ABSTRACT

O tratamento endodôntico frequentemente leva à perda de parte coronária de um elemento dentário, seja pelo enfraquecimento de sua estrutura, por lesões traumáticas ou por cárie. Desde que em condições favoráveis, esses dentes podem ser restaurados por meio de retenção adequada provida por meio de retentores intrarradiculares. Os pinos de quartzo utilizados como retentor intrarradicular surgiram com os avanços da odontologia adesiva, sendo uma opção relevante com inúmeras vantagens. A literatura odontológica aponta que a adaptação do pino às paredes do conduto radicular, somada a técnica de cimentação adesiva, são fatores essenciais para a longevidade do tratamento, sendo necessário em alguns casos sua personalização, para garantir melhor assentamento e promover prognóstico favorável. Objetivou-se neste artigo apresentar a técnica de preparo, personalização e cimentação do pino de quartzo na reconstrução de um dente tratado endodonticamente ilustrado por meio de caso clínico. Através de um protocolo ordenado e meticuloso da técnica de aplicação, foi possível alcançar o resultado desejado(AU)


Endodontic treatment often leads to loss of the coronary part of a tooth, either by weakening its structure, by traumatic lesions or by caries. Since under faborable conditions, these teetn can be restored by means of the biomechanical reinforcement promoted by the intraradicular retainers. The quartz posts have appeared with the advances of restorative dentistry, being a relevant option with numerous advantages. The dental literature indicates that the adaptation of the post to the root canal walls, together with the adhesive cementation technique, are essential factors for the longevity of the treatmet, being necessary in some cases its personalization, which guarantees its better settlement, avoiding the impression of undesirable forces to the tooth anf promoting better prognosis. The objective was to present the technique of preparation, personalization and cementation of the quartz post in the reconstruction of an endodontically treated tooth. Through an orderly and meticulous protocol of the application technique, it was possible to achieve the desired result(AU)

ABSTRACT

Abstract Nowadays, demand for esthetic restorations has risen considerably; thus, nonmetal esthetic posts made of either high-strength ceramics or reinforced resins, such as fiber-reinforced resin posts, have become more and more popular. Important characteristics of fiber-reinforced posts involve a modulus of elasticity similar to dentin and their ability to be cemented by an adhesive technique. A total of 36 maxillary incisors were divided into four groups. In this study, four adhesively luted fiber-reinforced (glass fiber, quartz glass fiber, zirconia glass fiber and woven polyethylene fiber ribbon) post systems were used. Post spaces were prepared by employing drills according to the protocol established for each group, and each post was adhesively luted with one of three adhesive systems. Three segments per root apical to the cementoenamel junction (CEJ) were obtained by sectioning the root under distilled water with a carbon spare saw. The samples (total of 108 sections) were 2.0±0.1 mm in thickness and they were stored individually in black film canisters with sterile distilled water. In order to determine the bond strength, the bonding area of each specimen was measured, and specimens were attached to a device to test microtensile strength at a speed of 1 mm/min. The analyses revealed no statistically significant differences between the adhesive systems and fiber-reinforced posts. (P> 0.05). However, the coronal portion of the root dentin had the highest bond strength. Adhesive systems used along with fiber-reinforced resin posts demonstrated reliable bonding.

ABSTRACT

Objetivo: Mostrar una alternativa de tratamiento para las fracturas coronorradiculares complicadas, que restablece la función y la estética de las piezas traumatizadas. Caso clínico: Una paciente de 16 años se presentó en la guardia del Centro Odontológico OSEP tras haber sufrido un accidente vial que le produjo un traumatismo dentoalveolar. El diagnóstico fue fractura coronaria sin exposición pulpar de la pieza dentaria 12 y fractura coronorradicular complicada de la pieza 11. El tratamiento incluyó restauración con resina compuesta de la pieza 12 y endodoncia, extrusión ortodóntica, alargamiento coronario y rehabilitación con perno de fibra de cuarzo y corona de resina compuesta de la pieza 11. Conclusiones: El tratamiento de las fracturas coronorradiculares representa un desafío para el odontólogo. Los resultados del tratamiento, en este paciente, fueron excelentes, ya que lograron devolver estética y función (AU)


Aim: To show an alternative treatment for complicated crown-root fractures, which restores function and aesthetics of the traumatized teeth. Case report: This paper reports the case of a 16-yearold girl who attends the OSEP Dental Center, stating that she had suffered a traffic accident resulting in a dentoalveolar trauma. The diagnosis was a coronary fracture without pulp exposure of tooth 12, and complicated crown-root fracture of tooth 11. Treatment included composite resin restoration of tooth 12 and endodontic, orthodontic extrusion, crown lengthening and rehabilitation with quartz fiber post and composite resin crown of tooth 11. Conclusions: The treatment of crown-root fractures poses a challenge for the dentist. The results of the treatment in this patient were excellent, and aesthetics and function were restored (AU)

ABSTRACT

BACKGROUND: Occupational exposure to crystalline silica is a potential risk factor for various systemic autoimmune diseases including systemic sclerosis. The etiology of systemic sclerosis is not conclusively known, but there are epidemiological studies that show the relationship between exposure to crystalline silica and risk of systemic sclerosis. Here we report, for the first time, two cases of crystalline silica-related systemic sclerosis in patients who worked in crystal processing in the jewelry-manufacturing field. CASE PRESENTATION: Case 1 is a 57-year-old man who had worked mainly in crystal processing for multiple jewelry-processing companies for 17 years, since the age of 15 years. He contracted tuberculosis at the age of 25 years and showed Raynaud's phenomenon of both the hands and feet at age 32 years. Digital cyanosis and sclerosis developed at approximately age 41 years. The patient was diagnosed with systemic sclerosis at age 48 years. Case 2 is a 52-year-old man who worked in crystal processing for various jewelry-processing companies for 7 years, since the age of 23 years. He first showed signs of cyanosis in the third and fourth digits of both hands at age 32 years, was diagnosed with Raynaud's syndrome at age 37 years, and was diagnosed with systemic sclerosis at age 38 years. Crystal processing is a detailed process that involves slabbing and trimming the selected amethyst and quartz crystals, which requires close proximity of the worker's face with the target area. In the 1980s and 1990s, the working hours were 12 h per day, and the working environment involved 15 workers crowded into a small, 70-m2 space with poor ventilation. CONCLUSION: Two workers who processed crystals with a maximum crystalline silica content of 56.66% developed systemic sclerosis. Considering the epidemiological and experimental evidence, exposure to crystalline silica dust was an important risk factor for systemic sclerosis. An active intervention is necessary to reduce exposure in similar exposure groups in the field of jewelry processing.

ABSTRACT

Lateritic crusts developed upon the Guiana Shield basement rocks (e.g. granites of the Água Branca Intrusive Suite) was recognized in the Vila Nova Colina region, located in south Roraima state. The objectives of this work were to study the mineral and geochemical characteristics of lateritic crusts and to evaluate the relationship between them and the rocks of the region, in order to understand the evolution of laterization and provide information about the economic potential. This study comprises petrography, mineralogy and litogeochemical investigations of the granites, the quartz-hornblende gabbro and the lateritic crusts formed from these rocks. The granites and the quartz-hornblende gabbro has developed, respectively, a specific type of lateritic crust: (i) the ferruginous vermiform one with high amounts of kaolinite, goethite, hematite, magnetite and anatase, higher concentration of Al2O3, Sc, Th e Zr and, (ii) the iron-titaniferous massive one with high amounts of magnetite, hematite, maghemite, goethite, kaolinite, anatase, gibbsite and ilmenite, and consequently in higher amounts of Fe2O3, TiO2, V e Cu. Each crust type indicates the effect of the lateritization in the south portion of Roraima and their topographic position. The higher topographic position of the massive crust (220 m), its mineral and geochemical composition, indicate the maturity (i.e. needed more time to be formed) and may represent a relict of an erosion phase in the region, probably associated with a subsidence of the Tacutu basin in the Miocene. Otherwise, the vermiform crust formed at a lower topographic position (140 m) with no gibbsite, suggests that it is younger than the massive one and represents the immature laterization of the Plio-Pleistocene.


Crostas lateríticas desenvolvidas sobre as rochas do embasamento do Escudo das Guianas (p.e. granitos da Suíte Intrusiva Água Branca) são reconhecidas na região de Vila Nova Colina, localizada ao sul do estado de Roraima. Os objetivos deste trabalho foram estudar as características minerais e geoquímicas dessas crostas lateríticas e avaliar a relação entre elas e as rochas da região, a fim de entender a evolução da lateritização e fornecer informações sobre seu potencial econômico. Este estudo compreende dados de petrografia, mineralogia e litogeoquímica de granitos, quartzo-hornblenda gabro e crostas lateríticas formadas a partir dessas rochas. Os granitos e o quartzo-hornblenda gabro desenvolveram, respectivamente, um tipo específico de crosta laterítica: (i) vermiforme ferruginosa com elevados teores de caulinita, goethita, hematita, magnetita e anatásio e maior concentração de Al2O3, Sc, Th, e Zr e, (ii) maciça ferro-titanífera com altos teores de magnetita, hematita, maghemita, goethita, caulinita, anatásio, gibbsita e ilmenita e de Fe2O3, TiO2, V e Cu. Cada tipo de crosta indica o efeito da lateritização na porção sul de Roraima e sua posição topográfica. O posicionamento topográfico mais elevado da crosta maciça (220 m) e sua composição mineralógica e geoquímica, indicam sua maturidade (isto é, precisou de mais tempo para ser formada) e, portanto relicto de uma fase erosiva na região, provavelmente associada a subsidência da bacia do Tacutu no Mioceno. Enquanto a crosta vermiforme em uma posição mais rebaixada (140 m) sem gibbsita, sugere que ela é mais jovem que a maciça e representa a lateritização imatura do Plio-Pleistoceno.

ABSTRACT

Statement of Problem: One of the factors affecting the degree of polymerization of light-cured composites is the type of light-curing unit used. In addition, physicomechanical properties of the composite resins depend on the degree of conversion and polymerization


Objectives: Since the type of initiator in new composite resins is not explained by manufacturers, this study is an attempt to compare the depth of hardening, with two LED and QTH light-curing units


Materials and Methods: Fifteen samples prepared from Gradia Direct and Filtek Z250, both of which being universal, were cured with QTH [Astralis 7] and LED [Bluephase C8] light-curing units. All the samples were molded in polyester resin and cut from the middle by a disk. The hardness of the cut area was evaluated at 0, 0.5, 1, 1.5, 2, 2.5, 3, 3.5 and 4-mm depth intervals and also at the same interval as the width of the sample, with Vickers hardness machine, while the samples were placed in a darkroom. Data were statistically analyzed using one-way ANOVA, two-way ANOVA, t-test and post Hoc Tukey's tests in SPSS, version 16


Results: Filtek Z250 was harder than Gradia Direct at all the depth with both light-curing units. The hardness of Filtek Z250 sample cured with Astralis 7 was higher than that cured with LED, but with Gradia Direct the LED unit resulted in higher hardness. Curing depth was not significantly different between the groups [p = 0.109]


Conclusions: Vickers hardness number for both composites used in this study is in an acceptable range for clinical implications. The composites' composition is important to be considered for selection of light unit. Based on the findings of the present study, LED did not present more curing depth compared with QTH

ABSTRACT

<p><b>OBJECTIVE</b>To compare the effects of oral treatment with tetrandrine (TD) and N-acetylcys-teine (NAC) separately or jointly on silica-exposed rats.</p><p><b>METHODS</b>40 sprague-Dawly (SD) rats were randomly divided into normal saline group, quartz group, TD treatment group (50 mg/kg), NAC treatment group (500 mg/kg) and combined treatment group (TD: 50 mg/kg + NAC: 500 mg/kg). Rats in normal saline group and other groups received intratracheal instillation of normal saline and quartz dust suspension respectively. Treatment groups were given TD, NAC separately or jointly via esophagus the next day after instillation, once a day and six times a week for 30 consecutive days. At the end of experiment, the pathological changes of lung tissues were evaluated by the methods of Foot, HE and Masson staining, the level of hydroxyproline (HYP), malondjalde-hyde (MDA), tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) in lung tissues were measured by alkaline hydrolysis method, the barbituric acid method and enzyme-linked immunosorbent assay (ELISA) respectively.</p><p><b>RESULTS</b>Compared with the quartz group, lymph nodes/body coefficients in all treatment groups and lung/body coefficient in combined treatment group were significantly decreased (P < 0.05). Pathology results showed that the normal saline group demonstrated no obvious evidence of lung damage. The quartz group lungs silicotic lesions focused on II~III level, the TD treatment group was mainly with I level, the NAC treatment group was mainly with I~II level, and the combined treatment group only showed little silicotic nodule, no obvious fibrosis. HYP content in TD treatment group and combined treatment group were significantly lower than that in the quartz group (P < 0.05), while it showed no obvious change in NAC treatment group. MDA content in lung tissues of each treatment group (TD treatment group, NAC treatment group and combined treatment group) were 18.80 ± 2.94, 20.13 ± 4.01 and 17.05 ± 3.52 nmol/ml respectively, which lower than in the quartz group (23.99 ± 3.26 nmol/ml). The level of IL-6 in lung tissues of the quartz group were 89.57 ± 8.78 pg/ml. After TD and NAC monotherapy, the IL-6 content decreased to 79.22 ± 9.65 pg/ml and 81.63 ± 5.72 pg/ml, and it decreased more significantly after combined medication (74.37 ± 3.17 pg/ml). The level of TNF-α in the quartz group were 59.05 ± 4.48 pg/ml. After TD and NAC monotherapy, the TNF-α content decreased to 50.48 ± 2.76 pg/ml and 54.28 ± 4.30 pg/ml, and it decreased more significantly after combined medication (49.10 ± 4.98 pg/ml).</p><p><b>CONCLUSION</b>NAC and TD could reduce MDA, TNF-α and IL-6 levels in lung tissue, and alleviate SiO2-induced pulmonary fibrosis in rats. Combined treatment with TD and NAC was more effective than TD or NAC treatment separately.</p>

ABSTRACT

The development and validation of a method for the determination of concentrations of thiocyanate in human plasma are described here. A modified colorimetric method of Bowler was used with the following alteration in Monica Manual, Part III. In order to obtain the same sensitivity in low amounts of clinical samples, quartz SUPRASIL(R) micro cuvettes have been used. The quantitation range was between 25-500 microM. Accuracy and precision of the quality control samples, linearity of the calibration curve, dilution, spike recovery and stability under various conditions were evaluated in the validation of the method and all demonstrated acceptable results. All validation results met good laboratory practice acceptance and FDA requirements to be acceptable for application in clinical trials. The validated method has been used for a Phase I clinical study in cancer patients orally administered with either 60 mg or 80 mg of GDC-0425 containing a cyanide (CN-) group. The thiocyanate levels from patients before and after drug administration showed no clinically significant differences.

ABSTRACT

In Latin America, people have largely abandoned the practice of wearing hats and traditional clothing that provided skin protection. Sunscreen application has therefore become essential to protect against the increased sun exposure. The physician-prescribed medical-grade sunscreens provide sufficient sun protection but the requirement for regular use puts a financial burden on the patient that is often not sustainable. An appropriate sunscreen should provide a high and broad ultraviolet (UV) protection against UVB and UVA. Several over-the-counter (OTC) sunscreens have been developed for sale at affordable prices and are available for purchase in convenient locations, such as local grocery stores. The aim of this study was to assess the in vitro UV protection of 34 popular OTC sunscreens found in the Latin American market. UV absorbance/transmittance was quantified by diffusion transmission spectroscopy using coarse silica plaques. Photostability was tested by irradiating them with simulated solar light and calculating the sun protection factor (SPF), critical length of absorption (C λ ), UVA/UVB ratio, and the spectral uniformity index (SUI). The results indicated that the in vitro SPFs were significantly lower than the value declared on the labels, particularly for those claiming high SPF values; however, the majority of these sunscreens offered high levels of UV protection. Considering the advantages of low cost and ample accessibility, we concluded that this sample of OTC sunscreens can be beneficial to the general public by providing some level of skin protection from solar radiation, and may be promoted to improve compliance with recommended photoprotection behavior.


En Latinoamérica, la población ha abandonado la costumbre de usar sombrero y ropa tradicional para protegerse del sol. En consecuencia, es básico el uso de protectores solares si se realizan actividades bajo sol. Los protectores solares que se usan en la práctica médica son adecuados, pero su uso frecuente condiciona una carga económica que muchos pacientes no pueden solventar debido a sus costos considerables. Un protector apropiado contiene una amplia y elevada protección ultravioleta (UV) A y B. En las tiendas de conveniencia, existen numerosos protectores solares a precios más accesibles. El objetivo del estudio fue determinar la protección UV in vitro de 34 protectores solares con amplia presencia comercial (de venta sin prescripción médica) en el mercado latinoamericano. La absorbancia/transmitancia de la radiación UV se cuantificó mediante espectroscopía de transmisión difusa. Placas de sílice esmerilado fueron recubiertas con el producto y expuestas a radiación solar simulada para conocer su fotoestabilidad. Se calcularon índices como el factor de protección solar (SPF), longitud crítica de absorción (C λ), relación UVA/UVB y el índice de uniformidad espectral (SUI). Se encontró que el SPF in vitro fue inferior al establecido en las etiquetas, especialmente en aquellos con valores altos. No obstante, la mayoría de los protectores incluidos ofrecen niveles de protección UV elevados. Considerando su amplia accesibilidad y menor costo, concluimos que esta muestra comercial de protectores solares podría utilizarse en el entorno clínico para favorecer su apego junto a las otras medidas de fotoprotección sugeridas.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the role of oxidative stress in the endoplasmic reticulum stress-induced apoptosis of alveolar macrophages triggered by quartz dust.</p><p><b>METHODS</b>Seventy-two healthy adult Wistar rats were randomly divided into control group, quartz dust group, quartz dust plus N-acetyl cysteine (NAC) group, and NAC group, with 18 rats in each group. One milliliter of sterile saline (for the control and NAC groups) or 1 ml of saline with 5%ultrafine quartz dust (for dust group and dust plus NAC group) was given to each rat by non-exposed endotracheal infusion. From the second day after dust infusion, rats in dust plus NAC group and NAC group received intragastric administration of NAC (100 mg/kg). In each week, the treatment with NAC lasted for 5 consecutive days, followed by 2 days' interval. For each group, 6 rats were randomly selected on the 14th, 28th, or 56th day after dust exposure; they were sacrificed by bloodletting from the femoral artery, and the lungs were collected. Bronchoalveolar lavage fluid was collected to separate macrophages. The protein expression of caspase-12 in alveolar macrophages, the apoptosis rate and reactive oxygen species (ROS) content of alveolar macrophages, and the protein carbonyl content of alveolar macrophages were determined by Western blot, flow cytometry, and colorimetry, respectively.</p><p><b>RESULTS</b>Increased protein expression of caspase-12, apoptosis rate, and content of ROS and protein carbonyl were discovered on the 14th day in the dust group, in comparison with the control group (P < 0.05), and the increase lasted till the 28th and 56th days. (P < 0.05). Compared with the dust group, the dust plus NAC group showed significant decreases in the content of ROS on the 14th, 28th, and 56th days (P < 0.05), significant decreases in the content of protein carbonyl on the 28th and 56th days (P < 0.05), and significant decreases in the protein expression of caspase-12 and apoptosis rate (P < 0.05).</p><p><b>CONCLUSION</b>Oxidative stress is potentially involved in the endoplasmic reticulum stress-induced apoptosis of alveolar macrophages triggered by quartz dust. Oxidative damage of protein in the endoplasmic reticulum may play an important role in the process.</p>

ABSTRACT

The contribution of particles to cardiovascular mortality and morbidity has been enlightened by epidemiologic and experimental studies. However, adverse biological effects of the particles with different sizes on cardiovascular cells have not been well recognized. In this study, sub-cultured human umbilical vein endothelial cells (HUVECs) were exposed to increasing concentrations of pure quartz particles (DQ) of three sizes (DQPM1, <1 μm; DQPM3-5, 3-5 μm; DQPM5, 5 μm) and carbon black particles of two sizes (CB0.1, <0.1 μm; CB1, <1 μm) for 24 h. Cytotoxicity was estimated by measuring the activity of lactate dehydrogenase (LDH) and cell viability. Nitric oxide (NO) generation and cytokines (TNF-α and IL-1β) releases were analyzed by using NO assay and enzyme-linked immunoabsorbent assay (ELISA), respectively. It was found that both particles induced adverse biological effects on HUVECs in a dose-dependent manner. The size of particle directly influenced the biological activity. For quartz, the smaller particles induced stronger cytotoxicity and higher levels of cytokine responses than those particles of big size. For carbon black particles, CB0.1 was more capable of inducing adverse responses on HUVECs than CB1 only at lower particle concentrations, in contrast to those at higher concentrations. Meanwhile, our data also revealed that quartz particles performed stronger cell damage and produced higher levels of TNF-α than carbon black particles, even if particles size was similar. In conclusion, particle size as well as particle composition should be both considered in assessing vascular endothelial cells injury and inflammation responses induced by particles.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the role of calcium activated-potassium channels (KCa) in the injury to rat alveolar macrophages induced by quartz.</p><p><b>METHODS</b>The experiments were conducted on a rat alveolar macrophage cell line (NR8383) in vitro, where crystal silica (100 üg/ml) and amorphous silica (100 üg/ml) were used as the test substances and the cells without any treatment as negative controls. At first the effects of two kinds of quartz were compared. Then KCa special inhibitors (Paxilline for BK, Tram-34 for IK, Apamin for SK) were added in different doses to the in vitro test system with 100 üg/ml crystal quartz as matrix, to observe the function of such channels. Cell viability, lactate dehydrogenase (LDH), interleukin-1β (IL-1β) and tumor necrosis factor-a (TNF-α) were tested.</p><p><b>RESULTS</b>Comparing to the negative control group, cell viability reduced, LDH leakage, IL-1β and TNF-α release increased significantly in the amorphous quartz group, furthermore, the effects by crystal quartz were much more serious than those by amorphous quartz, with a statistical significance (P < 0.01). Comparing to the crystal quartz group, IK blockers (Tram-34) led to increase in cell viability significantly, with a statistical significance (P < 0.01); all the KCa specific blockers (Paxilline, Tram-34, Apamin) could reduce LDH leakage and IL-1β release, with a statistical significance (P < 0.05); meanwhile, BK and IK blockers (Paxilline,Tram-34) were able to reduce TNF-α release,with a statistical significance (P < 0.05).Reduction of IL-1β and TNF-α by Tram-34 was dose-dependent, but not so in the other two blockers.</p><p><b>CONCLUSION</b>Blocking calcium-activated potassium channels (KCa) could reduce cell membrane damage as well as IL-1β and TNF-α release induced by crystal quartz in the rat alveolar macrophages cell line in vitro, which might serve as a signal in the early regulation of inflammatory responses by quartz.</p>

ABSTRACT

Teeth that have short clinical crown, which are not alone enough to support the definitive restoration can be best treated using the post and core system. The advantages of fiber post over conventional metallic post materials have led to its wide acceptance. In addition to that the combination of aesthetic and mechanical benefits of fiber post has provided it with a rise in the field of dentistry. Also the results obtained from some clinical trials have encouraged the clinicians to use the fiber posts confidently. Fiber posts are manufactured from pre-stretched fibers impregnated within a resin matrix. The fibers could that be of carbon, glass/silica, and quartz, whereas Epoxy and bis-GMA are the most widely used resin bases. But recently studies are also found to be going on for polyimide as possible material for the fiber post resin base as a substitute for the conventional materials.

ABSTRACT

OBJECTIVES: Light-curing of resin-based materials (RBMs) increases the pulp chamber temperature, with detrimental effects on the vital pulp. This in vitro study compared the temperature rise under demineralized human tooth dentin during light-curing and the degrees of conversion (DCs) of three different RBMs using quartz tungsten halogen (QTH) and light-emitting diode (LED) units (LCUs). MATERIALS AND METHODS: Demineralized and non-demineralized dentin disks were prepared from 120 extracted human mandibular molars. The temperature rise under the dentin disks (n = 12) during the light-curing of three RBMs, i.e. an Ormocer-based composite resin (Ceram. X, Dentsply DeTrey), a low-shrinkage silorane-based composite (Filtek P90, 3M ESPE), and a giomer (Beautifil II, Shofu GmbH), was measured with a K-type thermocouple wire. The DCs of the materials were investigated using Fourier transform infrared spectroscopy. RESULTS: The temperature rise under the demineralized dentin disks was higher than that under the non-demineralized dentin disks during the polymerization of all restorative materials (p 0.05). CONCLUSIONS: Although there were no significant differences in the DCs, the temperature rise under demineralized dentin disks for the silorane-based composite was higher than that for dimethacrylate-based restorative materials, particularly with QTH LCU.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the roles of Rho/Rock signaling pathway in silica-induced Epithelial-mesenchymal transition (EMT) in human bronchial epithelial cells (BEC) in vitro.</p><p><b>METHODS</b>Human BEC were incubated with silica with various concentrations for indicated times. Cell viability was assayed by MTT test. Morphologic Changes were observed by microscope. Mesenchymal marker α-smooth muscle actin (α-SMA), vimentin (Vim), and epithelial marker E-cadherin (E-cad) were analyzed by Western Blot. The pull-down assay was used to measure Rho activity. In the prevention experiments, the specific inhibitor for Rho effector ROCK (Y27632) was used to inhibit the activity of Rho.</p><p><b>RESULTS</b>Human BEC stimulated with silica were converted from a "cobblestone" epithelial structure into an elongated fibroblast-like shape structure. Incubation of human BEC with silica induced de novo expression of α-SMA and Vim, and loss of E-cad. Also, silica treatment resulted in Rho activation in human BEC. Y27632 up-regulated the E-cad expression but attenuated α-SMA and Vim expression in silica-stimulated cells.</p><p><b>CONCLUSION</b>The activation of Rho/ROCK signaling pathways is most likely involved in Silica-induced EMT in human bronchial epithelial cells.</p>

ABSTRACT

This study aimed to evaluate the influence of quartz fiber post placement on the fracture resistance of endodontically treated premolars with different dental defects under dynamic loading. Fifty extracted single-rooted mandibular premolars were randomized into five groups. Each group was prepared according to numbers of residual walls ranged from 0 to 4. Then each group was divided into two subgroups with one restored with quartz fiber posts and the other without posts. In no-post groups, gutta percha point 2 mm below cemento-enamel junction was removed. Composite resin was adapted to the well and used to shape the core directly. Each tooth was restored with a complete metal crown. Dynamic loading was carried out in a masticatory simulator with a nominal load of 50 N at 2 Hz for 300 000 loading cycles. Then a quasi-statically load was applied in a universal testing machine 30° to the long axis with a crosshead speed of 1 mm⋅min(-1) until fracture. Data were analyzed with one-way analysis of variance and pairwise comparison (P<0.05). No specimens failed during dynamic loading. The fracture resistance enhanced with the increase of numbers of coronal walls and the differences were significant (P<0.05). Placement of fiber posts had a significant effect when fewer than two walls remained (P<0.05), but it had no significant influence in groups with two, three or four walls (P>0.05). Fiber post did not change failure mode, and the fracture pattern was mainly favorable. More dentin walls need to be retained in clinic. When no less than two walls remained, a fiber post is not always necessary.

ABSTRACT

A 46-year-old man who had worked as a bumper spray painter in an automobile body shop for 15 years developed lung cancer. The patient was a nonsmoker with no family history of lung cancer. To determine whether the cancer was related to his work environment, we assessed the level of exposure to carcinogens during spray painting, sanding, and heat treatment. The results showed that spray painting with yellow paint increased the concentration of hexavalent chromium in the air to as much as 118.33 microg/m3. Analysis of the paint bulk materials showed that hexavalent chromium was mostly found in the form of lead chromate. Interestingly, strontium chromate was also detected, and the concentration of strontium chromate increased in line with the brightness of the yellow color. Some paints contained about 1% crystalline silica in the form of quartz.

ABSTRACT

Here, we present a case of lung cancer in a 48-year-old male horse trainer. To the best of our knowledge, this is the first such case report to include an exposure assessment of respirable crystalline silica (RCS) as a quartz. The trainer had no family history of lung cancer. Although he had a 15 pack/year cigarette-smoking history, he had stopped smoking 12 years prior to his diagnosis. For the past 23 years, he had performed longeing, and trained 7-12 horses per day on longeing arena surfaces covered by recycled sands, the same surfaces used in race tracks. We investigated his workplace RCS exposure, and found it to be the likely cause of his lung cancer. The 8-hour time weight average range of RCS was 0.020 to 0.086 mg/m3 in the longeing arena. Horse trainers are exposed to RCS from the sand in longeing arenas, and the exposure level is high enough to have epidemiological ramifications for the occupational risk of lung cancer.

ABSTRACT

BACKGROUND: Iron and steel foundry workers are exposed to various toxic and carcinogenic substances including crystalline silica, polycyclic aromatic hydrocarbons, and arsenic. Studies have been conducted on lung cancer in iron and steel founding workers and the concentration of crystalline silica in foundries; however, the concentration of crystalline silica and cases of lung cancer in a single foundry has never been reported in Korea. Therefore, the authors report two cases of lung cancer and concentration of crystalline silica by the X-ray diffraction method. CASE PRESENTATION: A 55-year-old blasting and grinding worker who worked in a foundry for 33 years was diagnosed with lung cancer. Another 64-year-old forklift driver who worked in foundries for 39 years was also diagnosed with lung cancer. Shot blast operatives were exposed to the highest level of respirable quartz (0.412 mg/m3), and a forklift driver was exposed to 0.223 mg/m3. CONCLUSIONS: The lung cancer of the two workers is very likely due to occupationally related exposure given their occupational history, the level of exposure to crystalline silica, and epidemiologic evidence. Further studies on the concentration of crystalline silica in foundries and techniques to reduce the crystalline silica concentration are required.
