ABSTRACT

In order to obtain nucleotides aptamers bind to IgE, 80 bp nucleotides single-stranded DNA library containing 40 random nucleotides was designed and synthesized. Oligonucleotides that bind to human Cepsilon3-Cepsilon4 protein were isolated from ssDNA pools by the systematic evolution of ligands by exponential enrichment (SELEX) method using nitrocellulose filters as screening medium. Through the optimization of critical PCR and asymmetric PCR parameters including annealing temperature, cycles, and molar ratios of target protein and ssDNA etc, a suitable screening system was established. The aptamers of Cepsilon3-Cepsilon4 protein with high affinity and high specificity were identified by ELISA with biotin-streptavidin-horseradish peroxidase system, and its primary sequence and second structure were analyzed by DNAMAN package and DNA folding sever after being cloned and sequenced. Moreover, target protein was bound to one aptamer and another aptamer modified with biotion together forming a sandwich-like complex, which was captured in microwell to detect IgE concentration using the optimal combination in the sandwich method named enzyme-linked aptamers sorption assay (ELASA). The method could be used for the quantitative detection of human IgE, and whose sensitivity reached to 120 ng x mL(-1).

ABSTRACT

Serum IgE and beta 2 microglobulin levels were determined in 31 patients suffering from acute myocardial infarction and 30 patients with other forms of ischemic heart disease. The levels of these parameters were studied on the first, third and seventh day after the onset of disease. The immunological method used for the determinations was ELISA Patients with myocardial infarction showed an elevated level of both parameters. The peak value of IgE was observed on the seventh day [P<0.05] but that of beta 2 microglobulin on the third day [P<0.05] after the onset of myocardial infarction

ABSTRACT

The relationship between intradermal skin tests reactivity and serum level of total IgE was evaluated in 200 atopic subjects selected from a general population sample randomly [having strong family history of allergy] attending ENT OPD of Sir Ganga Ram Hospital, Lahore. Intradermal test results were related to the history of allergy status of the patients and total serum IgE levels. Intradermal test reactivity to dust and pollen correlated with the presence of allergy symptoms. Similarly mean total serum IgE levels [Behring method] were significantly higher [P<0.001] in allergic [333.3 I.U./ml] than in non-allergic subjects [75.34 I.U./ml]. Positive association was found between the degree of intracutaneous test results and serum IgE levels

ABSTRACT

El CD23, descripto como una glucoproteína de 45 kDa, está relacionado con la activación de los linfocitos B y la regulación de la síntesis de IgE. Se degrada en fragmentos solubles (CD23s). Es una multicitocina involucrada en patologías con predominio de linfocitos TH2, tales como las enfermedades alérgicas, leucemia B crónica o en injerto de médula ósea. El 70 por ciento de los asmáticos presenta niveles de IgE aumentados. El eosinófilo es la célula central de la inflamación asmática. Objetivo: se evaluó la corrrelación entre niveles de CD23s, IgE sérica, eosinofilia sanguínea y esputo en asmáticos atópicos y por aspirina, frente a un grupo control de individuos normales. Pacientes: se estudiaron 27 pacientes atópicos: 14m, 13f, 10 asmáticos por AAS: 4m, 6f. Un grupo control de 10 sujetos: 5m, 5f. Nadie recibió corticoides en los dos meses previos. Se determinaron IgE total por ELISA Kabi-Pharmacia, CD23s por ELISA, Binding Site, MK112. Recuentos de eosinófilos en esputo y sangre periférica. Resultados: IgE:asmáticos atópicos: 500Ï252kU/l. Asmáticos por AAS: 68Ï27 kU/l. Controles: 51Ï20 kU/l<0.0005. CD23s: asmáticos atópicos: 7.65Ï2.25 ng/ml. Asmáticos por AAS: 3.4Ï0,95 ng/ml. Controles: 3.73Ï1.08 ng/ml; p<0.0005. Eosinófilos en esputo: asmáticos atópicos: 25Ï13 por ciento. Asmáticos por AAS: 23Ï13 por ciento. NS. Eosinofilia sanguínea: asmáticos atópicos: 685Ï316/mm3. Asmáticos por AAS: 555Ï190/mm3 NS. Control: 255Ï90mm3; p<0.0005. Conclusiones: nuestro estudio demuestra un incremento de la IgE y CD23s en los asmáticos atópicos. Los sensibles a aspirina no presentaron cambios significativos respecto del grupo control. La eosinofilia positiva se encontró en ambos grupos de asmáticos. Esto sugiere diferentes mecanismos inflamatorios en asma bronquial, con una vía común representada por los eosinófilos

ABSTRACT

The present study included 20 patients with atopic dermatitis [AD]. These patients comprised 2 groups; the first group included 10 patients with only AD [pure AD], the second group included 10 patients with AD and respiratory manifestations of bronchial asthma [AD with BA]. These patients as well as 10 normal individuals as a control group were subjected to HLA typing using lymphocyte microcytotoxicity testing. Serum total IgE was determined in the patients and control group. The results of the present study showed no significant difference between patients and control. The results of HLA typing of patients subgroups [only AD, AD with BA] were compared to control. It was found that HLA-A1 was significantly higher in patient groups in comparison to control group as P <0.05 and P <0.01, respectively. HLA-B8 was not significantly higher in patient groups in comparison to control as RR was 0.58 and 1, respectively. HLA-A3 was significantly increased in patient groups in comparison to control as P <0.05 and P <0.05, respectively. HLA-B7 was significantly increased in patient groups in comparison to control as P <0.01 and P <0.01, respectively. The present study also revealed the serum total IgE to be significantly increased in both groups of patients in comparison to control group. It was evidented that the amount of serum total IgE in cases of AD is much less than that in cases suffering from AD with BA. These results were discussed

ABSTRACT

Immunization of mice against T. spiralis infection using T. spiralis larval glycoproteins as homogenous antigen and crude hydatid fluid as heterogenous antigen showed a marked reduction in larva burden and a significant rise of serum IgE in immunized mice infected with T. spiralis larvae as compared with control group. More reduction was observed with immunization with homogenous T. spiralis larval antigen compared with heterogenous hydatid fluid antigen. The reduction in larval burden with rise of serum IgE begins after the third week of immunization

ABSTRACT

The present study was designed to evaluate the importance of IgE- antibody level estimation in patients with hydatid disease of the liver, before and after surgical treatment. Serum IgE level was estimated, preoperatively, 3 and 6 months postoperatively in 20 patients suffering from hydatid disease of the liver. The results were compared with 10 healthy control cases with no past or present history of any other affection that could affect the results in the diseased or control group. All diseased cases showed highly significant increase [p <0.001], when preoperative mean level of IgE antibody which was 285.6 +/- 26 IU/ml, compared with the mean level in the control group which was 110.4 +/- 6.6 IU/ml. However, in postoperative curable cases, i.e. those cases without any evidence of recurrence up to 6 months after the operation, no significant change [p >0.05] was observed when the mean level of IgE antibody, 3 and 6 months after the operation was compared with control group. On the other hand, and in those curable cases also, the results showed a demonstrated highly significant fall [p <0.001] in the mean level of IgE antibody from preoperative mean of 285 +/- 25 IU/ml, to postoperative mean of 112.7 +/- and 111.5 +/- 7.4 IU/ml, respectively, for 3 and 6 months after the operation. Recurrence was detected 6 months after the operation in 3 cases, in these recurrent cases the mean average of IgE antibody level was 323.3 +/- 25.2 IU/ml, which is of high significant increase when compared with the mean level in normal control group or postoperative curable cases [p <0.001]. So, estimation of serum IgE antibody level in patients with hydatid disease of the liver plays a very important role from diagnostic and prognostic point of view, as its level is closely related to primary development of the disease or its recurrence after surgical treatment

ABSTRACT

Empleando la tecnica de inmunofluorescencia directa se estudio la presencia de receptores de tipo IgE en la membrana de los linfocitos de 30 pacientes atopicos comparativamente con un grupo control constituido por 37 personas. Los resultados mostraron que el grupo atopico presenta un porcentaje de linfocitos de 9,63 mas o menos 2,66 que portan tal receptor frente el 1.77 mas o menos 0,37 presentado por el grupo control. Como hecho interesante se encontro una disminucion estadisticamente significativa para receptores IgM en atopicos, en cambio no hubo diferencia para IgA. La cuantificacion serica de IgE e IgA mostro un aumento para IgE y una disminucion para IgA
