ABSTRACT

The marine environment is a rich source of biologically active compounds with pharmacological properties. Marine organisms often produce secondary metabolites with structural features different from those produced by terrestrial ones, and the Phylum Porifera seems to be one of the most productive in this sense. This study was undertaken to provide data on mutagenic and antimutagenic activities from an acetone (Areac) and an ethanol (Areet) extract obtained from Arenosclera brasiliensis, an endemic Brazilian sponge. A qualitative Salmonella reverse mutation test was performed with the TA97, TA98, TA100, and TA102 strains by incubating cells with Areac and Areet in the presence and absence of a known mutagen. A cytotoxic evaluation of the extracts was also performed. A. brasiliensis did not display any mutagenic activity, but Areac showed significant toxicity against test strains. In the antimutagenic assay, a reduction in the number of his+ revertants was observed for the TA97, TA100 and TA102 strains treated with Areac when compared to the positive controls. Areet treatment showed protective activity against DNA lesions only for the TA100. These results are in agreement with those obtained previously with other A. brasiliensis extracts, suggesting an antimutagenic activity.

ABSTRACT

Se comprobó el efecto inductor mutagénico de la luz uv, obteniendo mutantes auxotróficos y morfológicos de salmonella typhimurium. El aislamiento de mutantes auxotróficos mediante la técnica de réplica en placa mejora con el uso de penicilina mediante el método de Hollyday, para la identificación de los requerimientos específicos de cada mutante, se caracterizó 3 mutantes auxotróficos de 24 mutantes escogidos al azar. De 24 mutantes auxotróficos, el 30 por ciento resultó ser metronina dependientes, y 25 por ciento revirtieron la características de mutantes auxotróficos.

ABSTRACT

El objetivo de este estudio fue comparar tres métodos utilizados en el control de calidad de medios de cultivo: a)el método original de Miles-Mirsa; b)el ecométrico y c)el método de estrías. Para comparar estos métodos, se evaluaron la productividad (crecimiento de un microorganismo que habitualmente se desarrolla en un medio de cultivo) y la selectividad (supresión del crecimiento de un microorganismo, que se espera sea inhibido en un medio de cultivo) de seis medios sólidos de cultivo usados para el aislamiento de Salmonella spp. y Shigella spp. Los resultados que se obtuvieron al comparar los 3 métodos, mediante la prueba de Chi2, reflejan que existe diferencia significativa (p<0.05) entre el método de estrías y los otros 2 en lo referente a la productividad, no existiendo diferencia significativa entre los 3 métodos, en cuanto a selectividad. De los 3 métodos ensayados, el ecométrico resultó el de más fácil y rápida ejecución

ABSTRACT

O uso das radiaçöes ionizantes na destruiçäo de microrganismos responsáveis pela deterioraçäo de alimentos e causadores de toxinfecçöes alimentares constitui aplicaçäo para fins verdadeiramente pacíficos da energia nuclear. As toxinfecçöes alimentares em nosso meio säo causadas em grande parte por Salmonella enteritidis var. typhimurium. Devido à escassez de informaçöes sobre a resistência da Salmonella enteritidis var. typhimurium à radiaçäo gama, o presente trabalho teve objetivo determinar a dose letal para esta bactéria por irradiaçäo gama do Cobalto-60, em leite contaminado experimentalmente com amostras isoladas em nosso meio. Foram irradiadas 119 amostras de leite tipo B contendo cerca de 150.000 salmonelas por ml, com dose entre 100 a 1.100 Gray (Gy), sendo dois lotes de sobreviventes re-irradiados com doses até 2.500 Gy. As bactérias näo irradiadas previamente foram totalmente destruídas com doses de 1.100 Gy. Salmonella enteritidis var. typhimurium descendentes dos sobreviventes de 1.100 Gy sobreviveram à dose de 1.200 Gy. Estas, re-irradiadas, mínima para Salmonela enteritidis var. typhimurium para radiaçäo gama do Cobalto-60 é de 1.200 Gy. A re-irradiaçäo de sobreviventes a doses menores induz ao aparecimento de germes mais rádio-resistentes
