ABSTRACT

Snow trout (Schizothorax plagiostomus) is an economically important freshwater fish, mostly found in northern areas of water reservoirs of Pakistan. The current study was conducted in River Swat to analyze the bioaccumulation of heavy metals (Pb, Cr, Ni, and Zn) in tissues of Schizothorax plagiostomus. Tissues were extracted and dissolved in perchloric acid (HClO4) and nitric acid (HNO3) along with hotplate. The heavy metals, zinc (Zn), lead (Pb), chromium (Cr), and Nickel (Ni) were determined using Perkin Elmer 2380 atomic absorption spectrophotometer. Results shows great variation in the content of the metal related to tissue type and sampling sites. A high concentration of bioaccumulation was reported at Charbagh, whereas lowest at Odigram: Charbagh>Landakai>Odigram. In the same way, Cr was the most accumulated heavy metal followed by lead, nickel, and Zinc.


A truta das neves (Schizothorax plagiostomus) é um peixe de água doce economicamente importante, encontrado principalmente nas áreas ao norte de reservatórios de água do Paquistão. O presente estudo foi realizado em River Swat para analisar a bioacumulação de metais pesados (Pb, Cr, Ni e Zn) em tecidos de Schizothorax plagiostomus. Os tecidos foram extraídos e dissolvidos em ácido perclórico (HClO4) e ácido nítrico (HNO3) com placa de aquecimento. Os metais pesados zinco (Zn), chumbo (Pb), cromo (Cr) e níquel (Ni) foram determinados usando espectrofotômetro de absorção atômica Perkin Elmer 2380. Os resultados mostram grande variação no conteúdo do metal relacionado ao tipo de tecido e locais de amostragem. Uma alta concentração de bioacumulação foi relatada em Charbagh, enquanto a mais baixa, em Odigram: Charbagh > Landakai > Odigram. Da mesma forma, Cr foi o metal pesado mais acumulado, seguido por Pb, Ni e Zn.

El hombre ha cambiado el ambiente para sostener la demanda global de recursos naturales como el agua. La gestión de los cuerpos hídricos tiene que ser constante, con el propósito preventivo y correctivo, dependiendo del estado de antropización de cada sistema. El objetivo de este artículo fue analizar la toxicidad del sedimento y los metales Mn, Zn, Pb, Ni, Cd y Cu, en el reservorio Paiva Castro, que abastece la Region Metropolitana de São Paulo. Se realizaron 2 muestreos (Mayo 2011, estación seca y Enero de 2012, estación húmeda) y se analizaron 5 puntos próximos a la captación de agua por la Companhia de Saneamento Básico do Estado de São Paulo. Se realizaron ensayos de toxicidad aguda y crónica en sedimento, a través de ensayos biológicos con el cladócero Daphnia similis y el insecto Chironomus xanthus. El tratamiento de datos se realizó con el test de Fisher (mortalidad). El nivel de asociación entre las variables en sedimento y en los test ecotoxicológicos fueron evaluados por test no-paramétricos, a través del coeficiente de correlación de Spearman's. Los resultados del presente trabajo señalaron bajas concentracones de metales en el sedimento del área de estudio y ausencia de toxicidad en los organismos ensayados. Se puede concluir que área estudiada del reservorio Paiva Castro se encuentra poco impactada por los metales, sin efectos directos sobre la calidad de vida los organismos bentónicos: D. similis y C. xanthus.


Man had changed the natural environment in an attempt trying to supply the global demand for resources. The management of the hydric bodies has to be constant, with preventive and corrective purpose, depending on the eutrophization state of each one. The objective of this article was to analyze the sediment toxicity and the metals Mn, Zn, Pb, Ni, Cd and Cu, in the Paiva Castro reservoir, that supply the Metropolitan Region of São Paulo. It was made 2 collections (May 2011, dry season and January 2012, wet season). It was analyzed 5 points next to the water captation station by the Companhia de Saneamento Básico do Estado de São Paulo. The sediment was analyzed as for acute and chronic toxicity through bioassays with the cladocerans Daphnia similis and the insect Chironomus xanthus. Data treatment was done with Fisher Exact Test (mortality). The association level between the variables in sediment and ecotoxicological tests was available in non-parametric tests, through the Spearman's correlation coefficient. Oriented on the results presented in this work, pointing low concentrations of heavy metals in the sediments presented in the collect local, and the absence of toxicity, we can say that at this reservoir, at least in the collect area, it's low impacted, not implicating in direct interferences in the quality of life of benthonic organisms.

ABSTRACT

Zinc (Zn) is an essential trace element for cellular viability, but concentrations above physiologic level may lead to cellular damage. The purpose of the present study was to evaluate the in vitro ZnCl2 genotoxicity and cytotoxicity in human leukocyte cells. This was assessed in an unprecedented way that correlated the level of intracellular Zn after cell exposition with the cellular damage. The exposure to increased Zn concentrations (2.5-20 µg mL-1), showed significantly reduced cellular leukocyte viability. However, significant DNA damages were observed only when the Zn exposure concentrations were from 10-20 µg mL-1. The Zn intracellular levels found in leukocytes was from 72.25-268.9 ρ g cell-1, starting to induce cytotoxicity and genotoxicity at concentrations of 95.68 and 126.2 ρg cell-1, respectively. The relationship between the exposure concentration and intracellular levels of Zn suggests that the influx of Zn, in the form of ZnCl2, occurs in human leukocytes under zero-order kinetics.


O Zinco (Zn) é um elemento traço essencial para a viabilidade celular, mas em concentrações acima dos níveis fisiológicos pode conduzir a danos celulares. A proposta do presente estudo foi avaliar a citotoxicidade e genotoxicidade do ZnCl2 em leucócitos humanos in vitro. De maneira sem precedentes, foi acessado o nível de Zn intracelular após exposição e relacionado com o nível de dano celular. A exposição a crescentes concentrações de Zn (2,5-20 µg mL-1), mostraram significante redução da viabilidade celular dos leucócitos. Entretanto, danos significativos ao DNA foram encontrados somente a partir das concentrações de exposição ao Zn de 10-20 µg mL-1. Os níveis intracelulares de Zn encontrados nos leucócitos foram de 72,25-268,9 ρg célula-1, começando a induzir citotoxicidade e genotoxicidade nas concentrações de 95,68 and 126,2 ρg célula-1, respectivamente. A relação entre a concentração de exposição e os níveis intracelulares de Zn sugerem que o influxo de Zn, sob a forma de ZnCl2, ocorre em cinética de ordem zero em leucócitos humanos.

ABSTRACT

The effects of sublethal concentrations of cadmium (0.64 µg/L), iron (0.043 mg/L) and zinc (0.31 mg/L) and a mixture of these metals on succinate dehydrogenase (SD) and alkaline phosphatase (AP) activity and on structural changes in the mitochondria of epithelium cells of the digestive tract were examined in the oligochaete Limnodrillus hoffmeisteri after 96 h of exposure in artificial sediments. SD activity was significantly inhibited, particularly in treatments with Cd alone (92.57 percent), while AP increased its activity with Cd alone (73.23 percent). However, when this metal was mixed with Fe and Zn, the inhibition of SD activity was lower (67.82 percent) than with Cd alone, showing an antagonistic effect and AP increased its activity (73.26 percent). Mitochondria were structurally damaged by exposure to Cd alone. However, in the metal mixtures, the toxic effects may exert interactive effects eliciting a less structural damage in the mitochondria of epithelium cells of the digestive tract than when Cd is alone.


Se estudió el efecto de las concentraciones subletales de Cd (0,64 µg/L), Fe (0,043 mg/L) y Zn (0,31 mg/L) en forma aislada y en mezcla sobre la actividad de la succinato deshidrogenasa (SD) y la fosfatasa alcalina (AP) en las mitocondrias de las células epiteliales del tracto digestivo en el oligoqueto Limnodrillus hoffmeisteri después de 96 h de exposición en sedimentos artificiales. La SD se inhibió significativamente, particularmente en los tratamientos con Cd en forma aislada (92,57 por ciento), mientras que la AP se incrementó con Cd en forma aislada (73,23 por ciento). Sin embargo, cuando este metal se mezcló con Fe y Zn, la inhibición de la SD fue menor (67,82 por ciento) que con Cd en forma aislada, lo que mostró un efecto antagonístico y la AP incrementó su actividad (73,23 por ciento). ABSTRACT

The influence of increased zinc concentrations (0.1, 0.5, 1.0, 1.5 and 2.0 mg(-1) ZnSO4 x 7H2O) on the total number and the morphology of the erythrocytes, as well as the processes related to their formation and destruction in the spleen of Carassius gibelio were investigated ex situ. It was found that zinc concentrations caused pathological alterations in the erythrocytes that were not identical in the different concentrations-poikilocytosis; ruptures in cell membranes in the concentrations of 0.5mg(-1) and 1.5 mg(-1); cells with double nuclei (symplasts); in the concentration of 1.0 mg(-1); in the highest concentrations (1.5 mg(-1) and 2.0 mg(-1)) presence of erythrocytes at initial stage of atypical mitotic division. Against the background of those various alterations, the total number of the erythrocytes in the peripheral blood increased simultaneously with the increase of zinc concentrations (p < 0.001). Morphological alterations in the spleen were also observed, indicating a compensational tendency against the toxic influence of zinc upon the fish erythrocytes-hyperplasia of the red pulp and lack of hemosiderin. These results show that the alterations in the total number and the morphology of the erythrocytes are connected with the relevant compensatory histopathological alterations in the spleen. The use of the ascertained alteration could be valuable in monitoring zinc-polluted waters.

ABSTRACT

The present investigation was undertaken to assess the effects of sublethal concentrations of zinc (0.1 and 0.2 mgl(-1)) on the nutritional value of fish Channa punctatus after exposure for 135 days. The parameters studied were, levels of total proteins, glycogen, total lipids, cholesterol and vitamins (A and D) in the muscle. Total proteins, glycogen, total lipids and vitamin D show highly significant decline from day 90 to 135, while cholesterol and vitamin A, show no significant changes upto 105 days. This study signifies a drastic reduction in the nutritive value of fish, which is compounded by the fact that under natural conditions fish are exposed to these levels of zinc for prolonged periods.

ABSTRACT

The effects of different concentrations of Zn, Pb and Cd singly as well as in the combination of zinc-lead, zinc-cadmium and lead-cadmium on the shoot root length and dry mass as well as chlorophyll contents of barley plants during early seedling growth were studied. The effects of different concentrations of Zn x Pb and Pb x Cd in combinations showed a synergistic relationship, while at higher concentrations the relationship was antagonistic. Lower as well as higher Zn x Cd concentrations showed antagonistic effects.

ABSTRACT

The diversity of phosphobacteria in Manakudi mangrove ecosystem of Tamil Nadu was carried out in root and rhizosphere soil samples. The counts of phosphobacteria were found higher in root samples than in soil samples particularly in Hymenachene acutigluma. The abundance of phosphobacterial diversity in Manakudi mangrove showed high degree of positive correlation with the content of phosphate in rhizosphere soil of all the mangrove and associated plant species. Nine phosphobacterial species belonging to 7 genera were reported from Manakudi mangrove ecosystem. All the identified bacterial species are sensitive to both the heavy metals (mercury and zinc) in terms of growth and physiology even at lower concentrations. The content of protein and total sugars were increased by the higher concentrations of heavy metals whereas decreased trend was noticed in lower concentrations of heavy metals.

ABSTRACT

O alginato ou hidrocolóide irreversível é um dos materiais de moldagem mais aceitos e utilizados na Odontologia. Muitas substâncias como zinco, cádmio, silicato de chumbo e fluoretos foram adicionadas em algumas marcas de alginatos, com o objetivo de melhorar suas propriedades físicas, químicas, mecânicas e se tornaram causa de preocupação no que se refere à toxicidade desses materiais. Em algumas marcas de alginatos relatou-se a presença de fluoretos, cádmio, silicatos de chumbo e zinco potencialmente tóxicos, isoladamente ou em conjunto, consequentemente, cuidados especiais devem ser tomados na preparação desses materiais. É necessário que haja um controle contínuo de metais e substâncias potencialmente tóxicas nos alginatos para se evitar a contaminação dos profissionais da área odontológica e pacientes. Nesta revisão analisou-se o potencial tóxico de alginatos usados em odontologia.

ABSTRACT

Zinc, cobre y selenio son cofactores de enzimas antioxidantes, por lo que una disminución de sus niveles séricos, compromete su papel preventivo de enfermedades degenerativas. En este estudio se planteó evaluar la influencia del hábito de fumar cigarrillo sobre las concentraciones séricas de estos cofactores en adultos jóvenes. La muestra estuvo conformada por 40 individuos sanos del sexo masculino, con edades entre 18 y 25 años, índice de masa corporal normal y consumo de nutrientes acorde con sus requerimientos nutricionales, divididos en grupo control (n=20) y grupo de fumadores crónicos (n=20), a los cuales se les cuantificó los niveles séricos de zinc, cobre, selenio y se les estimó la ingesta de nutrientes a través del recordatorio de 24 horas y la frecuencia de consumo de alimentos. En los fumadores los niveles séricos de zinc se mostraron severamente disminuidos (0,36 + 0,10 mg/L) y significativamente inferiores a los encontrados en los controles (0,71 + 0,09 mg/L), mientras que los niveles sérico de cobre y selenio no mostraron diferencia significativa. Los resultados indican que aún cuando el consumo de minerales en los fumadores fue adecuado, la dieta habitual no logró mantener la concentración sérica de zinc en los rangos normales, haciéndolos más susceptibles al estrés oxidativo.

ABSTRACT

Most laboratory assessments of toxicity and bioaccumulation of heavy metals have been concentrated on the accumulation of these metal ions when exposed singly to the test organisms. However, under the natural environmental settings, the metals are never present in isolation and may interact with each other, therefore justifying the need to study the influence of joint application of metals on accumulated levels in exposed animals. In this study, exposure of the periwinkle Tympanotonus fuscatus to sublethal concentrations (equivalent to 0.1 and 0.01 of 96 h LC50) of heavy metals revealed that they were bioaccumulative varying amounts, depending on the type of metal, exposure period and concentration in the test media. While Zn and Pb ion accumulation increased steadily with exposure time, the amounts of Cu accumulated fluctuated regularly over the 30-day experimental period. The levels of Zn, Cu and Cd bioaccumulated over the 30-day experimental period were reduced by over 2-6 folds (with bioaccumulation ratio values ranging from 0.15 to 0.81) when compared to concentrations of the respective metals accumulated during single bioaccumulation studies. However, Pb concentrations accumulated during the joint action studies increased nearly 2-fold (bioaccumulation ratio range 1.36 to 2.0-fold).


La mayoría de los análisis de laboratorio sobre toxicidad y bioacumulación de metales pesados se han concentrado en la acumulación individual de los iones metálicos en los organismos de prueba expuestos a ellos. Sin embargo, bajo condiciones naturales del ambiente, los metales nunca se presentan en forma aislada y deben interactuar unos con otros, justificando así la necesidad de estudiar el efecto de la aplicación conjunta de metales en animales de prueba. En este estudio la exposición de Tympanotonus fuscatus a concentraciones subletales (0.1-0.01 de 96 h LC50) de metales pesados reveló que la bioacumulación varía según el tipo de metal, el período de exposición y la concentración en el medio de prueba. Mientras los iones de Zn y Pb incrementaron gradualmente su acumulación al pasar el tiempo, las cantidades de Cu fluctuaron regularmente durante los 30 días del experimento. Los niveles de Zn, Cu y Cd bioacumulados durante los 30 días del experimento son de dos a seis veces menores (con un rango de bioacumulación de 0.15-0.81) que los que se obtienen por la exposición a los metales en forma individual. Por el contrario, la acumulación de Pb aumentó al doble (rango de bioacumulación 1.36-2.0).

17-day-old bean plants (Phaseolus vulgaris cv. Strike) were used to analyze the effects of Co2+ and Zn2+ on the time course of proline, total protein, chlorophyll and abscisic acid (ABA) levels in leaves. Controls, Co2+ and Zn2+-treated plants were grown for 8 days in Hoagland solution. Samples were taken at 2 day intervals. Proline, chlorophyll (a+b) and total protein contents of 17 day old primary leaves were determined by a spectrophotometer. ABA contents in roots and leaves of the seedlings were measured by high-pressure liquid chromatography. The presence of Zn2+ and Co2+ significantly increased the ABA contents in roots and leaves (p < 0.05 or p < 0.01). The increase of the abscisic acid content in the leaves was related to the content of the roots. This was further substantiated by enhanced accumulation of proline in the leaves of seedlings exposed to zinc and cobalt. The contents of chlorophyll (a+b) and total protein decreased with the concentration of both metals (p < 0.05 or p < 0.01). Cobalt proved to be comparatively more toxic than zinc.

ABSTRACT

Labeo rohita fingerlings were exposed to zinc metal toxicity (5 and 10 ppm) for duration of 5 and 15 days. The histological changes were studied in brain and liver of the treated fish. The brain tissues showed enlarged pyramidal cells with extensive vacoulation while severe necrosis, haemorrhage and degeneration of hepatocytes were witnessed in the liver tissues.

Metalotioneínas (MTs) são proteínas citosólicas cuja função biológica está relacionada à regulação de metais essenciais e a detoxificação de metais tóxicos. O conhecimento da compartimentalização intracelular de metais, da função fisiológica das MTs e dos fatores que controlam seus níveis em diferentes espécies de peixes é essencial para compreensão dos efeitos tóxicos induzidos por metais. Geophagus brasiliensis (acará) é uma espécie nativa do Brasil, com ampla distribuição e aspectos ecológicos que favorecem sua utilização como espécie bioindicadora dos efeitos da poluição ambiental por metais. Este trabalho teve por objetivo estudar MT e metais em acarás, analisando a influência dos parâmetros biológicos do animal, além das alterações produzidas pela exposição a um metal tóxico como cádmio (Cd). Acarás coletados no rio Guandu foram divididos em dois grupos. (...) Neste estudo foi verificado que MT e metais em acarás são influenciados por alguns parâmetros biológicos. A exposição ao Cd também promove uma alteração nos níveis de MT e de metais bem como na distribuição intracelular. Estes fatores estão relacionados com à toxicidade e a tolerância à exposição a metais e devem ser considerados para que esta espécie possa ser utilizada como bioindicadora da exposição ambiental a metais.

ABSTRACT

Sub-lethal concentrations of zinc administered to freshwater teleost, Channa punctatus for a period of 135 days brought about significant haematological alterations. The RBC count, haemoglobin and haematocrit content progressively decreased while WBC count, MCV, MCH and MCHC increased. Alterations in the haematological parameters were dose and duration dependent and can thus serve as a useful physiological index.

ABSTRACT

Alginate is one de materials most employed in practice to make dental impressions. Substances like zinc, cadmium and lead silicate, which are included in several alginate brands with the aim of improving their physical, chemical and mechanical properties, are a source of serious concern regards their toxicity. The most serious chronic effect of oral exposure to cadmium is renal toxicity. Assimilation of lead has deleterious effects on the gastrointestinal tract, hematopoietic system, cardiovascular system, central and peripheral nervous systems, kidneys, immune system, and reproductive system. Chronic oral exposures to zinc have resulted in hypochromic and microcyte anemia in some individuals. The aim of the present study was to measure the cadmium, lead and zinc contents of seven brands of alginate for dental use on sale in Brazil. The samples were weighed and placed in the Teflon cups of a closed-system microwave oven. Aqua regia (4 mL concentrated HCl:HNO3, 3:1 v/v) and hydrofluoric acid (2mL concentrated HF) were added to the samples, which were then subjected to heating. The samples were then cooled to room temperature and diluted to 25 mL in deionized water in a volumetric glass flask. The samples were diluted in duplicate and analyzed against a reagent blank. The analyses were performed in an atomic absorption flame spectrophotometer. Neither lead nor cadmium was detected. Zinc contents ranged from 0.001% to 1.36% by weight. The alginates exhibited low contents of the metals under study and gave no cause for concern regarding toxicity; even so, it is advisable to monitor potentially toxic materials continually and to analyze their plasmatic levels in the professionals working with them

ABSTRACT

Zinc is one of the important elements necessary for human growth. CNS utilizes zinc for its biological functions. Zinc is one of the important cofactors of many enzymes in the body and about 200 enzymes contain zinc as their integral part of their structure. If for any reason zinc is omitted from the enzyme structure, those enzymes can not catalyze any biological functions. The fluctuation of this trace element in the body is clinically important. The concentration change of this trace element can lead to some overt and unrecognized manifestations in haemodialysis patients. This study is to determine the possible fluctuation of serum level of zinc in haemodialysis patients before and after dialysis, to explain the differences in concentration of such trace element in these patients. This study is a comparative and nonexperimental study which was carried out on 50 haemodialysis patients chosen randomly from the patients referred to the department of haemodialysis in 5[th] Azar hospital in Gorgan university of medical sciences. The concentration of urea, creatinine, zinc of these patients were determined using laboratory kits and spectrophotometry tchnique before and after the dialysis. The findings were analyzed by wilkacson non parameter analytical method. Results of this study indicated that the average concenteration of zinc in haemodialysis patients after dialysis was 135.32 +/- 59.32 microgram / deciliter which markedly increased in comparison to the serum concentration of this trace element before the dialysis which was 78.38 +/- 37.46 microgram / deciliter [p < 0.05]. Uremia, dialysis membrane, contaminations, heavy metals, and the quality of consumed water in the dialysis procedure, all can pave the way for the abnormality of trace element and clinical manifestation in haemodialysis patients. The trace element variation concentrations in human body clinically are very important because the decrease and increase in level of each trace element has its own clinical manifestation. The increased concentration of this trace element leads to toxification accompanied with clinical syndromes such as diarrhea and vomiting. Therefore, it is suggested that before the process of dialysis the level of released zinc from the different part of dialysis machine [such as pipeline, consumed water and dialysis membrane] be determined and on the basis of such information the level of zinc in haemodialysis patients can be replaced by proper nutrition

ABSTRACT

OBJECTIVE: To determine the effect of exposure to metal dusts, fumes and high temperature levels among brass workers in comparison to a control group. STUDY DESIGN: Analytical cross-sectional study. METHODOLOGY: One hundred and fifty four brass workers were matched for age with 154 controls selected from the local population. An interviewer-administered questionnaire was used to determine the presence of acute and chronic symptoms and metal fume fever. Haemoglobin and blood zinc and copper levels were measured using the cyanmethaemoglobin technique and atomic absorption spectrophotometry respectively. Thermal environmental measurements were carried out by determining wet bulb and globe temperature (WBGT) levels and air velocity. RESULTS: Among the chronic symptoms anorexia (OR = 3.3), distaste (OR = 8.3), and aches and pains (OR = 4.0) were significantly higher in the study group. Among the acute symptoms at work, cough (OR = 4.2), dry nose (OR = 6.8), tearing (OR = 6.3), and itchy eyes (OR = 6.3) were significantly higher in the study group. Sweating was significantly higher in the control group. Metal fume fever was significantly higher among the study group with an OR of 7.6. Levels of both copper and zinc were significantly higher in the study group, although both median and mean values were lower than the normal reference ranges. The recommended WBGT level of 26.1 degrees C for an air velocity of less than 1.53 m/s for heavy work was exceeded only in two workshops. CONCLUSIONS: Prevalence of non-specific symptoms was higher among brass workers. It is necessary to take preventive measures.

ABSTRACT

The effects of insecticides (endosulfan, HCH) and heavy metals (copper and zinc) were studied on survival and duration of the moult cycle of larvae and megalopa of a common mangrove crab, Macrophthalmus erato, that inhabits Pitchavaram mangroves, southeast coast of India. In chronic toxicity test, percentage of survival from hatchling to megalopa stage at different concentrations of endosulfan (0.048 to 0.36 microg/L), HCH (0.32 to 2.4 microg/L), copper (9.0 to 67.5 microg/L) and zinc (15.2 to 139.0 microg/L) were 6, 12, 8 and 12 respectively. Endosulfan, HCH, copper and zinc considerably increased the total period in development from first zoea to megalopa stage from 18 days in the control to 23, 21, 20 and 22 days in the treated specimens. It is concluded that concentrations well below the observed chronic concentrations, may prove to be more deleterious to larva of this species in natural environment leading to reduction in the success of recruitment to the adult population.
