Abstract: Objective: The feasibility of the use of WHO impregnated paper and biochemical assays to determine lethal concentrations (LC50 and LC99) and insecticide metabolic enzyme levels of Triatoma dimidiata. Materials and methods: LC50 and LC99 were calculated with WHO papers impregnated at different concentrations of malathion, propoxur and deltamethrin; the percentage of insensitive acetylcholinesterase (iAChE); and the levels of esterases, glutathione S-transferases, and monooxygenases in laboratory nymphs of the first stage (5 to 7 days), were undertaken using the WHO biochemical assays. Results: Respectively the LC50 and LC99 µg/cm2 obtained for malathion were 43.83 and 114.38, propoxur 4.71 and 19.29, and deltamethrin 5.80 and 40.46. A 30% of the population had an iAChE, and only a few individuals had high P450 and β-eterase levels. Conclusion: Impregnated papers and biochemical tests developed by WHO for other insects, proved to be feasible methods in monitoring insecticide resistance and metabolic enzymes involved in T. dimidiata.
Resumen: Objetivo: La factibilidad de usar los papeles impregnados y ensayos bioquímicos según la OMS para determinar concentraciones letales (CL50 y CL99) y niveles enzimáticos en la resistencia a insecticidas en Triatoma dimidiata. Material y métodos: Se calcularon la CL50 y CL99 con papeles impregnados según la OMS a diferentes concentraciones de malatión, propoxur y deltametrina; el porcentaje de acetilcolinesterasa insensible (iAChE); y los niveles de esterasas, glutatión S-transferasas, y monooxigenasas en ninfas de laboratorio del estadio I (5-7 días) se determinaron usando los ensayos bioquímicos según la OMS. Resultados: Se obtuvieron las CL50 y CL99 µg / cm2 respectivamente para malatión 43.83 y 114.38, propoxur 4.71 y 19.29, y deltametrina 5.80 y 40.46. Un 30% de las chinches tuvo iAChE, y sólo pocos individuos tuvieron niveles superiores de P450 y β-eterasas. Conclusión: Los papeles impregnados y ensayos bioquímicos que describe la OMS para otros insectos demostraron ser métodos factibles para monitorear la resistencia a insecticidas y las enzimas metabólicas involucradas en T. dimidiata.

Resumen: Objetivo: Determinar la resistencia a insecticidas en Ae. aegypti y Ae. albopictus de Tapachula, Chiapas, México. Material y métodos: Se utilizaron ovitrampas para obtener huevos de mosquitos Aedes y se realizaron pruebas de susceptibilidad (CDC) y ensayos enzimáticos con la primera generación. Resultados: Aedes aegypti mostró resistencia a deltametrina, permetrina, malatión, clorpirifos, temefos y a bendiocarb (CARB), mientras que Aedes albopictus a malatión y en menor grado a cloripirifos, temefos, permetrina y deltametrina. Ambas especies mostraron altos niveles de enzimas como citocomo P450 y glutatión S-tranferasa, mientras que los niveles de esterasas variaron por especie y sitio muestreado. Se detectó acetilcolinesterasa insensible a insecticidas en ambas especies. Conclusión: En un hábitat urbano de Tapachula, Chiapas, México donde se aplica control con insecticidas Ae. aegypti y Ae. albopictus sólo son susceptibles al propoxur.
Abstract: Objective: To determine the insecticide resistance status of Ae. aegypti and Ae. albopictus from Tapachula, México. Materials and methods: Mosquito eggs were collected with the use of ovitraps and CDC susceptibility bioassays and biochemical assays were conducted to determine resistance levels and resistance mechanisms, respectively. Results: Ae. aegypti showed resistance to deltamethrin and permethrin (PYRs), malathion, chlorpyrifos and temephos (OP), and to bendiocarb (CARB), while Ae. albopictus showed resistance to malathion and to a lesser intensity to chlorypirifos, temephos, permethrin and deltamethrin. Both species showed high levels of P450 and GSTs, while levels of esterases varied by species and collection site. Altered acethilcholinesterase was detected in both species. Conclusion: In an urban habitat from Tapachula, Chiapas, Mexico where vector control using insecticides takes place, Ae. aegypti and Ae. albopictus are only susceptible to propoxur.

O fipronil é um inseticida de toxicidade seletiva amplamente empregado na agricultura e na medicina veterinária. Porém, há relatos de efeitos neurotóxicos dessa substância, que geram prejuízos para vertebrados. Avaliou-se a atividade locomotora, a coordenação motora e a atividade da enzima acetilcolinesterase cerebral em ratos expostos ao fipronil. Ratos Wistar machos adultos (n=15) receberam fipronil em dose de 30mg/kg, por via oral, durante 15 dias; o grupo controle (n=15) foi tratado com solução fisiológica, por via oral, no mesmo período. No 16° dia de experimentação, os animais foram submetidos aos testes de arena de campo aberto e hole board. No 17° dia, foram anestesiados e eutanasiados, procedendo-se à coleta de órgãos, e posteriormente foi feita a avaliação da AChE cerebral. A exposição ao fipronil não provocou alterações significativas sobre a coordenação motora e a atividade locomotora, porém gerou inibição significativa da atividade da acetilcolinesterase cerebral. Esses achados sugerem que o fipronil pode provocar efeitos neurotóxicos em curto prazo, os quais podem ser exacerbados caso a exposição seja prolongada.(AU)
Fipronil is a selective-toxicity insecticide widely used in agriculture and veterinary medicine. However, there are reports of neurotoxic effects of this substance, causing damages to vertebrates. We evaluated the locomotor activity, motor coordination and the activity of brain acetylcholinesterase in rats exposed to fipronil. Adult male Wistar rats (n= 15) received fipronil at a dose of 30mg/kg orally for 15 days; the Control group (n= 15) was treated with oral solution in the same period. On the 16th day of experimentation, the animals were submitted to the open field arena test and hole-board test. On the 17th day, they were anesthetized and euthanized, and organs were collected, and subsequently brain AChE was evaluated. Exposure to fipronil yielded no significant changes on motor coordination and locomotor activity but caused significant inhibition of brain acetylcholinesterase activity. These findings suggest that fipronil may cause short-term neurotoxic effects, which may be exacerbated if exposure is prolonged.(AU)

When exercises are done in intense or exhaustive modes, several acute biochemical mechanisms are triggered. The use of cryotherapy as cold-water immersion is largely used to accelerate the process of muscular recovery based on its anti-inflammatory and analgesic properties. The present study aimed to study the biochemical effects of cold-water immersion treatment in mice submitted to exercise-induced exhaustion. Swiss albino mice were divided into 4 treatment groups: control, cold-water immersion (CWI), swimming exhaustive protocol (SEP), and SEP+CWI. Treatment groups were subdivided into times of analysis: 0, 1, 3, and 5 days. Exhaustion groups were submitted to one SEP session, and the CWI groups submitted to one immersion session (12 min at 12°C) every 24 h. Reactive species production, inflammatory, cell viability, and antioxidant status were assessed. The SEP+CWI group showed a decrease in inflammatory damage biomarkers, and reactive species production, and presented increased cell viability compared to the SEP group. Furthermore, CWI increased acetylcholinesterase activity in the first two sessions. The present study showed that CWI was an effective treatment after exercise-induced muscle damage. It enhanced anti-inflammatory response, decreased reactive species production, increased cell viability, and promoted redox balance, which could decrease the time for the recovery process.

ABSTRACT Studies have shown that schizophrenic patients seem to have nutritional deficiencies. Ascorbic acid (AA) has an important antioxidant effect and neuromodulatory properties. The aim of this study was to evaluate the effects of AA on locomotor activity and the acetylcholinesterase activity (AChE) in an animal model of schizophrenia (SZ). Rats were supplemented with AA (0.1, 1, or 10 mg/kg), or water for 14 days (gavage). Between the 9th and 15th days, the animals received Ketamine (Ket) (25 mg/kg) or saline (i.p). After the last administration (30 min) rats were subjected to the behavioral test. Brain structures were dissected for biochemical analysis. There was a significant increase in the locomotor activity in Ket treated. AA prevented the hyperlocomotion induced by ket. Ket also showed an increase of AChE activity within the prefrontal cortex and striatum prevented by AA. Our data indicates an effect for AA in preventing alterations induced by Ket in an animal model of SZ, suggesting that it may be an adjuvant approach for the development of new therapeutic strategies within this psychiatric disorder.

Alzheimer's disease (AD) is a fast growing neurodegenerative disorder of the central nervous system and anti-oxidants can be used to help suppress the oxidative stress caused by the free radicals that are responsible for AD. A series of selected synthetic indole derivatives were biologically evaluated to identify potent new antioxidants. Most of the evaluated compounds showed significant to modest antioxidant properties (IC50 value 399.07 140.0±50 µM). Density Functional Theory (DFT) studies were carried out on the compounds and their corresponding free radicals. Differences in the energy of the parent compounds and their corresponding free radicals provided a good justification for the trend found in their IC50 values. In silico, docking of compounds into the proteins acetylcholinesterase (AChE) and butyrylcholinesterase (BChE), which are well known for contributing in AD disease, was also performed to predict anti-AD potential.
A doença de Alzheimer (DA) é uma doença neurodegenerativado sistema nervoso central, em rápido crescimento, e antioxidantes ajudam a suprimir o estresse oxidativo causado por radicais livres, responsávies pela DA. Avaliou-se, biologicamente, série de derivados sintéticos de indol selecionados para identificar novos antioxidantes. A maioria dos compostos avaliados apresentou de significativa a boa propriedade antioxidante (valor de IC50 399,07140.0 ± 50 µM). Eftuaram-se estudos de Teoria do Funcional de Densidade (DFT) com os compostos e os seus correspondentes radicais livres. As diferenças de energia entre os compostos protótipos e os radicais livres correspondentes proporcionaram boa justificativa para a tendência encontrada nos seus valores de IC50. O ancoramento in silico dos compostos com a acetilcolinesterase (AChE) e com a butirilcolinesterase (BChE), que contribuem para a DA, foi, também, realizado para prever o seu potencial anti-DA.

The extracts of shade dried leaves of A. tetracantha were tested for in vitro inhibitory activity on toxic venom enzymes like phosphomonoesterase, phosphodiesterase, acetylcholinesterase, hyaluronidase etc. from Bungarus caeruleus and Vipera russelli venoms.

Microcystins are secondary metabolites produced by different species of cyanobacteria, such as Microcystis aeruginosa (MA). In this study, the biochemical and genetic effects of lyophilized MA were evaluated in the neotropical fish Prochilodus lineatus exposed to 1 or 2 mg L-1 lyophilized MA (treated group) or only water (control group) in static toxicity tests for 24 and 96 h. The gills and liver were used in the analysis of biotransformation enzymes and antioxidant defenses, blood and gill cells in genetic analysis and in brain and muscle it was determined the activity of acetylcholinesterase (AChE). The results showed the biotransformation pathway activation due to the increase in hepatic CYP1A and in branchial and hepatic glutathione S-transferase (GST). The antioxidant defense proved to be greatly affected by MA exposure leading to changes, both in gills and liver, in the activities of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), glutathione reductase (GR) and in the content of tripeptide glutathione (GSH). Lipid peroxidation was not detected, but damage to DNA molecule was observed in blood cells. In conclusion, it can be state the lyophilized MA is able to promote changes in the biochemical and genetic parameters of P. lineatus.
As microcistinas são metabólitos secundários produzidos por diferentes espécies de cianobactérias, como a Microcystis aeruginosa (MA). Neste estudo, os efeitos bioquímicos e genéticos de liofilizado de MA foram avaliados para juvenis da espécie de peixe Neotropical Prochilodus lineatus expostos a 1 ou 2 mg L-1 de liofilizado de MA (grupo tratado) ou apenas à água (grupo controle), em testes de toxicidade estáticos, durante 24 e 96 h. As brânquias e o fígado foram usados para as análises das enzimas de biotransformação e defesas antioxidantes, células do sangue e das brânquias para análises genéticas e no cérebro e músculo foi determinada a atividade da acetilcolinesterase (AChE). Os resultados mostraram ativação da via de biotransformação devido ao aumento da atividade da CYP1A hepática e da atividade da glutationa S-transferase (GST) hepática e branquial. As defesas antioxidantes foram muito afetadas pela exposição a MA levando a alterações, tanto no fígado como nas brânquias, na atividade da superóxido dismutase (SOD), da catalase (CAT), da glutationa peroxidase (GPx), glutationa redutase (GR) e no conteúdo do tripeptídeo glutationa (GSH). Apesar dessas alterações a peroxidação lipídica não foi detectada em nenhum dos tecidos, mas danos na molécula de DNA foram observados nas células do sangue. Em conclusão, pode-se afirmar que o liofilizado de MA é capaz de promover alterações em parâmetros bioquímicos e genéticos de P. lineatus.

Introducción. La resistencia fisiológica de las poblaciones naturales de Aedes aegypti a los insecticidas de uso en salud pública, disminuye la eficacia del control químico utilizado para interrumpir la transmisión de dengue durante los brotes y epidemias. Objetivo. Determinar el estado de la susceptibilidad al carbamato propoxur, a los organofosforados adulticidas malatión y fenitrotión y al larvicida temefos, de 13 poblaciones naturales de Ae. aegypti en ocho municipios: una de Bucaramanga y dos de Sabana de Torres en el departamento de Santander; dos de Girardot y dos de La Mesa en Cundinamarca; dos de Villavicencio y dos de Puerto López en el Meta; una de San José del Guaviare, en Guaviare, y una de Florencia en Caquetá. Materiales y métodos. Las pruebas de susceptibilidad consistieron en bioensayos estandarizados de la Organización Mundial de la Salud (OMS), 1981, y botellas de los Centers for Disease Control and Prevention (CDC), 1998. Se aplicaron pruebas colorimétricas para determinar los niveles de enzimas como posibles responsables de la reducción de la susceptibilidad a insecticidas organofosforados y al carbamato propoxur. Resultados. Todas las poblaciones presentaron susceptibilidad a malatión y propoxur. Para el temefos, solo cuatro de las trece localidades evaluadas registraron susceptibilidad. Con respecto a los posibles mecanismos de desintoxicación enzimática, en siete de las once poblaciones naturales se registraron valores superiores al punto de corte para esterasas inespecíficas, solamente en Bucaramanga se presentó un incremento en las enzimas monooxigenasas del citocromo P450, pero en ninguna población se registró acetilcolinestarasa modificada. Conclusiones. La susceptibilidad generalizada a los organofosforados adulticidas evaluados, indica que el malatión, el insecticida más utilizado en Colombia para interrumpir la transmisión del dengue, sigue siendo eficaz. Las particularidades de los resultados de la resistencia fisiológica a insecticidas de las comunas de un mismo municipio, evidenciaron un fenómeno localizado de manera específica en espacio y tiempo.
Introduction. Physiological resistance of natural population of Aedes aegypti to insecticides contribute to the decreased efficacy of chemical control as a main control strategy during dengue outbreaks. Objective. The susceptibility status of Ae. aegypti was assessed for the carbamate propoxur, the adulticide malathion and the larvicide temephos on 13 natural populations of Ae. aegypti immature forms were taken from 8 Colombian localities. These included the following: Bucaramanga (1), Sabana de Torres (2), Girardot (2), La Mesa (2), Villavicencio (2), Puerto López (2), San José del Guaviare (1) and Florencia (1). Materials and methods. Susceptibility tests mainly consisted of the standardized bioassay outlined by WHO (1981) and CDC bottles (1998). Colorimetric tests were undertaken to determine enzyme levels possibly responsible for the reduction of susceptibility to organophosphate and carbamate insecticides. Results. All specimens demonstrated susceptibility to malathion and propoxur insecticides. Four of the 13 populations revealed susceptibility to the temephos larvicide. Seven of 11 populations showed a limited increase in values for nonspecific esterase enzymes. The Bucaramanga population was the only one which showed an increase in the cytochrome P450 monooxygenases enzymes. Neither population was found with modified acetilcolinesterase. Conclusions. The widespread susceptibility to organophosphates used as adulticides indicated that malathion, the most used insecticide in Colombia, remains effective in interrupting the transmission of dengue. Physiological resistance to insecticides occurring in communities of a single township proved to be a localized phenomenon.

Neste trabalho foi avaliada a atividade inibitória da acetilcolinesterase (AChE) pelo método de Ellman, modificado por Rhee, de extratos aquosos e etanólicos de oito plantas utilizadas na medicina popular da região Nordeste do Brasil. O extrato aquoso de E. velutina não apresentou atividade inibitória enquanto o extrato aquoso de Maytenus rigida apresentou baixa atividade inibitória (percentual de inibição de 4%). Detectou-se atividade inibitória moderada com o extrato aquoso de P. piperoides (percentual de inibição de 40 %), enquanto o extrato de V. agnus-castus L. inibiu 74% da atividade da AChE, caracterizando-se como potente atividade inibitória. A avaliação da inibição da AChE com os extratos etanólicos demonstrou que os extratos de Sideroxylon obtusifolium, Erythrina velutina, Vitex agnus-castus, Phoradendron piperoides, Chrysobalanus icaco, Bauhinia cheilantha e Orbignya phalerata não apresentaram atividade inibitória. Baixa atividade inibitória foi observada com os extratos etanólicos de Maytenus rigida (percentual de inibição de 7%) e de Hyptis fruticosa (percentual de inibição de 11%). O extrato etanólico de Moringa oleifera apresentou atividade inibitória moderada, inibindo 47% da atividade dessa enzima. Nenhum dos extratos etanólicos testados apresentou atividade inibitória potente da AChE. Os resultados dos estudos de inibição da acetilcolinesterase permitem concluir que o extrato aquoso de V. agnus-castus L. mostrou-se o mais eficaz quanto a inibição da AChE. Este resultado reforça a necessidade da continuidade do estudo desse extrato, de forma a realizar a partição do extrato e a purificação das frações para isolar a molécula responsável pela inibição observada.
In this study, we evaluated the inhibitory activity against acetylcholinesterase (AChE) according to Ellman's method, modified by Rhee, for ethanol and aqueous extracts from eight plants used in folk medicine in the northeast region of Brazil. E. velutina aqueous extract did not have inhibitory activity, while Maytenus rigida aqueous extract showed low inhibitory activity (percentage of inhibition of 4%). Moderate inhibitory activity was detected for Phoradendron piperoides aqueous extract (percentage of inhibition of 40%), while V. agnus castus L. extract inhibited 74% AChE activity, characterized as a potent inhibitory activity. Evaluation of AChE inhibition by ethanol extracts indicated that the extracts from Sideroxylon obtusifolium, Erythrina velutina, Vitex agnus-castus L., Phoradendron piperoides, Chrysobalanus icaco, Bauhinia cheilantha and Orbignya phalerata did not show inhibitory activity. A low inhibitory activity was observed for ethanol extracts from Maytenus rigida (percentage of inhibition of 7%) and Hyptis fruticosa (percentage of inhibition of 11%). Moringa oleifera ethanol extract showed moderate inhibitory activity, inhibiting 47% of the activity of this enzyme. None of the tested ethanol extracts showed potent inhibitory activity against AChE. Results of the studies of acetylcholinesterase inhibition allow the conclusion that V. agnus-castus L. aqueous extract showed to be most effective in inhibiting AChE. This result reinforces the need for continued study of this extract in order to make the partition and the purification of fractions to isolate the molecule responsible for the observed inhibition.

OBJETIVOS: Evaluar el estado de susceptibilidad a insecticidas piretroides deltametrina y lambdacialotrina y al organoclorado DDT, e identificar los mecanismos bioquímicos asociados con resistencia en 13 poblaciones naturales de Aedes aegypti recolectadas en localidades de Colombia donde el dengue es un grave problema de salud pública. MÉTODOS: Se recolectaron y criaron en condiciones controladas formas inmaduras de diferentes criaderos naturales del vector para cada localidad. Con la generación F2 se realizaron bioensayos utilizando las metodologías OMS 1981 (papeles impregnados) y CDC 1998 (botellas impregnadas). En las poblaciones con mortalidades compatibles con disminución de la susceptibilidad, se midieron los niveles de esterasas no específicas (ENE), oxidasas de función mixta (OFM) y acetilcolinesterasa modificada (ACEM) mediante pruebas colorimétricas. RESULTADOS: Todas las poblaciones del mosquito evaluadas evidenciaron resistencia al organoclorado DDT. En cuanto a los piretroides, se encontró resistencia generalizada a lambdacialotrina pero no a deltametrina. Los mecanismos bioquímicos de resistencia evaluados permitieron encontrar 7 de 11 poblaciones con ENE elevadas y una población con OFM incrementadas. CONCLUSIONES: Se descarta la resistencia cruzada de tipo fisiológico entre el DDT y lambdacialotrina en las poblaciones de A. aegypti evaluadas. La resistencia fisiológica a lambdacialotrina parece asociarse con el incremento de las ENE. El comportamiento diferencial en los niveles de susceptibilidad y los valores enzimáticos entre poblaciones se asociaron con la variabilidad genética y presión de selección química a nivel local.
OBJECTIVES: To assess the susceptibility status of 13 natural populations of Aedes aegypti (collected from sites in Colombia where dengue is a serious public health problem) to the pyrethroids, deltamethrin and lambda-cyhalothrin, and to the organochlorine, DDT, and to identify any biochemical mechanisms associated with resistance. METHODS: Immature forms of the vector were collected from natural breeding spots at each site and then raised under controlled conditions. Using the F2 generation, bioassays were performed using the World Health Organization's 1981 methodology (impregnated paper) and United States Centers for Disease Control and Prevention's 1998 methodology (impregnated bottles). In populations where mortality rates were consistent with decreased susceptibility, levels of nonspecific esterases (NSE), mixed-function oxidases (MFO), and acetylcholinesterase (AChE) were measured using colorimetric tests. RESULTS: All of the mosquito populations that were tested showed resistance to the organochlorine DDT. In the case of the pyrethroids, widespread resistance to lambda-cyhalothrin was found, but not to deltamethrin. Assessing the biochemical resistance mechanisms showed that 7 of the 11 populations had elevated NSE, and one population, increased MFO. CONCLUSIONS: Physiological cross-resistance between DDT and lambda-cyhalothrin in the A. aegypti populations tested was dismissed. Physiological resistance to lambda-cyhalothrin appears to be associated with increased NSE. The differences in susceptibility levels and enzyme values among the populations were associated with genetic variations and chemicals in use locally.

Os agrotóxicos são substâncias usadas para exterminar pragas ou doenças quecausam danos às plantações. Os organofosforados são agrotóxicos responsáveis por 70 por cento das intoxicações ocupacionais e atuam inibindo a enzima acetilcolinesterase por meio da fosforilação do aminoácido serina do sítio ativo. Existem diversos métodos de diagnóstico para monitorar a exposição a estes compostos, tais como: identificação e análise quantitativa de compostos livres noplasma; identificação e análise quantitativa de metabólitos na urina; determinação da atividade de colinesterases no plasma ou no sangue total. Entretanto, estas metodologias possuem algumas limitações, como por exemplo, a falta de especificidade, tecnologias de análise caras e variações interindividuais (sexo, idade, etc.). Uma tentativa de contornar estas limitações na quantificação da atividade de colinesterases é o desenvolvimento de metodologias baseadas na detecção da enzima fosforilada através de técnicas imunoquímicas. Este estudo teve como objetivo desenvolver um método de imunoensaio para o diagnósticorápido da exposição humana aos organofosforados pela detecção deacetilcolinesterase inibida no sangue humano por anticorpos aviários produzidos contra uma sequência de aminoácidos ao redor da serina fosforilada do centro catalítico da enzima. Para isto, foram adquiridos dois decapeptídeos sintéticos correspondentes ao sítio catalítico "não fosforilado" (SOH) e outro com o resíduo de serina fosforilado (SOP). Estes peptídeos foram conjugados com albumina bovina de soro (BSA), injetados em galinhas poedeiras da raça Isa-Brown e anticorpos IgY foram obtidas das gemas dos ovos das galinhas imunizadas. A especificidade dos anticorpos IgY anti-SOH e anti-SOP foi testada por meio de métodos ELISA constatando-se que estes anticorpos reconhecem os peptídeos livres, os peptídeos conjugados com BSA e a própria BSA...

The bivalve shellfish Pinctada radiate is a popular edible food for Alexandria local communities. In an attempt to track the quality of marine water of two different locations along Alexandria coast of Egypt we measured the RNA/DNA ratio and acetyl cholinesterase [AChE] activity in the haemolymph of this organism. The study showed an average ratio for RNA/DNA of 3.3, arranged as follows; 4.34 at El-Max > 2.8 at Miami localities. These results suggest that the coastal water of Alexandria is considered as unpolluted with examined insecticides in this research

The effects of various levels of oxygen saturation and ammonia concentration on NTPDase (ecto-nucleoside triphosphate diphosphohydrolase, E.C. 3.6.1.5) and acetylcholinesterase (AChE, E.C. 3.1.1.7) activities in whole brain of teleost fish (Rhamdia quelen) were investigated. The fish were exposed to one of two different dissolved oxygen levels, including high oxygen (6.5 mg.L-1) or low oxygen (3.5 mg.L-1), and one of two different ammonia levels, including high ammonia (0.1 mg.L-1) or low ammonia (0.03 mg.L-1) levels. The four experimental groups included the following (A) control, or high dissolved oxygen plus low NH3; (B) low dissolved oxygen plus low NH3; (C) high dissolved oxygen plus high NH3; (D) low dissolved oxygen plus high NH3. We found that enzyme activities were altered after 24 h exposure in groups C and D. ATP and ADP hydrolysis in whole brain of fish was enhanced in group D after 24 h exposure by 100 percent and 119 percent, respectively, compared to the control group. After 24 h exposure, AChE activity presented an increase of 34 percent and 39 percent in groups C and D, respectively, when compared to the control group. These results are consistent with the hypothesis that low oxygen levels increase ammonia toxicity. Moreover, the hypoxic events may increase blood flow by hypoxia increasing NTPDase activity, thus producing adenosine, a potent vasodilator.(AU)
No presente estudo, avaliou-se o efeito de diferentes níveis de saturação de oxigênio e amônia sobre a atividade das enzimas NTPDase (ecto-nucleosídeo trifosfato difosfohidrolase, E.C. 3.6.1.5) e acetilcolinesterase (AChE, E.C. 3.1.1.7) em encéfalo total de jundiás (Rhamdia quelen). Os peixes foram expostos a diferentes níveis de oxigênio dissolvido e amônia, níveis altos de oxigênio (6,5 mg/L) ou baixos de oxigênio (3,5 mg/L), e níveis altos de amônia (0,1 mg/L) ou baixos de amônia (0,03 mg/L). Os peixes foram divididos em quatro diferentes grupos: (A) controle ou alto nível de oxigênio dissolvido mais baixo nível de NH3; (B) baixo nível de oxigênio dissolvido mais baixo nível de NH3; (C) alto nível de oxigênio dissolvido mais alto nível de amônia; (D) baixo nível de oxigênio dissolvido mais alto nível de NH-3. As atividades de ambas as enzimas nos grupos C e D somente foram alteradas após 24 horas de exposição. A hidrólise do ATP e ADP em encéfalo total de jundiás foi aumentada após 24h de exposição para 104 por cento e 155 por cento no grupo D quando comparado ao grupo controle, respectivamente. A atividade da AChE apresentou após 24h de exposição um aumento de 37 por cento no grupo C e 27 por cento no grupo D, ambos comparados ao grupo controle. Os resultados obtidos corroboram com a hipótese que baixos níveis de saturação de oxigênio aumentam a toxicidade da amônia. Além disso, os eventos de hipóxia podem aumentar o fluxo sanguíneo, e este evento aumenta a atividade da NTPDase produzindo adenosina, um potente vasodilatador(AU)

OBJETIVOS: Diagnosticar la resistencia a insecticidas y sus mecanismos en Anopheles albimanus del sur de la Península de Yucatán (PY), México. MATERIAL Y MÉTODOS: La F1 de An. albimanus colectados durante noviembre-diciembre de 2005 en seis localidades de los municipios Othón P. Blanco en Quintana Roo y Calakmul en Campeche, fue sometida a pruebas de susceptibilidad con deltametrina, DDT, pirimifos metil y bendiocarb, y a pruebas bioquímicas para calcular los niveles de las enzimas involucradas en la resistencia. RESULTADOS: An. albimanus fue resistente al DDT y a deltametrina en las seis localidades con niveles elevados de glutatión S-transferasas (GSTs), monooxigenasas y esterasas, y a pirimifos metil en La Unión con una alta frecuencia de la acetilcolinesterasa (AChE) alterada. CONCLUSION: Las poblaciones de An. albimanus colectadas al sur de la PY son resistentes al DDT y deltametrina, y en La Unión además al pirimifos metil, con mecanismos basados en la AChE alterada para el pirimifos metil, GST para DDT, y monooxigenasas y esterasas para piretroides. Los resultados del presente estudio tienen importantes consecuencias prácticas para el control químico de An. albimanus en el sur de la PY.
OBJECTIVE: To diagnose susceptibility levels and insecticide resistance mechanisms in Anopheles albimanus from the southern Yucatan Peninsula (YP), Mexico. MATERIAL AND METHODS: F1 generation of An. albimanus females, collected from November to December 2005 in six villages in the Othon P. Blanco municipality in Quintana Roo and the Calakmul municipality in Campeche, were exposed to deltamethrin, DDT, pirimiphos-methyl and bendiocarb in susceptibility tests, as well as to biochemical assays in order to calculate the enzyme levels related to insecticide resistance. RESULTS: High levels of DDT and deltamethrin resistance were found in An. albimanus collected from the six villages, and a high resistance to pirimiphos-methyl was found in those from La Union, Quintana Roo. Biochemical assays showed high levels of glutathione S-transferase (GST), cytochrome P450 and esterases (with pNPA substrate) in all villages. The frequency of An. albimanus with altered acetylcholinesterase (AChE) was high in La Union (33 percent). CONCLUSIONS: The An. albimanus populations collected in the south of the YP are resistant to DDT and deltamethrin, whereas resistance to pirimiphos-methyl was significant only in those collected from La Union. The mechanisms explaining this resistance are based on high concentrations of GST, cytochrome P450 and esterasas, the former being responsible for DDT metabolism and the others for pyrethroid metabolism. The altered AChE was the mechanism correlated to pirimiphos-methyl resistance in La Union. The results of the present study have important practical consequences for the chemical control of An. albimanus in the south of the YP.

The bottle bioassay measuring the time-mortality rate is a simplified procedure for detecting insecticide resistance. It can be used with a biochemical microplate assay to identify the mechanism involved. This integrated approach was used to detect temephos resistance in Aedes aegypti from Nonthaburi (lowest use) and Roi Et (highest use). Ae. aegypti BKK1 laboratory strain was used as the susceptible reference strain. The appropriate concentration of insecticide for bottle bioassay was determined empirically for Ae. aegypti BKK1 strain and found to be in the range of 800-1,050 microg/bottle. The time-mortality rate at 800 microg/bottle was 170 +/- 8.66 minutes, significantly different from the time-mortality rates in the 850, 900, 950, and 1,050 microg/bottle (p = 0.008) concentrations, which were 135 +/- 15.00, 140 +/- 8.66, 135 +/- 15.00, and 125 +/- 8.66 minutes, respectively. The cut-off concentration selected for resistance detection was 850 microg/bottle. The time-mortality rate for the Roi Et strain was 382 +/- 26.41 minutes, significantly higher than the Nonthaburi (150 +/- 25.10 minutes) and BKK1 strains (145 +/- 20.49 minutes) (p < 0.001). The temephos resistance ratio (RR100) for the Ae. aegypti Roi Et strain was 2.64-fold higher at lethal time (LT100) than for the reference Ae. aegypti BKK1 strain. The mean optical density (OD) value from the biochemical microplate assay for the non-specific esterase of the Roi Et strain was higher than the mean OD for the non-specific esterase of both the Nonthaburi and BKK1 strains. Insensitive acetylcholinesterase was not found to be responsible for the resistance in the field-collected mosquitos. This study suggests that esterase detoxification is the primary cause of resistance in the Ae. aegypti population from Roi Et. Both the bottle bioassay and the biochemical microplate assay were proven to be promising tools for initial detection and field surveillance for temephos resistance.

The present study analyzes the ectopic development of the rat skeletal muscle originated from transplanted satellite cells. Satellite cells (10(6) cells) obtained from hindlimb muscles of newborn female 2BAW Wistar rats were injected subcutaneously into the dorsal area of adult male rats. After 3, 7, and 14 days, the transplanted tissues (N = 4-5) were processed for histochemical analysis of peripheral nerves, inactive X-chromosome and acetylcholinesterase. Nicotinic acetylcholine receptors (nAChRs) were also labeled with tetramethylrhodamine-labeled alpha-bungarotoxin. The development of ectopic muscles was successful in 86 percent of the implantation sites. By day 3, the transplanted cells were organized as multinucleated fibers containing multiple clusters of nAChRs (N = 2-4), resembling those from non-innervated cultured skeletal muscle fibers. After 7 days, the transplanted cells appeared as a highly vascularized tissue formed by bundles of fibers containing peripheral nuclei. The presence of X chromatin body indicated that subcutaneously developed fibers originated from female donor satellite cells. Differently from the extensor digitorum longus muscle of adult male rat (87.9 ± 1.0 æm; N = 213), the diameter of ectopic fibers (59.1 æm; N = 213) did not obey a Gaussian distribution and had a higher coefficient of variation. After 7 and 14 days, the organization of the nAChR clusters was similar to that of clusters from adult innervated extensor digitorum longus muscle. These findings indicate the histocompatibility of rats from 2BAW colony and that satellite cells transplanted into the subcutaneous space of adult animals are able to develop and fuse to form differentiated skeletal muscle fibers.

BACKGROUND: A diagnosis of Hirschsprung disease requires the demonstration of acetylcholinesterase fibres on frozen sections obtained from snap frozen biopsies of the rectum. This histochemical technique is generally not available in laboratories in developing countries. We improvised on the methodology of tissue preservation to make the staining technique more user-friendly, economical and reliable in demonstrating acetylcholinesterase activity in fresh rectal mucosal biopsies for the diagnosis of Hirschsprung disease. METHODS: Between June 1999 and May 2002 fresh rectal biopsies from 40 suspected cases of Hirschsprung disease were processed for routine frozen section (not snap frozen by liquid nitrogen) and stained by the Karnovsky and Roots method. These sections were assessed for the staining pattern of acetylcholinesterase fibres. The thickness of the nerve fibres and muscularis mucosa was assessed morphometrically. These were compared with biopsies obtained from 6 age-matched controls undergoing surgery for unrelated complaints. RESULTS: The sections stained for acetylcholinesterase by this improvised method of tissue fixation were good and crisp. A definite diagnosis of Hirschsprung disease was made in 25 cases and intestinal neuronal dysplasia in 1. The remaining 14 cases showed an equivocal staining pattern with no hypertrophic nerve bundles, thus excluding a diagnosis of Hirschsprung disease. The mean thickness of the submucosal nerve trunks measured in these enzyme-stained sections was found to be inversely proportional to the mean thickness of the muscularis mucosa. CONCLUSION: Our study on cryostat-cut sections suggests an inverse relationship between the thickness of the muscularis mucosa and the calibre of the nerve trunk--thinner the nerve trunk, thicker the muscularis mucosa and vice versa. Also, routine frozen sections, instead of snap frozen ones taken from a fresh rectal biopsy and stained by the Karnovsky and Roots method for acetylcholinesterase activity, are reliable for the diagnosis of Hirschsprung disease and are within the capability of a simple histopathology laboratory in a developing country.
