydrophila, Escherichia coli, Staphylococcus aureus and Micrococcus luteus were identified as co-isolates with P. fluorescens as dominant (0.7x10²cfu/mL) co-isolates in pond water. Clinical signs of five white spots with red periphery appeared on the external surface of infected fish. All the fish sampled, died after 4 to 9 days. There was no significant difference in the bacterial counts between different ponds, but the difference between fish organs/parts examined was significant. Fish from these ponds are therefore potentially dangerous to consumers and highly devalued, with the economic impact to producers. Preventive methods to avoid these infections are recommended. Rev. Las infeccines bacterianas son comunes en el pez de cultivo Clarias gariepinus, el cual es el más cultivado en Africa y se han convertido en una causa de preocupación, ya que constituye la mayor pérdida económica en las granjas piscícolas. Se proporcionan datos biológicos de los agentes patógenos con el fin de proporcionar información útil para buenas prácticas de gestión en las granjas. Las muestras fueron recolectadas mensualmente entre Enero 2008 y Diciembre 2009 en tres viveros de estanques de monocultivo, situados en tres posiciones diferentes: río arriba (A, pastizales), mitad del río (B, bosque mixto y pastos) y aguas abajo (C, bosque) a lo largo de 200km de longitud en las llanuras de inundación del río Cross, Nigeria. Un total de 720 alevines de entre 15.1 y 20.7g fueron analizados para determinar el grado de infección. Los patógenos bacteriales fueron tomados de las superficies externas, y fueron aislados e identificados por métodos estándar. Las aletas caudales de los alevines del estanque A tuvieron la mayor carga bacteriana (5.8x10³cfu/g), mientras el menor conteo de bacterias (1.2x103cfu/g) fue identificado en la cabeza de los peces del estanque C, con Flexibacter columnaris como el agente etiológico más importante. Pseudomonas fluorescens, Aeromonas hydrophila, Escherichia coli, Staphylococcus aureus y Micrococcus luteus se identificaron como co-aislamientos con P. fluorescens, como dominantes (0.7x102cfu/mL) co-aislados en el agua del estanque. Los signos clínicos fueron cinco puntos blancos con la periferia roja y aparecieron en la superficie externa de los peces infectados. Todos los peces de la muestra, murieron después de 4 a 9 días. No hubo diferencia significativa en los recuentos bacterianos entre los diferentes estanques, pero la diferencia entre los órganos y las partes de los peces examinados fue significativa. Objetivando o delineamento do perfil de sensibilidade dos agentes bacterianos causadores de enfermidades em peixes, 51 isolados bacterianos provenientes de Jundiá e pertencentes aos gêneros Acinetobacter spp. (8), Aeromonas spp. (15), Edwardsiella spp. (2), Enterobacter spp. (2), Klebsiella spp. (1), Plesiomonas spp. (5), Pseudomonas spp. (1), Staphylococcus spp.(11) e Vibrio spp. (6) foram testados frente aos antimicrobianos utilizados no tratamento de enfermidades em peixes. Dos 51 isolados bacterianos obtidos de exemplares de Jundiá (Rhamdia quelen) 51 (100 por cento) foram sensíveis a gentamicina, 49 (96,08 por cento) ao sulfazotrim, 47 (92,16 por cento) ao cloranfenicol, 43 (84,31 por cento), a tetraciclina, 43 (84,31 por cento) ao ácido nalidíxico, 31 (60,78 por cento) à nitrofurantoina, 22 (43,14 por cento) à eritromicina, 22 (43,14 por cento) à ampicilina, 15 (29,41 por cento) à espiramicina, 13 (25,50 por cento) à colistina e 5 (3 por cento) foram sensíveis a penicilina G. Com exceção de um isolado do gênero Staphylococcus spp., as bactérias analisadas no presente estudo foram resistentes a um ou mais agentes antimicrobianos testados. O conhecimento do perfil de sensibilidade das bactérias envolvidas em processos infecciosos nos peixes permitirá aos técnicos à adoção de uma antimicrobianoterapia racional, que contribuirá para o controle das enfermidades em Rhamdia quelen, sem causar grandes riscos à saúde pública e ao meio ambiente.(AU)


Aiming the evaluation of sensitivity profiles of pathogen bacteria responsible for fish diseases, 51 bacterial isolates from Jundiá (Rhamdia quelen) belonging to the genus Acinetobacter spp. (8), Aeromonas spp. (15), Edwardsiella spp. (2), Enterobacter spp. (2), Klebsiella spp. (1), Plesiomonas spp. (5), Pseudomonas spp. (1), Staphylococcus spp. (11), and Vibrio spp. (6), were tested against antimicrobial agents used for treatment of bacterial fish diseases. All samples were processed at the Laboratory of Bacteriology, Department of Preventive Veterinary Medicine, UFSM. From 51 bacteria isolated from jundiá fishes (Rhamdia quelen) 51 (100 percent) were sensitive to gentamycin, 49 (96,08 percent) to sulphazotrin, 47(92,16 percent) to chloramphenicol, 43 (84,31 percent) to tetracylin, 43 (84,31 percent) to naldixic acid, 31 (60,78 percent) to nitrofurantoin, 22 (43,14 percent) to erytromycin, 22 (43,14 percent) to ampicillin, 15 (29,41 percent) spiramycin, 13 (25,50 percent) to cholystin, and 5 (3 percent) to penicillin G. With exception of an isolate of Staphylococcus spp., the bacteria analyzed in the present study were resistant to one or more antimicrobial agents tested. Knowledge of the sensitivity profile of bacteria involved in infectious processes in fish will allow rational antimicrobial treatment that will contribute to the control of fish diseases in Rhamdia quelen without causing great risks to public health and the environment.(AU)

ABSTRACT

Pathogenic Aeromonas hydrophila (strain VB21), a multiple-drug resistance strain contains a plasmid of about 21 kb. After curing of plasmid, the isolates became sensitive to antimicrobials, to which they were earlier resistant. The cured bacteria exhibited significant alterations in their surface structure, growth profile and virulence properties, and failed to cause ulcerative disease syndrome (UDS) when injected into the Indian catfish Clarias batrachus. Routine biochemical studies revealed that the plasmid curing did not alter the biochemical properties of the bacteria. After transformation of the plasmid into cured A. hydrophila the bacterium regained its virulence properties and induced all the characteristic symptoms of UDS when injected into fish. Thus, the plasmid plays a pivotal role in the phenotype, growth and virulence of A. hydrophila and pathogenesis of aeromonad UDS.
