ABSTRACT

The urethral muscle of diabetic pregnant rats is affected by long-term mild diabetes and short-term severe diabetes, which plays a crucial role in the pathogenesis of pelvic floor disorders. We hypothesized that muscles outside the pelvis are subject to similar changes. The current study aimed at analyzing the effects of long-term mild and short-term severe diabetes on the structure and ultrastructure of fiber muscles and collagen in rats' rectus abdominis (RA) muscle. Therefore, the RA muscle of virgin, pregnant, long-term mild diabetic, short-term severe diabetic, long-term mild diabetic pregnant and short-term severe diabetic pregnant 3-month-old Wistar rats were collected. The structure was analyzed by picrosirius red staining, immunohistochemistry for fast and slow muscle fibers and transmission electron microscopy. We investigated two levels of STZ- induced diabetes: long-term mild diabetes (blood glucose level: 120-200 mg/dL) and short-term severe diabetes (blood glucose level >300 mg/dL). Long-term mild diabetic pregnant and short-term severe diabetic pregnant rats had decreased fast fibers and increased slow fibers, disrupted areas of sarcomere, intermyofibrillar mitochondria and myelin figures in the RA muscle. Both groups enabled us to analyze the specific influence of pregnancy, separately from diabetes. The current study demonstrated that diabetes and pregnancy induced intramuscular transformation and reorganization of RA muscle with a switch of fiber type adjusting their architecture according to intensity and duration of hyperglycemic insult within pregnancy.

Se realizó un estudio de corte transversal, donde se realizó el seguimiento de pacientes embarazadas con diagnóstico de diabetes, que se ingresaron a las salas del Hospital Berta Calderón Roque durante el Perído de 01 de enero del 2004- 31 de diciembre del 2007. El universo estuvo constituido por todas las pacientes embarazadas con diagnóstico de diabetes a las cuales se les brindó atención en las salas de esta unidad hospitalaria con un total de 163 pacientes. La muestra fue selecionada de forma probabilistica en las pacientes que cumplieron como requisito tener completo todos los datos en el expediente clínico con un total de 147 pacientes...

ABSTRACT

A hipoplasia de esmalte é a mais comum dentre as alterações de desenvolvimento do dente humano, e ocorre com freqüência em dentes decíduos de filhos de mães diabéticas. O presente estudo experimental analisou, por meio de microscopia óptica e morfometria, o órgão do esmalte de incisivos inferiores de filhotes de ratas com diabetes aloxânico, induzido previamente à gestação. Os resultados mostraram que não foram observadas pela microscopia óptica alterações significantes nos germes dentais dos animais descendentes de ratas diabéticas, com exceção de um caso. A análise morfométrica dos órgãos do esmalte de ratos nascidos de mães diabéticas tratadas e não tratadas evidenciou as seguintes diferenças estatisticamente significantes: menor espessura da matriz de esmalte, menor altura dos ameloblastos e área de seus núcleos. Nos animais nascidos de ratas diabéticas tratadas, observou-se núcleos dos ameloblastos mais elípticos e aumento da área correspondente ao interstício do retículo estrelado. Estes resultados indicam que há alterações estruturais no órgão do esmalte de descendentes de ratas com diabetes aloxânico as quais poderiam induzir a hipoplasia do esmalte dental visto por microscopia eletrônica de varredura, possivelmente refletindo as alterações metabólicas observadas nesta condição. Estudos futuros devem ser realizados a fim de determinar se estas alterações são transitórias ou permanentes.

ABSTRACT

Diabetic nephropathy is associated with increased urinary albumin and reduce kallikrein excretion. Increased activity of the renal kallikrein-kinin system has been suggested as one of the possible mechanisms underlying diabetic hyperfiltration. The present study shown that kallikrein-kinin system is progressively increased in the diabetic-pregnant rats at 7, 14, 21 days; 48 and 7 days after pregnancy (P<0.05 vs Control). However, this increase during diabetic pregnancy did not reached the levels of control pregnancy. On the other hand albumin excretion shown a significant and progressive renal damage in the diabetic state. These findings suggest that the diabetic pregnancy could impair the renal hemodynamic, but, on the other side could modulate the vasodilatador system at pregnancy in the attempt protect the fetus.

ABSTRACT

La presencia de diabetes en el embarazo afecta a un número significativo de mujeres y es considerada como un factor con importante papel en la morbilidad y mortalidad perinatal. Con el objetivo de determinar los efectos de esta enfermedad sobre el recién nacido, se realizó un estudio retrospectivo en 35 casos de mujeres con diabetes mellitus o diabetes gestacional que habían tenido un parto previamente. Se escogieron además al azar, 26 controles de partos de mujeres normales para establecer las comparaciones respectivas. Se observó un efecto de la enfermedad en variables como el Apgar, la talla y los perímetros cefálico y torácico de los recién nacidos. No se encontraron efectos sobre el peso como tampoco diferencias sobre la presencia de malformaciones. Por la importancia de esta situación tanto para la madre como para el recién nacido, se recomiendan medidas de control estricto durante el embarazo de estas mujeres, así como en la atención del recién nacido buscando minimizar los riesgos
