PURPOSE: To evaluate the effects of mesenchymal stem cells (MSC) from eight mice C57BL/6 gfp+ bone marrows expanded in cultures associated with platelets rich plasma (PRP) deriving from another eight mice, in the repair of critical defects in calvarial bone produced in twenty-four adult isogenic mice C57BL/6. METHODS: The animals were submitted to a cranial defect of 6.0mm in diameter and divided into two equal experimental groups. Control group did not receive treatment and the treated group received a MSC pellet containing 1.0 x 10(7) cells/mL associated with 50.0µL of plasma gel containing 1.0 x 10(9) autologous platelets within the defect. RESULTS: In the treated group was observed process of angiogenesis and bone repair better than control group. CONCLUSION: Mesenchymal stem cells derived from bone marrow of C57BL/6 gfp+ mice associated with PRP gel applied in bone critical defects produced in calvarial contributes positively to the process of bone repair.

OBJETIVO: Avaliar os efeitos da associação das células-tronco mesenquimais (MSC) oriundas da medula óssea de oito camundongos jovens C57BL/6 gfp+ e expandidas em culturas, com Plasma Rico em Plaquetas (PRP) provenientes de outros oito camundongos, na reparação de defeitos críticos confeccionados em calvária de 24 camundongos adultos C57BL/6. MÉTODOS: Os animais foram submetidos a um defeito craniano de 6,0mm de diâmetro e separados em dois grupos experimentais iguais. O grupo controle não recebeu tratamento e no grupo tratado foi administrado, no interior do defeito, pellet de MSC contendo 1,0 x 10(7) células/mL associado com 50,0µL de plasma em gel autólogo contendo 1,0 x 10(9) plaquetas. RESULTADOS: No grupo tratado verificou-se processo de angiogênese e reparação óssea superior ao grupo controle. CONCLUSÃO: A associação das células-tronco mesenquimais (MSC) derivadas da medula óssea de camundongos C57BL/6 gfp+ com gel de PRP aplicadas em defeitos ósseos críticos confeccionadas em calvária de camundongos C57BL/6 jovens, contribuiu positivamente para o processo de reparação óssea.

Mesenchymal stem cells [MSCs] have been studied and applied extensively because of their ability to self-renew and differentiate into various cell types. To isolate, increase the expansion rate and define rat bone marrow MSCs by immunohistochemical and electron microscopic studies. Twenty adult male albino rats were used and divided equally into two groups. The bone marrow was harvested and flushed out from the long bones of each group. In group 1, the cells were cultured using complete medium containing Dulbecco's modified Eagle's medium, 1% antibiotics, and 10% fetal bovine serum. In group 2, the cells were cultured in complete medium supplemented with 8% fetal bovine serum and 2% autologous rat serum. When the primary culture became nearly confluent, the adherent cells were subcultured. After 12 days from the primary culture, aliquots of the cells were prepared for Giemsa stain, transmission electron microscopy, and immunohistochemical staining for CD44, CD105, and CD34. The adherent cells in both groups were heterogeneous in appearance, and most of them were spindle or star-shaped with vesicular nuclei. Some cells were binucleated. The MSCs in group 2 reached confluency more rapidly than those in group 1. After passaging of group 2, the adherent cells became more homogeneously fibroblast-like in appearance. Immunostaining of MSCs revealed a positive reaction for CD44 and CD1 05 and a negative reaction for CD34. Ultrastructural examination revealed that the native MSCs appeared with many pseudopodia, the nucleus was eccentric, and the inner zone of the cytoplasm was rich in free ribosomes, many mitochondria, few rough endoplasmic reticulum, with obvious Golgi complex, and some lysosomes. Some MSCs showed no pseudopodia with central nucleus. Others appeared with two euchromatic elliptical nuclei and a nucleolus in one of them. MSCs can be easily purified, cultured, and expanded more rapidly using autologous rat serum

A rare case of Behcet's disease associated with myelodysplastic syndrome (MDS) is described. A 50-year-old Korean female suffering recurrent oral ulcer, genital ulcer, fatigue, arthralgia in both knees and fever was diagnosed as Behcet's disease. The findings of bone marrow aspirates were consistent with refractory anemia, a subtype of myelodysplastic syndrome. Chromosomal analysis of bone marrow cells revealed 46,XX,-8,-20,+der(8)t(8;20)(p23;p10),+der(8) t(8;20)(p23;q10)[30]. The chromosomal changes found in this patient were different from those of previous reports, which mostly revealed trisomy 8. If anemia, low reticulocyte count and dyspoietic cells are sustained in Behcet's disease, physicians should be alert to the possibility of MDS with aberration in chromosome 8 and perform a bone marrow study for the proper diagnosis and treatment of the disease. We presented a case of Behcet's disease associated with MDS, which is the first Korean case.
