ABSTRACT
Abstract This study was performed under laboratory conditions to identify isolates of the fungus Beauveria spp. that can control Polyphagotarsonemus latus in the greenhouse and field. Thirty Beauveria spp. isolates were tested by spraying 1 mL conidia (1 × 108 conidia/mL) on pepper leaf discs containing 15 mites. Evaluations were performed on the 3rd and 6th day post application by counting the number of dead mites. Vegetative growth and conidial production were measured from the selected isolates, and bioassays were conducted in the greenhouse on bean seedlings in plastic pots. The isolate Unioeste 53 was selected, and a conidial suspension (1 × 108 conidia/mL) was applied with a backpack sprayer. The evaluation consisted of pre- and post-treatment counts of the number of live mites on ten leaflets in both the plots treated with the fungus and control plots, and the same procedure was followed for the field experiment. In the laboratory, the Unioeste 53 isolate resulted in total and confirmed mortality rates of 70% and 57.7%, respectively. In the greenhouse, the population decreased by 76.71% by the 16th day after application. In the field, the population decreased by 66% by the 12th day after application, demonstrating the potential of this fungus for mite management.
Resumo Este trabalho teve por objetivo selecionar isolados do fungo Beauveria bassiana em condições de laboratório com potencial de uso em casa de vegetação e campo no controle do ácaro branco. Foi realizada uma seleção com 30 isolados de Beauveria spp. através de pulverização direta de conídios (1 × 108 conídios/mL) sobre discos foliares de pimenta contendo 15 ácaros. As avaliações foram realizadas no terceiro e sexto dia contando-se o número de mortos, e confirmação do patógeno em câmara úmida. Parâmetros de crescimento vegetativo e produção de conídios foram avaliados. No bioensaio em casa de vegetação foram preparados vasos com plantas de feijão e fez-se a aplicação do Unioeste 53 (1 × 108 conídios/mL), utilizando pulverizador costal, a avaliação constou da contagem prévia e posterior à aplicação do número de ácaros vivos em 10 folíolos, tanto nas parcelas destinadas ao tratamento com fungo, quanto na testemunha. A fase de campo seguiu os mesmos padrões, porém, com área experimental total de 225 m2, com oito parcelas de 10,24 m2, sendo 4 testemunhas e 4 onde foi aplicado o isolado Unioeste 53 seguindo metodologia de aplicação e avaliação já descritas para casa de vegetação. Em laboratório o isolado Unioeste 53 causou mortalidade total de 70% e 57,7% de mortalidade confirmada. Em casa de vegetação, apresentou redução da população de 76,71% 16 dias após aplicação, já em campo, a redução da população foi de 66% após 12 dias da aplicação, demonstrando o potencial do ácaro pelo fungo.

Este trabajo tuvo por objetivo aislar, identificar y caracterizar morfológicamente hongos entomopatógenos presentes en suelos cultivados con yerba mate (Ilex paraguariensis St. Hil.). Se realizó una prospección de hongos entomopatógenos nativos a partir de 40 muestras de suelos cultivados con yerba mate ubicados en la provincia de Misiones, República Argentina, desde mayo de 2008 hasta junio de 2010. Las muestras de suelo se obtuvieron en cercanías de plantas de yerba mate, se aplicó la técnica de dilución seriada en medios de cultivos selectivos. Después de obtenidos los cultivos puros, los hongos fueron caracterizados macroscópica y microscópicamente, y se los identificó mediante el uso de claves taxonómicas específicas. Se aislaron 29 cepas fúngicas pertenecientes a las especies Beauveria bassiana (n = 17), Metarhizium anisopliae (n = 2) y Purpureocillium lilacinum (n = 10)
This study aimed to morphologically isolate, identify and characterize entomopathogenic fungi present in soils cultivated with Paraguay tea (Ilex paraguariensis). A survey of native entomopathogenic fungi was conducted from 40 soil samples grown with Paraguay tea in the province of Misiones, Argentina, from May 2008 to June 2010. The soil dilution plate methodology on selective culture media was used to isolate microorganisms. Taxonomic identification was performed using macroscopic and microscopic characters and specific keys. Twenty nine strains, belonging to the species Beauveria bassiana (n = 17), Metarhizium anisopliae (n = 2) and Purpureocillium lilacinum (n = 10) were isolated and identified

The entomopathogenic fungus Beauveria bassiana (Balsamo 1835) Vuillemin is an effective alternative control agent against some agricultural pests and biological vectors of important diseases such as Chagas disease. In this work we studied an isolate of Beauveria bassiana from of the town of San Antonio Rayón, Puebla, Mexico and its entomopathogenic effects on Meccus pallidipennis (Stal 1872). Phylogenetic analysis using molecular comparison of the ITS and EF1α genes, showed that the resulting cladogram places the BUAP 04 strain with a relationship closer to the AFAO 9-6 strain, within the diversity of the B. bassiana sensu lato group. Although there was the possibility that BUAP 04 strain was a direct descendant of strains used in campaigns of biologic control, molecular study allowed us to recognize that it was a different fungus due to numerous inserts. A strain isolated from a T. dimiata was evaluated for pathogenicity against another triatoma (Meccus pallidipennis) species obtaining an LC50 of 4.16 x 10(6) spores/mL, confirming that the BUAP 04 strain is virulent for M. pallidipennis and could be a good prospect for formulations to control M. pallidipennis.

Se evaluó a concentración de 1x10(8) conidias/mL por vía oral en ratones albinos NMRI, la patogenicidad del aislamiento fúngico LF14 de Beauveria bassiana, el cual posee una elevada virulencia hacia Rhodnius prolixus y Triatoma maculata (Triatominae), vectores de enfermedad de Chagas en Venezuela. Se hicieron observaciones clínicas diarias. Se estimó el aclaramiento fúngico mediante examen directo y cultivo de muestras de heces, y de infectividad con la toma de muestras de tejidos y estudio histopatológico. No se registró muertes ni alteraciones clínico-patológicas, y el comportamiento en todos los animales fue activonormal. El peso promedio corporal, tanto en los animales expuestos a LF14 B. bassiana como los controles, se incrementó con el tiempo, siendo sólo las diferencias entre sexos estadísticamente significativas (P<0,0000). Se obtuvieron conidias viables en heces hasta el día 7 después del consumo del hongo. No se detectó anormalidades en la inspección anatomopatológico ni conidias germinadas en ninguno de los órganos estudiados, sin ninguna reacción tisular patológica, sugiriendo que no existe evidencia de multiplicación fúngica. Los resultados obtenidos permiten concluir que el aislamiento LF14 de B. bassiana no es patogénico cuando se administró por vía oral en ratones albinos NMRI.
This study evaluated concentrations of 1x10(8) conidia/mL orally administered in white mice (NMRI strain, males and females), pathogenicity of a native isolate LF14 of Beauveria bassiana (Fungi: Ascomycota), highly virulent to Rhodnius prolixus and Triatoma maculata (Triatominae), vectors of Chagas disease in Venezuela. Clinical evaluations were done daily. Clearance of fungus in the faeces was estimated by direct examination and culturing, and infectivity by performing mycological and histopathological tests.Animals showed no discernible pathological changes or death and had 100% active-normal behaviour. In all the mice groups, including control ones, there was an increase in the mean weight gained through time, with statistically significance difference between the sexes (P<0,0000). Viable conidia were observed in faeces until 7 days after consuming the fungus. Anatomopathological changes or germinated conidia were not detected in analysed organs, with normal tissue reactions, suggesting no evidence for fungal multiplication. Results obtained allowed us to conclude that isolate LF14 B. bassiana orally administered is safe and non pathogenic in white mice (NMRI strain).

Beauveria bassiana genetic diversity and ability to synthesize quercetin 2,3-dioxygenase (quercetinase) were analyzed. B. bassiana isolates, obtained from Brazilian soil samples, produced quercetinase after induction using 0.5 g/L quercetin. B. bassiana ATCC 7159 (29.6 nmol/mL/min) and isolate IP 11 (27.5 nmol/ml/min) showed the best performances and IP 3a (9.5 nmol/mL/min) presented the lowest level of quercetinase activity in the culture supernatant. A high level of polymorphism was detected by random amplified polymorphic DNA (RAPD) analysis. The use of internal-transcribed-spacer ribosomal region restriction fragment length polymorphism (ITS-RFLP) did not reveal characteristic markers to differentiate isolates. However, the ITS1-5.8S-ITS2 region sequence analysis provided more information on polymorphism among the isolates, allowing them to be clustered by relative similarity into three large groups. Correlation was tested according to the Person's correlation. Data of our studies showed, that lower associations among groups, level of quercetinase production, or geographical origin could be observed. This study presents the production of a novel biocatalyst by B. bassiana and suggests the possible industrial application of this fungal species in large-scale biotechnological manufacture of quercetinase.

The activity of Metarhizium anisopliae and Beauveria bassiana towards eggs and larvae of Tuta absoluta (Meyrick) was evaluated. Our data showed that the isolates were pathogenic to both developmental stages tested and the eggs were more susceptible than the 1st instars. The isolates URPE-6 and URPE-19 of M. anisopliae were more pathogenic to eggs and larvae, respectively. The compatibility of these two isolates with the insecticides chlorfenapyr, spinosad, indoxacarb, abamectin, and neem were evaluated. Spinosad and indoxacarb were compatible with the two M. anisopliae isolates in all tested concentrations. At the average recommended concentration, chlorfenapyr was compatible to URPE-6 and abamectin to UFPE-19. The use of entomopathogenic fungi associated with compatible insecticides may be a useful alternative to control T. absoluta.

O bicudo-do-algodoeiro, Anthonomus grandis, é considerado uma das principais pragas da cultura do algodão, demandando uma grande quantidade de inseticidas para o seu controle. O objetivo do presente trabalho foi avaliar isolados deBeauveria bassianapara o controle deA. grandis. Para tanto, foram testados nove isolados provenientes de diferentes hospedeiros na concentração de 1 x 108 conídios/mL, em condições de laboratório. Todos os isolados testados foram patogênicos ao bicudo-do-algodoeiro, causando entre 50 e 85% de mortalidade confirmada. Os tempos letais para matar 50% dos insetos (TL50) variaram entre 2,30 e 8,66 dias. O isolado IBCB 241 destacou-se como o mais virulento, apresentando a maior percentagem de mortalidade confirmada e o menor TL50, podendo assim ser considerado um promissor agente de controle biológico para o bicudodo-algodoeiro A.grandis.
The cotton boll weevil, Anthonomus grandis, is one of the most important pests of the cotton crop, requiring an elevated quantity of insecticides for its control.The objective of this work was to evaluate the effectiveness ofBeauveria bassianastrains for A. grandis control. In this assay, 9 strains from different hosts were tested in the concentration of 1 x 108 conidia/mL, under laboratory conditions. All strains evaluated were pathogenic to the cotton boll weevil, causing between 50 and 85% confirmed mortality. The lethal time to kill 50% of insects (LT50) ranged between 2.30 and 8.66 days. The IBCB 241 strain stood out as the most virulent, presenting the highest percentage of confirmed mortality and the shortest LT50, and can therefore be considered as a promising agent for the biological control of the cotton boll weevil A. grandis.

O presente estudo foi conduzido com o objetivo de selecionar isolados do fungo entomopatogênico Beauveria bassiana com potencial patogênico para utilização no controle do carrapato Rhipicephalus (Boophilus) microplus. Numa primeira etapa, foi avaliada a eficiência de trinta isolados na concentração de 5 × 10(8) conídios.mL-1. Destes, oito (IBCB01, IBCB02, IBCB07, IBCB17, IBCB21, IBCB74, IBCB149, IBCB165) apresentaram eficiência entre 90 e 99%; treze (IBCB03, IBCB14, IBCB16, IBCB24, IBCB95, IBCB97, IBCB102, IBCB141, IBCB146, IBCB147, IBCB150, IBCB154, IBCB157), indicaram percentuais de eficiência entre 80 e 89,5%; seis (IBCB47, IBCB75, IBCB84, IBCB145, IBCB161, IBCB164), entre 70 e 79% e apenas dois, (IBCB13 e IBCB143) revelaram-se pouco virulentos com porcentagens abaixo de 70%. Na segunda etapa do trabalho, foram analisados comparativamente dados de mortalidade acumulada dos cinco melhores isolados obtidos na primeira fase (IBCB01, IBCB07, IBCB21, IBCB66, IBCB165). Analisando-se os resultados obtidos in vitro, pode-se imputar que os isolados IBCB21 e IBCB66 são os que apresentam maior potencial para utilização a campo, tendo em vista o controle de R. (B.) microplus. Os isolados selecionados, na primeira fase, também foram testados quanto à potencialidade de produção massal em meio de arroz pré-cozido. A melhor produção massal foi obtida com o isolado IBCB66.
This study was carried out to select isolates of the entomopathogenic fungus Beauveria bassiana with pathogenic potential to control the Rhipicephalus (Boophilus) microplus tick. The effectiveness of thirty isolates was first tested at a concentration of 5 × 10(8) conidia.mL-1. Of these, eight were evaluated (IBCB01, IBCB02, IBCB07, IBCB17, IBCB21, IBCB74, IBCB149, IBCB165) and showed an effectiveness between 90 and 99%; thirteen (IBCB03, IBCB14, IBCB16, IBCB24, IBCB95, IBCB97, IBCB102, IBCB141, IBCB146, IBCB147, IBCB150, IBCB154, IBCB157) between 80 and 89,5%; six (IBCB47, IBCB75, IBCB84, IBCB145, IBCB161, IBCB164) between 70 and 79%, and only two (IBCB13 and IBCB143) had lower pathogenicity (70% or below). In the second step of the study, the five more effective strains in the first phase of the experiment (IBCB01, IBCB07, IBCB21, IBCB66, IBCB165) were analyzed comparatively. Based on in vitro results, it can be concluded that IBCB66 and IBCB21 are the isolates with higher potential for field control of R. (B.) microplus. IBCB01, IBCB07, IBCB21, IBCB66 e IBCB165 isolates were submitted to a conidial production test using a rice-based substrate. The best mass production of the entomopathogenic fungus was obtained with the IBCB66 strain.

O presente trabalho teve por objetivo avaliar a patogenicidade e a virulência do isolado IBCB66 de Beauveria bassiana para larvas de Rhipicephalus (Boophilus) microplus. O isolado IBCB66 foi utilizado como padrão, com a finalidade de determinar a CL50 (Concentração Letal), CL90, TL50 (Tempo Letal) e TL90. O isolado IBCB66 foi testado em seis concentrações diferentes (5 × 10(6), 10(7), 5 × 10(7), 10(8), 5 × 10(8) e 10(9)) para determinar a porcentagem de mortalidade. A mortalidade total de larvas foi observada 18 dias após o início do teste no grupo tratado com 5 × 10(9) conídios.mL-1. A análise de Probit dos dados consignados apontou a CL50 e CL90 concentrações de 3 × 10(7) e 5 × 10(8) conídios.mL-1 e para TL50 e TL90, foram 10 e 16 dias, respectivamente.
The objective of this research was to evaluate the pathogenicity and the virulence of the IBCB66 isolate of Beauveria bassiana on infected larvae of Rhipicephalus (Boophilus) microplus. The IBCB66 fungus strain was used as standard isolates of B. bassiana against R. (B.) microplus larvae. The larval bioassay tests using the IBCB66 isolate were carried out to determine the (Lethal Concentration) LC50, LC90, (Lethal Time) LT50 and LT90. The IBCB66 fungus strain was tested at six different concentrations (5 × 10(6), 10(7), 5 × 10(7), 10(8), 5 × 10(8) and 10(9) conidia.mL-1) to determine the percentage of larval mortality. In addition, a Probit analysis was also performed. Total larval mortality was observed eighteen days after the beginning of the test in the group treated with 5 × 10(9) conidia.mL-1. The LC50 and LC90 were 3 × 10(7) and 5 × 10(8) respectively and the LT50 and LT90 were 10 and 16 days.

Foram selecionados 20 equídeos naturalmente infestados por Anocentor nitens, dos quais seus pavilhões auriculares foram submetidos a quatro tratamentos distintos: gel associado a Beauveria bassiana, apenas gel, apenas B. bassiana e um grupo-controle. Procederam-se à contagem e ao registro do número total de fêmeas adultas (>3mm) em cada pavilhão auricular, nos dias 0, 1, 4, 7, 11, 14, 18, 21, 25 e 28, para cálculo do percentual de controle. Foram coletadas e mantidas em laboratório 20 fêmeas pertencentes a cada tratamento, para cálculo dos períodos de pré-postura, postura, incubação e eclosão, e peso médio das posturas. No grupo tratado com o composto gel associado a B. bassiana, observou-se percentual de controle maior que 50 por cento, entre o 4º e o 25º dia, assim como decréscimo progressivo do peso médio da postura das fêmeas coletadas no período entre o 7º e 14º dia. Para os outros tratamentos, o percentual de controle manteve-se abaixo de 20 por cento, e o peso da postura não apresentou diferença significativa. Os percentuais de controle, assim como as reduções no peso médio da postura, sugerem que a utilização do composto gel associado a B. bassiana potencializou a virulência do entomopatógeno.
Twenty horses naturally infected by Anocentor nitens were selected and their auricular pinna were submitted to four treatments: gel associated with Beauveria bassiana, gel only, B. bassiana only, and a control group. The count and the register of the total number of grown up tick females (>3mm) were performed in each auricular pinna on days 0, 1, 4, 7, 11, 14, 18, 21, 25, and 28 for the calculus of percentage of control. Twenty females from each treatment were collected and maintained in laboratory, to calculate the periods of pre-oviposition, oviposition, incubation, and hatching and mean weight of oviposition. In the group treated with the gel associated to B. bassiana, it was observed a control percentage higher than 50 percent from the 4º to the 25º day, such as a progressive decrease of mean weight of the ovipositions in the period from the 7º to 14º day. For the other treatments, the control percentage was kept below 20 percent and the oviposition weight showed no significant difference among the treatments. The control percentages, such as reductions in mean weight of oviposition, suggest that the use of gel associated with B. bassiana potentialized the virulence of the entomopathogen.

A Brazilian isolate of Beauveria bassiana (CG425) that shows high virulence against the coffee berry borer (CBB) was examined for the production of subtilisin-like (Pr1) and trypsin-like (Pr2) cuticle-degrading proteases. Fungal growth was either in nitrate-medium or in CBB cuticle-containing medium under both buffered and unbuffered conditions. In unbuffered medium supplemented with cuticle, the pH of cultures dropped and Pr1 and Pr2 activities were detected in high amounts only at a pH of 5.5 or higher. In buffered cultures, Pr1 and Pr2 activities were higher in medium supplemented with cuticle compared to activities with nitrate-medium. The Pr1 and Pr2 activities detected were mostly in the culture supernatant. These data suggest that Pr1 and Pr2 proteases produced by strain CG425 are induced by components of CBB cuticle, and that the culture pH influences the expression of these proteases, indicating the occurrence of an efficient mechanism of protein secretion in this fungus. The results obtained in this study extend the knowledge about protease production in B. bassiana CG425, opening new avenues for studying the role of secreted proteases in virulence against the coffee berry borer during the infection process.
O isolado brasileiro de Beauveria bassiana (CG425) que apresenta alta virulência contra a broca do café (CBB) foi analisado quanto à produção de proteases degradadoras de cutícula, tipo-subtilisina (Pr1) e tipo-tripsina (Pr2). O crescimento fúngico foi realizado em meio contendo nitrato e em meio contendo cutícula da broca em condições de pH tamponado e não tamponado. Em meio não tamponado, suplementado com cutícula, o pH da cultura caiu e as atividades de Pr1 e Pr2 foram detectadas somente em valores de pH igual ou superior a 5,5. Em culturas tamponadas, as atividades Pr1 e Pr2 foram superiores em meio suplementado com cutícula, comparativamente as atividades em meio contendo nitrato. As atividades Pr1 e Pr2 ocorreram predominantemente no sobrenadante de cultivo. Os dados obtidos sugerem que Pr1 e Pr2 produzidas pelo isolado CG425 são induzidas por componentes da cutícula da broca do café (CBB), e que o pH da cultura influencia a expressão destas proteases, indicando a ocorrência de um mecanismo eficiente de secreção por este fungo. Os resultados obtidos neste estudo aumentam o conhecimento a respeito da produção de proteases por B. bassiana CG425, abrindo novos caminhos para o estudo do papel de proteases na virulência contra a broca do café durante o processo de infecção.

Microbial biotransformations constitute an important alternative as models for drug metabolism study in mammalians and have been used for the industrial synthesis of chemicals with pharmaceutical purposes. Several microorganisms with unique biotransformation ability have been found by intensive screening and put in commercial applications. Ten isolates of Beauveria sp genus filamentous fungi, isolated from soil in the central Brazil, and Beauveria bassiana ATCC 7159 were evaluated for their capability of quercetin biotransformation. Biotransformation processes were carried out for 24 up to 96 hours and monitored by mass spectrometry analyses of the culture broth. All strains were able to metabolize quercetin, forming mammalian metabolites. The results were different from those presented by other microorganisms previously utilized, attrackting attention because of the great diversity of reactions. Methylated, sulphated, monoglucuronidated, and glucuronidated conjugated metabolites were simultaneously detected.
Biotransformações microbianas constituem uma alternativa importante como modelo para o estudo do metabolismo de medicamentos em mamíferos e são empregadas em processos sintéticos industriais com propósitos farmacêuticos. Diversos microrganismos com potencial para biotransformação têm sido encontrados através de screening intensivo e aplicados comercialmente. Dez cepas de fungos filamentosos do gênero Beauveria, isolados na região central do Brasil, e a cepa Beauveria bassiana ATCC 7159 foram avaliadas quanto à capacidade de biotransformação da quercetina. As reações de biotransformações foram realizadas por um período de 24 a 96 horas, e monitoradas através de espectrometria de massas do meio reacional. Todas as cepas foram capazes de metabolizar a quercetina, formando metabólitos encontrados nos mamíferos. Os resultados foram diferentes dos resultados apresentados por outros microrganismos utilizados anteriormente e chamam a atenção devido à diversidade das reações evidenciadas. Diferentes metabólitos conjugados foram detectados simultaneamente: metilados, sulfatados, monoglicuronados, e glicuronados.

Este trabalho teve por objetivo selecionar isolados de fungos entomopatogênicos, visando sua utilização no controle do cascudinho, Alphitobius diaperinus (Panzer). Larvas e adultos foram inoculados com suspensões de conídios de 99 isolados de Beauveria bassiana (Bals.) Vuill. e Metarhizium anisopliae (Metsch.) Sorok. em concentrações variáveis de 10(5) a 10(9) conídios/ml. Avaliou-se também o crescimento vegetativo em meio-de-cultura, produção de conídios em colônias, arroz e em cadáveres de larvas e adultos do inseto. Os isolados de B. bassiana foram mais eficientes que os isolados de M. anisopliae e as larvas foram mais suscetíveis que os adultos. Os isolados UNIOESTE 04 e UNIOESTE 02, ambos obtidos em cadáveres de cascudinho, foram mais eficientes. Além disso, UNIOESTE 04 apresentou elevada produção em diferentes meios-de-cultura, evidenciando o seu potencial para aplicação em aviários no controle do cascudinho.
