ABSTRACT

Background: Potato peel extract has demonstrated the ability to reduce platelet aggregation in vitro, suggesting its potential as a dietary intervention for preventing atherothrombotic disorders. Objective: This study aims to evaluate the impact of a potato peel-rich diet on platelet aggregation. Methods: A randomized, crossover-controlled, open two-period study was carried out with the participation of 12 healthy volunteers. Platelet aggregation was assessed before and after a seven-day dietary intervention. Participants consumed either a diet rich in potato peel (2 g/kg/d) or acetylsalicylic acid (ASA) as a reference (100 mg/d). Platelet aggregation percentages were measured following stimulation with arachidonic acid (AA, 150 µg/mL), adenosine diphosphate (ADP, 10 µM), and collagen (COL, 10 µg/mL). Results: The potato peel-rich diet resulted in a slight but significant reduction in platelet aggregation when stimulated with arachidonic acid compared to baseline values (85.0±2.0% vs. 91.3±1.7%, p<0.05). This effect was less pronounced than the reduction achieved with ASA (16±1.9%, p<0.001). Conclusion: The administration of a diet rich in potato peel reduces platelet aggregation induced by arachidonic acid, suggesting its potential role in the prevention of atherothrombotic disorders.


Introducción: El extracto de cáscara de patata ha demostrado su capacidad para reducir la agregación plaquetaria in vitro, lo que sugiere su potencial como intervención dietética para prevenir trastornos aterotrombóticos. Objetivo: Evaluar el impacto de una dieta rica en cáscara de patata en la agregación plaquetaria. Materiales y métodos: Se llevó a cabo un estudio aleatorizado, controlado, cruzado y abierto con la participación de 12 voluntarios sanos. Se evaluó la agregación plaquetaria antes y después de una intervención dietética de siete días. Los participantes consumieron una dieta rica en cáscara de patata (2 g/kg/d) o ácido acetilsalicílico (ASA) como referente (100 mg/d). Se midieron los porcentajes de agregación plaquetaria después de la estimulación con ácido araquidónico (AA, 150 µg/mL), difosfato de adenosina (ADP, 10 µM) y colágeno (COL, 10 µg/mL). Resultados: La dieta rica en cáscara de patata resultó en una ligera pero significativa reducción en la agregación plaquetaria cuando se estimuló con ácido araquidónico en comparación con los valores iniciales (85,0 ± 2,0% vs. 91,3 ± 1,7%, p <0,05). Este efecto fue menos pronunciado que la reducción lograda con ASA (16 ± 1,9%, p <0,001). Conclusión: La administración de una dieta rica en cáscara de patata reduce la agregación plaquetaria inducida por ácido araquidónico, lo que sugiere su papel potencial en la prevención de trastornos aterotrombóticos.

Potato is a plant that has high nutrient demand during its cycle. Given the concern with the environment, due to the large amount of synthetic mineral fertilizer used in the crops, the objective of this work was to evaluate the efficiency of organomineral fertilizers in the cultivation of potato, cultivar Ágata, under cerrado conditions. The experimental design was in subdivided plots, consisted of four doses of organomineral fertilizer, corresponding to 40, 60, 80, 100 and 120% of the mineral fertilizer dose in the organomineral source (2800 kg ha-1 of formulated 3-32-6) and the subplots consisted of four collection seasons (61, 74, 89 and 110 days after planting) and four repetitions. Organomineral fertilizer with 40% of the recommended dose for potato cultivation provides higher yield of tubers in the Especial potato class and higher accumulation of total dry mass, in addition to providing satisfactory productivity for the tubers of higher commercial value. The use of organomineral fertilizers promotes the same behavior as mineral fertilizers, not interfering with potato development.

ABSTRACT

En Guatemala, la producción del cultivo de papa se ve afectada por los nematodos Globodera rostochiensis y Globo-dera pallida. La capacidad de ambas especies para formar quistes complica su control y provoca el aumento de sus poblaciones. En Guatemala se reporta la presencia de ambas especies de nematodos por identificación morfológica, sin embargo, no se ha realizado una confirmación molecular. Este es el primer estudio para validar la presencia de ambas especies de nematodos por PCR múltiple y la determinación de la diversidad y estructura genética de las poblaciones utilizando marcadores moleculares. Se realizaron muestreos en cuatro departamentos productores de papa del país. La identificación por PCR se realizó con el cebador común ITS5 y los cebadores PITSr3 específico para G. rostochiensisy PITSp4 para G. pallida. La caracterización molecular se realizó con el marcador AFLP. Se confirmó la presencia de las dos especies de nematodos en los cuatro departamentos. Los índices de diversidad Shannon y heterocigosidad esperada revelaron mayor diversidad genética en G. rostochiensis (H = 0.311, He = 0.301) que en G. pallida (H = 0.035, He = 0.223). Los métodos NJ, DAPC y PCA exhibieron una débil estructura entre las poblaciones de ambas especies de nematodos. Los resultados sugieren un patrón de dispersión desde Quetzaltenango hacia el resto del país, atribuido a la comercialización de semilla contaminada con nematodos. Se sugiere promover programas de socialización sobre los beneficios del uso de semilla certificada, además de constantes monitoreos moleculares para un diagnóstico certero de ambas especies de nematodos.


In Guatemala, potato crop production is affected by the nematodes Globodera rostochiensis and Globodera pallida. The ability of both species to form cysts complicates their control and causes an increase in their populations. In Guatemala, both species of nematodes have been reported by morphological identification; however, molecular confirmation has not been carried out. It is the first study to validate the presence of both nematode species by multiplex PCR and determine the diversity and genetic structure of the populations using molecular markers. Sampling was carried out in four pota-to-producing departments of the country. PCR identification was performed with the common primer ITS5 and the primers PITSr3 specific for G. rostochiensis and PITSp4 for G. pallida. We performed molecular characterization with the AFLP marker. We confirmed the presence of the two nematode species in the four departments. Shannon diversity and expected heterozygosity indices revealed higher genetic diversity in G. rostochiensis (H = 0.311, He = 0.301) than in G. pallida (H = 0.035, He = 0.223). The NJ, DAPC, and PCA methods exhibited weak structure among populations of both nematode species. The results suggest a dispersal pattern from Quetzaltenango to the rest of the country, attributed to the commer-cialization of seed contaminated with nematodes. We suggest promoting socialization programs on the benefits of using certified seeds and constant molecular monitoring for an accurate diagnosis of both species of nematodes.

Aims@#Potatoes are considered one of the most strategic vegetable crops all over the world. Alternaria alternata has recently contaminated certain potatoes farms in different regions in Egypt. Among thirteen samples from fifteen regions were studied in a precedent study. Our study was aimed to investigate the effect of Bacillus thuringiensis subsp. Kurosaki suspension on inhibiting the growth of the three tested isolates of A. alternata and minimizing their mycotoxins production in vitro using three isolates with three levels of highly, moderate and low pathogenicity with unequal amounts of dual mycotoxins production.@*Methodology and results@#Three isolates of A. alternata from three regions, Kom Hamada (KH3), Alamin (Alam1) and Nobaria (NO3), which were determined as a producer of tenuazonic acid (TeA) and alternariol monomethyl ether (AME) toxins. Bacillus thuringiensis (Bt) use as commercial fungicide was applied with three suspension concentrations (75, 150 and 300 μg/mL) as inhibitor for the two mycotoxins. Our results illustrated that the three tested isolates recorded high TeA and AME inhibition efficacies by increasing the Bt suspension concentration. The highest inhibitory concentration of Bt was at concentration 75 μg/mL for isolated from Nobaria third region (NO3) and Alam1 it was (99.83 and 99.74%) for mycotoxin (AME) while, TeA mycotoxin had the most inhibition percentage (99.58%) at concentration 150 μg/mL for the isolate (NO3).@*Conclusion, significance and impact of study@#The preliminary results of the study suggest that B. thuringiensis spores’ suspension with different concentrations can be used as anti-mycotoxigenic agents to inhibit the (TeA) and (AME) mycotoxins produced by Alternaria alternata.

Resumen La gota de la papa o tizón tardío es una de las enfermedades más agresivas que atacan los cultivos de papa y en poco tiempo los destruye, ocasionando enormes pérdidas económicas, es producida por el fitopatógeno Phytophthora infestans, oomyceto que ha tomado gran importancia debido a sus efectos devastadores y las dificultades para lograr su erradicación. Los fungicidas de naturaleza química representan un problema debido al uso inadecuado, lo que hace muy difícil eliminar la enfermedad por la aparición de nuevas especies resistentes. Existen nuevas alternativas para su control, basadas en el uso de sustancias de naturaleza vegetal, y la aplicación de nuevas herramientas capaces de realizar edición de genes, reprogramar o eliminar secuencias de ADN/ARN, favoreciendo así, la obtención de cultivos libres de sustancias tóxicas. En esta revisión, se presentan los métodos existentes de biocontrol: como aceites esenciales, metabolitos microbianos, herramientas moleculares y el uso de sustancias biodegradables que favorecen el manejo y prevención de plagas, que ayudan a mitigar la problemática ambiental generada por el uso de fungicidas de naturaleza química.


Abstract The drop of the potato or late blight is one of the most aggressive diseases that attack the potato crops and in a short time destroys them, causing great economic losses, it is produced by the phytopathogen Phytophthora infestans, oomyceto that has taken great importance due to its devastating effects and difficulties in achieving its eradication. Fungicides of a chemical nature represent a problem due to the affected use, which makes it very difficult to eliminate the disease due to the appearance of new resistant species. There are new alternatives for its control, based on the use of substances of the plant nature, and the application of new specific tools to perform gene editing, reprogram or eliminate DNA / RNA sequences, thus favoring the obtaining of cultures free of toxic substances . In this review, specific biocontrol methods are presented, such as essential oils, microbial metabolites, molecular tools and the use of biodegradable substances that favor the management and prevention of pests, which help mitigate the environmental problems generated by the use of fungicides from chemical nature.

Potato scab caused by different species of phytopathogenic Streptomyces is considered one of the main bacterial diseases of economic crop importance worldwide. Several studies are being carried out in order to control the disease, but until now, there is no efficient way to do this. Some management strategies have been investigated including application of chemical and biological products and utilization of resistant cultivars of potato but there are few reports about the impact of pH and irrigation regimes on the disease. The present study aimed to evaluate the effects of these last two factors on the incidence and severity of potato scab caused by S. scabiei, S. acidiscabies, Streptomyces sp., S. caviscabies and S. europaeiscabiei in assays at pH 4.0, 4.5, 5.0, 5.5, 6.5 and 7.5; and irrigation regimes of once a week, alternate days and daily in greenhouse conditions. The experimental design for the pH tests was randomized blocks arranged in a 5x2 factorial scheme, with 5 replications and 3x2 for the irrigation regimes with 5 replications. The pH tests showed significant differences between the treatments and pH 4,0 - 4,5 presented lower incidence and severity of the disease for the most species tested but no significant differences were observed between the irrigation regimes. The soil acidification is considered a classic strategy for management of the disease and the results obtained herein corroborated this hypothesis.

HIGHLIGHTS Chips from orange-fleshed sweet potato have a good acceptability. Drying process showed retention of carotenoids total content. Chips from drying or frying process showed high resistant starch content.


Abstract There is currently a great demand for industrialized products with functional properties, together with the increase in consumption of roots and sweet potato products. Sweet potatoes have a high content of resistant starch, while only the orange-fleshed roots also have a high content of carotenoids. Due to these, this work aimed to produce orange-fleshed sweet potato chips, by two processes: drying oven and immersion frying. The chips were evaluated for the content of resistant starch and carotenoids in nature and chips sweet potatoes, and evaluations of the physical attributes and sensory analysis of the chips. The drying process retained a greater content of total carotenoids. Fried chips can be considered high resistant starch content, even with a decrease in the content after this processing; they also showed more intense coloring and pleasant texture. There was a statistical difference between the varieties only regarding the content of carotenoids and resistant starch. Thereby, it can be concluded that the chips of both processing have good technological and functional qualities, and that the frying process presented best hardness which led to greater acceptability and purchase intention.

ABSTRACT

Minituber production is an important step to obtain high-quality seed potatoes. This production is carried out in a greenhouse using a large volume of commercial substrate acquired in specialized trade. An alternative to reduce the volume of substrate purchased for each production of minitubers would be to reuse it, which would be a way to optimize its use. The objective of this study was to evaluate the efficiency of reused substrates in the minituber production. The experiment was conducted in Tapira-MG with the Atlantic potato cultivar. The experimental design was a randomized block with six treatments and four replications, where each plot was represented by a box (0.15 x 0.4 x 0.5m: height, width, and length, respectively) containing substrates. Treatments consisted of formulation 04-14-08 rates (0, 25, 50, 75, and 100 g box-1) added to reused substrate and a control consisting of a new virgin substrate. At 45 days after transplanting, the following characteristics were evaluated: stem size (cm), and quantities of fresh and dry matter of aerial part of plants. Harvesting was performed manually, counting, and calculating the yield of tubers according to class. The reuse substrate with enriched nutrient regardless of the rate, when compared to the use of a new virgin substrate, did not interfere in potato plant development and in the total productivity of minitubers of classes I, II, III, and IV. This can be a feasible alternative for optimizing the use of the virgin substrate. The rate of 25 g box-1 of 4-14-8 is sufficient to provide the needs of potato in nutrients, ensuring high productivity.


A produção de minitubérculos é uma etapa importante para obtenção de batata-semente de alta qualidade. Esta produção é realizada em casa de vegetação e utiliza grande volume de substrato comercial adquirido no comércio especializado. Uma alternativa para diminuir o volume de substrato adquirido a cada produção de minitubérculos seria a reutilização do mesmo, o que seria uma forma de otimizar o seu uso. Neste sentido, objetivou-se avaliar a eficiência de substratos reutilizados na produção de minitubérculos de batata. O experimento foi conduzido no município de Tapira-MG, com a cultivar Atlantic. O delineamento experimental foi o de blocos casualizados com seis tratamentos e quatro repetições, onde cada parcela foi representada por uma caixa (0,15 x 0,4 x 0,5 m: altura, largura e comprimento, respectivamente) contendo substratos. Os tratamentos foram constituídos por doses do formulado 04-14-08 (0, 25, 50, 75 e 100 g caixa-1) adicionadas ao substrato reutilizado e uma testemunha com substrato virgem. Aos 45 dias após o transplantio foram avaliadas as características: tamanho de haste (cm), matéria fresca e matéria seca da parte aérea. A colheita foi realizada manualmente e procedeu a contagem e a produtividade de tubérculos, em classes. O substrato reutilizado enriquecido com nutrientes, independentemente da dose, quando comparado ao substrato virgem, não interferiu no desenvolvimento da planta de batata e na produtividade total dos minitubérculos das classes I, II, III e IV. Esta pode ser uma alternativa viável para otimizar o uso do substrato virgem. A taxa de 25 g box-1 de 4-14-8 é suficiente para atender às necessidades de batata em nutrientes, garantindo alta produtividade.

The selection of potato plants (Solanum tuberosum L.) that are efficient in the use of phosphorus (P) plays an important role in increasing crop productivity, reducing the cost of production due to the high price of phosphate fertilizers, as well as reducing the pollution of the environment due to the better use of the applied fertilizers. The objective of this work was to compare the method of selection of potato clones for the efficiency of use and response to P between in vitro and off - soil systems with the use of sand as substrate. To that end, potato clones SMIC 148-A, Dakota Rose, SMINIA 793101-3, SMIB 106-7, SMIF 212-3, SMIJ 319-1 and P 150 were cultivated at low and high levels of P in the culture systems in vitro (1,935 and 19,346 mg P L-1) and off-soil (2.32 and 23.2 mg P L-1). The selection of potato clones using only as a criterion the accumulation of P under low nutrient level is not adequate, both in off-soil and in vitro cultivation. Clones selected as being more efficient in the use of P in in vitro cultivation do not prove to be necessarily more efficient in off-soil cultivation. No clone remains in the same classification group regarding the efficiency of use and response to P, based on the production of dry mass, in the two cropping systems.


A seleção de plantas de batata (Solanum tuberosum L.) que são eficientes no uso de fósforo (P) desempenha um papel importante no aumento da produtividade das culturas. reduzindo o custo de produção devido ao alto preço dos fertilizantes fosfatados. além de reduzir a poluição do meio ambiente devido ao melhor uso dos fertilizantes aplicados. O objetivo deste trabalho foi comparar o método de seleção de clones de batata quanto à eficiência de uso e resposta ao P entre sistemas in vitro e fora do solo com o uso de areia como substrato. Para o efeito. os clones de batata SMIC 148-A. Dakota Rose. SMINIA 793101-3. SMIB 106-7. SMIF 212-3. SMIJ 319-1 e P 150 foram cultivados a baixos e altos níveis de P nos sistemas de cultivo in vitro (1.935 e 19.346 mg P L-1) e fora do solo (2.32 e 23.2 mg P L-1). A seleção de clones de batata utilizando apenas como critério o acúmulo de P em baixo nível de nutrientes não é adequada. tanto no cultivo fora do solo quanto no in vitro. Clones selecionados como mais eficientes no uso de P em cultivo in vitro não se mostraram necessariamente mais eficientes no cultivo fora do solo. Nenhum clone permanece no mesmo grupo de classificação quanto à eficiência de uso e resposta ao P. baseado na produção de massa seca nos dois sistemas de cultivo.

ABSTRACT This study describes the development of mixed flour added of maca for gluten-free bread (GFB) employing a Simplex Centroid Design. Four pseudo-components were evaluated in order to improve the GFB specific volume and texture profile analysis: maca (MC; 0.25 to 0.75), potato starch (PS; 0.25 to 0.75), sweet manioc flour (SEM; 0 to 0.25), and sour manioc flour (SOM; 0 to 0.25). The best GFB formulation - by desirability results - was evaluated regarding physio-chemical, microbiological aspects, and sensory acceptance. The responses specific volume and hardness were adjusted with a special cubic model (R2= 0.973 and 0.913), while cohesiveness, springiness and chewiness was adjusted with a linear model. The significant contribution of maca into GFB dough was evident in the models, and the desirability results indicated an appropriate mixed flour composed by MC (0.25), PS (0.5), and SEM (0.25). The GFB elaborated with this mixed flour generated a product well accepted (acceptability index higher than 73.9%) for attributes of aroma, colour, texture, flavour, and overall acceptance. The values of physio-chemical parameters obtained in the present article was in the range described for GFB in the literature. In addition, the processing method applied allowed for obtaining a GFB that was microbiologically stable.


RESUMEN Este estudio describe el desarrollo de la harina mezclada (MF) agregada de maca para la elaboración de pan sin gluten (GFB) empleando un Diseño Centroide Simplex. Se evaluaron cuatro pseudo-componentes para mejorar el volumen específico y el perfil de textura de GFB: maca (MC; 0,25 a 0,75), almidón de patata (PS; 0,25 a 0,75), harina de yuca dulce (SEM; 0 a 0,25), y harina de yuca agrio (SOM; 0 a 0,25). La mejor formulación de GFB - por la deseabilidad - se evaluó en relación con los aspectos fisicoquímicos, microbiológicos, y sensoriales. El volumen específico y la dureza se ajustaron a un modelo cúbico especial (R2= 0,975 y 0,913). La cohesión, elasticidad y masticabilidad se ajustaron a un modelo lineal. La contribución de la maca a la masa de GFB fue evidente en los modelos, y los resultados indican una MF apropiada compuesta por MC (0,25), PS (0.5) y SEM (0,25). El GFB elaborado con esta MF genera un producto aceptado (índice de aceptabilidad mayor que 73,9%) para aroma, color, textura, sabor y aceptación general. Los parámetros físico-químicos obtenidos estaban en el rango descrito para GFB en la literatura y GFB se mostró microbiológicamente estable.

Pectobacterium is a complex taxon of strains with diverse characteristics. It comprises several genera, including Erwinia, Brenneria, Pectobacterium, Dickeya, and Pantoea. Pectobacterium and Dickeya cause diseases in a wide range of plants, including potatoes, where they are causative agents of soft rot in tubers and blackleg in field-grown plants.Characterizing Pectobacterium species allows for the analysis of the diversity of pectinolytic bacteria, which may support control strategies for plant bacterial diseases. The aim of this study was to perform biochemical, physiological, and molecular characterizations of Pectobacteriumspp. from different sites and host plants. The isolated strains were characterized by the glucose fermentation test, Gram staining, catalase activity, oxidase activity, growth at 37 ºC, reducing substances from sucrose, phosphatase activity, indole production, acid production from different sources (sorbitol, melibiose, citrate, and lactose), pathogenicity in potato, and hypersensitivity reactions. Molecular characterization was performed with species-specific primers ECA1f/ECA2r and EXPCCF/EXPCCR, which identify P.atrosepticum and P.carotovorum subsp. carotovorum (Pcc), respectively, and with primers 1491f/L1RA/L1RG and Br1f/L1RA/L1RG that differentiate Pcc from Dickeya chrysanthemi and from P. carotovorum subsp. brasiliensis. The strains were identified as belonging to the genus Pectobacterium, though they did not fit the biochemical nor the molecular classification standards for subspecies differentiation, indicating significant diversity among the strains.


Pectobacterium é um táxon complexo de isolados bacterianos com características diversas. Compreende vários gêneros como Erwinia, Brenneria, Pectobacterium, Dickeya e Pantoea. Pectobacterium e Dickeya causam doenças em ampla variedade de plantas, incluindo a batateira, na qual são os agentes etiológicos da podridão mole dos tubérculos e da canela-preta de plantas cultivadas em campo.A caracterização de espécies de Pectobacterium permite a análise da diversidade de bactérias pectolíticas, podendo auxiliar estratégias de controle de doenças bacterianas em plantas. O objetivo deste trabalho foi caracterizar bioquímica, fisiológica e molecularmente isolados de Pectobacterium sp. provenientes de diferentes locais e hospedeiros. Os isolados foram caracterizados pelos testes de fermentação de glicose, Gram, catalase, oxidase, crescimento à 37 ºC, redução de substâncias a partir de sacarose, atividade da fosfatase, produção de indol, produção de ácido a partir de sorbitol, melibiose, citrato e lactose, patogenicidade em batata e reação de hipersensibilidade. Para a caracterização molecular, foram utilizados os pares de primersECA1f/ECA2r e EXPCCF/EXPCCR [específicos para P. atrosepticum e P.carotovorum subsp. carotovorum(Pcc), respectivamente] e as tríades de primers 1491f/L1RA/L1RG e Br1f/L1RA/L1RG, para diferenciar Pcc de Dickeya chrysanthemi e de P. carotovorum subsp. brasiliensis. Os isolados foram identificados como pertencentes ao gênero Pectobacterium, no entanto, não se enquadraram na classificação bioquímica e tampouco molecular para diferenciação das subespécies, demonstrando a grande diversidade dos mesmos.

ABSTRACT

Abstract During the last years Phytophthora infestans sensu lato (Mont. De Bary) has caused epidemics in Colombia in Andean fruit crops such as Solanum quitoense and Solanum betaceum. Establishment of new or modified experimental procedures to study this pathogen is a mandatory subject for scientists. Twelve isolates of Phytophthora spp. obtained from six different Solanum hosts in Colombia were used to evaluate the effect of five different solid media for growth and ability to produce sporangia and liberate zoospores. Determination of the best media culture and optimal growth temperature were necessary to perform measurements and correlate the provenance of isolates with phenotypic traits. Modifications were made to use ingredients available in local markets on the following media: lime bean agar (LBA), Tree tomato or tree tomato agar (TA), carrot agar (AZ), Rye A modified agar and 32% non-clarified V8 agar. Cardinal temperature determination was performed at 10, 15, 20, and 25 °C. Morphometric traits were measured once the optimal media culture and temperature were defined. Correlation analysis showed that there is a relationship between the host and isolate's preferences for media culture and optimal growth temperature. In addition, the production of characteristic sporangia, sporangiophore and mycelia was related with the media type used and host from which the isolate was collected. In this work useful information was provided to make studies about the biology and development of isolates gathered from cultivated and wild non-traditional hosts.


Resumo Durante os últimos anos Phytophthora infestans sensu lato causou epidemias na Colômbia em lavouras de frutos andinos, como Solanum quitoense e Solanum betaceum. Estabelecimento de procedimentos experimentais novos ou modificados para estudar este patógeno é um assunto obrigatório para os cientistas. Doze isolados de Phytophthora spp. obtidos de seis diferentes hosts Solanum na Colômbia foram usados para avaliar o efeito de cinco diferentes mídias sólidas para o crescimento e a capacidade de produzir esporângios e libertar zoósporos. Determinação da melhor cultura de mídia e temperatura de crescimento ideal foram necessárias para realizar medições e correlacionar a proveniência de isolados com traços fenotípica. Foram feitas modificações para usar os ingredientes disponíveis nos mercados locais nos seguintes meios: ágar do feijão de cal (LBA), tomate da árvore ou ágar do tomate da árvore (TA), ágar da cenoura (AZ), centeio um ágar modificado e 32% de ágar-V8 não-esclarecido. A determinação da temperatura Cardeal foi realizada em 10, 15, 20 e 25 °C. Traços morfométricos foram medidos uma vez que a cultura de mídia ideal e temperatura foram definidos. Análise de correlação mostrou que há uma relação entre o hospedeiro e isolar as preferências para a cultura de mídia e temperatura de crescimento ideal. Além disso, a produção de esporângios característica, esporangióforo e mycelia foi relacionada com o tipo de mídia utilizado e hospedeiros a partir do qual o isolado foi coletado. Neste trabalho foram fornecidas informações úteis para fazer estudos sobre a biologia e o desenvolvimento de isolados recolhidos de hospedeiros não-tradicionais cultivados e selvagens.

ABSTRACT

La punta morada es una enfermedad que afecta la producción de algunas especies de solanáceas como la papa y el tomate, causando enrollamiento en las puntas de las hojas con una marcada coloración morada, decaimiento temprano de la planta y en la papa se observa tuberización aérea. Como patógenos asociados a la enfermedad se consideran al fitoplasma BLTVA y la bacteria Candidatus Liberibacter solanacearum. Dada la similitud en la sin-tomatología foliar que generan ambos patógenos, es difícil precisar cuál de ellos está implicado en la enfermedad. En Guatemala, existen reportes de la sintomatología típica de punta morada en las principales zonas productoras de papa y tomate, desconociéndose el agente asociado. La investigación determinó cuál de los dos patógenos reportados está asociados a la enfermedad en 12 municipios productores de papa y/o tomate en el país. Se realizaron ampli-ficaciones de ADN con cebadores específicos para cada patógeno asociado a la enfermedad. Por la alta incidencia del fitoplasma BLTVA en las muestras de papa (73.9%), en comparación a C. Liberibacter solanacearum (26%), este es considerado como el patógeno asociado más importante en papa. En las muestras de tomate, la incidencia del fitoplasma BLTVA (29.8%) y C. Liberibacter solanacearum del (27.6%) fue similar. Además, sobresale el primer reporte de la detección del fitoplasma BLTVA afectando el cultivo de tomate en Guatemala. Se sugiere un monitoreo constante, mediante métodos moleculares, para un diagnóstico certero y establecer medidas de manejo de la enfermedad para evitar su diseminación hacia zonas aún no afectadas.


The potato purple top is a disease that affects the production of some solanaceous species such as potatoes and tomatoes, causing curl at the tips of the leaves with a marked purple coloration, early decay of the plant, and aerial tuberization is observed in the potato. BLTVA phytoplasma and Candidatus Liberibacter solanacearum are considered as pathogens associated with the disease. Given the similarity in foliar symptoms generated by both pathogens, it is difficult to determine which one is involved in the disease. There are reports of the typical potato purple top symptoms in the main potato and tomato producing areas in Guatemala, being unknown the associated agent. The research determined which of the two reported pathogens is associated with the disease in 12 potatoes and/or tomato producing areas in the country. We performed DNA amplification with specific primers for each disease-associated pathogen. Due to the high incidence of BLTVA phytoplasma in potato samples (73.9%), com-pared to C. liberibacter solanacearum (26%), this is considered the most important associated pathogen in potatoes. In tomato samples, the incidence of BLTVA phytoplasma (29.8%) and C. liberibacter solanacearum (27.6%) was similar. Besides, the first report of the detection of the BLTVA phytoplasma affecting tomato cultivation in Gua-temala stands out. Using molecular methods, constant monitoring is suggested for an accurate diagnosis and to establish management measures for the disease to prevent its spread to areas not yet affected.

Abstract Nitrogen (N) and potassium (K) in potato crop planting synergistically increase tuber yield, but there are no studies on this interaction in sidedressing. In two experiments with 'Atlantic' potato combinations of four N rates (0, 50, 100, and 150 kg ha-1) with four K2O rates (0, 100, 200, and 300 kg ha-1) were applied in sidedressing in a 4×4 factorial scheme with three replications in a completely randomized design. Adjacent commercial fields were sampled to economic comparisons with experimental results. Significant interaction between N and K sidedressing rates with tuber yields increase also was confirmed and classified as Liebig-synergism. Compared to the isolated N and K applications in sidedressing, joint N and K fertilizations, respectively, increases by 11% and 48% marketable tuber yields in the summer-fall experiment, and 12% and 7% in the spring experiment. Joint N and K applications as sidedressing was more profitable than planting fertilization, mainly at higher N and K rates. The response of specific gravity of 'Atlantic' potato tubers to the N and K sidedressing rates was mediated by interactions between edaphoclimatic conditions and inputs of N and K. The combined application of N and K sidedressing rates increased specific gravity in the summer-fall experiment, but had a negative effect in the spring experiment. Therefore, our results provide strong evidence that the fertilization management for potato crop in Brazil can be modified by applying higher amounts of N and K in sidedressing to match nutritional needs of the crop.

RESUMEN Con el objetivo de determinar el efecto del consumo de harina de cáscara o harina de pulpa de dos variedades de Solanum tuberosum (papa Yungay y papa Canchán) sobre la acumulación de tejido adiposo, peso de órganos y estrés oxidativo en hígado de ratas realizamos un estudio experimental en 24 ratas Holtzman obesas, divididos en cuatro grupos y, sometidas a dietas que contenían 10% de harina de las dos variedades de papa. Los grupos fueron T1: cáscara Yungay, T2: pulpa Yungay; T3: cáscara Canchán; y T4: pulpa Canchán. Al finalizar, se sacrificaron todos los animales para registrar los pesos de órganos y tejido adiposo, y extraer muestras para determinar la actividad enzimática de superóxido dismutasa y catalasa en el hígado. El grupo de ratas obesas que consumió pulpa de variedad Yungay tuvo menor estrés oxidativo en el hígado; además, independientemente de la parte de tubérculo consumido, esta variedad redujo el peso de los riñones.


ABSTRACT This study aimed to determine the effect of the consumption of peel flour or pulp flour from two varieties of Solanum tuberosum (Yungay potato and Canchán potato) on the accumulation of adipose tissue, organ weight, and oxidative stress in the liver of rats. We carried out an experimental study in 24 obese Holtzman rats, divided into four groups and subjected to diets containing 10% flour from both varieties of potato. The groups were T1: Yungay peel; T2: Yungay pulp; T3: Canchán peel; and T4: Canchán pulp. When the study was completed, all the animals were slaughtered to record the weights of organs and adipose tissue and to extract samples to determine the enzyme activity of superoxide dismutase and catalase in the liver. The group of obese rats that consumed the pulp of the Yungay variety had less oxidative stress in the liver. Also, regardless of the tuber part consumed, this variety reduced the weight of the kidneys.

Abstract A strain isolated from potato common scab superficial lesions in El Fuerte Valley in northern Sinaloa, Mexico, was identified by 16S rRNA and morphological methods. Moreover, the effects of the crude extract of strain V2 was evaluated on radish and potato. The isolate was similar to Streptomyces acidiscabies in its morphological properties; however, the 16S rRNA gene sequence of strain V2 was neither 100% identical to this species nor to the streptomycetes previously reported in Sinaloa, Mexico. Strain V2 did not amplify any specific PCR products for genes necl and tomA, which have been found and reported in S. acidiscabies. Strain V2 produced a PCR product for the txtAB operon, which is related to the production of thaxtomin. In vitro assays using crude thaxtomin extract and a spore suspension of the organism caused necrotic symptoms on radish and potato, which were highly virulent in potato. This study reports that Streptomyces sp. V2 has a toxigenic region (TR) that is associated with the thaxtomin gene cluster.


Resumen Se aisló una cepa de una lesión superficial de sarna común de la papa en un ejemplar procedente del Valle del Fuerte, en el norte de Sinaloa, México. La cepa fue identificada por secuenciación del gen 16S ARNr, y por sus características morfológicas. Los efectos del extracto crudo de dicha cepa, llamada V2, fue evaluado en papa y rábano. El aislado fue similar a Streptomyces acidiscabies en sus características morfológicas, pero la secuencia del gen 16S ARNr de la cepa V2 no fue 100% idéntica a la de dicha especie, ni tampoco a las de cepas identificadas dentro de este taxón previamente en Sinaloa, México. La cepa V2 no amplificó los productos específicos de PCR de los genes nec1 y tomA, los cuales sí se han reportado en S. acidiscabies. La cepa V2 amplificó el producto de PCR para del operón txtAB, relacionado con la producción de taxtomina. A través de ensayos in vitro usando un extracto crudo de taxtomina y una suspensión de esporas del organismo aislado se verificó la producción de síntomas necróticos en rábano y papa, con mayor virulencia en esta última especie. Este estudio indica que Streptomyces sp. V2 tiene una región toxigénica (TR) asociada con el cluster de genes de taxtomina.

Yacon is a functional food with great potential for horticultural exploitation and its cultivation demands the generation of agronomic information. To address these needs, this study was performed to evaluate the production of yacon planted in different planting seasons under two growing conditions in Brazil. The experiments were conducted in two different locations, one in the mountain region (tropical microclimate at altitude with milder temperatures) and the other in the lowland region (warm tropical microclimate with higher temperatures). At both sites, yacon was planted in four seasons: autumn (April), winter (July), spring (September) and summer (December). At harvest, the tuberous roots were evaluated for leaf area index and the variables of vegetative development and productivity of plants (for total and standard commercial roots). It was concluded that yacon cultivation in mountain conditions (tropical climate with milder temperature) promotes the development and production of yacon. Additionally, planting in the autumn for both growing conditions (mountain or lowland) promotes the development and production of yacon, including increased production of roots of better commercial grade, allowing greater economic profitability. These results indicate that temperature has an influence on the growth and yield of the tuberous roots of yacon, and milder temperatures were more favourable.


A yacon é um alimento funcional com grande potencial para exploração hortícola e seu cultivo exige a geração de informações agronômicas. Para atender a essas necessidades, este estudo foirealizado com o objetivo de avaliar a produção de yacon plantada em diferentes estações de plantio sob duas condições de crescimento no Brasil. Os experimentos foram conduzidos em dois locais diferentes, um em região montanhosa (microclima tropical de altitude com temperaturas mais amenas) e o outro em região de baixada (microclima tropical quente com temperaturas mais elevadas). Em ambos os locais, a yacon foi plantada em quatro estações: outono (abril), inverno (julho), primavera (setembro) e verão (dezembro). Por ocasião da colheita das raízes tuberosas foram avaliados o índice de área foliar e variáveis de desenvolvimento vegetativo e produtividade das plantas (raízes tuberosas totais e por padrão). Concluiu-se que o cultivo de yacon em condições de montanha (clima tropical com temperatura mais suave) promove melhor desenvolvimento e produção de yacon. Além disso, plantar no outono para ambas as condições de crescimento (montanha ou baixada) promove melhor desenvolvimento e produção de yacon, incluindo o aumento da produção de raízes tuberosas de melhor qualidade comercial, permitindo maior rentabilidade econômica. Estes resultados indicam que a temperatura influenciou o crescimento e o rendimento das raízes tuberosas de yacon e as temperaturas mais amenas foram mais favoráveis.

Objetivou-se com este trabalho avaliar a qualidade pós-colheita de batatas, submetidas ao tratamento hidrotérmico. Tubérculos de batatas foram submetidos a água nas temperaturas de 50 e 60ºC, por 1, 6, 11 e 21 minutos, e avaliados quanto ao coração preto, brotação, esverdeamento, mancha preta e exsudação aos 1, 5, 10, 15 e 20 dias de armazenamento. Os dados foram analisados por meio do teste Dunnett (5%). Não houve presença de coração preto e esverdeamento. A incidência de brotação foi menor nos tratamentos de 50°C por 21 minutos e 60°C por 6 minutos. No 1° dia de armazenamento, os tratamentos de 50°C por 21 minutos, 60°C por 12 e 21 minutos e 70°C por 1, 6, 12 e 21 minutos desencadearam a presença de mancha preta. Os tratamentos de 50°C, por 1, 6 e 11 minutos e 60°C, por 1 e 6 minutos proporcionaram maior vida pós-colheita dos tubérculos.

ABSTRACT

Peru is not only megadiverse in flora and fauna, but also in "native varieties of potatoes" that have been used by pre-Inca civilizations. In this context, "native potatoes" not only have a high nutritional value in antioxidants, but also a high capacity to cure and/or alleviate diseases; providing quality of life that is reflected in the longevity of the Andean population. However, in the absence of documentation and dissemination, many varieties and their ethnobotanical knowledge is being lost. To preserve and disseminate this valuable ancestral knowledge, a taxonomic, phytogeographic and ethnobotanical inventory was carried out, based on periodic botanical explorations during the period 1997-2017. The information obtained was complemented with other researches and reports by other authors. 317 "varieties of native potato" are reported for the North of Peru, of which 86, are "native varieties of colored pulp".


El Perú no sólo es megadiverso en flora y fauna, sino también en "variedades nativas de papas" que han sido utilizadas desde las civilizaciones preincas. En este contexto las "papas nativas" no sólo poseen un elevado valor nutricional en antioxidantes, sino también una alta capacidad de curar y/o aliviar enfermedades; brindando calidad de vida que se refleja en la longevidad de la población andina. Sin embargo, ante la falta de documentación y difusión, muchas variedades y su conocimiento etnobotánico se está perdiendo. Para preservar y difundir este valioso conocimiento ancestral, se llevó a cabo un inventario taxonómico, fitogeográfico y etnobotánico, en base a exploraciones botánicas periódicas durante el período 1997-2017. La información obtenida se complementó con otras investigaciones y reportes de otros autores. Se reportan para el Norte del Perú 317 "variedades de papa nativa", de las cuales 86, son variedades "nativas de pulpa de color".
