ABSTRACT

Introduction: Monitoring changes in the levels of immune markers is of great significance in evaluating the effectiveness of treatment in patients with allergic rhinitis. Objectives: Determine the change in the concentration of immune markers after treatment in patients with allergic rhinitis caused by cotton dust. Methods: A descriptive, single-group, comparative before and after intervention study on 52 patients with allergic rhinitis caused by cotton dust. Comparison of immunological markers results before and after 36 months of treatment. Results: Total IgE concentration after treatment decreased, the median decreased from 1227.756 U/mL to 676.805 UI/mL. Serum levels of IgG, IgG4, and IgG1 in patients after treatment increased compared to before (p< 0.001). The cytokines also changed in the direction of no longer responding toward allergy. Median IL-17 decreased from 1.752 mg/dL to 0.417 mg/dL. Conclusion: In patients with allergic rhinitis after specific sublingual desensitization treatment, IgE levels and cytokines such as IL-6 and IL-17 are significantly reduced and IgG, IgG4 and IgG1 levels are increased after treatment(AU)


Introducción: El monitoreo de los cambios en los niveles de marcadores inmunes es de gran importancia para evaluar la efectividad del tratamiento en pacientes con rinitis alérgica. Objetivos: Determinar el cambio en la concentración de marcadores inmunes después del tratamiento, en pacientes con rinitis alérgica causada por polvo de algodón. Métodos: Estudio descriptivo, monogrupo, comparativo antes y después de la intervención, en 52 pacientes con rinitis alérgica por polvo de algodón. Se compararon resultados de marcadores inmunológicos antes y después de 36 meses de tratamiento. Resultados: La concentración de IgE total después del tratamiento disminuyó, la mediana disminuyó de 1227,756 U/mL a 676,805 UI/mL. Los niveles séricos de IgG, IgG4 e IgG1 en pacientes, después del tratamiento, aumentaron (p< 0,001). Las citocinas también cambiaron en dirección a ausencia de respuesta a la alergia. La mediana de IL-17 disminuyó de 1,752 mg/dL a 0,417 mg/dL. ABSTRACT

Application of different fertilizers to check the efficiency of expression of Bt (Bacillus thuringiensis) gene in one of the leading commercialized crops (cotton) against Lepidopteran species is of great concern. The expression of Cry protein level can be controlled by the improvement of nutrients levels. Therefore, the myth of response of Cry toxin to different combinations of NP fertilizers was explored in three Bt cotton cultivars. Combinations include three levels of nitrogen and three levels of phosphorus fertilizers. Immunostrips and Cry gene(s) specific primer based PCR (Polymerase Chain Reaction) analysis were used for the presence of Bt gene that unveiled the presence of Cry1Ac gene only. Further, the ELISA (enzyme-linked immunosorbent assay) kit was used to quantify the expression of Cry1Ac protein. Under various NP fertilizers rates, the level of toxin protein exhibited highly significant differences. The highest toxin level mean was found to be 2.3740 and 2.1732 µg/g under the treatment of N150P75 kg ha-¹ combination while the lowest toxin level mean was found to be 0.9158 and 0.7641 µg/g at the N50P25 kg ha-¹ level at 80 and 120 DAS (Days After Sowing), respectively. It was concluded from the research that the usage of NP fertilizers has a positive relation with the expression of Cry1Ac toxin in Bt cotton. We recommend using the N150P50 kg ha-1 level as the most economical and practicable fertilizer instead of the standard dose N100P50 kg ha-¹ to get the desired level of Cry1Ac level for long lasting plant resistance (<1.5). The revised dose of fertilizer may help farmers to avoid the cross-resistance development in contradiction of insect pests.


A aplicação de diferentes fertilizantes para verificar a eficiência da expressão do gene Bt (Bacillus thuringiensis) em uma das principais culturas comercializadas (algodão) contra espécies de lepidópteros é uma grande preocupação. A expressão do nível de proteína Cry pode ser controlada pela melhoria dos níveis de nutrientes. Portanto, o mito da resposta da toxina Cry a diferentes combinações de fertilizantes NP foi explorado em três cultivares de algodão Bt. As combinações incluem três níveis de nitrogênio e três níveis de fertilizantes de fósforo. A análise de PCR (reação em cadeia da polimerase) específica para o gene (s) Immunostrips e Cry (s) foi usada para a presença do gene Bt que revelou a presença do gene Cry1Ac apenas. Além disso, o kit ELISA (ensaio de imunoabsorção enzimática) foi usado para quantificar a expressão da proteína Cry1Ac. Sob várias taxas de fertilizantes NP, o nível de proteína de toxina exibiu diferenças altamente significativas. A média do nível mais alto de toxina foi de 2,3740 e 2,1732 µg / g sob o tratamento da combinação N150P75 kg ha-¹, enquanto a média do nível mais baixo de toxina foi de 0,9158 e 0,7641 µg / g no nível de N50P25 kg ha-¹ em 80 e 120 DAS (dias após a semeadura), respectivamente. Concluiu-se com a pesquisa que o uso de fertilizantes NP tem relação positiva com a expressão da toxina Cry1Ac no algodão Bt. Recomendamos o uso do nível de N150P50 kg ha-¹ como o fertilizante mais econômico e praticável em vez da dose padrão N100P50 kg ha-¹ para obter o nível desejado de nível de Cry1Ac para resistência de planta de longa duração (<1,5). A dose revisada de fertilizante pode ajudar os agricultores a evitar o desenvolvimento de resistência cruzada em contradição com as pragas de insetos.

ABSTRACT

The cotton boll weevil, Anthonomus grandis grandis Boheman (Coleoptera: Curculionidae), is a key cotton crop pest in Brazil. Adverse climatic factors, such as high temperatures and low soil moisture, dehydrate oviposited cotton squares (bud flowers) on the ground and cause high mortality of its offspring within these plant structures. The objective of this research was to evaluate the mortality of the cotton boll weevil in drip and sprinkler irrigated cotton crops. The experimental was in included randomized blocks with two treatments: drip (T1) and sprinkler (T2, control) irrigated cotton crops with sixteen replications. Each parcel had one emergence cage, installed between two cotton rows per irrigation system, with 37 cotton squares with opened oviposition punctures and yellowish bracts, to capture adult cotton boll weevils. The average number of boll weevils that emerged from the cotton squares and the causes of mortality at different development stages were determined per treatment. Third-generation life tables of the boll weevil were prepared using the natural mortality data in drip and sprinkler irrigation treatments and plus actual, apparent and indispensable mortality rates and the lethality of each mortality cause. We conclude that the application of water directly to the root zone of the plants in a targeted manner, using the drip irrigation system, can cause high mortality of the cotton boll weevil immature stages inside cotton squares fallen on the ground. This is because the cotton squares fallen on the drier and hotter soil between the rows of drip-irrigated cotton dehydrates causing the boll weevils to die. This is important because it can reduce its population density of the pest and, consequently, the number of applications of chemical insecticides for its control. Thus, contributing to increase the viability of cotton production, mainly in areas of the Brazilian semiarid region where the cotton is cultivated in organic system.


O bicudo-do-algodoeiro, Anthonomus grandis grandis Boheman (Coleoptera: Curculionidae), é uma praga-chave da cultura do algodão no Brasil. Fatores climáticos adversos, como altas temperaturas e baixa umidade do solo, desidratam os botões florais de algodão ovipositados caídos ao solo e causam alta mortalidade de seus descendentes dentro dessas estruturas vegetais. O objetivo desta pesquisa foi avaliar a mortalidade do bicudo-do-algodoeiro em lavouras de algodão irrigadas por gotejamento e aspersão. O delineamento experimental foi em blocos casualizados com dois tratamentos: cultivo de algodão irrigado por gotejamento (T1) e aspersão (T2, testemunha) com dezesseis repetições. Cada parcela possuía uma gaiola de emergência, instalada entre duas fileiras de algodão por sistema de irrigação, com 37 botões florais de algodão com orifícios de oviposição, brácteas abertas e amarelecidas, para captura do bicudo adulto. O número médio de bicudos que emergiu dos botões florais de algodão e as causas de mortalidade, em diferentes estágios de desenvolvimento, foram determinados por tratamento. As tabelas de vida de terceira geração do bicudo-do-algodoeiro foram preparadas usando os dados de mortalidade natural em tratamentos de irrigação por gotejamento e aspersão e as taxas de mortalidade reais, aparentes e indispensáveis e a letalidade de cada causa de mortalidade. Concluímos que a aplicação de água diretamente na zona radicular das plantas de forma direcionada, utilizando o sistema de irrigação por gotejamento, pode causar altas mortalidade dos estágios imaturos do bicudo-do-algodoeiro nos botões florais de algodão caídos no solo. Isso ocorre porque os botões florais de algodão caídos no solo mais seco e mais quente entre as fileiras do algodão irrigado por gotejamento desidratam, causando a morte dos bicudos. ABSTRACT

Interaction among nitrogen fertilization using bovine manure, poultry manure, Jatropha curcas seed cake and urea, and the diseases Ramularia leaf spot (RLS) and Boll rot (BR), caused by Ramulariopsis pseudoglycines and Diplodia gossypina, respectively, in cotton plants (Gossypium hirsutum L.), was studied under field conditions. Intensity (incidence and severity in percentage) of RLS and incidence (%) of BR were evaluated over time, starting in reproductive stage B1 (first visible flower bud). A randomized complete block design with a 4x4 factorial arrangement was used (fertilizers x doses), totaling 16 treatments with four replications. Disease progress was analyzed with the nonlinear Logistic and Gompertz models, obtaining the epidemiological parameters amount of initial disease (Y0) and progress rate (r). Plants fertilized with 50 kg N ha-1, presented an incidence twice greater than those obtained with other fertilizers. The Logistic model better fits RLS, but no model could represent BR. Only the epidemiological parameters of RLS were affected differently in this experiment compared to BR disease. The possible role of organic and inorganic nitrogen fertilization in the RLS and BR management is discussed.

Sucking pests are major threat to cotton field crop which cause unbearable losses to the crop yield. Aim of the current study was to record seasonal dynamics of major sucking insect pests including whitefly, jassid, thrips and their natural arthropod predators i.e. green lacewings and spiders in cotton field plots. The effects of surrounding field crops on pests' density and predatory efficiency of predators were also recorded. For sampling and survey of insects, the visual counting was found to be the most efficient method for recording the abundance of insects, trailed by net sweeping and tapping. Whitefly was the most dominant sucking pest found on the vegetative stage of cotton, followed by jassid and thrips. Fluctuated populations of predatory arthropods, spiders and green lacewings were also recorded during whole cropping season however, the densities of pests and predators varied with crop phenology. Spiders' population was encouraging at both vegetative and flowering stage and also the same trend of jassid and whitefly were observed at both stages of the crop. Surrounding habitats showed non-significant effect on population densities of insect pests and predators. For abiotic factors, the spiders showed strong positive correlation with humidity and temperature. However, green lacewing was only positively correlated with humidity. On the other hand, the populations of whitefly, jassid and thrips showed non-significant correlation with both temperature and humidity. Overall densities of sucking insect pests were found above economic threshold level. The plant age, crop stage and surrounding habitats effect on the population fluctuation of pests as well as the predators' abundance. The future studies are also warranted to investigate the altered habitats and multiple trap cropping to find out their impact on unattended insect predators and parasitoids in cotton crop.


As pragas sugadoras são uma grande ameaça para a cultura do algodão, causando perdas insuportáveis no rendimento da cultura. O objetivo do estudo atual foi registrar a dinâmica sazonal das principais pragas de insetos sugadores, incluindo mosca-branca, jassid, tripes e seus artrópodes predadores naturais, ou seja, crisopídeos e aranhas verdes em parcelas de algodão. Os efeitos das plantações circundantes na densidade de pragas e na eficiência predatória de predadores também foram registrados. Para amostragem e pesquisa de insetos, a contagem visual foi considerada o método mais eficiente para registrar a abundância de insetos, seguido por varredura e batida de rede. A mosca-branca foi a praga sugadora mais dominante encontrada na fase vegetativa do algodoeiro, seguida pelo jassid e tripes. Populações flutuantes de artrópodes predadores, aranhas e crisálidas também foram registradas durante toda a safra, no entanto as densidades de pragas e predadores variaram com a fenologia da cultura. A população de aranhas foi encorajadora tanto na fase vegetativa como na floração e também a mesma tendência de jassid e mosca-branca foi observada em ambas as fases da cultura. Os habitats circundantes mostraram efeito não significativo nas densidades populacionais de insetos-praga e predadores. Para os fatores abióticos, as aranhas apresentaram forte correlação positiva com umidade e temperatura. No entanto, lacewing verde foi apenas positivamente correlacionado com a umidade. Por outro lado, as populações de mosca-branca, jassid e tripes apresentaram correlação não significativa com temperatura e umidade. As densidades gerais de pragas sugadoras de insetos foram encontradas acima do nível do limiar econômico. A idade da planta, o estágio da cultura e os habitats circundantes afetam a flutuação populacional de pragas, bem como a abundância de predadores. Os estudos futuros também são necessários para investigar os habitats alterados e cultivo com armadilhas múltiplas [...].

Optimum planting arrangement is an important attribute for efficient utilization of available resources and to obtain high yield of cotton. Application of plant growth promoter and retardant on cotton in improved planting density are the innovative techniques in the establishment of more productive cotton crop. Therefore, we planned a field study to assess the role of bio-stimulant and growth retardant in the resource utilization efficiency of cotton cultivars planted under variable row spacing at Agronomic Research Area Bahauddin Zakariya University and Usmania Agricultural Farm Shujabad during Kharif 2012. Experimental treatments consisted of cotton genotypes viz. CIM-573 and CIM-598, cultivated under conventional (75 cm), medium (50 cm) and ultra-narrow row spacing (25 cm) with foliar spray of bio-stimulant (moringa leaf extract) and growth retardant (mepiquate chloride), either sole or in combination, keeping distilled water as a control. Exogenously applied MLE alone and MLE + MC significantly enhanced the number of squares, flowers and green bolls per plant leading to higher cotton seed and lint yield of CIM 598 cultivar cultivated under conventional row spacing. While application of MC alone and MLE + MC produced maximum micronaire value, fiber strength and fiber uniformity ratio of CIM 573 cultivar cultivated under conventional row spacing. The results suggested that application of MLE is a possible approach to enhance the cotton productivity and the use of MC to enhance the fiber quality attributes under conventional row spacing.


A configuração ideal de plantio é um atributo importante para a utilização eficiente dos recursos disponíveis e para obter alto rendimento de algodão. A aplicação de promotores de crescimento de plantas e reguladores de crescimento no algodão em uma densidade de plantio adequada são técnicas inovadoras na obtenção de safras de algodão mais produtivas. Portanto, foi planejado um estudo de campo para avaliar o papel de um bioestimulante e um regulador de crescimento na eficiência da utilização de recursos de cultivares de algodão plantadas com espaçamento variável entre linhas na Área de Pesquisa Agronômica Universidade Bahauddin Zakariya e Usmania Agricultural Farm Shujabad durante Kharif 2012. Os tratamentos experimentais consistiram em genótipos de algodão viz. CIM-573 e CIM-598, cultivadas sob espaçamento convencional (75 cm), médio (50 cm) e ultraestreito (25 cm) e pulverização foliar de bioestimulante (extrato de folha de moringa) e regulador de crescimento (cloreto de mepiquato)), sozinho ou combinado, mantendo a água destilada como controle. O MLE aplicado exogenamente sozinho e o MLE + MC aumentaram significativamente o número de quadrados, flores e cápsulas verdes por planta, levando a um maior rendimento de sementes e fibra de algodão da cultivar CIM 598 cultivada sob espaçamento convencional entre fileiras. Enquanto a aplicação de MC sozinho e MLE + MC produziu valor máximo de micronaire, resistência da fibra e razão de uniformidade da fibra da cultivar CIM 573 cultivada sob espaçamento convencional entre linhas. Os resultados sugeriram que a aplicação do MLE é uma abordagem possível para aumentar a produtividade do algodão e o uso de MC para aprimorar os atributos de qualidade da fibra no espaçamento convencional entre linhas.

ABSTRACT

A field study was carried out to determine the influence of foliage applied plant growth promoter and retardant in improving soil applied sulphur fertilizer use efficiency in cotton during two consecutive summers 2014 and 2015. Experimental trial comprised of three different sources of sulphur (ammonium sulphate, potassium sulphate and elemental sulphur) and foliar spray of plant growth promoter and growth retardant including tap water was taken as control. Among treatments soil applied ammonium sulphate with foliage applied amino acid produced maximum plant height, sympodial branches, pods per plant, seed cotton yield, fiber yield, biological yield, protein contents, oil contents and leaf nitrogen uptake as compared to the other treatments. Whereas, soil applied potassium sulphate with foliar spray of mepiquat chloride on cotton significantly improved the boll weight and leaf potassium uptake. We conclude that soil applied ammonium sulphate and foliage spray of amino acid was more effective in improving the productivity and quality attributes of cotton.


Foi realizado um estudo de campo para determinar a influência do promotor de crescimento das plantas e retardador da folhagem em algodão, para melhora da eficiência do uso de fertilizantes à base de enxofre aplicados no solo durante dois verões consecutivos (2014 e 2015). O ensaio experimental foi composto de três fontes diferentes de enxofre (sulfato de amônio, sulfato de potássio e enxofre elementar) e pulverização foliar do promotor de crescimento de plantas e retardador de crescimento, incluindo água da torneira que foi tomada como controle. Entre os tratamentos, o sulfato de amônio aplicado no solo com aminoácido aplicado na folhagem produziu o máximo na altura da planta, ramos simodiais, capulhos por planta, rendimento de algodão em caroço, rendimento de fibra, rendimento biológico, conteúdo de proteínas, conteúdo de óleo e absorção de nitrogênio nas folhas quando comparado a outros tratamentos. Enquanto o solo fertilizado com sulfato de potássio e aplicação foliar de cloreto de mepiquat no algodão melhorou, significativamente, o peso do capulho e a absorção de potássio nas folhas. Sulfato de amônio aplicado no solo e a aplicação foliar de aminoácidos foram mais eficazes na melhora dos atributos de produtividade e qualidade do algodão.

Vegetable oils have their specific physicochemical properties due to which they are playing vital role in human nutritional diet for health benefits. Cottonseed oil is obtained from various species of cotton seeds that are famous to be grown mainly for their fiber quality. The most prominently used specie is Gossypium hirsutum. It is obvious that the seeds of different variety of cotton vary as grown in diverse agroclimatic conditions with respect to oil, fats and protein contents. Cottonseed oil is routinely used for cooking and food manufacturing products. Cottonseed oil obtained after proper extraction/processing steps from crude state to refined oil in a variety of ways. Cotton crop is considered for their dual-use purpose, for fiber quality and oil production to promote health benefits in the world. Keeping in view the above facts, this review clearly demonstrated an overview about physicochemical and functional properties of cottonseed oil to promote health benefits associated with the use of this oil. The overall characteristics and all concerned health benefits of CSO will further improve their usefulness is a compact way. We have summarized a brief multi-dimensional features of CSO in all aspects up to the best of our knowledge fort he end researchers who can further research in the respective aspect.


Os óleos vegetais têm propriedades físico-químicas específicas que desempenham um papel vital na dieta nutricional humana em benefício à saúde. O óleo de semente de algodão, utilizado rotineiramente no preparo e na fabricação de alimentos, é obtido através de várias espécies de sementes de algodão, famosas pela alta qualidade de sua fibra, cuja espécie mais utilizada é Gossypium hirsutum. As sementes variam, em relação ao teor do óleo, da gordura e das proteínas, de acordo com o cultivo e as diversas condições agroclimáticas. O óleo de algodão é obtido após etapas adequadas de extração das sementes e processamento do estado bruto ao refinado. Assim, a presente revisão demonstra, de maneira geral, as propriedades físico-químicas e funcionais do óleo de semente de algodão e seus benefícios à saúde humana, resumindo suas características multidimensionais. As características gerais e todos os benefícios do composto podem melhorar ainda mais se utilizadas de forma compacta, auxiliando futuros pesquisadores.

As sementes variam, em relação ao teor do óleo, da gordura e das proteínas, de acordo com o cultivo e as diversas condições agroclimáticas. O óleo de algodão é obtido após etapas adequadas de extração das sementes e processamento do estado bruto ao refinado. Assim, a presente revisão demonstra, de maneira geral, as propriedades físico-químicas e funcionais do óleo de semente de algodão e seus benefícios à saúde humana, resumindo suas características multidimensionais. As características gerais e todos os benefícios do composto podem melhorar ainda mais se utilizadas de forma compacta, auxiliando futuros pesquisadores.

ABSTRACT

Understanding the genetic diversity and overcoming genotype-by-environment interaction issues is an essential step in breeding programs that aims to improve the performance of desirable traits. This study estimated genetic diversity and applied genotype + genotype-by-environment (GGE) biplot analyses in cotton genotypes. Twelve genotypes were evaluated for fiber yield, fiber length, fiber strength, and micronaire. Estimation of variance components and genetic parameters was made through restricted maximum likelihood and the prediction of genotypic values was made through best linear unbiased prediction. The modified Tocher and principal component analysis (PCA) methods, were used to quantify genetic diversity among genotypes. GGE biplot was performed to find the best genotypes regarding adaptability and stability. The Tocher technique and PCA allowed for the formation of clusters of similar genotypes based on a multivariate framework. The GGE biplot indicated that the genotypes IMACV 690 and IMA08 WS were highly adaptable and stable for the main traits in cotton. The cross between the genotype IMACV 690 and IMA08 WS is the most recommended to increase the performance of the main traits in cotton crops.


Compreender a diversidade genética e contornar os problemas causados pela interação genótipos por ambientes é uma etapa importante em programas de melhoramento. Este estudo teve como objetivo estimar a diversidade genética e aplicar a metodologia de biplot genótipo + genótipo por ambiente (GGE biplot) em doze genótipos de algodão avaliados quanto ao rendimento da fibra, comprimento da fibra, resistência da fibra e micronaire. A estimativa dos componentes de variância e dos parâmetros genéticos foi feita através do método da máxima verossimilhança restrita e a predição dos valores genotípicos por meio da melhor predição linear não enviesada. Os métodos de Tocher modificado e análise de componentes principais (PCA) foram utilizados para quantificar a diversidade genética entre os genótipos. O método GGE biplot foi conduzido para encontrar os melhores genótipos em relação à adaptabilidade e estabilidade. As técnicas de Tocher e PCA permitiram a formação de clusters de genótipos semelhantes com base em uma estrutura multivariada. O GGE biplot indicou que os genótipos IMACV 690 e IMA08 WS foram altamente adaptáveis e estáveis para as principais características do algodão. O cruzamento dentre os genótipos IMACV 690 e IMA08 WS é o mais recomendado para aumentar o desempenho das principais características na cultura do algodão.

BACKGROUND: Cytokinin signal transduction is mediated by a two-component system (TCS). Two-component systems are utilized in plant responses to hormones as well as to biotic and abiotic environmental stimuli. In plants, response regulatory genes (RRs) are one of the main members of the two-component system (TCS). METHOD: From the aspects of gene structure, evolution mode, expression type, regulatory network and gene function, the evolution process and role of RR genes in the evolution of the cotton genome were analyzed. RESULT: A total of 284 RR genes in four cotton species were identified. Including 1049 orthologous/paralogous gene pairs were identified, most of which were whole genome duplication (WGD). The RR genes promoter elements contain phytohormone responses and abiotic or biotic stress-related cis-elements. Expression analysis showed that RR genes family may be negatively regulate and involved in salt stress and drought stress in plants. Protein regulatory network analysis showed that RR family proteins are involved in regulating the DNA-binding transcription factor activity (COG5641) pathway and HP kinase pathways. VIGS analysis showed that the GhRR7 gene may be in the same regulatory pathway as GhAHP5 and GhPHYB, ultimately negatively regulating cotton drought stress by regulating POD, SOD, CAT, H2O2 and other reactive oxygen removal systems. CONCLUSION: This study is the first to gain insight into RR gene members in cotton. Our research lays the foundation for discovering the genes related to drought and salt tolerance and creating new cotton germplasm materials for drought and salt tolerance.

BACKGROUND: The internal NAD(P)H dehydrogenase (NDA) gene family was a member of the NAD(P)H dehydrogenase (ND) gene family, mainly involved in the non-phosphorylated respiratory pathways in mitochondria and played crucial roles in response to abiotic stress. METHODS: The whole genome identification, structure analysis and expression pattern of NDA gene family were conducted to analyze the NDA gene family. RESULTS: There were 51, 52, 26, and 24 NDA genes identified in G. hirsutum, G. barbadense, G. arboreum and G. raimondii, respectively. According to the structural characteristics of genes and traits of phylogenetic tree, we divided the NDA gene family into 8 clades. Gene structure analysis showed that the NDA gene family was relatively conservative. The four Gossypium species had good collinearity, and segmental duplication played an important role in the evolution of the NDA gene family. Analysis of cis-elements showed that most GhNDA genes contained cis-elements related to light response and plant hormones (ABA, MeJA and GA). The analysis of the expression patterns of GhNDA genes under different alkaline stress showed that GhNDA genes were actively involved in the response to alkaline stress, possibly through different molecular mechanisms. By analyzing the existing RNA-Seq data after alkaline stress, it was found that an NDA family gene GhNDA32 was expressed, and then theGhNDA32 was silenced by virus-induced gene silencing (VIGS). By observing the phenotype, we found that the wilting degree of silenced plants was much higher than that of the control plant after alkaline treatment, suggesting that GhNDA32 gene was involved in the response to alkaline stress. CONCLUSIONS: In this study, GhNDAs participated in response to alkaline stress, especially NaHCO3 stress. It was of great significance for the future research on the molecular mechanism of NDA gene family in responding to abiotic stresses.

Sucking pests are major threat to cotton field crop which cause unbearable losses to the crop yield. Aim of the current study was to record seasonal dynamics of major sucking insect pests including whitefly, jassid, thrips and their natural arthropod predators i.e. green lacewings and spiders in cotton field plots. The effects of surrounding field crops on pests' density and predatory efficiency of predators were also recorded. For sampling and survey of insects, the visual counting was found to be the most efficient method for recording the abundance of insects, trailed by net sweeping and tapping. Whitefly was the most dominant sucking pest found on the vegetative stage of cotton, followed by jassid and thrips. Fluctuated populations of predatory arthropods, spiders and green lacewings were also recorded during whole cropping season however, the densities of pests and predators varied with crop phenology. Spiders' population was encouraging at both vegetative and flowering stage and also the same trend of jassid and whitefly were observed at both stages of the crop. Surrounding habitats showed non-significant effect on population densities of insect pests and predators. For abiotic factors, the spiders showed strong positive correlation with humidity and temperature. However, green lacewing was only positively correlated with humidity. On the other hand, the populations of whitefly, jassid and thrips showed non-significant correlation with both temperature and humidity. Overall densities of sucking insect pests were found above economic threshold level. The plant age, crop stage and surrounding habitats effect on the population fluctuation of pests as well as the predators' abundance. The future studies are also warranted to investigate the altered habitats and multiple trap cropping to find out their impact on unattended insect predators and parasitoids in cotton crop.


As pragas sugadoras são uma grande ameaça para a cultura do algodão, causando perdas insuportáveis no rendimento da cultura. O objetivo do estudo atual foi registrar a dinâmica sazonal das principais pragas de insetos sugadores, incluindo mosca-branca, jassid, tripes e seus artrópodes predadores naturais, ou seja, crisopídeos e aranhas verdes em parcelas de algodão. Os efeitos das plantações circundantes na densidade de pragas e na eficiência predatória de predadores também foram registrados. Para amostragem e pesquisa de insetos, a contagem visual foi considerada o método mais eficiente para registrar a abundância de insetos, seguido por varredura e batida de rede. A mosca-branca foi a praga sugadora mais dominante encontrada na fase vegetativa do algodoeiro, seguida pelo jassid e tripes. Populações flutuantes de artrópodes predadores, aranhas e crisálidas também foram registradas durante toda a safra, no entanto as densidades de pragas e predadores variaram com a fenologia da cultura. A população de aranhas foi encorajadora tanto na fase vegetativa como na floração e também a mesma tendência de jassid e mosca-branca foi observada em ambas as fases da cultura. Os habitats circundantes mostraram efeito não significativo nas densidades populacionais de insetos-praga e predadores. Para os fatores abióticos, as aranhas apresentaram forte correlação positiva com umidade e temperatura. No entanto, lacewing verde foi apenas positivamente correlacionado com a umidade. Por outro lado, as populações de mosca-branca, jassid e tripes apresentaram correlação não significativa com temperatura e umidade. As densidades gerais de pragas sugadoras de insetos foram encontradas acima do nível do limiar econômico. A idade da planta, o estágio da cultura e os habitats circundantes afetam a flutuação populacional de pragas, bem como a abundância de predadores. Os estudos futuros também são necessários para investigar os habitats alterados e cultivo com armadilhas múltiplas para descobrir seu impacto sobre predadores de insetos e parasitoides desacompanhados na cultura do algodão.

ABSTRACT

RESUMEN El neotrópico es sitio de origen de gran variedad de plantas que actualmente son cultivadas con éxito en diferentes regiones del mundo. Sin embargo, condiciones climáticas adversas, que se pueden ver acrecentadas por efectos del cambio climático antropogénico, pueden afectar su rendimiento y productividad debido a las situaciones de estrés abiótico que se pueden generar. Como alternativa para contrarrestar estos efectos, se ha experimentado con modificaciones genéticas, particularmente en genes relacionados con la producción de osmolitos y factores de transcripción que han llevado a que estas plantas, a nivel experimental, tengan mayor tolerancia a estrés oxidativo, altas y bajas temperaturas y fotoinhibición, sequía y salinidad, mediante la acumulación de osmoprotectores, la regulación en la expresión de genes y cambios en el fenotipo. En este trabajo se presentan y describen las estrategias metodológicas planteadas con estos fines y se complementan con ejemplos de trabajos realizados en cultivos de origen neotropical de importancia económica, como maíz, algodón, papa y tomate. Además, y debido a la novedad y potencial que ofrece la edición génica por medio del sistema CRISPR/Cas9, también se mencionan trabajos realizados en plantas con origen neotropical, enfocados en comprender e implementar mecanismos de tolerancia a sequía. Las metodologías aquí descritas podrían constituirse en opciones prácticas para mejorar la seguridad alimentaria con miras a contrarrestar las consecuencias negativas del cambio climático antropogénico.


ABSTRACT The neotropics are the site of origin of a large variety of plants that are currently successfully cultivated in different regions of the world. However, adverse climatic conditions, which can be exacerbated by the effects of anthropogenic climate change, can affect their yield and productivity due to the abiotic stresses that can be generated. As an alternative to counteract these effects, genetic modifications have been experimentally implemented, particularly in genes related to osmolyte production and transcription factors, which have ultimately led to increased tolerance of these plants to oxidative stress, high and low temperatures and photoinhibition, drought and salinity, through the accumulation of osmoprotectants, regulation of gene expression and changes in phenotype. In this work, the methodological strategies proposed to these aims are presented and described, and they are complemented with examples of studies carried out in economically important crops of neotropical origin, such as corn, cotton, potato and tomato. In addition, and due to the novelty and potential that gene editing offers through the CRISPR/Cas9 system, works conducted in plants with neotropical origin, focused on understanding and implementing drought tolerance mechanisms, are also mentioned. The methodologies described here could become practical options to achieve food security in the frame of the adverse effects caused by anthropogenic climate change.

ABSTRACT

The objective of this work was to analyze the genetic diversity using conventional methods and artificial neural networks among 12 colored fiber cotton genotypes, using technological characteristics of the fiber and productivity in terms of cottonseed and cotton fiber yield. The experiment was conducted in an experimental area located at Fazenda Capim Branco, belonging to the Federal University of Uberlândia, in the city of Uberlândia, Minas Gerais. Twelve genotypes of colored fiber cotton were evaluated, 10 from the Cotton Genetic Improvement Program (PROMALG): UFUJP - 01, UFUJP - 02, UFUJP - 05, UFUJP - 08, UFUJP - 09, UFUJP - 10, UFUJP - 11, UFUJP - 13, UFUJP - 16, UFUJP - 17 and two commercial cultivars: BRS Rubi (RC) and BRS Topázio (TC). The experimental design used was complete randomized block (CRB) with three replications. The following evaluations were carried out at full maturation: yield of cottonseed (kg ha-1) and the technological characteristics, which include, fiber length, micronaire, maturation, length uniformity, short fiber index, elongation and strength, using the HVI (High volume instrument) device. Genetic dissimilarity was measured using the generalized Mahalanobis distance and after obtaining the dissimilarity matrix, the genotypes were grouped using a hierarchical clustering method (UPGMA). A discriminant analysis and the Kohonen Self-Organizing Map (SOM) by Artificial Neural Networks (ANN's) were performed through computational intelligence. SOM was able to detect differences and organize the similarities between accesses in a more coherent way, forming a larger number of groups, when compared to the method that uses the Mahalanobis matrix. It was also more accurate than the discriminant analysis, since it made it possible to differentiate groups more coherently when comparing their phenotypic behavior. The methods that use computational intelligence proved to be more efficient in detecting similarity, with Kohonen's Self-Organizing Map being the most adequate to classify and group cotton genotypes.

A very limited amount of information is available in relation both to the residual effect of herbicides destinated to the destruction of cotton stalks and to the time interval required to prevent that development and yield of the following crop be affected. This work aimed to identify the residual activity of herbicides intended to eliminate cotton stalks and to estimate the safety interval (SI) of time for the next cotton sowing. Two trails were simultaneously carried out, the first one for a single application and a second one for the two sequential applications of herbicide treatments, in a 15×5 factorial design organized in randomized complete blocks with four replications. Levels of first factor were constituted by herbicide treatments and the levels of the second factor were composed five periods of time for cotton sowing after herbicide application (0, 30, 60, 90, and 120 days). Herbicides were 2,4-D, glyphosate, saflufenacil, [imazapic + imazapyr], dicamba, fluroxypyr and sulfentrazone. Results provided an indication of residual activity of herbicide treatments in soil and indicated that a single application or two sequential applications of glyphosate + dicamba + saflufenacil have a considerable potential to affect cotton and a period exceeding 100 days for a single application and exceeding 120 days for two sequential applications was necessary. Treatments with 2,4-D and 2,4-D + glyphosate provided the shortest safe interval and may be used for cotton stalk destruction with no risks for the crop sowing after the withdrawal period.

Plant parasitic nematodes are major threats to Brazilian and world agriculture. Among them, Pratylenchus brachyurus and Meloidogyne incognita stand out as major pests for several crops, including corn, cotton, soybean among others, which can be components of integrated crop-livestock-forestry systems (ICLFs). In this context, information about the host status of eucalypts (Corymbia spp. and Eucalyptus spp.) to plant parasitic nematodes becomes more relevant in Brazil, due to the use of eucalypts in ICLFs. If tree components used on this system increase the population density of P. brachyurus and M. incognita, it is possible that these pathogens could damage shorter-cycle crops. Since information about the host status of eucalypts to plant parasitic nematodes is scarce, this study evaluated the host status of some eucalypt species to P. brachyurus and M. incognita. Two greenhouse trials were done to evaluate the reproduction of P. brachyurus and one to M. incognita, using some of the most cultivated species and hybrids of eucalypts in Brazil. The population growth of P. brachyurus increased on Corymbia citriodora, Eucalyptus grandis, Eucalyptus dunnii × Eucalyptus urophylla, and E. grandis × E. urophylla after ~90 days of inoculation. Conversely, despite M. incognita reproducing well in the control plants, no individuals were recovered from C. citriodora, E. urophylla and E. grandis × E. urophylla, which were classified as resistant plants. Based on both obtained and available data, M. incognita poses no threat to eucalypt species today. However, P. brachyurus is suggested to be a threat to eucalypts.

Genetically modified plants are one of the tactics used in integrated pest management - IPM. There is great concern about the impact of these plants on non-target organisms. On the other hand, there is little information in the literature on the effects of transgenics (Bacillus thuringiensis) Bt on populations of phytophagous mites, and the physiological responses that this attack promotes on plants. The objective of this work was to evaluate the biology of the T. ludeni mite in Bt cotton, expressing the Cry1F and Cry1Ac proteins. To evaluate the behavior of food and oviposition preference of the T. ludeni with Bt cotton and isohybrid. Verify if the physiological stress caused by T. ludeni's attack is differentiated in Bt cotton. The mites were reared in Bt cotton and isohybrid, in a total of 40 replicates in the completely randomized design and the biological cycle was evaluated. The food preference and oviposition analysis were done with 10 replicates, with choice. The physiological stress was evaluated through chlorophyll fluorescence, under greenhouse conditions. The data of the T. ludeni biology were analyzed by Student's t-test, for food and oviposition preference the chi-square test was performed. Regression models were fitted for the fluorescence parameters. The model identity test was used to evaluate the differences between Bt and isohybrid treatments. Cry1F and Cry1Ac proteins have not affected the biology of T. ludeni. The photosynthetic parameters in Bt cotton plants were less influenced by T. ludeni infestation.


O uso de plantas geneticamente modificadas é uma das táticas utilizadas no manejo integrado de pragas - MIP. Observa-se grande preocupação com o impacto dessas plantas sobre organismos não alvos. Por outro lado, existe pouca informação na literatura sobre efeitos dos transgênicos (Bacillus thuringiensis) Bt em populações de ácaros fitófagos, e as respostas fisiológicas que esse ataque promove nas plantas. Objetivou-se com esse trabalho avaliar a biologia do ácaro T. ludeni em algodoeiro Bt, expressando as proteínas Cry1F e Cry1Ac. Avaliar se há comportamento de preferência alimentar e postura de T. ludeni em relação ao algodoeiro Bt e seu iso-híbrido. E verificar se o estresse fisiológico causado pelo ataque de T. ludeni é diferenciado em algodoeiro Bt. Os ácaros foram criados em algodoeiro Bt e iso-híbrido, em um total de 40 repetições no delineamento inteiramente casualizado, onde foi avaliado o ciclo biológico. A análise de preferência alimentar e de posturas foi feita com 10 repetições, com escolha. O estresse fisiológico foi avaliando através da fluorescência da clorofila, em casa de vegetação. Os dados da biologia de T. ludeni foram analisados pelo teste t Student, para preferência alimentar e postura foi realizado o teste qui-quadrado. Para os parâmetros da fluorescência, foram ajustados modelos de regressão. Para testar as diferenças entre Bt e iso-híbrido foi utilizado o teste de identidade de modelos. As proteínas Cry1F e Cry1Ac não afetaram a biologia de T. ludeni. Os parâmetros fotossintéticos em plantas de algodoeiro Bt foram menos influenciados pela infestação de T. ludeni.

BACKGROUND: Melatonin 2-hydroxylase (M2H) is the first enzyme in the catabolism pathway of melatonin, which catalyzes the production of 2-hydroxymelatonin (2-OHM) from melatonin. The content of 2-hydroxymelatonin in plants is much higher than that of melatonin. So M2H may be a key enzyme in the metabolic pathway of melatonin. METHOD: We conducted a systematic analysis of the M2H gene family in Gossypium hirsutum based on the whole genome sequence by integrating the structural characteristics, phylogenetic relationships, expression profile, and biological stress of the members of the Gossypium hirsutum M2H gene family. RESULT: We identified 265 M2H genes in the whole genome of Gossypium hirsutum, which were divided into 7 clades (clades I-VII) according to phylogenetic analysis. Most M2H members in each group had similar motif composition and gene structure characteristics. More than half of GhM2H members contain ABA-responsive elements and MeJA-responsive elements. Under different stress conditions, the expression levels of the gene changed, indicating that GhM2H members were involved in the regulation of abiotic stress. Some genes in the GhM2H family were involved in regulating melatonin levels in cotton under salt stress, and some genes were regulated by exogenous melatonin. CONCLUSION: This study is helpful to explore the function of GhM2H, the downstream metabolism gene of melatonin in cotton, and lay the foundation for better exploring the molecular mechanism of melatonin improving cotton's response to abiotic stress.

Abstract The mealybug, Phenacoccus solenopsis Tinsley (Hemiptera: Pseudococcidae) is a cotton pest widespread in several cotton growing regions of Brazil, particularly in the semi-arid region of southwestern Bahia. The impact of kaolin on survival, development and reproduction of P. solenopsis was evaluated in the laboratory. The experiment was developed in a completely randomized design with two treatments: immature or newly emerged adults of P. solenopsis sprayed with kaolin and fed with cotton leaf discs treated with kaolin suspension (with kaolin) (T1) and immature or newly emerged adults of P. solenopsis sprayed with distilled water and fed with cotton leaf discs treated with distilled water (without kaolin) (T2). The kaolin suspension shortens the life cycle, increases the reproductive potential and population growth of the cotton mealybug, P. solenopsis and, therefore, it should be used with caution on cotton plants in regions with a history of occurrence of this pest.


Resumo A cochonilha Phenacoccus solenopsis Tinsley (Hemiptera: Pseudococcidae) é uma praga de algodão, amplamente, difundida em várias regiões produtoras do Brasil, particularmente, na região semiárida do sudoeste da Bahia. O impacto do caulim na sobrevivência, desenvolvimento e reprodução de P. solenopsis foi avaliado em laboratório. O delineamento experimental foi inteiramente casualizado com dois tratamentos: imaturos ou adultos recém-emergidos de P. solenopsis alimentados com discos foliares de algodão tratados com caulim (com caulim) e pulverizadas com este produto (T1) e imaturos ou adultos recém-emergidos de P. solenopsis alimentados com discos foliares de algodão tratados com água destilada (sem caulim) e pulverização dos insetos com água destilada (T2). A suspensão do caulim encurtou o ciclo de vida, aumentou o potencial reprodutivo e o crescimento populacional da cochonilha-do-algodoeiro, P. solenopsis e, portanto, deve ser utilizado com cautela em plantas de algodão em regiões com histórico de ocorrência desta praga.
