ABSTRACT

Glycyrrhizin, a triterpenoid glycoside and Licorice from Glycyrrhiza glabra and Ammonium salt of Glycyrrhizic acid (Sigma) were tested for antiviral activity on three strains of Japanese encephalitis virus (JEV), Nakayama, P-20778 and 821564 XY48. Purified glycyrrhizin (M.w. 822.92) inhibited plaque formation in all the three strains of JEV at a concentration of 500 micrograms/ml at 96 hrs, Similar effect was observed at 1000 micrograms/ml concentration with Licorice and Ammonium salt of glycyrrhizic acid. The minimal inhibitory concentrations were not toxic to porcine stable kidney (PS) and human cervical carcinoma (HeLa) cell lines. Cyctotoxicity of these chemicals was evaluated by trypan blue dye exclusion test which indicated subtoxic concentrations at 5,000 micrograms/ml at 96 hrs and toxic concentrations were 10,000 micrograms/ml at the same time period for the host cells PS. Thus the indigenously purified glycyrrhizin seems to be more potent antiviral agent than Licorice and ammonium salt of glycyrrhizic acid (Sigma) for JEV 'in vitro'.

ABSTRACT

The preliminary results of our study in vitro on the effect of Interferon on Flaviviruses showed that Interferon alpha and Interferon beta were more effective on JE viruses Vip local strain and JaGAr strain, but less on Dengue virus type 2 strain. However, the effect of these 2 interferons on the 2 strains of JE viruses were still variable which need further investigations. The JE virus Vip local strain seemed to be more susceptible to interferon than the Japanese prototype JaGAr strain. Thus, the in vivo trial on JE disease in Thailand is strongly recommended.

ABSTRACT

Human serum lipoproteins were purified by ultracentrifuging and their concentrations adjusted as required to be within the normal male/female serum range for all assays. The activities in inhibition of hemagglutination (HAI) for Japanese encephalitis virus were--low density lipoprotein (LDL) greater than very low density lipoprotein (VLDL) greater than high density lipoprotein (HDL). Heating (56 degrees C/30 minutes) caused the LDL titer to fall and freeze-thawing (20 degrees C/room temperature) the VLDL titer to rise slightly, possibly as a result of alteration in lipoprotein structure. The additon of lipoprotein depleted serum appeared to dampen these effects and there was no nett change in titer when it was added to a lipoprotein mixture. Similarly, unfractionated normal serum showed no significant change in titer after these treatments. The lipoproteins lacked significant virus neutralizing (VN) activity and this remained so in spite of fluctuations in HAI titer after heating and freeze-thawing.
