ABSTRACT
Foot and mouth disease is a highly contagious viral disease that poses a significant economic threat to cloven-hoofed animals, including cattle and sheep. The emergence of a novel foot and mouth disease virus-A isolate, FMDV-A-Egy-AHRI-RL385-Ven-2022, in Egypt in 2022 has raised concerns about its potential impact on existing vaccination programs. Given that vaccination is a key strategy for foot and mouth disease virus control, the present study was aimed to assess the cross-protective efficacy of both local and imported inactivated vaccines against this new threat. Through challenge experiments and serum neutralization tests, we observed limited effectiveness of both vaccine types. The calculated r1-values at 28 days post-vaccination indicated a minimal immune response to FMDV-A-Egy-AHRI-RL385-Ven-2022 (0.176 and 0.175 for local and imported vaccines, respectively). Challenge experiments further confirmed these findings, revealing 0percent protection from the local vaccine and only 20percent rotection from imported vaccines by day 7 post-challenge. These results underscore the urgent need to update existing foot and mouth disease virus vaccines in Egypt by incorporating the newly circulating FMDV-A-Egy-AHRI-RL385-Ven-2022 strain. This proactive measure is crucial to prevent future outbreaks and ensure effective disease control(AU)

Cattle breeding has great importance in the Brazilian economy, however, diseases affect production, causing economic losses, such as myiasis. The causative agent of myiasis is Cochliomyia hominivorax, the screwworm fly. The agents are suitable for tropical and subtropical regions in Brazil, and exhibit parasitic drug resistance. The objective of this work was to verify the occurrence of C. hominivorax in cattle in the state of Maranhão, Brazil. In the first stage, 19 dairy and beef cattle properties were chosen, distributed in 11 municipalities in Maranhão. Questionnaires asked about the management, the drugs used and the cost of prevention related to the disease. The cattle were examined by inspection, partially collecting the larvae found on the body surface. The number of larvae per bovine, location of lesions, origin and complications were observed and noted. Then, the collected specimens were placed in individual flasks, per sampled host, containing alcohol at 70°Gay-Lussac (GL). In the second stage, the fly larvae were identified in the laboratory, examined under a stereomicroscope and identified by their morphology and analysis of perithematics plaques and respiratory spiracles. It was found more that 60% (12/19) of the properties had parasitized cattle. Among the 4.509 animals observed, 0.57% (26/4509) were parasitized with the larvae of C. hominivorax. Infestations were found in the navel (50%), as well as in the neck plate (42%), vulva (4%) and horn (4%). It is concluded that there was a low prevalence of myiasis caused by C. hominivorax in cattle in the state of Maranhão, but prevention and health care should always be adopted by livestock farmers.

CRISPR/Cas (clustered regularly interspaced short palindromic repeats/CRISPR-associated protein) is widely used in the field of livestock breeding. However, its low efficiency, untargeted cutting and low safety have greatly hampered its use for introducing single base mutations in livestock breeding. Single base editing, as a new gene editing tool, can directly replace bases without introducing double strand breaks. Single base editing shows high efficiency and strong specificity, and provides a simpler and more effective method for precise gene modification in livestock breeding. This paper introduces the principle and development of single base editing technology and its application in livestock breeding.

Foot and mouth disease virus is a highly infectious and contagious pathogen. Recently the topotype VII, Lib‐12 lineage of serotype SAT2 was reported through outbreaks in Egypt during 2018. Vaccination is an effective way to control and combat the foot and mouth disease virus outbreaks especially in endemic areas like Egypt. The present study was aimed to evaluate the efficacy of the current produced foot and mouth disease vaccine, against the recently isolated field strain foot and mouth disease virus SAT2 topotype VII, Lib-12 lineage (SAT2 Libya), by applying in vitro and in vivo studies. Two batches of the current foot and mouth disease virus vaccine were inoculated in calves. At the 28th day post-vaccination serum samples were collected and tested against tissue culture adapted foot and mouth disease virus SAT2 Libya and SAT2/EGY/2/2012 using virus neutralization test to determine serological relationship (r1-value). The challenge test for vaccinated calves was carried out against the virulent foot and mouth disease virus SAT2 Libya. It was found that neutralizing antibody titers induced by the two vaccine batches (1 and 2) and those in unvaccinated animals were 0.48, 0.39 and 0.15 log10 TCID50/mL, respectively, while the challenge revealed protection values of 20 percent, 0 percent and 0 percent, respectively. Furthermore, the r1 values were 0.195 and 0.186 for vaccine batches (1 and 2), respectively. It was concluded that the available local commercial inactivated foot and mouth disease virus vaccine batches (SAT2 SAT2/EGY/2/2012) are unable to protect calves against the current circulating foot and mouth disease virus field isolate SAT2 topotype VII, Lib-12 lineage, thus it is highly recommended to update the existing vaccines with the present isolated strain(AU)

ABSTRACT Fasciolosis and paramphistomosis are a major health problem for the livestock economy worldwide. In Colombia, both distomatosis are reported in cattle, particularly in high Andean tropics, with annual economic losses close to COP 40 billion. The goal of this study was to determine the prevalence of fasciolosis and paramphistomosis in cattle from La Candelaria farm, Caucasia (Colombia), and the presence of intermediate host snails. A descriptive cross-sectional study was carried out with non-probability convenience sampling. Stool samples were collected from the animals and the diagnosis was made using the modified Dennis technique. Digenea prevalence were determined according to sex, weight, age, and race. Freshwater snails were collected in the studied area and were identified by morphology. 400 and 66 fecal samples from 178 bovines of the Zebu (Bos indicus), BON (white-eared white) breeds and the cross between them were analyzed. Fasciola hepatica and Paramphistomidae were diagnosed with a prevalence of 2,2% and 30,9%, respectively. Coinfection was diagnosed in 1,1% of the cattle. Both trematodosis were most frequent in females (p = 0,03). Freshwater molluscs Ampullariidae, Physidae and Planorbidae without digenea larval stages were identified. It was concluded that dual-purpose cattle from La Candelaria farm are exposed to F. hepatica and Paramphistomidae and are probably infected on the farm grounds. Paramphistomidae was more prevalent than F. hepatica, which agrees with other studies in herds from the Colombian high Andean tropics. Paramphistomidae was found in all age groups.

The objective of this study was to estimate (co)variances and genetic parameters and to predict genetic trends for weight at 120 (W120), 210 (W210), 365 (W365), and 450 (W450) days of age in Nelore cattle raised in the northern region of Brazil. The database comprised records of 30,387 animals born between 2000 and 2013 in the Brazilian North. Estimates were calculated by the Restricted Maximum Likelihood (REML) method, in single- and multi-trait analyses in an animal model. Heritability as obtained using single- and multi-trait models for W120 (0.22 and 0.31), W210 (0.20 and 0.34), W365 (0.51 and 0.51), and W450 (0.49 and 0.51) indicated moderate to high magnitudes, with the possibility of genetic selection and incorporation into the herd. Genetic correlations between growth traits were favorable, ranging from 0.78 to 0.96. Genetic trends for W120 and W210 varied largely, from -0.31 to 4.68 and -0.53 to 7.62 kg/year, respectively. Smaller fluctuations were observed in genetic trends for W365 and W450, which ranged from -1.08 to 10.90 and -1.29 to 12.51 kg/year, respectively. Selection for W365 and W450 proved to be the criterion of choice for Nelore herds raised in the region; however, it may compromise adult performance because of higher costs and time for production. A thorough analysis of mattings is recommended to allow the selection of earlier-developing animals.(AU)

This study aims to determine the prevalence of Fasciola hepatica in cattle from the Abancay, Curahuasi, and Tamburco districts in the Abancay province, Apurímac, Peru, during the 2018 rainy season, and the association between prevalence and age, breed, and district of origin. In total, 295 stool samples were collected, namely 34 from Tamburco, 193 from Curahuasi and 68 from Abancay. For coproparasitological evaluation, the four-sieve sedimentation technique described by Girão and Ueno was used. The total prevalence of F. hepatica in the cattle sampled in this study was 50.8% (150/295), and the prevalence by district was 42.6% (29/68) in Abancay, 53.8% (104/193) in Curahuasi, and 50% (17/34) in Tamburco. No significant association was found with the variable district of origin (p<0.05). However, using a bivariate logistic regression analysis, a significant association was found between F. hepatica prevalence and the breed variable (p=0.008). A similar significant association with the breed variable (p=0.007) was also found using a multiple logistic regression analysis. The high prevalence of F. hepatica identified in this study is consistent with previous reports made in the Apurímac Region, an area considered hyperendemic for the parasite, thus highlighting the need for effective health programs to control disease distribution, which may have an economic and, because of its zoonotic character, public health impact.

The use of blood metabolites (BM), fecal starch (FS), and apparent digestion of starch, (ATTSD) as indicators of feed efficiency (FE) in beef cattle in the feedlot was studied. Fourteen bulls were used, originating in an industrial cross, without a defined racial group, with mean body weight of 284.86kg, individually fed, being evaluated in a 42-day confinement system. After the evaluation, the animals were divided into two groups according to the individual FE: high feed efficiency (HE) and low feed efficiency (LE). There was a difference between the groups in the variables FE, feed conversion (FC), final weight (FW), and daily weight gain (DWG). The FE had a positive correlation with DWG, FC, and FW. There was no difference between the groups for the variables BM, FS, and ATTSD, nor was there any correlation between these variables and FE. Considering the feed cost, the HE animals proved more profitable. BM, FS, and ATTSD did not statistically show potential to be used as indicators of FE, despite the evidence of numerical differences of these variables between the different groups, tendency of correlations with FE, and discriminating function with potential assertiveness.(AU)

ABSTRACT Serological controls for diseases of viral origin in animal production systems and the identification of factors associated with infections are decisive elements to establish prevention and control measures. The aim of this study was to establish the serological status for Bovine Viral Diarrhea (BVD) and Enzootic Bovine Leukemia (EBL) viruses in Blanco Orejinegro (BON) cattle from Colombia, and to identify the factors associated with seropositivity. A cross-sectional study was conducted with a total of 498 animals of all age groups and physiological states of the BON breed were selected, belonging to 14 herds located in 6 states of Colombia, in which a survey with 27 questions was conducted. By means of the chi-square test, possible factors associated with seropositivity against the 2 viruses were identified. A sample of 4 ml of blood was taken from each animal to extract plasma and make indirect Elisa tests to detect antibodies against both pathogens. General seropositivity of 27,1% was obtained for EBL, finding as factors associated with seropositivity the inadequate disposition of placental tissues after delivery of the cows and the non-performance of serological tests on new animals entering the herd. For BVD, seropositivity obtained was 50,6%, and the factors associated with seropositivity identified were having had a history of the disease in the herd, and using semen from bulls that are not known to be free for the infection. We suggest establishing control measures considering the factors associated with each viral infection to limit their expansion in the BON cattle production systems of Colombia.

This report aims to discuss the occurrence of protozoan infestation in a Girolanda cattle herd in the state of Bahia / BA. Bovine trypanosomiasis is a severe disease that is harmful to cattle, inducing large productive and economic losses. The animals are affected by protozoa of the genus Trypanosoma spp., giving rise to infestations in the vast majority of them in a silent and lasting way. In the Bahian rural property described, 10% of adult cows died and the rest of the animals showed clinical signs of anemia, drop in milk production, low body score, apathy, among others. Among the information obtained through the farm manager, who helped in the diagnosis and in ordering tests for the respective disease were: purchase of cattle from the region of the state of Minas Gerais and dubious management of oxytocin for improvements in milk production (suspected shared needles). Faced with the suspicion and in conjunction with the information obtained, laboratory tests and molecular tests were carried out to confirm the suspicion. After the examinations, it was found that the animals were affected by protozoa of the genus Trypanossoma vivax. The animals were treated with volume replenishers and drugs such as isometamidium chloride in a single dose calculated according to the body weight.

BACKGROUND: This study aimed to explore genetic polymorphisms of the CCKAR gene and their relationship with the growth and development of Qinchuan cattle which could be used as molecular markers for the improvement of the breeding of Qinchuan cattle. RESULTS: Here, we have identified seven single nucleotide polymorphisms (SNPs) at loci g. 1463 C>G; g. 1532 T>A; g. 1570 G>A; g. 1594 C>A; g. 1640 T>C; g. 1677 G>C; and g. 1735 C>T in the coding region of the bovine CCKAR gene. The frequencies identified on allelic and genotypic characteristics have shown that all seven SNPs diverged from the Hardy-Weinberg-Equilibrium. The SNP2, SNP3, SNP6 and SNP7 had the lowest polymorphism information content values, and remaining SNPs were found to be moderate (0.25 < PIC < 0.50). The genotype CG in SNP1 at loci g.1463 C>G had the greatest association with WH, HW, CD and CCF, while the genotype TA at the very same loci was associated with BFT, ULA and IMF content in Qinchuan cattle. The CCKAR gene expression level in adipose tissue, small intestine, liver and skeleton muscle was found to be higher, whereas, the expression level of mRNA in organs of other digestive system including reticulum, abomasum and omasum was moderate. Some expression of CCKAR mRNA was found in the large intestine, kidney and rumen. CONCLUSIONS: In summary, our finding suggested that the CCKAR gene could be used as a potential candidate for the improvement of carcass quality and body measurements of Qinchuan cattle.

BACKGROUND: Transmembrane protein 95 (TMEM95) plays a role in male fertility. Previous studies showed that genes with a significant impact on reproductive traits can also affect the growth traits of livestock. Thus, we speculated that the genetic variation of TMEM95 gene may have effects on growth traits of cattle. RESULTS: Two SNPs were genotyped. The rs136174626 and rs41904693 were in the intron 4 and 30 -untranslated region, respectively. The linkage disequilibrium analysis illustrated that these two loci were not linked. The rs136174626 was associated with six growth traits of Nanyang cattle, four traits of Luxi cattle, and three traits of Ji'an cattle. For rs41904693 locus, the GG individuals had greater body height and abdominal girth in Ji' an cattle than TT and TG individuals. In Jinnan cattle, GG and TT individuals had greater body height, height at hip cross, body length, and heart girth than TG individuals. The potential splice site prediction results suggest that the rs136174626 may influence the splicing efficiency of TMEM95, and the miRNA binding site prediction results showed that the rs41904693 may influence the expression of TMEM95 by affecting the binding efficiency of Bta-miR-1584 and TMEM95 30 -UTR. CONCLUSIONS: The findings of the study suggested that the two SNPs in TMEM95 could be a reliable basis for molecular breeding in cattle.

Abstract Objective: To isolate STEC in stool samples from cattle in Ulloa, Valle del Cauca, and to detect virulence factors associated with its pathogenesis. Materials and methods: We took 21 samples from cattle, which were taken directly from the rectum of the animal using swabs. The samples were processed until obtaining pure colonies and evaluated for the presence of the stx1, stx2, eae, saa and hlyA genes by PCR. Afterward, the cytotoxic effect of positive samples were evaluated on Vero cells (ATCC-CCL- 81.4). Results: We observed that from the 21 stools samples, 12 presented bacteria with one or both stx genes. A total of 106 isolates of STEC were obtained and differences among each other were observed regarding the presence and absence of the virulence genes, obtaining five combinations of genes. We found that 48 isolates presented only stx2 gene and 58 presented both the stx1 and stx2 gene. Regarding the other virulence genes, the hlyA gene was detected in 44 isolates and the saa gene was detected in 57 isolates. Conclusions: All the STEC supernatants showed cytotoxic activity on Vero cells, while in its absence the cells formed monolayer after 48 h of incubation. This work is the first report in Colombia that provides information about the presence of STEC in stool cattle, virulence genes and its potential cytotoxic effect in native strains.

The reproductive efficiency of livestock is the basis for the success of livestock, dairy or beef, and having high reproductive performance depends on several factors within the production system and the presence of infectious diseases of the reproductive sphere in the herd is one of the factors that can compromise that efficiency. The aim of this study was to use molecular biology as a diagnostic tool for the detection of Leptospira spp. DNA in cows with reproductive disorders on a rural property in the municipality of Boca do Acre, Amazonas, Brazil. Vaginal mucus was collected from nine Nelore breeding cows with a history of abortion and birth of weak calves submitted to DNA extraction and nested-PCR technique for 16S gene amplification at the bacterial genus level. Of the nine samples analyzed, five (55.55%) amplified a product of 331bp. The municipality of Boca do Acre is bordered by Peru and Bolivia, and knowledge of the prevalence of the disease, serovars, and circulating Leptospira species is essential for the adoption of measures related to animal husbandry, as well as health education for ranchers and their workers to avoid a possible occupational infection since this disease is considered an important zoonosis. New molecular studies using primers that allow the identification of the Leptospira species and mainly pathogenic species should be conducted in this region in order to elucidate the possible species of this etiological agent and the possible reservoirs of the disease to begin the understanding of the epidemiology of this disease in cattle in this region of border.

The application of spatial and temporal analysis in epidemiology aims to understand the causes and consequences of spatial and temporal heterogeneity in studies of infectious diseases. Bovine tuberculosis (bTB) is a chronic and progressive infectious disease caused by Mycobacterium bovis and is an important zoonosis worldwide. The aim of this study was to conduct spatial and temporal analyses of a secondary database of bTB-positive cases registered by the Federal Inspection Service in two slaughterhouses from the West of Minas mesoregion of Minas Gerais state, Brazil, from 2008 to 2012 and to suggest its use in epidemiological surveillance. The culled cattle with bTB macroscopic lesions during post mortem inspection were considered positive in this study. The data used were the positive cases registered on the "Sistema de Informações Gerenciais do Serviço de Inspeção Federal" (SIG/SIF - Management Information System of the Federal Inspection Service) of the "Ministério da Agricultura, Pecuária e Abastecimento" (MAPA - Ministry of Agriculture, Livestock and Supplies), the number of animals slaughtered monthly and their municipalities of origin, per slaughterhouse. The prevalence (%) of cases per cattle slaughtered was calculated, and the relationship between these cases of bTB and cases of human tuberculosis (hTB) and the GDP per capita of the mesoregions comprising the municipalities surveyed was evaluated. The prevalence was 1,030 cases of bTB (0.28%). The lowest case numbers and prevalence rates were obtained in 2010, while the highest rates were observed in 2011. Slaughterhouse 1 showed stationarity, while Slaughterhouse 2 showed an increasing annual trend for data since October 2009. A statistical correlation between the numbers of cases of bovine and human TB (p=0.006, r=0.148) was observed. The prevalence for Slaughterhouses 1 and 2 were not correlated (p>0.05). The municipalities of origin demonstrated randomness in their spatial distribution. SIG/SIF-MAPA may be used for the analysis of spatial and temporal distribution to contribute to the monitoring of animal health services with information on the prevalence of culling for bTB in the West of Minas, South/Southwest of Minas, Campo das Vertentes and Zona da Mata mesoregions of Minas Gerais, Brazil.(AU)

At present, there is a concern about the quality of milk and diseases related to its consumption, as it can generate discomfort and allergic reactions in some individuals due to its protein components. Thus, the present study was developed to identify the allele and genotype frequencies of genes for ß-casein, A1 and A2, in dairy herds in the region of Araguaína-TO, Brazil. Genetic material from 421 animals (crossbred dairy cattle in lactation) was used. All animals were numbered for identification, and DNA samples were extracted from hair bulbs. Samples for two markers from the polymorphic regions were characterized and confirmed by real-time PCR using the ABI Prism® 7500 Sequence Detection System (Applied Biosystems). Allele and genotype frequencies were determined using the TaqMan™ detection system, where the primer and probe release different fluorescence signals for each allele of the polymorphism. The sampled herd showed frequencies of 28.27% for the A1 allele and 71.73% for the A2 allele. Genotype frequencies were 52.96% (223/421) for A2A2; 37.53% (158/421) for the A1A2 genotype; and 9.50% (40/421) for the A1A1 genotype. The frequency of the A1 allele for ß-casein in dairy herds from the northern region of Tocantins was low and is per the results of previous studies. Although the A2A2 genotype of ß-casein had a high relative frequency, the A1A2 genotype is still rather frequent, warranting greater selection pressure.(AU)

This study aimed to establish the prevalence of animals persistently infected (PI) with bovine viral diarrhea virus (BVDV) in dairy farms at Parana State, Brazil. Samples were collected from 6,465 female Holstein Friesian Dairy Cattle, including animals less than two years old, females over two years old who had not given birth at the farm, and mothers of calves diagnosed as persistently infected. The cattle came from 40 dairy herds distributed in 10 municipalities in the State of Paraná. The samples were obtained from May 2015 to August 2018. The diagnosis of PI animals was made with an antigen-capture ELISA test. We detected PI animals in fifteen herds sampled (37.5%), ranging from one to sixteen animals per herd. The prevalence in Parana State's municipalities was 1.78%, ranging from 0.3 to 8.9% at positive herds. The analysis of the individual herds shows significant dissemination of the BVDV in Parana's municipalities, including endemic areas. With this, we highlight the need for measures to raise awareness among producers about the existence and importance of bovine viral diarrhea (BVD) in dairy herds, reinforcing the PI animals' role in disease epidemiology and the economic impact caused by the maintenance of these farm animals.(AU)

Fipronil was registered in Uruguay in 1997, and, since then, it has been used for the control of Haematobia irritans irritans and Rhipicephalus microplus. The susceptibility of H. irritants to this drug has not been evaluated. Therefore, the goal of the present study was to evaluate the resistance of H. irritans to fipronil. Additionally, a survey was carried out with the farmers to evaluate the use of fipronil for H. irritans control in the ranches where the flies came from. For the bioassays, 31 field populations of H. irritans were exposed to 10 concentrations of fipronil (3.2-16.0μg.cm2), and their LC50 values were calculated using probit analysis. A bioassay was performed with horn flies from the susceptible colony maintained at the USDA-ARS Knipling-Bushland U.S. Livestock Insects Research Laboratory for comparison and calculation of resistance ratios (RRs). All 31 field populations surveyed in the study were susceptible to fipronil, with resistance ratios ranging from <0.5 to 2.2. Four populations with RRs >1 did not differ significantly from the susceptible strain. A single population showed an RR >2.2. Overall, the survey shows that fipronil was mostly used for R. microplus control, and in only three ranches, which were free of R. microplus, was fipronil used for horn fly control. Seventeen farmers did not use fipronil at all in the last three years. It is concluded that, in Uruguay, field populations of horn flies remain susceptible to fipronil.(AU)

Diseases of the bovine digestive system make up an important group of diseases, often being responsible for significant economic losses in the livestock sector. The current work aimed to carry out a retrospective study of the diseases of the digestive system in cattle diagnosed at the "Clínica de Bovinos de Garanhuns", "Universidade Federal Rural de Pernambuco", from January 1999 to December 2018. In this period, a total of 9,343 cattle were admitted to the CBG-UFRPE, of which 2,238 (24.0%) were diagnosed with diseases of the digestive system. In two decades, the total number of cattle admitted annually by the institution almost doubled as well as the number of cases of digestive diseases. The diseases categorized as mechanical/motor were the most prevalent (33.6%), followed by fermentative (28.4%), gastroenteritis (16.0%), esophageal diseases (9.7%), and diseases of the oral cavity (4.5%). Lesions of the rectum and anus, congenital alterations, and other digestive diseases showed relative frequencies below 4%. Traumatic reticulitis and its sequelae (14.5%) are the most prevalent disorders, followed by simple indigestion (10.1%), esophageal and intestinal obstructive disorders (9.0%), non-specific gastroenteritis (8.6%), displaced abomasum (RDA and LDA) (5.5%), and frothy bloat, ruminal lactic acidosis, and impaction of the forestomach and abomasum, which represented approximately 5% each. In general, these diseases presented a lethality rate of 46.0%, with emphasis on mechanical/motor diseases with a lethality rate of 73.8%. In general, diseases were more prevalent in females, crossbreeds, aged over 24 months, and raised in a semi-intensive system. Diseases of the digestive system increased over the years studied, representing a considerable portion of the diseases that affect cattle raised in this region, which makes up the main milk basin in the State of Pernambuco, confirming its economic and social impact in the region.(AU)
As lesões do reto e ânus, as alterações congênitas e outras enfermidades digestórias apresentaram frequências relativas inferiores a 4%.
