ABSTRACT

Angiokeratoma corporis diffusum [ACD] is a variety of angiokeratoma, characterized by diffuse cutaneous hyperkeratotic vascular lesions. ACD is usually associated with many lysosomal enzyme deficiencies, though it is not a rule. It is associated with systemic features along with skin lesions. It may also present as isolated cutaneous form. X-linked recessive form usually manifests in males and females act as carriers. We report a case of ACD without systemic features in a young Kashmiri female.

ABSTRACT

Various oxidases and hydrolytic enzymes were analyzed to investigate the relationship between these enzymes and the skin pathogenicity of 18 Mucorales strains. Each strain was cultured in a nutrient medium containing starch as a carbon source. The cells grew quickly and were at a good state of growth after incubation for three days. Oxidase activity was not detected in any strain, whereas Mucor spp. including Mucor racemosus IFM47053 typically had high alcohol dehydrogenase (ADH) activity and all the strains had catalase activity. The culture filtrate and the cell free extract of each strain were applied to APIZYM test system, which revealed that all the strains examined produced many hydrolytic enzymes both inside and outside their mycelia. In the case of Absidia corymbifera strains, lipase activity was comparatively high, and polysaccharide hydrolytic enzymes such as alpha-glucosidase, beta-glucosidase, N-acetyl-beta-glucosaminidase, alpha-mannosidase, and alpha-fucosidase were produced.

ABSTRACT

The aim of the present study was to demonstrate the presence of alpha-L-fucosidase in Trypanosoma cruzi. Immunocytochemical and biochemical techniques were used to localize and characterize a membrane-associated, neutral-pH-optimum, alpha-L-fucosidase from the parasite. Light and electron microscopy localized the alpha-L-fucosidase specifically on the surface of the parasite and on membranes in the posterior region of the epimastigote stage. Although much less intense, labeling was also detected on the surface of trypomastigotes. At least 50 percent of the alpha-L-fucosidase activity was associated with epimastigote membrane solubilized with 1 M NaCl or 1 percent Triton X-100, suggesting that alpha-L-fucosidase is peripherally associated with membranes. The enzyme from epimastigotes had a neutral pH optimum (near 7) but displayed low specific activity when p-nitrophenyl-alpha-L-fucoside was employed as substrate (0.028 U/mg protein for epimastigotes and 0.015 U/mg protein for tissue culture-derived trypomastigotes). Polyacrylamide gel electrophoresis and Western blotting analysis both showed an expected 50-kDa polypeptide which was immunoreactive with anti-alpha-L-fucosidase antibodies

ABSTRACT

To evaluate the role of serum alpha-L-fucosidase (AFU) in the diagnosis of hepatocellular carcinoma (HCC), we simultaneously studied both AFU activity and alpha-fetoprotein (AFP) level in 60 patients with HCC, 60 patients with cirrhosis and chronic hepatitis each, 30 patients with other liver tumors and 60 healthy subjects. Serum AFU activity in patients with HCC (1,418.62 +/- 575.76 nmol/ml/hr) was significantly higher than that found in cirrhosis (831.25 +/- 261.13 nmol/ml/hr), chronic hepatitis (717.71 +/- 205.86 nmol/ ml/hr) or other tumors (706.68 +/- 197.67 nmol/ml/hr) and in controls (504.18 +/- 121.88 nmol/ml/hr, p < 0.05). With 870 nmol/ml/hr (mean value of controls plus 3 standard deviations) considered as the cut-off point, AFU was more sensitive (81.7 vs 39.1%) but less specific (70.7 vs 99.3%) than AFP at a level of > 400 ng/ml as a tumor marker of HCC. With both markers combined, the sensitivity was improved to as much as 82.6%. AFU activity in HCC patients was correlated to tumor size (r = 0.3529, p = 0.006) but not associated with tumor staging classified by Okuda's criteria (p = 0.1). The AFU activity in the viral hepatitis group (hepatitis B or C) was also significantly higher than in the non-viral group (p = 0.0005). We conclude AFU to be a useful marker, in conjunction with AFP and ultrasonography, for detecting HCC, particularly in patients with underlying viral hepatitis and cirrhosis.

ABSTRACT

Se describen 3 pacientes: 2 del sexo masculino y 1 del femenino, con enfermedad por almacenamiento lisosomal del tipo fucosidosis. En los 3 casos existió el antecedente de retardo psicomotor y posteriormente se hizo evidente el deterioro progresivo de las capacidades psíquicas motoras adquiridas. Todos tenían facies toscas, retardo mental profundo y alteraciones morfológicas en vértebras lumbares. Desde el punto de vista bioquímico, los 3 pacientes presentaron excreción urinaria de oligisacáridos ricos en fucosa, así como disminución de la actividad de la enzima alfa-L-fucosidasa en leucocitos. Se hace énfasis en la demostración del defecto enzimático como único criterio diagnóstico confirmatorio

ABSTRACT

Se comunica el caso de un paciente de sexo masculino de 24 años de edad, que presenta angioqueratomas múltiples en todo el tegumento y mucosas, con afectación oftalmológica típica. Se realizó ultramicroscopía de una lesión de piel, donde pudieron verse inclusiones citoplasmáticas en células endoteliales, fibroblastos, células periteliales y perineurales. Bioquímicamente se comprobó la deficiencia enzimática en plasma de la * galactosidasa "A". Se realiza una revisión histórica y se consideran además los aspectos clínicos, histopatológicos, ultramicroscópicos y enzimáticos, como así los distintos diagnósticos diferenciales. También se dan algunas pautas terapéuticas novedosas y otras paliativas. Para finalizar, sugerimos el estudio genético de los familiares directos del paciente para detectar posibles futuros casos y poder brindarles consejo genético adecuado

ABSTRACT

Con el propósito de conocer los niveles séricos de ácido siálico, 1-fucosa y fucosidasa, se cuantificaron sus valores en 107 adultos (25 a 90 años) sometidos a endoscopia oral. Con los resultados obtenidos se analiza su utilidad como método de pesquisa en el diagnóstico de cáncer gástrico. La muestra estudiada estuvo integrada por cuatro grupos: sin alteraciones endoscópicas (n=25); úlcera duodenal (n = 25); úlcera gástrica (n = 25) y cáncer gástrico (n = 32). Los valores promedio obtenidos para el ácido siálico fueron: 1,96; 1,19; 20,5 y 2,80 micronmol/ml; para la 1-fucosa: 9,32; 9,00; 10,8 y 13,0 mg/dl y para la actividad fucosidasa: 11,5; 10,2; 9,06 y 10,6 U/l respectivamente. El coeficiente de correlación ácido siálico-1-fucosa fue 0,80 (p < 0,01) en los grupos sin alteración y cáncer gástrico. La fucosidasa no tuvo variaciones significativas entre los diferentes grupos estudiados. Las determinaçöes de ácido siálico y 1-fucosa presentaron una sensibilidad del 81 y 74% respectivamente en pacientes portadores de cáncer gástrico pero ella fue de 97% cuando se consideran pacientes que tienen ácido siálico o 1-fucosa positivos. Porcentajes importantes de pacientes sin alteraciones endoscópicas o con otras afecciones gástricas cursan con valores positivos de ácido siálico y 1-fucosa. Por tratarse de procedimientos simples y sensibles, las ácido siálico y 1-fucosa. Por tratarse de procedimientos simpsles y sensibles, las determinaciones séricas de ácido siálico y 1-fucosa pudiera ser de utilidad para la detección de cáncer gástrico o de su control evolutivo

ABSTRACT

Los niveles de alfa-fucosidasa y n-acetil-hexosaminidasa fueron dosadas en el suero de 190 pacientes portadores de cancer de diversos tipos. Del total estudiado, 109 pacientes habian sido sometidos a tratamiento mientras que 81 habian recibido tratamiento adecuado.El 87% de las pacientes con carcinoma de mama y el 80% de las portadoras de carcinoma de cuello uterino que no habian sido sometidas a tratamiento mostraron niveles enzimaticos sericos por enzima de los valores maximos normales, mientras que el 95% de las pacientes con carcinomas de utero, ovario u otros carcinomas invasivos, presentaron actividad enzimatica serica elevada.Una disminucion de la actividad enzimatica durante el tratamiento parece predecir una respuesta positiva al mismo, mientras que el progresivo incremento de los niveles enzimaticos durante o despues del tratamiento, indicaria una respuesta negativa al mismo o la recurrencia de la enfermedad
