ABSTRACT

Canine Distemper is a disease caused by Canine morbillivirus (CM), a pantropic virus that can affect the central nervous system (CNS), causing demyelination. However, the pathogenesis of this lesion remains to be clarified. Brain samples of 14 naturally infected dogs by CM were analyzed to evaluate the presence of oxidative stress and demyelination. RT-PCR assay was performed to confirm a diagnosis of canine distemper in the brain, immunohistochemistry anti-CM was used to localize the viral proteins in the tissue, and anti-4-hydroxy-2-nonenal (4-HNE) was a marker of a product of lipid peroxidation. The results showed the presence of viral proteins in the demyelinated area with the presence of 4-HNE. Our results suggest that the CM virus infection causes oxidative stress leading to lipid peroxidation, which causes tissue damage and demyelination. In conclusion, oxidative stress plays a significant role in canine distemper pathogenesis in the CNS.(AU)


A cinomose canina é uma doença causada pelo Morbilivírus canino (CM), um vírus pantrópico que pode afetar o sistema nervoso central (SNC), causando desmielinização. No entanto, a patogênese dessa lesão não está totalmente esclarecida. RT-PCR e imuno-histoquímica foram realizadas para confirmação do diagnóstico de cinomose em amostras de encéfalo de 14 cães naturalmente infectados. Após confirmação, foi realizada uma avaliação do estresse oxidativo por imuno-histoquímica com uso de anti-4-hidroxi-nonenal (4HNE) como marcador de produtos resultantes da peroxidação lipídica. Os resultados sugerem que a infecção pelo CM causa estresse oxidativo no tecido, levando a peroxidação lipídica, a qual causa danos ao tecido, culminando com desmielinização. Conclui-se que o estresse oxidativo tem papel importante na patogênese da cinomose canina no sistema nervoso central.(AU)

Objetivou-se através deste trabalho, determinar a prevalência de cinomose canina no semiárido da Paraíba, através de testes rápidos imunocromatográficos, correlacionando-a com os principais achados clínicos e hematológicos. Foram analisadas 67 fichas de animais em que foram realizados testes rápidos para pesquisa de antígeno em amostras nasais e oculares no período de janeiro a dezembro de 2019. Observou-se que 47% (32/67) dos cães analisados foram positivos para cinomose canina. As variáveis que apresentaram diferença estatística significativa (p<0,05) para a infecção foram animais sem raça definida 60% (21/35), animais não vacinados 70% (29/42), e período seco do ano, sendo o mês de agosto (40%; 13/32), com maior ocorrência. Os principais sistemas afetados foram o respiratório 61% (17/28), oftalmológico 70% (22/31), nervoso 69% (13/19), dermatológico 45% (9/20), e gastrintestinal 42% (6/14). As principais alterações hematológicas foram anemia 66% (23/32), leucopenia 76% (19/25) e linfopenia 48% (15/31). Concluiu-se que foi elevada a ocorrência de cinomose canina em animais com suspeita clínica no Semiárido Paraibano, e animais sem raça definida, não vacinados, no período seco do ano foram mais diagnosticados com a enfermidade.

ABSTRACT

Descrevem-se os principais aspectos epidemiológicos, clínicos e anatomopatológicos de casos de hepatite infecciosa canina diagnosticados no Laboratório de Patologia Animal da Universidade Federal de Campina Grande, durante o período de janeiro de 2003 a dezembro de 2016. Dos 1.640 cães necropsiados, 15 foram diagnosticados como hepatite infecciosa canina (0,91%). Dos cães acometidos nove eram machos e seis fêmeas. As idades variaram de 45 dias a sete anos, sendo a maioria filhotes. Dez animais não apresentavam raça definida, quatro eram Poodles e um Rottweiler. A maioria dos cães não recebeu nenhum tipo de protocolo vacinal. Os cães eram oriundos dos municípios de Patos, São Mamede e Teixeira, pertencentes ao estado da Paraíba, Nordeste do Brasil. A maioria dos cães apresentou curso clínico variando de hiperagudo a agudo. Os principais sinais clínicos foram convulsão, apatia e hiporexia. Na necropsia, as principais alterações foram observadas no fígado que se apresentava de pálido a alaranjado e com áreas irregulares avermelhadas na superfície capsular, além de acentuação do padrão lobular e edema na parede da vesícula biliar. Hemorragias foram observadas em vários órgãos. Na histopatologia havia necrose centrolobular de hepatócitos associada a corpúsculos de inclusão viral intranucleares, hemorragia e infiltrado inflamatório misto. O diagnóstico foi estabelecido com base nas características lesões histopatológicas e foi confirmado por imuno-histoquímica. A hepatite infecciosa canina ocorre ocasionalmente na Paraíba, acometendo particularmente cães jovens e não vacinados.(AU)


We described the main epidemiological, clinical and pathological aspects of canine infectious hepatitis diagnosed in the Laboratory of Animal Pathology of the Federal University of Campina Grande during the period from January 2003 to December 2016. Of the 1,640 necropsied dogs, 15 were diagnosed as infectious canine hepatitis (0.91%). Of the dogs affected nine were males and six females. The ages ranged from 45 days to seven years, being most of them young. Ten animals were mixed breed, four were Poodles and one Rottweiler. Most of the dogs do not received any vaccine protocol. The dogs came from the municipalities of Patos, São Mamede and Teixeira, from Paraiba, northeastern of Brazil. Most of the dogs presented clinical course varying from hyperacute to acute. The main clinical signs were seizure, apathy and hyporexia. At necropsy, the major alterations were observed in the liver, which was pale to orange and with irregular reddish areas on the capsular surface, besides accentuation of the lobular pattern and edema in the wall of the gallbladder. Hemorrhages were observed in several organs. In the histopathology there was centrolobular necrosis of hepatocytes associated with intranuclear viral inclusion bodies, hemorrhages and mixed inflammatory infiltrate. The diagnosis was established based on the characteristic histopathological lesions and was confirmed by immunohistochemistry. Infectious canine hepatitis occurs occasionally in the Paraiba, affecting particularly young and unvaccinated dogs.(AU)

ABSTRACT

O objetivo do presente trabalho foi determinar a presença de anticorpos para Leishmania infantum, Neospora caninum e Toxoplasma gondii, por meio da reação de imunofluorescência indireta (RIFI), em cães (n=78) provenientes da região central do Rio Grande do Sul, necropsiados no Hospital Veterinário da Universidade Federal de Santa Maria (UFSM), bem como avaliar os dados epidemiológicos, sazonais e anátomo-histopatológicos. Do total de animais avaliados, 67,9% (53/78) apresentaram soropositividade para ao menos um agente. A ocorrência de anticorpos para L. infantum, N. caninum e T. gondii foi de 33,3 (26/78), 37,1 (29/78) e 43,5% (34/78), respectivamente. Detectaram-se monoinfecções em 9,4% (5/53) para L. infantum, 18,8% (10/53) para N. caninum e 20,7% (11/53) para T. gondii. As coinfecções foram observadas em 27/53 (50,9%) dos animais. As infecções ocorreram independentemente de idade, sexo, procedência ou raça (P>0,05). Não se verificaram lesões anatomo-histopatológicas relacionadas aos agentes pesquisados, caracterizando-os como animais assintomáticos. Os resultados confirmaram a exposição de cães a esses protozoários na região central do RS e, em especial, demonstraram a circulação do agente causador da leishmaniose em uma área considerada indene para a enfermidade.(AU)


The present paper is aimed to determine the presence of antibodies for Leishmania infantum, Neospora caninum and Toxoplasma gondii by indirect immunofluorescence (IIF) in dogs (n=78) from the central region in the state of Rio Grande do Sul necropsied in the Veterinary Hospital from Universidade Federal de Santa Maria (UFSM). The data was evaluated regarding epidemiological, anatomic, and histopathologic findings. Of the total animals evaluated, 67.9% (53/78) showed seropositivity for at least one agent. The occurrence of antibodies to L. infantum, N. caninum and T. gondii was 33.3% (26/78) 37.1% (29/78) and 43.5% (34/78), respectively. The mono infections were detected in 9.4% (5/53) of L. infantum, 18.8% (10/53) for N. caninum and 20.7% (11/53) T. gondii. The coinfections occurred in 50.9% (27/53) of animals. There were not anatomical and histopathological lesions regarding these surveyed agents, characterizing them as subclinical animals. The results confirmed the exposition of dogs to these protozoa in the central region of the RS, highlighting the circulation of the causer agent of leishmaniasis in an area considered harmless for the disease.(AU)

ABSTRACT

ABSTRACTRabies virus (RABV) isolated from different mammals seems to have unique characteristics that influence the outcome of infection. RABV circulates in nature and is maintained by reservoirs that are responsible for the persistence of the disease for almost 4000 years. Considering the different pattern of pathogenicity of RABV strains in naturally and experimentally infected animals, the aim of this study was to analyze the characteristics of RABV variants isolated from the main Brazilian reservoirs, being related to a dog (variant 2),Desmodus rotundus (variant 3), crab eating fox, marmoset, and Myotis spp. Viral replication in brain tissue of experimentally infected mouse was evaluated by two laboratory techniques and the results were compared to clinical evolution from five RABV variants. The presence of the RABV was investigated in brain samples by fluorescent antibody test (FAT) and real time polymerase chain reaction (qRT-PCR) for quantification of rabies virus nucleoprotein gene (N gene). Virus replication is not correlated with clinical signs and evolution. The pattern of FAT is associated with RABV replication levels. Virus isolates from crab eating fox and marmoset had a longer evolution period and higher survival rate suggesting that the evolution period may contribute to the outcome. RABV virus variants had independent characteristics that determine the clinical evolution and survival of the infected mice.

ABSTRACT

O estudo tem o objetivo de identificar efeitos indesejáveis da ribavirina, prednisona e DMSO em cães naturalmente infectados com o vírus da cinomose. Foram utilizados 60 cães apresentando quadro neurológico da cinomose com evolução de 10 dias. Os animais foram internados e receberam tratamento de suporte; foram avaliados diariamente e realizados hemograma, dosagem bioquímica e exame de urina tipo I. Os grupos 1 e 2 foram tratados com ribavirina e sua associação com DMSO; os grupos 3 e 4 com DMSO e prednisona e o grupos 5 com ribavirina e prednisona e o grupo 6 com ribavirina, prednisona e DMSO. Os animais foram anestesiados para a colheita de líquor, medula óssea e sangue, antes do tratamento para diagnóstico através da RT-PCR. As amostras negativas foram analisadas pela técnica de hn-PCR. Todos os animais apresentaram resultado positivo em pelo menos uma das duas reações. O efeito adverso da ribavirina e a sua associação com a prednisona foi a anemia hemolítica, que foi confirmada pela observação de bilirrubina na urina apenas dos cães tratados com ribavirina.


The present study aims at the identification of undesirable effects of ribavirin, predinisone and DMSO in dogs naturally infected by canine distemper virus. The research analyzed 60 dogs with clinical neurological signs and 10 days of evolution. The animals were hospitalized for the appropriate support treatment; were daily observed, and complete blood cells count, biochemical analysis, and urine exam type I were conducted. Groups 1 and 2 were treated with ribavirin and its combination with DMSO; Groups 3 and 4 treated with prednisone and DMSO, Group 5 treated with ribavirin and prednisone, while Group 6 with ribavirin, prednisone and DMSO. Before the treatment, animals were anesthetized for the cerebrospinal fluid, bone marrow and blood samples collection for the diagnosis based on RT-PCR. The negative samples were analyzed using the hn-PCR technique. All the animals presented positive results in at least one of the 2 tests. The adverse result of ribavirin and its association with prednisone was characterized by haemolytic anemia, confirmed by the evaluation of bilirrubin occurrence only in the urine of dogs treated with ribavirin.

ABSTRACT

Despite the widespread prevalence of canine parvovirus disease [CPV] in Iranian dog population, molecular diagnosis of CPV variants, and investigation of the trends of its genetic changes is a new effort. In this study 50 samples from dogs suspicious of infection with clinical signs of diarrhea and vomiting, and 25 samples from dogs suspected of infection with general symptoms such as depression and anorexia were collected from dogs presented to the veterinary clinic of Shiraz University, Shiraz, Iran. Viral DNA was extracted from feces. Three specific pairs of primers, P2, Pab, and Pb, were used in a PCR assay for differential diagnosis of the virus type. Pab primer pairs detect the new type-strains, CPV-2a and 2b. The primer pairs P2 and Pb detect CPV types 2 and 2b, respectively. Our results showed that 44 individuals with clinical signs of diarrhea and vomiting were positive for CPV-2. 39 individuals [89%] were positive for CPV-2b and 5 individuals [11%] for CPV-2a. Therefore, the CPV-2b was identified as the predominant virus type. All dogs without symptoms of diarrhea and vomiting were CPV-negative. The relationship of breed, age and sex with PCR results was not significant [P>0.05]. For the first time in the country, the causative agent of CPV-2 was identified, and presence of new antigenic variants, CPV-2a and CPV-2b was confirmed

Investigou-se a ocorrência de leptospirose em cães da cidade de Patos, semiárido paraibano, e realizou-se um estudode fatores de risco associados à infecção. Foram examinadas 152 amostras de soro sanguíneo de cães atendidos emhospital veterinário no período de julho a novembro de 2008. O diagnóstico da leptospirose foi realizado pela técnica desoroaglutinação microscópica, utilizando-se uma coleção de 24 sorovariedades. A frequência de animais soropositivosencontrada foi de 19,73% (IC 95% = 13,73% - 26,96%), e as sorovariedades mais frequentes foram Autumnalis (13,16%)e Grippotyphosa (1,97%). A análise de regressão logística múltipla mostrou que os fatores de risco para a leptospiroseforam a não definição da raça (odds ratio = 3,67; P = 0,009) e o contato com caprinos/ovinos (odds ratio = 10; P = 0,034).


The occurrence of leptospirosis was investigated in dogs from Patos city, semiarid of Paraíba State, Northeast region ofBrazil, and the risk factors associated with the infection were analyzed. One hundred and fifty two blood samples werecollected from dogs attended in a veterinary hospital during July to November 2008. The diagnostic method carriedout for leptospirosis was the microscopic agglutination test, using a batch of 24 leptospiral serovars. The frequency ofseropositive animals was 19.73% (95% CI = 13.73% - 26.96%) and most frequent reactant serovars were Autumnalis(13.16%) and Grippotyphosa (1.97%). The multivariate logistic regression analysis showed that the risk factors forleptospirosis were mixed breed (odds ratio = 3.67; P = 0.009) and the contact with caprine/ovine (odds ratio = 10; P =0.034).

ABSTRACT

Relatam-se o primeiro isolamento de herpesvirus canino 1 (CaHV-1) e a localização atípica das lesões vesiculares associadas a este vírus na Argentina. A amostra foi recuperada de lesões vesiculares, localizadas na parte interna da coxa direita, em uma fêmea de raça Labrador. A cadela tinha quatro anos de idade e era de propriedade privada. O primeiro diagnóstico foi realizado pela reação em cadeia da polimerase e, posteriormente, o vírus foi isolado e sua identificação confirmada por imunofluorescência indireta e pelo teste de neutralização viral.

ABSTRACT

Two male dogs at the age of 3 and 5 months of Doberman pinscher and German shepherd breeds were referred to the Veterinary Hospital of Ahvaz University with clinical signs of depression, vomiting, haemorrhagic diarrhoea, profound dehydration, fever and anorexia. The affected dogs had not the history of vaccination. Concurrent infection of canine parvovirus and coronavirus was detected in dogs by means of immunochromatography assay. The haemogram showed lowered white blood cell counts as leukopenia, neutropenia and lymphopenia. The dogs received supportive treatment to correct the life-threatening dehydration and prevention of secondary bacterial infections. Despite treatment, one dog [Doberman pinscher] died within 3 days after the onset of clinical signs, and the second dog recovered after 4 days. To the best of our knowledge, this is the first report of concurrent infection of canine parvovirus and coronavirus in diarrhoeic dogs in Iran

ABSTRACT

Empregou-se a técnica de reação em cadeia pela polimerase precedida de transcrição reversa para detecção do vírus da cinomose canina (CC). Para a padronização da técnica foram selecionados quatro pares de oligonucleotídeos (P1, P2, N1, H1), baseados em seqüências dos genes da fosfoproteína, neuraminidase e hemaglutinina, sendo utilizadas três cepas vacinais de vírus da CC como controles positivos. Foram analisadas três amostras isoladas de cães com cinomose e quatro amostras provenientes de cães com suspeita clínica de cinomose. Não houve amplificação nas amostras com suspeita clínica da doença. Os resultados obtidos com os oligonucleotídeos P1 e N1 foram superiores aos de H1. Os oligonucleotídeos P2 foram considerados inapropriados para a detecção do vírus da CC. Os amplicons obtidos com os oligonucleotídeos P1, N1 e H1 foram clivados com endonucleases de restrição, sendo os perfis das amostras virais comparados aos da amostra vacinal Lederle, utilizada como referência. Um padrão similar de restrição foi observado em todas as amostras analisadas


The reverse transcription-polymerase chain reaction was used to detect canine distemper virus (CDV). Four oligonucleotide pairs were selected (P1, P2, N1, H1), based on the sequences of the phosphoprotein, hemagglutinin and nuraminidase genes for assay standardization, and three CDV vaccine strains were used as positive controls. Three viral isolates from dogs with canine distemper and four samples from animals clinically suspected of distemper were analysed. No amplification was detected in suspected samples. Results obtained by using P1 and N1 oligonucleotides were superior to those with H1 ones. P2 oligonucleotides were considered inadequate for CDV detection. Amplicons resulting from amplification of P1, N1 and H1 oligonucleotides were submitted to cleavage by restriction endonucleases and restriction patterns of viral samples were compared to that of Lederle strain used as reference. A similar restriction pattern was observed in all analysed samples

ABSTRACT

O parvovirus canino pode estar associado a parasitas intestinais, vírus ou bactérias enteropatogênicas, os quais podem predispor ou contribuir para o agravamento do quadro mórbido e, nos casos de diarréia grave, na instalaçäo do choque endotóxico. Com a finalidade de analisar a existência de infecçäo bacteriana concomitante ou de bacteremia, em cäes com enterite hemorrágica aguda e grave, causada por parvovírus, procedeu-se à coprocultura e à hemocultura, em 34 animais atendidos no Hospital Veterinário da FMVZ/USP. O diagnóstico clínico e etiológico de parvovirose baseou-se no perfil hematológico e na presença de partículas virais nas fezes dos cäes, observadas por meio de microscopia eletrônica. Näo foram isoladas Salmonella sp ou E. coli enteropatogênica, ao contrário de Campylobacter jejuni, que foi isolado das fezes de cinco dos pacientes estudados. Em relaçäo à hemocultura, Pseudomonas sp, E. coli, Alcaligenes odorans e bacilo Gram-negativo näo fermentador, näo identificado, foram isolados de cinco cäes, dos quais quatro sofreram evoluçäo fatal. Aparentemente, a infecçäo bacteriana concomitante näo exerce papel de destaque na evoluçäo do processo, mas existe uma invasäo da corrente sangüínea por bactérias pertencentes à flora intestinal normal, o que resulta no agravamento do quadro mórbido, contribuindo para a instalaçäo do choque endotóxico e o óbito do animal
