ABSTRACT
Six compounds were isolated from aqueous extract of wine-processed Corni Fructus through silica gel, ODS column chromatography, Sephadex LH-20 gel column chromatography, reverse phase preparative HPLC and other chromatographic separation technologies. Their structures were identified with multiple spectroscopical methods including HR-ESI-MS, UV, IR, NMR and ECD and so on. Their structures were established as pinoresinoside B(1), cornusgallicacid A(2),(+)-isolariciresinol-9'-O-β-glucopyranoside(3),(-)-isolariciresinol 3α-O-β-D-glucopyranoside(4),(7R,8S)-dihydrodehydrodiconiferyl alcohol 9-O-β-D-glucopyranoside(5), and(-)-seco isolariciresinol-9'-O-β-D-glucopyranoside(6). Among them, compounds 1 and 2 were two new compounds. The biological activity evaluation results showed that compounds 2 and 6 had strong DPPH free radical scavenging ability, with EC_(50) values of(4.18±1.96) and(21.45±1.19) μmol·L~(-1), respectively. Compounds 1 and 2 had protective effects on H_2O_2-induced oxidative damage in NRK-52E cells in a dose-dependent manner, and the cell survival rate of compound 2 at 100 μmol·L~(-1) was 96.09%±1.77%.

Polyynes, such as facarindiol (FAD) and oplopandiol (OPD), are responsible for anticancer activities of Oplopanax elatus (O. elatus). A novel approach to pharmacokinetics determination of the two natural polyynes in rats was developed and validated using a liquid chromatography-electrospray ionization-mass spectrometry (LC-MS) method. Biosamples were prepared by liquid-liquid extraction using ethyl acetate/n-hexane (V : V = 9 : 1) and the analytes were eluted on an Agilent ZORBAX Eclipse Plus C18 threaded column (4.6 mm × 50 mm, 1.8 μm) with the mobile phase of acetonitrile-0.1% aqueous formic acid at a flow-rate of 0.5 mL·min(-1) within a total run time of 11 min. All analytes were simultaneously monitored in a single-quadrupole mass spectrometer in the selected ion monitoring (SIM) mode using electrospray source in positive mode. The method was demonstrated to be rapid, sensitive, and reliable, and it was successfully applied to the pharmacokinetic studies of the two polyynes in rat plasma after oral administration of polyynes extract of O. elatus.

This study aimed to conduct measurement uncertainty assessment of a new method for determination of Sudan colorants (Sudan I, II, III and IV) in food by high performance liquid chromatography (HPLC). Samples were extracted with organic solvents (hexane, 20% acetone) and first purified by magnesium trisilicate (2MgO·3SiO2). The Sudan colorants (Sudan I-IV) were also initially separated on C8 by gradient elution using acetonitrile and 0.1% (v/v) formic acid aqueous solution as the mobile phases and detected with diode-array detector (DAD). The uncertainty of mathematical model of Sudan I, II, III and IV is based on EURACHEM guidelines. The sources and components of uncertainty were calculated. The experiment gave a good linear relationship over the concentration from 0.4 to 4.0 μg/mL and spiked recoveries were from 74.0% to 97.5%. The limits of determination (LOD) were 48, 61, 36, 58 μg/kg for the four analytes, respectively. The total uncertainty of Sudan colorants (Sudan I, II, III and IV) was 810±30.8, 790±28.4, 750±27.0, 730±50.0 μg/kg, respectively. The recovery uncertainty was the most significant factor contributing to the total uncertainty. The developed method is simple, rapid, and highly sensitive. It can be used for the determination of trace Sudan dyes in food samples. The sources of uncertainty have been identified and uncertainty components have been simplified and considered.

This paper analyzes the parental care dimension and the relationship with the decisions on (non)-vaccination of their children in the context of high-income and well-schooled families in São Paulo city/state. The research adopts the qualitative approach, using in-depth interviews conducted with 15 couples that were divided into three groups: vaccinators, selective vaccinators and non-vaccinators. The analytic-interpretative study of the data was performed by means of content analysis and in line with the benchmarks of care of health and family. For all the couples analyzed, the option of (non)-vaccination of their children is perceived as parental care and protection of the child. However, for the vaccinators, protection is to vaccinate their children; for the selective vaccinators, protection is to study case by case; and for the non-vaccinators, protection is not to vaccinate their children, but to protect them against the risks of vaccination. The study also revealed that the reasons for non-vaccination, selection and/or postponement of the vaccination schedule were similar to those found in the international literature. The study highlights the importance of socio-cultural comprehension of (non)-acceptance of vaccination in the context of parental care.

The study was conducted to evaluate the status of worker exposure to polycyclic aromatic hydrocarbons (PAHs) through the measurement of urinary metabolites such as 1-hydroxypyrene (OHP) and 2-naphthol. A survey using a questionnaire involving 326 workers with measurement of urinary metabolites of 1-OHP and 2-naphthol was conducted. The differences in urinary 1-OHP and 2-naphthol concentrations, and changes in work, smoking habits and lifestyle were analyzed. The number of male subjects was 314 (96.3%), the largest age group was the fifth decade (170 cases, 52.1%). The urinary 1-OHP and 2-naphthol concentrations were significantly higher in the production workers. The urinary 1-OHP and 2-naphthol concentrations were significantly higher in smokers. In a multiple regression model, log (1-OHP) increased in smokers and production workers, while log (2-naphthol) only increased in smokers. Our results suggest that workers in this factory were exposed to PAHs from non-occupational as well as occupational sources. The occupational exposure to PAHs can be reduced through the improvement of the process, but the exposure due to smoking can be prevented only by giving up smoking.

The postural risk factors for dentists include the ease of vision in the workplace, cold, vibration and mechanical pressure in tissues, incorrect posture, functional fixity, cognitive requirements and work-related organizational and psychosocial factors. The objective was to analyze the posture of endodontists at the workplace. Eighteen right-handed endodontists aged 25 to 60 years (34±3) participated in the study. Electromyography, kinemetry, ergonomic scales (RULA and Couto's checklist) and biophotogrammetry were used to analyze the posture of endodontists during root canal treatment of the maxillary right first and second molars using rotary and manual instrumentation. The variations observed in the electromyographic activities during the performance of rotary and manual techniques suggest that the fibers of the longissimus region, anterior and medium deltoid, medium trapezium, biceps, triceps brachii, brachioradialis and short thumb abductor muscles underwent adaptations to provide more accurate functional movements. Computerized kinemetry and biophotogrammetry showed that, as far as posture is concerned, rotary technique was more demanding than the manual technique. In conclusion, the group of endodontists evaluated in this study exhibited posture disorders regardless of whether the rotary or manual technique was used.

Sirtuin1 is an enzyme that deacetylates histones and several non-histone proteins including P53 during the stress. P300 is a member of the histone acetyl transferase family and enzyme that acetylates histones. Hereby, this study describes the potency combination of Salermide as a Sirtuin1 inhibitor and cholera toxin B [CTB] as a P300 activator to induce apoptosis Michigan Cancer Foundation-7 [MCF-7] and MRC-5. Cells were cultured and treated with a combination of Salermide and CTB respectively at concentrations of 80.56 and 85.43 micro mol/L based on inhibitory concentration 50 indexes at different times. The percentage of apoptotic cells were measured by flow cytometry. Real-time polymerase chain reaction was performed to estimate the messenger ribonucleic acid expression of Sirtuin1 and P300 in cells. Enzyme linked immunosorbent assay and Bradford protein techniques were used to detect the endogenous levels of total and acetylated P53 protein generated in both cell lines. Our findings indicated that the combination of two drugs could effectively induced apoptosis in MCF-7 significantly higher than MRC-5. We showed that expression of Sirtuin1 and P300 was dramatically down-regulated with increasing time by the combination of Salermide and CTB treatment in MCF-7, but not MRC-5. The acetylated and total P53 protein levels were increased more in MCF-7 than MRC-5 with incubated combination of drugs at different times. Combination of CTB and Salermide in 72 h through decreasing expression of Sirtuin1 and P300 genes induced acetylation of P53 protein and consequently showed the most apoptosis in MCF-7 cells, but it could be well-tolerated in MRC-5. Therefore, combination of drugs could be used as an anticancer agent

Influences of phenol, alpha-naphtol and beta-naphtol, which are toxic chemicals, on citric acid biosynthesis and biomass in the artificial culture setting of Aspergillus niger using batch fermenter are examined in the most favorable fermentation conditions, and a model is proposed. Addition of certain concentrations of phenol, alpha-naphtol and beta-naphtol to the culture increases the citric acid production. According to this model, maximum citric acid concentration is 48.3 g L-1 in a culture that does not contain any toxic chemicals, whereas the maximum concentrations obtained in cultures containing 25 mg L-1 phenol, 25 mg L-1 alpha-naphtol and 15 mg L-1 beta-naphtol are 62.5 g L-1, 78.1 g L-1 and 86.0 g L-1, respectively. Moreover, addition of toxic chemicals to the culture reduces fermentation time by 24 hrs.

OBJECTIVE: The objetive of this study is to assess the level of oxidative stress by the exposure to volatile organic compounds (VOCs) and polycyclic aromatic hydrocarbons (PAHs) in volunteers who cleaned up the crude oil spilled from the Hebei Spirit on the Coast of Taean County, Korea. METHODS: The study subjects were 46 volunteers and 37 controls from the National Institute of Environmental Research. A self-administered questionnaire was used for volunteers to examine their working environment and health effects. Urinary concentrations of hippuric acid, 1-hydroxypyrene, and 2-naphthol were measured as exposure markers for VOCs and PAHs, and urinary thiobarbituric acid reactive substances (TBARS) and 8-hydroxydeoxyguanosine (8-OHdG) levels were measured as oxidative stress markers. RESULTS: Sixty-four percent of the respondents presented a variety of symptoms, including sore eyes and throat, nausea, dizziness, headache, low back pain, and leg pain after participating in the cleanup operation. Urinary 2-naphthol and TBARS concentrations appeared higher in the volunteers. Univariate and multivariate analyses showed that urinary TBARS and 8-OHdG levels were positively correlated with urinary 2-naphthol and 1-hydroxypyrene concentrations respectively, especially among the volunteers. CONCLUSIONS: The result implies that participating in cleanup work of oil spills may have a possibility to induce oxidative damage by exposure to PAHs in crude oil.
