ABSTRACT

This study was investigated the effects of dietary supplementation with prebiotics beta-glucan, MOS 500 and Bio MOS [registered sign] on the broilers performance, biochemical parameters and immune response for 5 weeks. One hundred and twenty 1-day-old broiler chickens were randomly assigned to 4 dietary treatment groups as follows: control, beta-glucan [1g/kg], MOS 500 [0.5g/kg] and Bio MOS [registered sign] [1g/kg]. Each treatment was divided into three replicates [ten birds/ replicate]. The overall BW gains of birds treated with prebiotics were significantly [P <0.01] higher than those of the control group. During the experiment, the birds fed supplemental Bio MOS [registered sign] gave the best FCR followed by beta-glucan MOS 500 and then the control group. Addition of these prebiotics were improved the levels of serum total protein and globulin which in turn support the immune function against NDV and IBDV. In addition to, supplementation of broiler chicks by beta-glucan, MOS 500 and Bio-MOS [registered sign] had no effect on the liver transmination enzymes and some kidney function which represent the non pathological metabolism of the liver and kidneys indicating their safety supplementation in broiler feed

ABSTRACT

The effect of carnosine administration for 15 and 30 days on 13 different serum proteins, total protein, albumin, prealbumin, alpha-and beta-lipoproteins, alpha-1-macroglobulin, alpha-1 acid glycoprotein, alpha-1 antitrypsin, cholinesterase, ceruloplasmin, hemopexin, haptoglobin and transferrin and 3 liver parameters [DNA, RNA and weight] were investigated in healthy [HC] and partially hepatectomized [PHC] mice. HC15 days animals presented 4 increased parameters, while the PHCl5 days mice presented 11 promoted parameters. In 30 days groups, HC30 days mice demonstrated increased 13 parameters, while the PHC30 days showed promotion of all parameters. These results pointed to the protein promoting effect of carnosine in both normal and partially hepatectomized mice. However, the more actively regenerating liver cells in the operated mice were more labile to stimulation by carnosine administration

ABSTRACT

The clinical diagnosis of ascites is an easy task but the etiological diagnosis of the type of ascites is occasionally a different problem. Many studies have investigated the possibility of using a single marker to detect cancer in ascitic patients; but no analysis of ascitic fluid including cytology has been shown to be specific and sufficiently sensitive for either hepatic or extrahepatic cancer. To evaluate the role of various biochemical parameters including cholesterol, LDH, total protein, albumin and glucose for the differentiation of cirrhotic ascites from malignancy related ascites, fifty ascitic patients were classified into three main groups: Group I: included 20 patients with cirrhotic ascites. Group II: included 15 ascitic patients with hepatocellular carcinoma on top of liver cirrhosis. Group III: included 15 ascitic patients with extrahepatic malignancy without liver cirrhosis. All patients were subjected to: clinical history and examination, various imaging, routine laboratory investigations, histopathological examination, cytological examination of ascitic fluid, and biochemical analysis of both serum and ascitic fluid. We found that, although cytological examination of ascitic fluid is specific, it is less sensitive than ascitic fluid biochemical analysis in determining the cause of ascites. The accuracy of ascitic fluid cholesterol LDH, SAAG, A/S LDH ratio, A/S cholesterol ratio, A/S protein ratio, AFTp and ascitic/blood glucose ratio in discrimination of exudative malignant ascites from transudative cirrhotic ascites are [91%, 91%, 91%, 86%, 83%, 83%, 80% and 71%] respectively. Ascitic fluid biochemical analysis may be useful than cytological examination in follow-up of cirrhotic ascitic patients aiming to early detection of complication

ABSTRACT

The effects of oral administration of phenobarbital [PB] and or L- tryptophan [TR] and their withdrawal on liver function, lipid metabolism and protein content of adult male albino rats were studied. Thirty-six rats were divided into six equal groups [control, PB group, TR group, PB and TR group, PB withdrawal group and PB and TR withdrawal group]. Rats received orally 50 mg/kg body weight PB and/or TR daily for four weeks. The results suggested that PB has a drastic effect on the estimated parameters that extends after its withdrawal. TR administration may, to some extent, decline, but not abolish this effect

ABSTRACT

Gallbladder bile from thirty patients undergoing elective cholecystectomy for calcular and noncalcular chronic cholecystitis were included in this study. Normal bile was also aspirated from gallbladders of 10 subjects undergoing surgery for surgical conditions not involving liver or biliary tract [control group]. The present work aimed at studying some proteins and some oxidative state parameters in gallbladder bile of normal subjects and patients with calcular and noncalcular cholecystitis and their possible contribution in gallbladder lithogenicity. Collected bile was examined for total cholesterol, bilirubin, proteins, mucoproteins and procollagen Ill peptide. Protein and glycoprotein fractionation was done by polyacrilamide gel electrophoresis. In addition some oxidative state parameters were done biliary ferritin, superoxide dismutase [SOD] glutathione and thiobarbituric acid reactive substances [TBARS]. It could be noticed that: there was an increase in total biliary proteins and mucoproteins in gall stone formers than in controls. Pronucleating glycoproteins of 130 KD and 42 KD were found in higher frequency in the calcular than noncalcular and control groups. Antinucleating 58 KD glycoprotein and 120 KD protein were found in higher frequency in controls than in calcular groups. As regards the oxidative state prameters studied, mean values for SOD, glutathione and ferritin were significantly lower in calcular and noncalcular groups when compared to control group and TBARS were significantly higher. It could be concluded that the state of increased oxidative stress in bile of calcular group and also the biliary protein pattern may contribute to the Iithogenicity of bile

ABSTRACT

Fifty neonates were included in this study, divided into two groups. Group 1[control group] consisted of 22 healthy neonates. Group 1A included 12 full term, 3 females and 9 males. Their gestational ages ranged between 37-39 weeks [mean 37.58 +/- 0.76 weeks]. Group 1B included 10 preterm neonates, 4 females and 6 males. Their gestational ages ranged between 28-34 weeks [mean 30.8 +/- 2.44 weeks]. Group II included 28 sick neonates admitted to the neonatal care unit of the pediatric department of El-Menoufiya University hospital and El-Menshawy general hospital. 10 had asphyxia, 5 had pneumonia, 7 had hyaline membrane disease and 6 had acute hemolysis. All the neonates were under antibiotic therapy. Group IIA consisted of 14 full term neonates, 2 females and 12 males. Their gestational ages ranged between 37-40 weeks [mean 37.64 +/- 0.81 weeks]. Group IIB consisted of 14 preterm neonates, 3 females and 11 males. Their gestational ages ranged between 27-35 weeks [mean 30.5 +/- 2.44 weeks]. All the neonates were subjected to thorough clinical history and examination, as well as measurement of serum sodium and potassium, blood urea, serum creatinine, urinary creatinine, total protein excretion, and measurement of urinary excretion of microalbumin and retinol binding protein [RBP] in untimed urine samples by ELIZA technique. The blood urea, microalbuminuria and total proteins were significantly elevated in sick neonates [Group II] compared to healthy neonates [Group I], however the difference was not influenced by the gestational age. No significant difference could be detected in serum sodium, potassium, serum creatinine and urinary creatinine between both groups. Measurement of RBP revealed that the healthy preterm neonates [Group IB] had a significantly higher level of RBP compared to the healthy full term [Group IA, P <0.05]. Also, the sick neonates [Group II] had a significantly higher level compared to the healthy ones [Group I, P<0.01]. RBP showed a significant negative correlation with the gestational age and the Apgar score at 1 and 5 minutes, and a significant positive correlation with blood urea and microalbuminuria. Evaluating the renal function by the excretion of RBP and microalbuminuria, showed that RBP was positive in 84% of the cases while microalbuminuria was detected in 30% of the cases only. It can be concluded that Retinol binding protein is an early and specific non-invasive marker of tubular functions. It has the advantage of being able to unmask even subtle cases of kidney injury when other parameters of kidney functions are still within the normal range

ABSTRACT

Administration of Dinothrombium tinctorium 100 mg/kg/day oral for 8 weeks caused no significant increase in weight, prostate glands, seminal vesicle, epididymis, testosterone level in serum as well as in testis tissue. The sperm concentration and sperm motility have also no significant increase, serum transaminase/phosphate, serum protein/cholestrol/lipid/bilirubin, blood urea and blood sugar remain unaltered. The histological observations of the testis indicated no significant changes in the diameter of the seminiferous tubules, Sertoli cells, Leydig's cells, spermatocyte, spermatid and spermatozoa

ABSTRACT

We report a 16 year old phenotypic female who presented with delayed puberty. The patient had a eunuchoid habitus, no breast development and little pubic hair. External genitalia were normal. Serum gonadotropin levels were elevated but the plasma testosterone level was normal for a prepubertal woman. Karyotype of the peripheral blood showed a 46XY pattern- At laparotomy the patient was found to have an infantile uterus, normal fallopian tubes and bilateral streak gonads. Histological examination revealed bilateral small streak gonads with fibrofatty tissue

ABSTRACT

B2-microglobulin [B2-m], as a low molecular weight protein, was evaluated in serum and synovial fluid of 10 patients with rheumatoid disease [RD], 10 patients with osteoarthritis [OA], and 10 age and sex matched controls. These investigations were done after excluding all patients with renal diseases. It was found that the serum level of B2-m was significantly elevated, in both RD and OA with more increase in the former, when compared with the control group. The synovial fluid/serum ratio of B2-m was a better diagnostic tool that could be used for differentiation between RD and OA, rather than using the serum level of B2-m in each disease alone

ABSTRACT

In the present study we have documented the use of the reagent p-benzoquinone (PBQ) for the spectrophotometric determination of total protein in blood plasma. Since the products of reaction are stable for several hours at room temperature after the 20-min boiling step, the time at which absorbance is measured is not a critical factor. Common anticoagulants such as EDTYA, citrate, or heparin do not interfere with the PBQ method at concentrations used in clinical laboratories. The products of the reaction between PBQ and either plasma (specific absorbance 2.33 x 10-3 ñ 0.20 x 10-3 ug cm -2) or purified proteins (specific absorbance 2.61 x 10-3 ñ 0.31 x 10-3 ug cm-2) show an absorption band at 350 nm, which follows Beer's law, and therefore can be used for analytical purposes. The PBQ method has a lower limit of detection (4 ug/ml) than that of biuret method (45 yg/ml) for a final reaction mixture of 5.0 and 4.2 ml, respectively

ABSTRACT

O proposito da implantacao do laboratorio de fracionamento de plasma humano para isolar varios componentes proteicos tais como albumina, imunoglobulina G endovenosa (IgG), Fator VIII e IX para uso interno do Hospital das Clinicas da Faculdade de Medicina da Universidade de Sao Paulo, foi solucionar parcialmente o grande problema da escassez destes produtos, alem de seus precos elevados. Iniciou-se com o isolamento da albumina, quanto a imunoglobulina G a mesma se encontra em adiantada fase experimental. A opcao pelo metodo de cromatografia em coluna foi devido a limitada area fisica disponivel e por ser este processo considerado uma tecnologia de ponta, e de provavel preferencia na aplicacao industrial no futuro. Os geis usados na cromatografia em coluna para essa finalidade foram: de troca ionica DEAE-Sepharose FF, CM-Sepharose FF, e de filtracao tipo Sephadex G-25 e Sephacryl S-200 HR. Para alcancar esse objetivo foi necessario introduzir certas modificacoes e adaptacoes inerentes a cada etapa de fracionamento, obtendo-se assim uma albumina de alta qualidade, comparavel as comercialmente conhecidas

Extensöes de sangue periférico de aves (macho e fêmea), foram submetidas a um estudo topoquímico que permitiu as seguintes conclusöes: com o PAS, a presença de glicogênio nos trombócitos e alguns heterófilos; com o azul de bromofenol (proteína) positividade em todos os elementos celulares, com intensa reatividade da regiäo equatorial dos grânulos fusiformes de heterófilos; com o método da orto-toluidina-água oxigenada (mieloperoxidase), reatividade discutível nos polimorfonucleares e negativa nos demais tipos de células e com Sudan black (lipídios) uma positividade em alguns heterófilos, eosinófilos e monócitos

ABSTRACT

En el presente trabajo describimos dos pacientes portadores de mieloma múltiple, de clase IgD (tipo Kappa y lambda), encontrados sobre 5 casos de mieloma múltiple, que hemos tenido oportunidad de estudiar en la provincia de Formosa (Argentina), entre los años 1985 y 1988. En todos los casos, además de los estudios hematológicos correspondientes, se utilizan métodos simples y relativamente poco costosos (Proteinograma, inmunoelectroforesis, electroinmunofijación), para arribar al diagnóstico. Los dos pacientes con MM IgD concurren a la consulta hematológica para investigación de anemia refractaria. Ninguno de ellos presentó hipercalcemia importante y sólo uno evidenció alteraciones óseas, al estudio radiológico. El tiempo de sobrevida desde el momento del diagnóstico fue de cuatro meses en ambos casos. Al igual que otros autores, concordamos en que la frecuencia del MM IgD, encontrada en la literatura, puede no ser la verdadera y que la misma se verá modificada por el mejoramiento de la metodología empleada.

ABSTRACT

Se realizó un estudio de algunos indicadores nutricionales en pacientes gerontogeriátricos diabéticos y de otro grupo de individuos supuestamente normales, con el objetivo de evaluar la necesidad de sugerir modificaciones dietéticas. Se seleccionaron 26 pacientes diabéticos de ambos sexos, hospitalizados en salas de Medicina, y 30 pacientes, posiblemente sanos, como grupo control. El estudio nutricional llevado a cabo en estos pacientes gerontogeriátricos diabéticos es de gran valor, por lo que se considera que debe ampliarse a otros indicadores bioquímicos

Se sintetizaron anfolitos carrier (AC) mediante la reacción entre tetraetilenpoentamina (TEPA) y ácido acrílico (AA). El producto obtenido se utilizó en la separación de proteínas séricas mediante isoelectroenfoque (IEF) en geles de agarosa y de poliacrilamida (PAA). Se determinó el gradiente de pH obtenido y se analizó el comportamiento isotacoforético de las distintas fracciones del anfolito carrier, obtenidas luego de su IEF sobre gel de agarosa

ABSTRACT

Se describe un procedimiento para la síntesis de anfolitos aplicables a la formación de gradientes de pH para isoelectroenfoque. Se utiliza tetraetilenpentamina a la cual se le agrega N, N'-metilenbisacrilamida como reactivo de condensación, con las finalidades de incrementar la diversidad de componentes anfotéricos, y/o de aumentar el número de grupos amino en la molécula resultante del acoplamiento de tetraetilenpentamina de a pares. En una operación posterior se hacen reaccionar estos productos con un ácido alfa, ß-insaturado. El procedimiento utilizando ácido acrílico se describe en detalle. Se estudia la uniformidad del gradiente de pH cortando el gel de poliacrilamida en fracciones de 1 x 1 cm y efectuando la medición del pH en cada una de ellas por el método de elución. La capacidad de formar gradientes estables de pH del producto sintetizado se demuestra realizando isoelectroenfoques de lisados eritrocitarios en geles de poliacrilamida de 1 mm de espesor

ABSTRACT

Se detallan todos los pasos a llevar a cabo para realizar isoelectroenfoque en geles de poliacrilamida de 1 mm de espesor adheridos a un celofán como soporte. El gel se adhiere firmemente al celofán durante todos las operaciones (corrida, tinción y desteñido) y el soporte lo protege de las roturas, razón por la cual es fácilmente manejable. Se discute la optimización de las condiciones de trabajo para anfolito no comercial. Se muestra la buena resolución obtenida con dicho anfolito de sueros humanos normales y patológicos

ABSTRACT

The sera of 25 patients suffering from advanced stages [T3 and T4] of carcinoma of the bilharzial urinary bladder were examined for serum proteins and immunoglobulin levels. In addition, sera of 16 bilharzial cystitis patients and of 16 normal healthy individuals were included in the study. While there was a reduction in the albumin level and an increase in alpha-1, alpha-2, and gamma globulin levels in the sera of the 25 cancer patients, the beta globulins remained apparently unchanged and the alpha-2/beta ratio had therefore inverted values [1.5] as compared to normal controls [0.9]. A definite increase in IgG, IgA, IgM and IgE fractions in bilharzial bladder cancer patients was found. The immunoglobulin levels were higher in patients with T3 than with T4 lesions. On the other hand, there was a marked decrease in the non-immunoglobulin component[s] in sera of the cancer patients. This decrease was noted more in patients with T4 than in those with T3 lesions. Serum proteins and immunoglobulins in the bilharzial cystitis patients had intermediate values more or less midway between the bladder cancer patients and the normal controls
