ABSTRACT

Abstract Crotalaria (Fabaceae) occurs abundantly in tropical and subtropical regions and has about 600 known species. These plants are widely used in agriculture, mainly as cover plants and green manures, in addition to their use in the management of phytonematodes. A striking feature of these species is the production of pyrrolizidine alkaloids (PAs), secondary allelochemicals involved in plant defense against herbivores. In Crotalaria species, monocrotaline is the predominant PA, which has many biological activities reported, including cytotoxicity, tumorigenicity, hepatotoxicity and neurotoxicity, with a wide range of ecological interactions. Thus, studies have sought to elucidate the effects of this compound to promote an increase in flora and fauna (mainly insects and nematodes) associated with agroecosystems, favoring the natural biological control. This review summarizes information about the monocrotaline, showing such effects in these environments, both above and below ground, and their potential use in pest management programs.


Resumo Crotalaria (Linnaeus, 1753) (Fabaceae) ocorre abundantemente em regiões tropicais e subtropicais e tem cerca de 600 espécies conhecidas. Estas plantas são amplamente utilizadas na agricultura, principalmente como cobertura e adubos verdes, além da sua utilização no manejo de fitonematoides. Uma característica marcante destas espécies é a produção de alcalóides pirrolizidinicos (APs), aleloquímicos secundários envolvidos na defesa das plantas contra os herbívoros. Nas espécies de Crotalaria, a monocrotalina é a AP predominante, que tem muitas atividades biológicas relatadas, incluindo citotoxicidade, tumorigenicidade, hepatotoxicidade e neurotoxicidade, além de uma vasta gama de interações ecológicas. Assim, estudos têm procurado elucidar os efeitos desse composto para promover um incremento na flora e fauna (principalmente insetos e nematoides) associados aos agroecossistemas, favorecendo o controle biológico natural. Esta revisão compila informações sobre a monocrotalina, mostrando tais efeitos nesses ambientes, tanto acima como abaixo do solo e a sua potencial utilização em programas de manejo de pragas.

ABSTRACT

Hepatic sinusoidal obstruction syndrome (HSOS) via exposure to pyrrolizidine alkaloids (PAs) is with high mortality and there is no effective treatment in clinics. Bear bile powder (BBP) is a famous traditional animal drug for curing a variety of hepatobiliary diseases such as cholestasis, inflammation, and fibrosis. Here, we aim to evaluate the protective effect of BBP against HSOS induced by senecionine, a highly hepatotoxic PA compound. Our results showed that BBP treatment protected mice from senecionine-induced HSOS dose-dependently, which was evident by improved liver histology including reduced infiltration of inflammatory cells and collagen positive cells, alleviated intrahepatic hemorrhage and hepatic sinusoidal endothelial cells, as well as decreased conventional serum liver function indicators. In addition, BBP treatment lowered matrix metalloproteinase 9 and pyrrole-protein adducts, two well-known markers positively associated with the severity of PA-induced HSOS. Further investigation showed that BBP treatment prevents the development of liver fibrosis by decreasing transforming growth factor beta and downstream fibrotic molecules. BBP treatment also alleviated senecionine-induced liver inflammation and lowered the pro-inflammatory cytokines, in which tauroursodeoxycholic acid played an important role. What's more, BBP treatment also decreased the accumulation of hydrophobic bile acids, such as cholic acid, taurocholic acid, glycocholic acid, as well. We concluded that BBP attenuates senecionine-induced HSOS in mice by repairing the bile acids homeostasis, preventing liver fibrosis, and alleviating liver inflammation. Our present study helps to pave the way to therapeutic approaches of the treatment of PA-induced liver injury in clinics.

ABSTRACT

This study aims to develop a UPLC-MS/MS method for simultaneous determination of six pyrrolizidine alkaloids(PAs)--intermedine N-oxide(ImNO), lycopsamine N-oxide(LyNO), seneciphylline(Sp), seneciphylline N-oxide(SpNO), senecionine N-oxide(SnNO), and senkirkine(Sk) in different parts of Emilia sonchifolia. UPLC conditions are as follows: ACQUITY UPLC HSS T3 column(2.1 mm×100 mm, 1.8 μm), mobile phase consisting of 0.05% formic acid and 2.5 mmol·L~(-1) ammonium formate in water(A)-0.05% formic acid and 2.5 mmol·L~(-1) ammonium formate in acetonitrile(B) for gradient elution. MS conditions are as below: electrospray ionization(ESI) in the positive ion mode, multiple reaction monitoring(MRM), and the content of the six PAs was calculated with the external standard method. The results suggested the differences in the six PAs among different parts of E. sonchifolia. Sk was detected in all the four parts, with similar content. SnNO also existed in all the four parts, but the content in roots was significantly higher than that in other parts. Sp and SpNO were found in both roots and flowers, with the content higher in the former than in the later. ImNO and LyNO were only found in leaves, and the content was low. Among the six components detected, ImNO, LyNO, and SpNO were found and determined for the first time, which enriched the toxic components and laid a scientific basis for the quality and safety evaluation of E. sonchifolia.

ABSTRACT

Pyrrolizidine alkaloids(PAs) are a kind of natural toxins, which can cause severe hepatotoxicity, pulmonary toxicity, genotoxicity, neurotoxicity, embryotoxicity and even death. Therefore, international organizations and countries such as World Health Organization have paid great attention to herbal medicines and preparations containing PAs. PAs are widely distributed in Chinese herb medicines and contained in hundreds of traditional Chinese medicine preparations. The content of adonifoline, the main PAs in Senecionis Scandentis Herba, shall be less than 0.004% in herbal medicines as described in Chinese pharmacopeia. However, there is no guidance in preparations which contain Senecionis Scandentis Herba. In this study, 14 preparations were analyzed by an UPLC-MS method. Among them, 8 preparations were found to contain adonifoline much higher than the content limits of such countries as Germany, Netherlands and New Zealand. And the highest contents of adonifoline were found in Qianbai Biyan Tablets and Qianbai Biyan Capsules, which are officially recorded in Chinese Pharmacopeia. The contents of adonifoline varied in different batches of the same preparations. The highest content was 156.10 μg·g~(-1) Qianbai Biyan Tablets, whose daily intake of adonifoline was up to 1 030.26 μg according to the recommended dosage of the preparation. Our results showed the potential risk of these preparations, and the content limit of adonifoline shall be inspected Chinese medicine preparations containing Senecionis Scandentis Herba.

ABSTRACT

Pyrrolizidine alkaloids(PAs) are a group of naturally occurring alkaloids with a pyrrolizidine skeleton which can be found in about 3% of the world's flowering plants. It is notorious that PAs are cause the hepatoxic and genotoxic-carcinogenic effects by taking PA-containing herbs, food and dietary supplements. In order to control the poisoning caused by PAs, European Medicines Agency has set a limit of intake of PAs from herbal medicinal products at 0.007 μg of 1,2-unsaturated PAs/kg body weight. Nonetheless, a systematic overview of the amount of PAs in the herb has not been provided. Therefore, this paper is to systematically review the current status of PAs content analysis of herbal medicines and foods reported in the literature, and to provide theoretical and experimental support for the safety risk assessment and control of PAs in Chinese herbal medicines.

ABSTRACT

A ingestão de Senecio spp. (maria-mole) é, possivelmente, a principal causa de morte de bovinos por agentes tóxicos nas regiões central e sul do Rio Grande do Sul. Ao considerar a limitação de informações acerca dessa condição no Oeste do Rio Grande do Sul, esse trabalho objetiva descrever os principais aspectos epidemiológicos e clínico-patológicos da seneciose em bovinos nessa região. O estudo foi realizado por meio da aplicação de questionários em 16 propriedades rurais de municípios da região que apresentaram casos suspeitos da intoxicação em bovinos, de agosto de 2011 a março de 2014. Durante as visitas as propriedades foram coletadas plantas do gênero Senecio para identificação botânica, bem como se procedeu a coleta de fragmentos de fígado através de biópsia transtorácica para confirmação da intoxicação. As espécies de Senecio mais frequentes nas propriedades foram S. brasiliensis e S. heterotrichius. De um total de 88 bovinos, de nove propriedades distintas, que apresentaram falha no ganho de peso, 69 animais (aproximadamente 80%) foram positivos para seneciose crônica por apresentarem lesões características da intoxicação, tais como fibrose periportal (78% dos casos), megalocitose (76% dos casos) e proliferação de ductos biliares (68% dos casos), classificadas entre discretas e acentuadas. O trabalho confirmou a ocorrência da doença, mesmo em bovinos sem sinais clínicos evidentes. Os resultados obtidos nessas avaliações foram fundamentais para orientar proprietários e técnicos quanto às principais características da doença e às formas de controle a serem adotadas. O emprego da biópsia hepática possibilitou o diagnóstico precoce da intoxicação e auxiliou os criadores quanto ao descarte mais criterioso de bovinos e a real situação da intoxicação no rebanho, minimizando as perdas econômicas.(AU)


The ingestion of Senecio spp. (ragwort) is perhaps the leading cause of death of cattle in central and southern Rio Grande do Sul, Brazil. Considering the limited information about this condition in the Western region of Rio Grande do Sul, Brazil, this paper describes main epidemiological clinical and pathological aspects of seneciosis in cattle. The assessments were made through questionnaires on 16 rural properties which had suspected cases of poisoning in cattle, from August 2011 to March 2014. During the visits were evaluated epidemiological aspects of poisoning and performed sample collection of Senecio plants for botanical identification, as well as collection of liver samples using transthoracic biopsy for confirmation of the poisoning. Senecio species most common on farms were S. brasiliensis and S. heterotrichius. From 88 cattle that failed to thrive on nine different farms, 69 animals (about 80%) were positive for chronic seneciosis with periportal fibrosis (78% of cases), megalocytosis (76% of cases) and with bile duct proliferation (68% of cases). Lesions were classified as mild, moderate or marked. The current study confirms the occurrence of this poisoning, even in cattle without evident clinical signs. The use of liver biopsy enabled the early diagnosis of poisoning and helped farmers carefully to dispose affectd cattle, as well as to recognize the real situation of poisoning in the herd and minimize economic losses.(AU)

ABSTRACT

Intoxicações por plantas do gênero Senecio representam uma importante causa de morte em animais de produção, sendo Senecio brasiliensis a espécie mais envolvida nos casos de intoxicação espontânea. Nesse trabalho, são descritos os aspectos epidemiológicos, clínicos e anatomopatológicos de um surto de intoxicação natural por Senecio brasiliensis em equinos. Dois equinos, de um total de dezoito, foram afetados. Os casos ocorreram em uma propriedade rural no município de São Martinho da Serra, Rio Grande do Sul, Brasil. Grande quantidade de exemplares de Senecio brasiliensis em estágio de brotação foi encontrada na área onde os equinos estavam. Os animais eram da raça Crioula, sendo uma égua com quatro anos e um potro com cinco meses. A égua apresentou evolução aguda da doença com duração de três dias e o potro evolução crônica de dois meses. As principais manifestações clínicas observadas em ambos os equinos incluíam apatia, anorexia, emagrecimento, desidratação e sinais neurológicos. Além disso, havia moderado edema subcutâneo na região ventral do abdômen e marcada icterícia nas mucosas ocular, oral e vulvar da égua e palidez das mucosas no potro. Na avaliação bioquímica do potro, havia acentuado aumento da atividade sérica da gamaglutamil transferase (119 U/L), aumento da fosfatase alcalina (434 U/L) e hipoalbuminemia (2,1g/dL). No hemograma do mesmo, observou-se leve anemia normocítica e normocrômica (30% de hematócrito; VCM: 39,5 fL; HCM: 15,3pg). As alterações presentes em ambas as necropsias foram observadas principalmente no fígado, que estava difusamente aumentado de tamanho, escuro e firme. Na superfície de corte, observaram-se acentuação do padrão lobular e hemorragia. Histologicamente, no fígado dos equinos, havia graus variados de necrose coagulativa, hemorragia, fibrose, proliferação de ductos biliares, hepatomegalocitose e bilestase. Nas diferentes seções analisadas do encéfalo, havia alterações astrocitárias restritas à substância cinzenta, predominantemente, no córtex telencefálico. Os astrócitos degenerados eram similares aos astrócitos de Alzheimer tipo II. O diagnóstico da intoxicação nesses casos foi baseado na epidemiologia, no quadro clínico, nas lesões de necropsia e nos achados histopatológicos. Cabe ressaltar que a necrose de coagulação e a discreta fibrose e proliferação de ductos biliares observados na égua, constituiu uma apresentação subaguda da intoxicação, enquanto que a fibrose, mais predominante no potro, caracterizou uma hepatopatia crônica.(AU)


Poisoning due to plants of the genus Senecio is considered an important cause of death in livestock. Senecio brasiliensis is the most common species involved in spontaneous cases. This paper describes the epidemiological, clinical and pathological aspects of a natural outbreak of intoxication with Senecio brasiliensis affecting two out of eighteen horses in a farm. The cases occurred in the municipality of Sao Martinho da Serra, Rio Grande do Sul State, Southern Brazil. A large amount of Senecio brasiliensis in its sprouting stage was found in the fields where the horses used to be kept. A four-year-old, Crioulo mare and a five-month-old, male, Crioulo foal were affected. The clinical course was acute in the mare, comprising three days and chronic in the foal, lasting for two months. The main clinical signs in both animals included apathy, anorexia, weight loss, dehydration and neurological signs. Moreover, subcutaneous edema was noted in the ventral abdomen and the mucous membranes were icteric in the mare and pale in the foal. The serum biochemistry in the foal showed hypoalbuminemia (2.1g/dL), increased activity of the serum alkaline phosphatase (434 U/L) and gamma glutamyl transferase (119 U/L). In the hemogram, a mild normocytic normochromic anemia was noted in the foal (Hct: 30%; MCV: 39.5 fL; MCH: 15.3pg). Necropsy findings observed in both horses included an enlarged, dark red and firm liver with extensive areas of hemorrhage and enhanced lobular pattern on cut surface. Histologically, the liver showed variable degrees of fibrosis, coagulative necrosis, biliary ducts hyperplasia, megalocytosis and cholestasis. In the brain, the astrocytes in the cerebrum were degenerated and similar to Alzheimer type II astrocytes. The diagnosis in this outbreak was established based on the epidemiology, clinical picture, gross and histologic findings. It is noteworthy that the coagulative necrosis, minimal fibrosis, and biliary duct hyperplasia observed in the mare are characteristic of a subacute presentation. On the other hand, the fibrosis that was more prominent in the foal characterizes a chronic hepatopathy.(AU)

ABSTRACT

Intoxicações por plantas do gênero Senecio representam uma importante causa de morte em bovinos no Rio Grande do Sul e estima-se que mais de 50% das mortes causadas por plantas tóxicas devem-se à intoxicação por Senecio spp. Nesse trabalho, são descritos os aspectos epidemiológicos, clínicos e anatomopatológicos de um surto de intoxicação natural por Senecio brasiliensis em bezerros e a caracterização e evolução das lesões hepáticas durante o surto. Um lote com 206 bovinos, fêmeas, sem raça definida, foi desmamado com aproximadamente quatro meses de idade e soltos em campo nativo com 25 hectares, contendo grande quantidade de Senecio brasiliensis em diferentes estágios. Os bovinos permaneceram nesse campo durante três meses e posteriormente foram transferidos para outra área. Na nova área, seis bezerras, com idades entre sete a oito meses, começaram a apresentar sinais clínicos de emagrecimento, fraqueza, diarreia e edema subcutâneo de declive na região submandibular. Os seis bovinos morreram em um período de 15 dias e um deles foi necropsiado. Macroscopicamente, foi visualizado o primeiro dos três padrões distintos de lesão hepática dos bovinos desse surto, que se caracterizava por um fígado aumentado de tamanho, com os bordos arredondados e com superfície capsular e de corte alaranjada. Histologicamente havia acentuada degeneração vacuolar hepatocelular difusa, bilestase, megalocitose e leve proliferação de ductos biliares e fibrose periportal. Dois meses após o início do surto, constatou-se que em um período de 20 dias adoeceram mais 28 bezerras e, dessas, 14 morreram. Os bovinos acometidos nessa segunda visita tinham idades entre nove a 10 meses. O principal sinal clínico observado foi fotossensibilização, e incluía graus variados de fotofobia, lacrimejamento, salivação excessiva, descarga ocular mucopurulenta, secreção nasal e glossite diftérica ventral. Das 14 bezerras mortas, realizou-se a necropsia de duas delas, sendo visualizado o segundo padrão de lesão hepática nesse surto (Padrão 2). Os fígados estavam com a superfície capsular lisa e esbranquiçada e moderadamente diminuídos de tamanho. Ao corte observou-se leve irregularidade do parênquima e áreas mais claras intercaladas com áreas avermelhadas. O aspecto histológico incluía acentuada fibrose periportal, marcada proliferação de ductos biliares, megalocitose, áreas multifocais de necrose de hepatócitos, hemorragia e lesões veno-oclusivas. A atividade sérica da GGT foi avaliada em 15 bovinos afetados nessa segunda visita à propriedade. Desses, 13 bezerras testadas apresentaram alterações nos valores, que variaram de 26-175 U/L. Em uma terceira visita à propriedade, sete meses após o início do surto, mais 27 bezerras adoeceram em um período de 40 dias e, dessas, 23 morreram, sendo três submetidas à necropsia. Evidenciou-se o terceiro padrão de lesão hepática. O fígado dos três bovinos estava diminuído de tamanho e acentuadamente firme. A superfície capsular era lisa e esbranquiçada, e a superfície de corte apresentava-se difusamente acastanhada. Lesões histológicas semelhantes ao segundo padrão foram também constatadas no fígado dessas três bezerras. Na quarta visita à propriedade, realizaram-se biópsias hepáticas nos 163 bovinos restantes do lote. Lesões hepáticas características da intoxicação estavam presentes em 103 dos 163 bovinos submetidos à biopsia hepática. Os bovinos afetados foram classificados em grupos de acordo com a severidade e os aspectos morfológicos observados. O diagnóstico de intoxicação nos bovinos desse estudo baseou-se nos achados epidemiológicos, clínicos e anatomopatológicos, associados ao histórico do consumo da planta e evolução das lesões.(AU)


Poisoning by Senecio spp. plants are an important cause of death in cattle in the State of Rio Grande do Sul, Brazil. It is estimated that over 50% of deaths caused by toxic plants are due to the ingestion of Senecio spp. The epidemiological, clinical and anatomopathological aspects of a spontaneous outbreak of Senecio brasiliensis in calves are described here. Characterization and evolution of liver lesions during the outbreak are also described. Two hundred and six 4-month-old female calves were weaned and placed in a 25-hectare pasture heavily infested by S. brasiliensis at of varying growth stages. The calves remained in this pasture for three months and were later transferred to another area where six calves aged 7-8- month-old developed clinical signs including unthriftiness, diarrhea, and subcutaneous dependent edema in the submandibular region. All these six affected calves died within 15 days from the onset of clinical signs and one of those was necropsied. Grossly the first of three patterns of hepatic lesions described in this report was observed and it was characterized by an enlarged liver, with round borders and an orange hue to the capsular and cut surfaces. Histologically there was marked diffuse vacuolar hepatocellular degeneration, bilestasis, hepatocellular megalocytosis, and mild bile duct proliferation and periportal fibrosis. Two months after the onset of the outbreak, in another visit to the farm, it was observed that an additional 28 calves got sick and 14 died. Affected calves observed in this second on-site visit were 9-10 month-old and their main clinical sign was photosensitization characterized by varying degrees of photophobia, tearing from the eyes, marked drooling, mucopurulent ocular discharge, increased nasal secretion, and ventral diphtheric glossitis. Two of the fourteen calves that died were necropsied and displayed the second pattern of hepatic lesions observed in the outbreak. Liver had smooth and whitish capsular surfaces and a moderate decrease in size. There was moderate irregularity to the cut surface of hepatic parenchyma and pale areas intercalated with red ones. Histologically there was marked periportal fibrosis, marked bile duct proliferation, hepatocellular megalocytosis, focal areas of hepatocellular necrosis, and hemorrhage and veno-occlusive changes. In this second on-site visit, GGT serum activity was determined in 15 affected calves and 13 of them had increased values (26-175 U/L). Seven months after the onset of the outbreak, a third visit was made to the farm when was learned that and additional 27 calves have been affected, 23 of which died within a period of 40 days. At the necropsy of three of these calves the third pattern of hepatic lesions were observed. The liver was decreased in size and markedly firm. The capsular surface was smooth and whitish and the capsular surface was diffusely tan. Histological lesions similar to those observed in the second pattern were seen in in the liver of these three calves. A fourth on-site visit to the farm was made and a liver biopsy was performed in each of the 163 calves remaining in the herd and hepatic lesions characteristic of Senecio poisoning were observed in the liver biopsy of 103 of them. Affected calves were classified in groups according to the severity and morphologic aspects of the observed hepatic lesions. The diagnosis of Senecio poisoning in calves of this study was based on epidemiology, clinical and anatomopathological findings associated with the history of consumption of the plant, and on the chronological development of the lesions.(AU)

Poisoning by Senecio spp. plants are an important cause of death in cattle in the State of Rio Grande do Sul, Brazil. The calves remained in this pasture for three months and were later transferred to another area where six calves aged 7-8- month-old developed clinical signs including unthriftiness, diarrhea, and subcutaneous dependent edema in the submandibular region. All these six affected calves died within 15 days from the onset of clinical signs and one of those was necropsied. Grossly the first of three patterns of hepatic lesions described in this report was observed and it was characterized by an enlarged liver, with round borders and an orange hue to the capsular and cut surfaces. Histologically there was marked diffuse vacuolar hepatocellular degeneration, bilestasis, hepatocellular megalocytosis, and mild bile duct proliferation and periportal fibrosis. Two months after the onset of the outbreak, in another visit to the farm, it was observed that an additional 28 calves got sick and 14 died. Affected calves observed in this second on-site visit were 9-10 month-old and their main clinical sign was photosensitization characterized by varying degrees of photophobia, tearing from the eyes, marked drooling, mucopurulent ocular discharge, increased nasal secretion, and ventral diphtheric glossitis. Intoxicações por plantas do gênero Senecio representam uma importante causa de morte em bovinos no Rio Grande do Sul e estima-se que mais de 50% das mortes causadas por plantas tóxicas devem-se à intoxicação por Senecio spp. Nesse trabalho, são descritos os aspectos epidemiológicos, clínicos e anatomopatológicos de um surto de intoxicação natural por Senecio brasiliensis em bezerros e a caracterização e evolução das lesões hepáticas durante o surto. Um lote com 206 bovinos, fêmeas, sem raça definida, foi desmamado com aproximadamente quatro meses de idade e soltos em campo nativo com 25 hectares, contendo grande quantidade de Senecio brasiliensis em diferentes estágios. Os bovinos permaneceram nesse campo durante três meses e posteriormente foram transferidos para outra área. Na nova área, seis bezerras, com idades entre sete a oito meses, começaram a apresentar sinais clínicos de emagrecimento, fraqueza, diarreia e edema subcutâneo de declive na região submandibular. Os seis bovinos morreram em um período de 15 dias e um deles foi necropsiado. Macroscopicamente, foi visualizado o primeiro dos três padrões distintos de lesão hepática dos bovinos desse surto, que se caracterizava por um fígado aumentado de tamanho, com os bordos arredondados e com superfície capsular e de corte alaranjada. Histologicamente havia acentuada degeneração vacuolar hepatocelular difusa, bilestase, megalocitose e leve proliferação de ductos biliares e fibrose periportal. Dois meses após o início do surto, constatou-se que em um período de 20 dias adoeceram mais 28 bezerras e, dessas, 14 morreram. Os bovinos acometidos nessa segunda visita tinham idades entre nove a 10 meses. O principal sinal clínico observado foi fotossensibilização, e incluía graus variados de fotofobia, lacrimejamento, salivação excessiva, descarga ocular mucopurulenta, secreção nasal e glossite diftérica ventral.[...]

A intoxicação por plantas é sabidamente uma das principais causas de morte de bovinos adultos no Brasil. Uma vez que pouco pode ser feito no que diz respeito ao tratamento destas intoxicações, os esforços devem se concentrar na adoção de medidas profiláticas alternativas, como a utilização de uma espécie menos sensível no local de ocorrência de determinadas plantas. Objetivou-se com esse trabalho verificar a sensibilidade de caprinos às crotalárias (Crotalaria spectabilis e Crotalaria juncea) para que estes possam ser utilizados na profilaxia da intoxicação por essas plantas em bovinos. Sementes de C. spectabilis e C. juncea foram fornecidas em doses únicas ou diárias a oito caprinos adultos jovens, de ambos os sexos, divididos aleatoriamente em dois grupos de quatro animais (um grupo para cada planta)...

ABSTRACT

Diversas espécies de Senecio estão amplamente difundidas nas pastagens de propriedades rurais do Sul do Brasil. Criadores dessa região relatam quedas nos índices reprodutivos dos rebanhos bovinos, muitas vezes de causas não determinadas. Várias plantas tóxicas são capazes de causar alterações reprodutivas diretas e indiretas em bovinos em diversos países, incluindo o Brasil, no entanto seus mecanismos patogenéticos ainda são pouco compreendidos. O objetivo desse trabalho é descrever lesões ovarianas em vacas com seneciose crônica proveniente de propriedades rurais da mesorregião Sudoeste Rio-grandense. Foram estudados 21 casos positivos de seneciose crônica diagnosticados entre 2011 e 2014. O estudo revelou que a seneciose crônica é a principal causa de morte de bovinos adultos na região. Quatro vacas prenhes apresentaram lesões hepáticas clássicas da intoxicação por Senecio spp. Essas vacas tiveram seus ovários avaliados histologicamente e células luteínicas grandes (CLG) desses ovários apresentavam megalocitose e pseudoinclusões nucleares. Algumas CLG apresentaram núcleos com até 23,69μm de diâmetro e o aumento no tamanho desses núcleos foi significativamente maior que os de vacas controle. Conclui-se que a intoxicação por Senecio spp. causa alterações ovarianas em vacas e é possível que a intoxicação cause perdas reprodutivas nos rebanhos bovinas da região...


Several species of Senecio are widely distributed on pasture lands in Southern Brazil. Farmers from this region are known to complain about declines in reproductive rates in cattl from undetermined causes. Several poisonous plants can cause direct and indirect reproductive disorders in cattle in several countries, including Brazil. However, their pathogenetic mechanisms are still poorly understood. The aim of this study is to describe ovarian lesions in cows with chronic seneciosis, from farms located in the Southwest Mesoregion of Rio Grande do Sul, a southern state in Brazil. Twenty-one cases of bovine chronic seneciosis diagnosed between 2011 and 2014 were analyzed. The study showed that chronic seneciosis is the major cause of death in adult cattle in the region. Four pregnant cows showing classical necropsy large luteal cells (LLG) from the ovaries of these four cows had marked megalocytosis and nuclear pseudo-inclusions. Some LLG showed nuclei with up to 23.69μm in diameter and the increase in size of these nuclei was significantly greater than measured those of control cows. It is concluded that the ingestion Senecio spp. induces ovarian changes in cows and the intoxication should be considered as a possible cause of reproductive failure in cattle herds from this region...

Descrevem-se dois surtos de intoxicação por Senecio madagascariensis Poir. diagnosticados em bovinos em outubro de 2013 na região sul do Rio Grande do Sul. A morbidade foi de 3,2% e de 6,1% respectivamente e a letalidade foi de 100%. Um terceiro caso da intoxicação ocorreu em uma propriedade na qual de 54 bovinos um morreu com sinais clínicos da intoxicação. Em todos os casos, os bovinos estavam em áreas altamente infestas por S. madagascariensis que se encontrava em floração. Os sinais clínicos caracterizaram-se por diarreia, tenesmo, opistótono e emagrecimento progressivo e a morte ocorreu entre 10 e 15 dias após o início dos sinais clínicos. Nas necropsias as lesões eram de edema do mesentério, das paredes do abomaso e do rúmen, e das paredes da vesícula biliar, além de fígado firme e com aspecto marmorizado. Histologicamente havia no fígado proliferação de tecido conjuntivo fibroso, principalmente nos espaços porta, megalocitose e hiperplasia de ductos biliares. A observação de grande quantidade de S. madagascariensis em várias propriedades nos municípios de Arroio Grande, Pedro Osório e Capão do Leão a partir do ano 2013 sugere que esta planta está em pleno processo de adaptação e disseminação nesta região e que outros surtos podem ocorrer nos próximos anos. Os surtos relatados aparentemente resultaram do consumo da planta durante o outono/inverno de 2013, quando a mesma estava já em floração. A quantificação dos alcaloides revelou a presença de 500 µg/g e 4000µg/g de planta seca de alcaloides pirrolizidínicos em duas das três propriedades com casos de seneciose. Acredita-se que a grande quantidade de planta existente nas áreas onde os animais estavam e a quantidade de alcaloides presentes na mesma foram fatores que determinaram a ocorrência dos surtos...


Two outbreaks of Senecio madagascariensis poisoning in cattle occurred in October 2013 in southern Brazil. Morbidity rate ranged were 3.2% and 6.1%, and lethality rate was 100%. A third case of poisoning occurred on a property in which out of 54 cattle one died with clinical signs of intoxication. In all cases, the cattle were in areas highly infested by S. madagascariensis that was in bloom. The clinical signs were diarrhea, tenesmus, opisthotonus, and progressive weight loss. Death occurred between 10 and 15 days after the onset of clinical signs. At necropsy, lesions were edema in the mesentery, in the wall of the rumen and abomasum, and in the walls of the gall bladder; the liver was firm with marbled aspect. Histologically, the liver had proliferation of fibrous connective tissue, especially in portal areas, hepatomegalocytosis, and bile duct hyperplasia. The observation of large number of S. madagascariensis in several farms in the counties of Arroio Grande, Pedro Osório and Capão do Leão reinforces that this plant is in the process of adaptation and dissemination in this region and that other outbreaks may occur in coming years. The cases reported here apparently resulted from the consumption of the plant during the fall/winter of 2013, when it was in bloom. The quantification of alkaloids in S. madagascariensis revealed the presence of 500µg/g and 4000µg/g of pyrrolizidine alkaloids in the dry plant from two properties. It is believed that the large amount of the plant in the areas where the animals were and the amount of pyrrolizidine alkaloids found were factors that contributed to the occurrence of outbreaks...

ABSTRACT

The purpose of this study is to evaluate the embryotoxicity of alkaloids in Senecionis Scandentis Hebra on in vitro cultured mouse embryos. Mouse whole embryo culture (WEC) was applied in this study. Post-implantation (8.5 d) mouse embryos were isolated from their mothers, and cultured in medium of immediately centrifuged serum (ICS) with different concentrations of seneciphylline (target concentrations were 100, 50, 25 and 12.5 μg x mL(-1)) or senkirkine (target concentrations were 50, 25 and 12.5 μg x mL(-1)) for 48 h. After culturing completed, the development and organic morphodifferentiation of the cultured embryos were evaluated microscopically. Treatment with seneciphylline and senkirkine had adverse effects on the development and organic morphodifferentiation of embryos. The effect also had clear dose-response. Alkaloidals in Senecionis Scandentis Hebra had embryotoxicity on cultured embryos, which indicated that pregnant people exposed to Senecionis Scandentis Hebra may get potential risk on fetus.

ABSTRACT

OBJECTIVE: To describe the CT and MRI features of hepatic sinusoidal obstruction syndrome (HSOS) caused by herbal medicine Gynura segetum. MATERIALS AND METHODS: The CT and MRI features of 16 consecutive Gynura segetum induced HSOS cases (12 men, 4 women) were analyzed. Eight patients had CT; three patients had MRI, and the remaining five patients had both CT and MRI examinations. Based on their clinical presentations and outcomes, the patients were classified into three categories: mild, moderate, and severe. The severity of the disease was also evaluated radiologically based on the abnormal hepatic patchy enhancement in post-contrast CT or MRI images. RESULTS: Ascites, patchy liver enhancement, and main right hepatic vein narrowing or occlusion were present in all 16 cases. Hepatomegaly and gallbladder wall thickening were present in 14 cases (87.5%, 14/16). Periportal high intensity on T2-weighted images was present in 6 cases (75%, 6/8). Normal liver parenchymal enhancement surrounding the main hepatic vein forming a clover-like sign was observed in 4 cases (25%, 4/16). The extent of patchy liver enhancement was statistically associated with clinical severity classification (kappa = 0.565). CONCLUSION: Ascites, patchy liver enhancement, and the main hepatic veins narrowing were the most frequent signs of herbal medicine induced HSOS. The grade of abnormal patchy liver enhancement was associated with the clinical severity.

ABSTRACT

<p><b>OBJECTIVE</b>To study the toxic effects of aqueous extract of Crotalariae Assamicae Semen (CAS), one of the pyrrolizidine alkaloid-containing Chinese herbal medicines, in rats and the possible mechanism in association with liver damage.</p><p><b>METHOD</b>The aqueous extract of CAS (CASE) was prepared by the conventional water extracting-alcohol precipitating method. The LD50 value of CASE in rats was determined by Kärber method. Rats were randomly divided into four groups in which three groups were orally administered with different doses of the CASE and one group with distilled water as control. Toxic effects were assessed by morphological, biochemical and histopathological changes. Moreover, in vitro metabolism using rat liver microsomes was also conducted and applied for the exploration of the underlying mechanism of liver damage.</p><p><b>RESULT</b>The LD50 value of CASE in Wistar rats was (2.36 +/- 0.26) g x kg(-1). The toxic effects were found in all groups of rats dosed with CASE, in which serum levels of ALT and AST were significantly elevated, and the obvious and dose-dependent damages in liver and lung were observed by histopathological examination. Moreover, the liver tissue-bound pyrroles were detected and generated in a dose-dependent manner, and the pyrrole metabolites observed in the in vitro microsomal metabolism. All the evidences suggested a strong correlation between metabolism and toxicity of CASE in rats.</p><p><b>CONCLUSION</b>CASE could induce the acute toxicity in rats, of which liver and lung were the major targets. Toxic effects were strongly correlated with pyrrolizidine alkaloids in CAS. The possible mechanism for its liver toxicity may be related to the formation of pyrrole metabolites as well as the corresponding tissue-binding products.</p>

ABSTRACT

Sheep are more resistant to poisoning by Senecio spp. than cattle and horses. To determine whether this resistance could be induced by ingestion of small and repeated doses of the plant and to know how long the resistance could last, three experiments with fresh green leaves and stalks of Senecio brasiliensis were conducted. In Experiment 1, to determine the minimum dose for acute poisoning, single doses of 60, 80, 90, 100 and 100g/kg body weight (bw) were administered to five 1 and a half-year-old male Corriedale sheep. The sheep fed 60g/kg bw and 80g/kg bw of S. brasiliensis did not get sick; however, the sheep that received 80g/kg bw, developed mild fibrosis and megalocytosis in liver biopsies performed 90, 120 and 150 days after the end of plant administration. The sheep that received 90 and 100g/kg bw showed clinical signs of anorexia, prostration, paddling movements and abdominal pain, and died 12-48 hours after the onset of clinical signs. At their necropsy, ascites, disseminated petechiae and accentuated lobular pattern of the liver were found, and histologicaly hemorrhagic centrilobular necrosis was observed. In Experiment 2, the dose of 100g/kg bw fractionated into 2, 5 and 10 daily doses was administered to three sheep respectively. The fractioned dosage did not cause clinical signs of poisoning in any of the sheep, but periportal fibrosis and moderate megalocytosis were observed in liver biopsy performed at day 60 after the end of the plant administration. No histological lesions were observed in hepatic biopsies of sheep that received 10 daily doses of 10g/kg bw. In Experiment 3, to determine whether the sheep become resistant to the acute form of poisoning, four sheep ingested daily doses of 15g/kg bw of S. brasiliensis during 30 days and daily doses of 30g/kg bw for 10 days. On the day following the last dose of 30g/kg bw, two sheep were challenged with 100g/kg bw of the plant in a single dose. No clinical signs were observed in both sheep and no histological lesions could be found in the liver biopsies obtained 15 and 30 days after administration of the challenge dose. The third and fourth sheep received the challenge dose (100g/kg bw) 15 and 45 days after the end of plant administration. These animals showed anorexia, abdominal pain, and died about 12 hours after the onset of clinical signs. Gross and microscopic lesions were similar to those observed in sheep from Experiment 1. The results of this study demonstrate that sheep are susceptible to acute S. brasiliensis poisoning at doses of 90 and 100g/kg bw, but they become resistant after the continuous ingestion of small daily doses. This resistance has been lost 15 days after the end of the plant ingestion. Natural acute poisoning by S. brasiliensis probably does not occur in sheep, because they would have to eat large amounts of the plant in a short period of time.


Ovinos são mais resistentes à intoxicação por Senecio spp. que bovinos e equinos. Para determinar se essa resistência é induzida pela ingestão de pequenas e repetidas doses da planta e se essa resistência é duradoura, foram realizados três experimentos com folhas e talos verdes de Senecio brasilienses. Para determinar a dose mínima que causa intoxicação aguda (experimento 1), foram administradas doses únicas de 60, 80, 90, 100 e 100g/kg de peso corporal (pc) a cinco ovinos, respectivamente. Os animais que receberam 60 e 80 g/kg de pc de S. brasiliensis não adoeceram, porém o ovino que recebeu 80g/kg de pc apresentava fibrose e megalocitose discretas nas biópsias realizadas aos 90, 120 e 150 dias do término da administração da planta. Os ovinos que receberam 90 e 100g/kg de pc apresentaram anorexia, prostração, movimentos de pedalagem, dor abdominal e morte 12-48 horas após o aparecimento dos sinais clínicos. Na necropsia havia ascite, petéquias disseminadas e acentuação do padrão lobular hepático. Histologicamente havia necrose hemorrágica centro-lobular. No Experimento 2 a dose capaz de causar a intoxicação aguda foi fracionada e administrada em duas, cinco e 10 doses diárias para 3 ovinos, respectivamente. A dose tóxica fracionada não provocou sinais clínicos de intoxicação em nenhum dos ovinos, porém havia fibrose periportal e megalocitose moderadas nas biopsias realizadas aos 60 dias do término da administração da planta, as quais não evoluíram. O ovino que recebeu a dose fracionada em 10 administrações não apresentou lesões histológicas nas biópsias. Para determinar se os ovinos tornam-se resistentes à forma aguda da intoxicação (experimento 3), foram administradas doses diárias de 15g/kg de pc por 30 dias e 30g/kg de pc por 10 dias a quatro ovinos. No dia seguinte à última administração dois ovinos receberam a dose única de 100g/kg de pc de S. brasiliensis, mas não adoeceram nem apresentaram lesões em biópsias realizadas 15 e 30 dias após o desafio. Entretanto, dois ovinos que receberam essa dose, 15 e 45 dias após a última administração da planta, respectivamente, apresentaram anorexia, dor abdominal, movimentos de pedalagem e morreram 12 horas após o início dos sinais clínicos. As lesões macro e microscópicas eram semelhantes às observadas nos ovinos do Experimento 1. Os resultados do presente trabalho demonstraram que, experimentalmente, ovinos intoxicam-se de forma aguda com doses únicas de 90-100g/kg de pc, mas são capazes de suportar essas doses após a ingestão contínua de pequenas quantidades da planta, no entanto, essa suposta resistência é perdida se os ovinos deixam de ingerir a mesma. Pode-se sugerir também que a intoxicação aguda natural, provavelmente, não ocorre devido à improbabilidade de ingestão, por um ovino, da quantidade necessária de planta em um período curto de tempo.

Foram realizados dois experimentos para determinar os efeitos tóxicos de diferentes doses de aflatoxinas em bezerros, considerando-se aspectos clínicos, produtivos e patológicos. ... No primeiro experimento, o ganho de peso dos bezerros recebendo AFB1 foi equivalente ao do grupo controle durante todo período experimental. ...Durante o período experimental, e três semanas após o término desse período, não foram observados sinais clínicos e alterações histopatológicas associadas ao consumo de aflatoxinas, em qualquer dos bezerros do grupo tratamento do primeiro experimento.... Níveis alterados da atividade sérica de FA e GGT foram observados em todos os bezerros do grupo tratamento durante grande parte do período experimental. Queda acentuada do nível da AS sérica foi observada na coleta do 49º dia do experimento no bezerro que recebia a maior dose de aflatoxina. Não foram observadas variações no hematócrito e na atividade sérica da AST, nem nos níveis séricos de proteína total, bilirrubina total e bilirrubina direta em qualquer dos bezerros desse experimento. Alterações histopatológicas nos bezerros intoxicados incluíram proliferação de ductos biliares, degeneração citoplasmática vacuolar consistente com acumulação hepatocelular de lipídios, fibrose periportal, ou em ponte, megalocitose, fibrose subendotelial das veias hepáticas terminais e edema. Achados de necropsia do bezerro recebendo a maior dose de AFB1 incluíram fígado levemente aumentado de tamanho, difusamente amarelo-claro e firme, discreta ascite, edema de mesentério e submucosa do abomaso. Os dados obtidos nesses experimentos permitem afirmar que doses de 500±100 ppb de AFB1 não causam alterações patológicas e produtivas em bezerros em condições experimentais, mas podem estar associadas à mínimas alterações bioquímicas, enquanto doses de 1.250, 2.500 e 5.000 ppb de aflatoxina B1 causam doença hepática crônica em bezerros em condições experimentais.


Two experiments were performed in order to determine the toxic effects of varying doses of aflatoxins in calves. Clinical, productive and pathologic aspects of affected calves were considered….During all the experimental period of the first experiment, the weight gain of the calves receiving AFB1 was equivalent to that of the control group.…During the whole experimental period and up to three weeks after the final of the experiment, no clinical signs or histopathological changes associated with the consumption of aflatoxins were observed in any of the calves of the first experiment….Increased activity of AF and GGT were observed in all the calves of the treated group during most part of the experimental period. A marked drop in the serum levels of SA was observed in the serum sampled on the 49º day of the experiment in the calf receiving. the largest dose of aflatoxin. No changes were observed regarding PCV, TP, total bilirubin, direct bilirubin and in the serum activity of AST in any of the calves of the second experiment. Histopathological changes in intoxicated calves included bile duct proliferation, cytoplasmic vacuolar hepatocelular degeneration consistent with hepatocelular deposit of lipids, periportal to bridging fibrosis, megalocytosis, subendothelial edema and fibrosis in terminal hepatic veins. Necropsy findings in the euthanatized calf which receive de largest doses of AFB1 included slight enlargement of the liver which was firm and diffusely light-yellow, mild ascites, and edema of the mesentery and of abomasal folds. Data stemmed from these two experiments allow to conclude that AFB1 doses of 500±100 in the ration do not cause pathologic changes or decrease in productivity in calves kept in experimental conditions, but can be associated to minimal serum biochemistry; while AFB1 doses of 1.250, 2.500 e 5.000 ppb in the ration cause chronic hepatic disease in calves in kept in experimental conditions.

ABSTRACT

<p><b>OBJECTIVE</b>To study the pyrrolizidine alkaloids of Senecio cannabifolius var. integrilifolius.</p><p><b>METHOD</b>Various chromatographic methods were used for the isolation and purification of chemical constitutents of the plant and their structures were elucidated on the basis of spectral analysis.</p><p><b>RESULT</b>Four compounds were determinated as jacobine(1), jacoline(2), jaconine(3), senecicannabine(4).</p><p><b>CONCLUSION</b>All compounds were isolated from this plant for the first time.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To develop an UPLC-MS method for the simultaneously quantitation of adonifoline and senecinine in Senecio herbs.</p><p><b>METHOD</b>UPLC-Micro 2000 was used for quantification in SIR mode under ESI+. Monocrotaline was used as the internal standard. Chromatography was performed on a Shiseido Capcell Pak MG (2.0 mm x 50 mm, 3 microm) column at 30 degrees C using an gradient elution of acetonitrile-0.5% formic acid in water at the flow rate of 0.3 mL x min(-1). The injection volume was 2 microL.</p><p><b>RESULT</b>Good linearity was obtained for quantitation of adonifoline over the range of 1.02-816.00 microg x L(-1) (r = 0.998 0). And recoveries at different concentration levels were between 95.73% and 103.0% with RSDs no more than 2.5%. For quantification of senecionine, the linear range was between 1.08-860.56 microg x L(-1) (r = 0.997 6). And recoveries at different concentration levels were between 95.67% and 101.5% with RSDs no more than 2.3%. Good reproducibility and precision were also achieved.</p><p><b>CONCLUSION</b>The new developed UPLC method is sensitive, accurate and reliable enough for the quantitation of adonifoline and senecionine in Senecio herbs thus can be used for the limit detection of pyrrolizidne alkaloids in S. scandens. It can also be used for the identification of fake drugs of S. scandens such as S. vulgaris. The developed method was served for the quality evaluation of Herba Senecionis Scandentis.</p>

ABSTRACT

It has been well-known that many medicinal plants used in traditional Chinese medicine contain hepatotoxic pyrrolizidine alkaloids (HPAs), and some even have been recorded in many editions of the Chinese Pharmacopoeia (ChP). In order to clarify the current status of these PAs-containing Chinese materia medica and proprietary Chinese formulae, the ChP 2010, the newest version, and the related safety issues were thoroughly investigated and analyzed on the current advances in research. Total nine crude drugs (not including the processed slices) were found to contain HPAs, which may be present in tens of Chinese proprietary drugs prepared with these crude drugs. Because of the lack of the alkaloid limitation in most monographs, their potential threats to human health may be underestimated. For this reason, attention should be drawn to the importance of the issue. The key point is to conduct the basic studies immediately on these PA-containing herbal plants or products, whose possible hazards need to be carefully assessed. Further efforts should also be made to elevate the criteria for quality control and ensure the drugs' safety in clinic for human health.
