ABSTRACT

This study aimed to investigate the effects of ethylene biosynthesis inhibitors on oxidative metabolisms and the in vitro conservation of Lippia filifolia, using the lipid peroxidation index (TBARS), antioxidative enzymes and pigments as biomarkers. We found that EDTA, sodium thiosulfate (STS) and especially Co had protective effects on oxidative stress in tissues cultured in vitro, resulting in a delay of the senescence and the reduction of subcultures frequency, con-tributing to the germplasm conservation of this species.


O objetivo deste estudo foi investigar os efeitos de inibidores da biossíntese do etileno no metabolismo oxidativo e na conservação in vitro de Lippia filifolia. Para isso, foram avaliados o índice de peroxidação lipídica (TBARS), a atividade de enzimas antioxidativas e o conteúdo de pigmentos fotossintéticos e de antocianinas. Os resultados evidenciaram que o EDTA, o tiossulfato de sódio (STS) e, especialmente, o Co apresentaram ação protetora sobre o estresse oxidativo nos tecidos, o que resultou em atraso no início da senescência das culturas e na redução da frequência dos subcultivos, contribuindo para a conservação do germoplasma dessa espécie.

ABSTRACT

Two independent parameters, epicotyl height (cm) and number of induced buds were studied on Pinus pinaster explants to analyse the effects of three phytohormones (6-benzylaminopurine, jasmonic acid, ethylene) which were combined or not in 11 different treatments. Epicotyle length diminished significantly in relation to the control medium (medium without exogen phytohormones) in presence of jasmonic acid, 6-benzylaminopurine or Ethephon (which is converted to ethylene in plants) in any of treatments. Concentrations of 100 microM of jasmonic acid and Ethephon had a greater inhibitory effect than the treatments with 10 microM. In addition to that, jasmonic acid was a stronger inhibitor than Ethephon in any of the tried combinations. There were no significant differences between the control treatment and the treatment s with only 10 microM of jasmonic acid or Ethephon. However, 10 microM 6-benzylaminopurine induced bud formation. The different combinations of 6-benzylaminopurine with jasmonic acid and Ethephon showed that concentrations of 10 to 100 microM did not affect the number of induced buds. Jasmonic acid had an inhibitory effect which Ethephon only showed when combined with 100 microM of jasmonic acid and 10 microM of 6-benzylaminopurine. Three response groups were defined by cluster analysis: group 1 produced the greatest mean number of buds (4 to 5) and a mean epicotyl growth of 1 to 1.5 cm; group 2 produced 2 to 4 buds and a mean growth of 0.5 to 1.2 cm; group 3 produced only one bud and a mean epicotyl length of 1.2 to 2 cm.

ABSTRACT

Com o objetivo de estabelecer relaçöes entre a maturaçäo pós-colheita e o estádio de desenvolvimento do maracujá-amarelo (Passiflora edulis f. flavicarpa) por ocasiäo da colheita, estudaram-se as modificaçöes em alguns constituintes químicos, físicos e na biossíntese do etileno e o efeito do tratamento com ethephon sobre o amadurecimento pós-colheita. A taxa de respiraçäo do fruto mantido na planta decresce gradualmente com o tempo mas sem mostrar o climatério respiratório. A produçäo de etileno e o climatério säo antecipados com a colheita dos frutos. Os frutos colhidos aos 65 dias após a antese apresentam elevaçäo no teor de ACC, na atividade ACC oxidase e na taxa de produçäo de etileno, indicando início da autocatálise. A aplicaçäo de ethephon, imediatamente após a colheita, näo antecipa o climatério respiratório dos frutos, mas aumenta as taxas de respiraçäo, de produçäo de etileno e de perda de massa, bem como, os sólidos solúveis totais e a acidez total titulável. Ao final do amadurecimento, o teor de sacarose aumenta e os de glicose e frutose pouco säo alterados. Os sólidos solúveis se estabilizam, enquanto a acidez total titulável aumenta, até sessenta dias após a antese e entäo decresce. Os resultados indicam que o maracujá-amarelo atinge a maturidade fisiológica entre 60 e 65 dias após a antese
