ABSTRACT

Abstract Under salt stress conditions, plant growth is reduced due to osmotic, nutritional and oxidative imbalance. However, salicylic acid acts in the mitigation of this abiotic stress by promoting an increase in growth, photosynthesis, nitrogen metabolism, synthesis of osmoregulators and antioxidant enzymes. In this context, the objective was to evaluate the effect of salicylic acid doses on the growth and physiological changes of eggplant seedlings under salt stress. The experiment was conducted in a greenhouse, where the treatments were distributed in randomized blocks using a central composite matrix Box with five levels of electrical conductivity of irrigation water (CEw) (0.50; 1.08; 2.50; 3.92 and 4.50 dS m-1), associated with five doses of salicylic acid (SA) (0.00; 0.22; 0.75; 1.28 and 1.50 mM), with four repetitions and each plot composed of three plants. At 40 days after sowing, plant height, stem diameter, number of leaves, leaf area, electrolyte leakage, relative water content, and total dry mass were determined. ECw and SA application influenced the growth and physiological changes of eggplant seedlings. Increasing the ECw reduced growth in the absence of SA. Membrane damage with the use of SA remained stable up to 3.9 dS m-1 of ECw. The relative water content independent of the CEw increased with 1.0 mM of SA. The use of SA at the concentration of 1.0 mM mitigated the deleterious effect of salinity on seedling growth up to 2.50 dS m-1 of ECw.


Resumo Em condições de estresse salino, o crescimento das plantas é reduzido, em virtude, do desequilíbrio osmótico, nutricional e oxidativo. Contudo, o ácido salicílico atua na mitigação desse estresse abiótico por promover incremento no crescimento, fotossíntese, metabolismo do nitrogênio, síntese de osmorreguladores e enzimas antioxidantes. Nesse contexto, objetivou-se avaliar o efeito de doses de ácido salicílico sobre o crescimento e alterações fisiológicas de mudas de berinjela sob estresse salino. O experimento foi conduzido em casa de vegetação, onde os tratamentos foram distribuídos em blocos ao acaso utilizando uma matriz composta central Box com cinco níveis de condutividade elétrica da água de irrigação (CEa) (0,50; 1,08; 2,50; 3,92 e 4,50 dS m-1), associada a cinco doses de ácido salicílico (AS) (0,00; 0,22; 0,75; 1,28 e 1,50 mM), com quatro repetições e cada parcela composta por três plantas. Aos 40 dias após a semeadura, foram determinados a altura da planta, diâmetro do caule, número de folhas, área foliar, vazamento de eletrólito, teor relativo de água e massa seca total. A CEa e a aplicação de AS influenciaram no crescimento e nas alterações fisiológicas das mudas de berinjela. O aumento da CEa reduziu o crescimento na ausência de AS. O dano de membrana com o uso de AS manteve-se estável até 3,9 dS m-1 de CEa. O conteúdo relativo de água independentemente da CEa aumentou com 1 mM de SA. O uso de AS na concentração de 1 mM mitigou o efeito deletério da salinidade no crescimento das mudas até 2,50 dS m-1 de CEa.

ABSTRACT

Salinity is one of the most critical environmental parameters regarding fish physiology, modifying food intake and growth performance in many fish species. The present study has investigated the effects of different salinity levels on growth performance, feeding and survival of Asian seabass Lates calcarifer juveniles. Asian seabass juveniles were reared at 0 (T1), 5 (T2), 22 (T3), 36 (T4), and 42 (T5) ppt salinity. Approximately eight hundred thirty fish individuals with an average weight of 1.24±0.52 g were randomly distributed (166 fish/Tank) in 5 concrete tanks (each tank 30×6×4 ft, volume 19,122 L) for forty days. Juveniles were initially fed 42% crude protein-containing diets at a rate of 6% of their body weight per day. The results showed that salinity level had a significant effect on the weight gain (WG), average daily weight gain (ADWG), specific growth rate (SGR), feed conversion ratio (FCR), survival rate (SR), total biomass and health indices (p<0.05). The highest WG (39.11±1.49 g), ADWG (1.00±0.12 g), SGR (8.74±0.03% d-1) and lowest FCR (0.96±0.20) were observed with T3 treatment, which was significantly higher compared to other treatment groups (p<0.05). Among the health indices, the highest hepatosomatic index and viscerosomatic index were found with T3 treatment, significantly higher than the other groups (p<0.05). No significant differences were found among the treatments in terms of survival rate (p>0.05), but the maximum survival rate (98.89±0.0%) was observed in the T3 and T2 treatments. The maximum level of crude proteins (19.99±1.4%) was found in the whole-body biochemical composition of Asian seabass juveniles in the T3 treatment group. The second-order polynomial regression showed that 20 ppt salinity is optimum for the best growth of Asian seabass. Thus, the present study recommends 20 to 36 ppt salinity for the commercial farming of Asian seabass under a closed aquaculture system.


A salinidade é um dos parâmetros ambientais mais críticos em relação à fisiologia dos peixes, modificando a ingestão de alimentos e o desempenho de crescimento em muitas espécies. O presente estudo investigou os efeitos de diferentes níveis de salinidade no desempenho de crescimento, alimentação e sobrevivência de jovens espécies de robalos asiáticos Lates calcarifer, as quais foram criadas sob salinidade 0 (T1), 5 (T2), 22 (T3), 36 (T4) e 42 (T5) ppt. Aproximadamente 830 indivíduos de peixes com peso médio de 1,24 ± 0,52 g foram distribuídos aleatoriamente (166 peixes / tanque) em cinco tanques de concreto (cada tanque tinha 30 × 6 × 4 pés (9,1 x 1,8 x 1,2 metros), com volume de 19,12 litros) por 40 dias. Os peixes foram inicialmente alimentados com uma dieta contendo 42% de proteína bruta a uma taxa de 6% do seu peso corporal por dia. Os resultados mostraram que o nível de salinidade teve um efeito significativo no ganho de peso (GP), ganho de peso médio diário (GPMD), taxa de crescimento específico (TCE), taxa de conversão alimentar (TCA), taxa de sobrevivência (TS), biomassa total e índices de saúde (p < 0,05). O maior GP (39,11 ± 1,49 g), GPMD (1,00 ± 0,12 g), TCE (8,74 ± 0,03% d-1) e o menor TCA (0,96 ± 0,20) foram observados com o tratamento T3, que foi significativamente superior em comparação com os outros tratamentos (p < 0,05). Entre os índices de saúde, os maiores índices hepatossomáticos e viscerossomáticos foram encontrados no tratamento T3, significativamente superior do que os demais grupos (p < 0,05). Não foram encontradas diferenças significativas entre os tratamentos quanto à TS (p > 0,05), mas a TS máxima (98,89 ± 0,0%) foi observada nos tratamentos T3 e T2. O nível máximo de proteína bruta (19,99 ± 1,4%) foi encontrado na composição bioquímica dos corpos dos jovens robalos asiáticos no grupo T3. A regressão polinomial de segunda ordem mostrou que a salinidade de 20 ppt é ótima para o melhor crescimento do robalo asiático. Assim, o presente estudo recomenda salinidade de 20 a 36 ppt para o cultivo comercial de robalo asiático em sistema de aquicultura fechado.

ABSTRACT

Tagetes genus of Composite family consider one of the most favorite floriculture plants. Therefore, of particular interest examine the salt tolerance of this bedding and coloring agent plant. In this research, was report the role of glycinebetaine (GB) in attenuating the adverse impacts of salt stress in African marigold plant, along with their anti-oxidative capacities and biochemical attributes. The salt stressed African marigold (100 and 150 mM NaCl) was treated with GB at 200 mM, beside untreated control plants. According to the obtained results, the growth characters were negatively in salt stressed plants but a mitigate impact of GB were observed in this respect. Obviously, the morphological as well as some physiological characters were reduced with salinity treatments while GB treatment reverses these effects. Overall, the alleviate impact of GB on the negative impact of salt stress was enhanced through improving total phenolic and antioxidant enzyme activity. Further, it is concluded that GB concentration induces the activities of antioxidative enzymes which scavenged ROS increased under saline conditions.


O Tagetes, da família Composite, é um dos gêneros mais apreciados de plantas de floricultura. Portanto, é de particular interesse examinar a tolerância ao sal desta planta de substrato e corante. Nesta pesquisa, foi relatado o papel da glicinebetaína (GB) na atenuação dos impactos adversos do estresse salino na calêndula africana, juntamente com suas capacidades antioxidantes e atributos bioquímicos. A calêndula africana estressada com sal (NaCl 100 e 150 mM) foi tratada com GB a 200 mM, ao lado de plantas de controle não tratadas. De acordo com os resultados obtidos, os caracteres de crescimento foram negativos em plantas estressadas por sal, mas um impacto mitigado de GB foi observado neste aspecto. Obviamente, os caracteres morfológicos e fisiológicos foram reduzidos com os tratamentos de salinidade, enquanto o tratamento com GB reverteu esses efeitos. No geral, o impacto de alívio do GB no impacto negativo do estresse salino foi aprimorado através da melhoria da atividade das enzimas fenólicas e antioxidantes totais. Além disso, conclui-se que a concentração de GB induz as atividades de enzimas antioxidantes que sequestraram ROS aumentadas em condições salinas.

ABSTRACT

Hypersaline estuaries are commonly found in the Semiarid Northeast Brazilian coast. However, the utilization of these estuaries by early life stages of fishes is less well documented. This study aimed to evaluate the effects of environmental factors on the ichthyoplankton, and to describe their spatial and temporal variation in two estuaries (Tubarão and Casqueira). The sampling were diurnally in four excursions carried out during two dry-season months (November and December 2017) and two rainy-season months (May and July 2018). A total of 423 larvae representing 30 species (14 families) and 2,762 eggs (five families) were collected. Although the community structures were similar in both estuaries, there was strong seasonal variation, with higher abundances observed during the dry season. Low rainfall was the primary environmental driver affecting ichthyoplankton in both estuaries. Other environmental factors, such as depth, water temperature, and channel width, were also predictor variables for the distribution of eggs and larvae. The ichthyoplankton was dominated by marine estuarine-dependent species in both estuaries, accounting for 76% of individuals. The presence of the early life stages of typically marine species suggests that hypersaline environments provide important nursery areas and play an essential role for some coastal fish species.(AU)


Os estuários hipersalinos são comumente encontrados na região semiárida da costa nordestina brasileira. No entanto, a utilização desses estuários pelos primeiros estágios de vida dos peixes é pouco documentada. O objetivo deste estudo foi avaliar os efeitos de fatores ambientais sobre o ictioplâncton e descrever sua variação espacial e temporal em dois estuários (Tubarão e Casqueira). As amostragens foram diurnas em quatro excursões realizadas em dois meses da estação seca (novembro e dezembro de 2017) e dois meses da estação chuvosa (maio e julho de 2018). Um total de 423 larvas representando 30 espécies (14 famílias) e 2.762 ovos (cinco famílias) foram coletadas. Embora as estruturas das comunidades fossem semelhantes em ambos os estuários, houve fortes diferenças de variação sazonal, com maiores abundâncias observadas durante a estação seca. A baixa precipitação foi o principal fator ambiental que afetou o ictioplâncton em ambos os estuários. Outros fatores, como profundidade, temperatura da água e largura do canal, também foram preditoras para a distribuição de ovos e larvas. O ictioplâncton foi dominado por espécies marinhas dependentes em ambos os estuários, representando 76% dos indivíduos. A presença de estágios iniciais de espécies tipicamente marinhas sugere que ambientes hipersalinos fornecem áreas de berçário importantes e desempenham um papel essencial para algumas espécies de peixes costeiros.(AU)

Abstract The primary objective of this study was to investigate the seasonal fish and crustacean variations concerning taxonomic composition, species richness, and diversity in sandy beach habitat. For this purpose, we investigated the Sonmiani Hor lagoon area during four distinct seasons, i.e., northeast (NE) monsoon, pre-monsoon, south-west (SW) monsoon, and post-monsoon for one year. During each haul, the net was pulled about 100m along the beach in 0.5m depth. The results showed a strong linear correlation between the diversity index and equitability in fishes (r = 0.978). The diversity index was strong negatively correlated with the abundance and biomass (r = -0.978, -0.972, respectively). The physical attributes like sea surface water temperature and salinity showed a strong negative effect on species assemblages (r = -0.981 and -0.943, respectively). The mean air and water temperature illustrated approximately 3°C difference during NE and pre-monsoon seasons. However, salinity, pH, and electrical conductivity did not show any significant seasonal variabilities. Under the ecological indices, the fish species displayed higher diversity (H' = 3.19) during SW monsoon, whereas the lowest diversity was observed during pre-monsoon (H' = 1.58). The equitability and species richness, however, remained more noticeable during SW monsoon (J' = 0.81). The total number of individuals of fish and crustaceans reached 4799 with 3813 fish individuals and 986 individuals of crustaceans. A total of 27 families of fish while five crustacean families comprising of 30 genera and 38 fish species while ten genera and 17 species of crustaceans were recorded. Liza subviridis displayed the highest abundance among the sampled fish species. In conclusion, fish species constituted a significant part of the coastal fauna in the study area. The seasonal variations displayed distinct variations in fish species composition and diversity.


Resumo O objetivo principal deste estudo foi investigar as variações sazonais de peixes e crustáceos em relação à composição taxonômica, riqueza de espécies e diversidade no habitat de praias arenosas. Para esse fim, investigamos a área da lagoa Sonmiani Hor durante quatro estações distintas, ou seja, monção nordeste (NE), pré-monção, monção sudoeste (SW) e pós-monção, por um ano. Durante cada transporte, a rede foi puxada cerca de 100 m ao longo da praia, a 0,5 m de profundidade. Os resultados mostraram uma forte correlação linear entre o índice de diversidade e a equitabilidade de peixes (r = 0,978). O índice de diversidade apresentou forte correlação negativa com a abundância e a biomassa (r = -0,978 e -0,972, respectivamente). Os atributos físicos, como temperatura da água da superfície do mar e salinidade, apresentaram forte efeito negativo sobre o conjunto de espécies (r = -0,981 e -0,943, respectivamente). A temperatura média do ar e da água teve uma diferença de aproximadamente 3 °C durante a monção NE e a pré-monção. No entanto, salinidade, pH e condutividade elétrica não apresentaram variabilidade sazonal significativa. A respeito dos índices ecológicos, as espécies de peixes apresentaram maior diversidade (H' = 3,19) durante a monção SW, enquanto a menor diversidade observada foi na pré-monção (H' = 1,58). A equitabilidade e a riqueza de espécies, no entanto, permaneceram mais perceptíveis durante a monção SW (J' = 0,81). O número total de peixes e crustáceos foi de 4.799, com 3.813 peixes e 986 crustáceos. Foram registradas 27 famílias de peixes e 5 famílias de crustáceos, com 30 gêneros e 38 espécies de peixes, além de 10 gêneros e 17 espécies de crustáceos. Liza subviridis apresentou a maior abundância entre as espécies de peixes amostradas. Em conclusão, as espécies de peixes constituíram uma parte significativa da fauna costeira na área de estudo. As variações sazonais apresentaram variações distintas na composição e diversidade das espécies de peixes.

ABSTRACT

The objective of this work was to evaluate the biochemical and hormonal variables of Moxotó and Canindé goats submitted to two temperatures - 26.0±0.6 (thermoneutral) and 32.0±1.2°C (above thermal comfort zone) - and consuming water with three levels of salinity (1.0, 6.0 and 12.0 dSm-1). Thirty-six animals (18 of each breed) were used, with an average age of 5.0±0.6months and an average weight of 20.0±2.3kg, housed in metabolic cages inside a climate chamber. The animals were distributed in a completely randomized design with a 2 × 2 × 3 factorial scheme (2 breeds, 2 temperatures and 3 levels of salinity) and three replications. The glucose and urea had a significant effect (P>0.05) according to water salinity. Glucose, cholesterol, protein, albumin, globulin, aspartate aminotransferase and hormones (T4, T3 and cortisol) varied according to temperature (P<0.05). There was a significant effect of time on hormonal variables (P<0.05). Biochemical and hormonal variables changed according to temperature and day shift, so that metabolism was reduced in the animals under thermal stress and accelerated when animals were in the thermal comfort zone.(AU)


O objetivo do trabalho foi avaliar as variáveis bioquímicas e hormonais de caprinos das raças Moxotó e Canindé, submetidos a duas temperaturas (26,0±0,6ºC e 32,0±1,2ºC), termoneutra e acima da zona de conforto térmico, respectivamente), consumindo água com três níveis de salinidade (1,0, 6,0 e 12,0dSm-1), utilizando-se 36 animais (18 de cada raça), com idade média de 5,0±0,6 meses e peso médio de 20,0±2,3kg, alojados em gaiolas metabólicas no interior de uma câmara climática. Os animais foram distribuídos em um delineamento inteiramente ao acaso, com esquema fatorial de 2 x 2 x 3 (2 raças, 2 temperaturas e 3 níveis de salinidade) e três repetições. A glicose e a ureia apresentaram efeito significativo (P<0,05) em função da salinidade da água. Glicose, colesterol, proteína, albumina, globulina, AST e hormônios (T4, T3 e cortisol) variaram conforme as temperaturas (P<0,05). Observou-se efeito significativo do horário sobre as variáveis hormonais (P<0,05). As variáveis bioquímicas e hormonais sofrem alterações em função da temperatura e do turno do dia, de modo que o metabolismo é reduzido em animais sob estresse térmico e acelerado quando os animais encontram-se na zona de conforto térmico.(AU)

Physalis peruviana L. is a solanacea that has been gaining prominence due to its fruits presenting good acceptance in the national and international market. However, several abiotic factors, such as salinity, can cause physiological disturbances in plants, and these changes may be of greater or lesser intent according to species. Therefore, the objective of the present work was to evaluate the physiological behavior of P. peruviana submitted to different fluxes of photosynthetically active photons (PPFD) and saline stress. The experimental design was a randomized block design with three saline levels (ECw) (0.5, 2.75 and 5.00 dS m-1) with four replications. Gas exchange measurements were performed with a portable infrared gas analyzer. Liquid CO2 assimilation, stomatal conductance, internal CO2 concentration, water use efficiency and instantaneous carboxylation efficiency were measured. Data were subjected to analysis of variance by F test and in cases of significance applied to regression analysis. The increase in PPFD provided reductions in stomatal conductance up to the density of approximately 400 µmol m-2s-1, being more pronounced in ECw of 2.75 and 5.0 dS m-1. The maximum CO2 assimilation rates in the three salinities are different according to the PPFD. The salinity of irrigation water reduced the quantum efficiency of photosynthesis in P. peruviana plants.

Abstract Studies in Salvelinus alpinus, Arctic charr, indicate that it has a low capacity to hyposmorregulatory or adaption to sea in winter periods in Arctic waters. The investigation finds to determinate the rank optimum of salinity to can cultivate this species at Chile. The weight adequate was determined to join on the sea by analysis of gill Na+, K+-ATPase activity, that it was found between the ranks 80-130 g, with 14.5 U/mg. It underwent evaluation of fish growth of 72 g salinities from 0 (control), 18, 25 and 33 g/L (sea water) for 94 days. The results indicate that the largest increases were obtained in brackish water. T18 g/L and T25 g/L achieved growth of 25% and 19% on day 94 and term sampling respectively. It is important to mention that the 8% that survived in seawater introduced percentages growth 16.6% equivalent to brackish water and control. These results suggest that Salvelinus alpinus can grow in seawater, with levels of Na+, K+-ATPase similar to those submitted by Salmo salar with a weight not less than 80 g.


Resumo Estudos em Salvelinus alpinus, Charr Ártico, indicam que tem baixa capacidade de hiposmorregulação ou adaptação ao mar em períodos de inverno nas águas do Ártico. A investigação determina o melhor nível de salinidade para cultivar esta espécie no Chile. Determinou-se o peso adequado para se unir ao mar pela análise da atividade da Na +, K + -ATPase das brânquias, que foi encontrada entre as faixas de 80 a 130 g, com 14,5 U/mg. Foi avaliado o crescimento de 72 g salinidades de 0 (controle), 18, 25 e 33 g/L (água do mar) por 94 dias. Os resultados indicam que os maiores aumentos foram obtidos em água salobra. T18 g/L e T25 g/L alcançaram crescimento de 25% e 19% no dia 94 e amostragem a termo, respectivamente. É importante mencionar que os 8% que sobreviveram na água do mar apresentaram percentuais de crescimento de 16,6% equivalentes a água salobra e controle. Estes resultados sugerem que o Salvelinus alpinus pode crescer em água do mar, com níveis de Na +, K + -ATPase semelhantes aos apresentados por Salmo salar com um peso não inferior a 80 g.

Abstract Salinity, of both soil and water, is one of the main causes of crop yield decline. Within this context, the objective of this study was to evaluate the influence of different salts on the germination of chia seeds. The experiment was conducted in a BOD chamber at a constant temperature of 20 °C and in the presence of light. The seeds were placed on paper soaked with aqueous solutions of calcium chloride (CaCl2), sodium chloride (NaCl), potassium chloride (KCl), and magnesium chloride (MgCl2), at the osmotic potentials zero, -0.10, -0.20, -0.30, and -0.40 MPa. The effect of the salinity was evaluated using a germination test, with counts on days 7 and 14 after sowing. Based on the results, chia seeds tolerate concentrations of NaCl to -0.4 MPa and KCl to -0.20 MPa. The salts CaCl2 and MgCl2 had a negative effect on the germination and vigor of the chia seeds for the osmotic potentials -0.30 MPa and -0.20 MPa, respectively.


Resumo A salinidade, tanto dos solos como das águas, é uma das principais causas da queda de rendimento das culturas. Neste contexto, o objetivo deste estudo foi avaliar a influência de diferentes sais na germinação de sementes de chia. O experimento foi conduzido em câmara BOD, na temperatura constante de 20 °C e em presença de luz. As sementes foram colocadas sobre papel embebido em soluções aquosas de cloreto de cálcio (CaCl2), cloreto de sódio (NaCl), cloreto de potássio (KCl) e cloreto de magnésio (MgCl2) nos potenciais osmóticos correspondentes a zero; -0,10; -0,20; -0,30 e -0,40 MPa. O efeito da salinidade foi avaliado através do teste de germinação com contagens aos 7 dias e 14 dias após a semeadura. De acordo com os resultados é possível concluir que as sementes de chia toleram concentrações de NaCl até -0,4 MPa e KCl até -0,20 MPa. Os sais CaCl2 e MgCl2 apresentam efeito negativo sobre a germinação e o vigor das sementes de chia a partir dos potenciais osmóticos de -0,30 MPa e -0,20 MPa, respectivamente.

ABSTRACT

The production of grafted passion fruit is an alternative for plant adaptation to saline environments. The objective of this study was to evaluate the effect of salt stress on physiology, biometry and fruit quality of P. edulis grafted on Passiflora spp. The experiment was conducted in completely randomized design, in a 3 x 2 factorial scheme, corresponding to three species of Passiflora (P. edulis, P. gibertii and P. cincinnata) with P. edulis scion and two levels of irrigation water salinity (0.5 - control and 4.5 dS m-1), with four repetitions. Water salinity compromises gas exchanges (CO2 assimilation raste and transpiration) and physiological variables (total chlorophyll and total water consumption) in grafted P. edulis. The interaction between the factors (water salinity x species) compromised only the growth in plant height and number of leaves. In relation to the species, auto-grafted P. edulis stood out from the other species, with higher internal CO2 concentration, number of leaves, stem dry mass, peel thickness, total soluble solids (TSS) of the pulp and TSS/TA ratio (titratable acidity). Auto-grafted P. edulis under saline conditions develops vital mechanisms (TSS and TSS/TA), which attenuates the effects of salt stress on the physico-chemical quality of the fruits.


A produção de maracujazeiro enxertado é uma alternativa para adaptação das plantas a ambientes salinos. Objetivou-se avaliar o efeito do estresse salino na fisiologia, biometria e qualidade de frutos de P. edulis enxertado em espécies de Passiflora spp. O delineamento utilizado foi inteiramente casualizado, em esquema fatorial 3 x 2, sendo três espécies de Passiflora (P. edulis, P. gibertii e P. cincinnata) tendo como copa P. edulis e dois níveis de salinidade de água de irrigação (0,5 ­ testemunha e 4,5 dS m-1), com quatro repetições. A salinidade da água compromete as trocas gasosas (taxa de assimilação de CO2 e transpiração) e variáveis fisiológicas (clorofila total e consumo hídrico total) em P. edulis enxertado. A interação entre os fatores (salinidade da água x espécie) compromete apenas o crescimento em altura de plantas e número de folhas. Em relação às espécies, o P. edulis auto enxertado se destaca em relação as demais espécies apresentando maior concentração interna de CO2, número de folhas, massa seca de caule, espessura da casca do fruto, sólidos solúveis totais (SST) da polpa e razão sólidos solúveis totais por acidez titulável (SST/AT). O P. edulis auto enxertado sob condições de salinidade, desenvolve mecanismos vitais (SST e SST/AT), que atenuam os efeitos do estresse salino na qualidade físico-química dos frutos.

ABSTRACT

Halophilic bacteria are microorganisms that grow optimally in the presence of the very high concentration of sodium chloride. Halophiles are vital sources of various enzymes including hydrolases, which are very stable and catalytically highly efficient at high salt concentration and other extreme conditions such as high temperature, pH and presence of organic solvents. Several hydrolases such as amylases, proteases, and lipases have been obtained from halophilic bacteria and are commonly used for various industrial applications. We initiated a screening to isolate and characterize the halophilic bacteria from the Red Sea, which is one of the saltiest bodies of water in the world. Water and soil samples, collected from the Red Sea coast, Jeddah, Saudi Arabia, were screened for isolation of halophilic bacteria. Ten bacterial isolates were obtained, which were characterized by biochemical tests and 16S rRNA gene sequencing. Hydrolase producing bacteria among the isolates were screened by plate assay on starch and gelatin agar plates for amylase and protease, respectively. Two bacterial isolates i.e. Bacillus glycinifermentans S3 and Enterobacter cloacae W1were found to possess significant amylase and protease activity.


Bactérias halofílicas são microrganismos que crescem de maneira ideal na presença de uma concentração muito alta de cloreto de sódio. Halófilos são fontes vitais de várias enzimas, incluindo hidrolases, que são muito estáveis e cataliticamente altamente eficientes em alta concentração de sal e outras condições extremas, como alta temperatura, pH e presença de solventes orgânicos. Várias hidrolases como amilases, proteases e lipases foram obtidas a partir de bactérias halofílicas e são comumente usadas para várias aplicações industriais. Iniciamos uma triagem para isolar e caracterizar as bactérias halofílicas do Mar Vermelho, que é um dos corpos de água mais salgados do mundo. Amostras de água e solo, coletadas na costa do Mar Vermelho, Jeddah, na Arábia Saudita, foram examinadas quanto ao isolamento de bactérias halofílicas. Foram obtidos dez isolados bacterianos, caracterizados por testes bioquímicos e seqüenciamento do gene 16S rRNA. As bactérias produtoras de hidrolase entre os isolados foram triadas por ensaio em placa em placas de amido e ágar de gelatina para amilase e protease, respectivamente. Verificou-se que dois isolados bacterianos, isto é, Bacillus glycinifermentans S3 e Enterobacter cloacae W1, possuíam significativa atividade de amilase e protease.

Background: Freeze-drying is known as one of the best methods to preserve bacterial strains. Protectant is the key factor affecting the survival rate of freeze-dried strains. In addition, salinity, bacterial suspension concentration, drying time, and other factors can also affect the survival rate of strains to varying degrees. At present, there are relatively few studies on freeze-drying preservation of marine bacteria. In the present study, we performed the freeze-drying protectant screening and optimized the preservation conditions for Pseudoalteromonas nigrifaciens, which is widely distributed in marine environment. The protective effects of the screened protectants were verified by 18 other marine bacterial strains. Results: The results indicated that the combination of 5.0% (w/v) lactose, 5.0% (w/v) mannitol, 5.0% (w/v) trehalose, 10.0% (w/v) skim milk powder, 0.5% (w/v) ascorbic acid and 0.5% (w/v) gelatin was the best choice for the preservation of P. nigrifaciens. The suggested salinity and concentration of initial cell suspension were 10 g/L NaCl and 1.0 × 109 CFU/mL, respectively. Furthermore, stationary-phase cells were the best choice for the freeze-drying process. The highest survival rate of P. nigrifaciens reached 52.8% when using 5­10% (w/v) skim milk as rehydration medium. Moreover, the other 18 marine strains belonging to Pseudoalteromonas, Vibrio, Photobacterium, Planomicrobium, Edwardsiella, Enterococcus, Bacillus, and Saccharomyces were freezedried under the abovementioned conditions. Their survival rates were 2.3­95.1%. Conclusion: Collectively, our results supported that the protectant mixture and parameters were beneficial for lyophilization of marine bacteria

In freshwater fish with external fertilization, sperm sampling can be contaminated with urine, which triggers motility and gives rise to decreased fertilization success. The maintenance of freshwater fish in hyperosmotic conditions may reduce urine production and improve sperm quality. Thus, the aim of this work was to verify if acute exposure to various NaCl concentrations improves sperm quality in the yellowtail tetra Astyanax altiparanae. Spermiation was induced using a single dose of carp pituitary gland (5 mg kg-1) and the males were maintained at various NaCl concentrations: NaCl 0.00% (control), NaCl 0.45% (hypoosmotic), NaCl 0.9% (isosmotic) and NaCl 1.0% (hyperosmotic) for 6 h at 26 °C. Sperm was collected and verified for activation by urine and motility traits. At 0.00%, 0.45%, and 0.90%, the sperm was motile just after sampling, indicating activation by urine. Surprisingly, at hyperosmotic conditions, no activation was observed. Other sperm and motility parameters did not show any statistical differences, including sperm viability (P = 0.7083), concentration (P = 0.9030), total motility (P = 0.6149), VCL (curvilinear velocity; P = 0.1216), VAP (average path velocity; P = 0.1231) and VSL (straight-line velocity; P = 0.1340). Our results indicate that acute maintenance at hyperosmotic conditions eliminates sperm activation by urine and maintains sperm quality. Such a new procedure is interesting for both basic and applied sciences, including reproductive practice in fish.(AU)


Em peixes de água doce com fertilização externa, a amostragem de espermatozoides pode ser contaminada pela urina, o que desencadeia motilidade e gera menor sucesso na fertilização. A manutenção de peixes de água doce em condições hiperosmóticas pode reduzir a produção de urina e melhorar a qualidade do esperma. Assim, o presente trabalho foi delineado para verificar se a exposição aguda a várias concentrações de NaCl melhora a qualidade do esperma no tetra-amarelo Astyanax altiparanae. A espermiação foi induzida usando uma dose única de hipófise da carpa (5 mg kg-1) e os machos foram mantidos em várias concentrações de NaCl: NaCl 0,00% (controle), NaCl 0,45% (hipoosmótico), NaCl 0,9% (isosmótico) e NaCl 1,0% (hiperosmótico) por seis horas a 26 °C. O esperma foi colhido e verificado quanto à ativação por urina e traços de motilidade. Em 0,00%, 0,45%, 0,90% os espermatozóides eram móveis logo após a amostragem, indicando ativação pela urina. Surpreendentemente, em condições hiperosmóticas, nenhuma ativação foi observada. Outros parâmetros espermáticos e de motilidade não mostraram diferenças estatísticas, incluindo viabilidade espermática (P = 0,7083), concentração (P = 0,9030), motilidade total (P = 0,6149), VCL (Velocidade Curvilinear; P = 0,1216), VMD (Velocidade Média de Deslocamento; P = 0,1230) e VLR (Velocidade em linha Reta; P = 0,1340). Nossos resultados indicam que a manutenção aguda em condições hiperosmóticas elimina a ativação do esperma pela urina e mantém a qualidade do esperma. Salinity is one of the factors that negatively impact plant growth and productivity; therefore, it is necessary to seek solutions that help mitigate the degenerative action of saline stress on crops. The objective of this study was to evaluate the effects of salicylic acid (SA) on the germination and vigor of Mesosphaerum suaveolens (L.) Kuntze seeds subjected to salinity. The experiment was carried out using a completely randomized design, in an incomplete 5 × 5 factorial scheme, with five levels of electrical conductivity of irrigation water (ECw - 0.0, 1.45, 5.00, 8.55, and 10.00 dS m-1) and five doses of SA (0.0, 0.29, 1.00, 1.71, and 2.00 mM), with four replications of 50 seeds, totaling nine combinations generated via a central composite design. The germination percentage, first germination count, germination speed index, average germination time, percentage of abnormal seedlings, shoot, root, and seedling lengths, and total seedling dry mass were evaluated. Salicylic acid attenuated the effect of saline stress on initial growth, mitigating damage to root and shoot lengths. Seed priming with salicylic acid improved germination and seed vigor.(AU)


A salinidade é um dos fatores que promove a inibição no crescimento e produtividade vegetal, sendo necessária a busca por alternativas que auxiliem na mitigação da ação degenerativa do estresse salino às culturas. Objetivou-se com este trabalho avaliar doses de ácido salicílico na germinação e vigor de sementes de Mesosphaerum suaveolens (L.) Kuntze submetidas à salinidade. O experimento foi conduzido em delineamento inteiramente casualizado, em esquema fatorial incompleto 5 x 5, com cinco níveis de condutividade elétrica da água de irrigação (CEa de 0,0; 1,45; 5,00; 8,55 e 10,00 dS m-1) e cinco doses de AS de (0,0; 0,29; 1,00; 1,71 e 2,00 mM L-1), com quatro repetições de 50 sementes, totalizando nove combinações geradas via matriz Composto Central de Box. As variáveis avaliadas foram: porcentagem de germinação, primeira contagem de germinação, índice de velocidade de germinação, tempo médio de germinação, porcentagem de plântulas anormais, comprimento da parte aérea, radícula e plântula e massa seca total de plântulas. O ácido salicílico atenua o efeito do estresse salino sobre o crescimento inicial, mitigando os danos sobre o comprimento de radícula e da parte aérea. O tratamento das sementes com ácido salicílico melhora a germinação e o vigor das sementes.(AU)

The influence of salinity and temperature on larval fish assemblages, as well as, temporal and vertical patterns on larval fish assemblages off the inlet of the Presidio River, southeast coast of the Gulf of California were evaluated. Samplings for ichthyoplankton and environmental parameter measurements were carried out at three depths, in five sampling sites, during September and December 1994, and April and June 1995. Anchovies and herrings (Clupeiformes) were the most abundant larval fish accounting for 77% of the total abundance. A significant relationship between salinity and the abundance of larval herrings (Opisthonema medirastre and O. dovii) and between the water temperature and the abundance of the weakfish (Cynoscion reticulatus) was found. Anchovies (Anchoa lucida, A. walkeri, and A. nasus) were present in all sampled months, while O. medirastre and O. dovii occurred in December and June, and C. reticulatus in September. Larvae of pelagic fish were more abundant near the surface, while larvae of demersal fish were more abundant near the bottom. The present study, by emphasising the importance of considering temporal and vertical changes in larval fish assemblages in coastal environments with ecological and economic importance, will be useful for designing more efficient sampling programs.(AU)


Se evaluó la influencia de la salinidad y la temperatura, así como patrones temporales y verticales en la comunidad de larvas de peces enfrente del río Presidio, costa sudeste del Golfo de California. Los muestreos de ictioplancton y las mediciones de los parámetros ambientales fueron realizados a tres profundidades, en cinco sitios de muestreo, durante septiembre y diciembre de 1994, y abril y junio de 1995. Las anchoas y sardinas (Clupeiformes) fueron las larvas de peces más abundantes representando el 77% de la abundancia total. Se encontró una relación significativa entre la salinidad y la abundancia de larvas de sardinas (Opisthonema medirastre and O. dovii), y entre la temperatura del agua y la abundancia de la corvina rayada (Cynoscion reticulatus). Las anchoas (Anchoa lucida, A. walkeri y A. nasus) estuvieron presentes en todos los meses muestreados, mientras que O. medirastre y O. dovii ocurrieron en diciembre y junio, y C. reticulatus en septiembre. Las larvas de peces pelágicos fueron más abundantes cerca de la superficie, mientras que las larvas de peces demersales fueron más abundantes cerca del fondo. El presente estudio, al enfatizar la importancia de considerar los cambios temporales y verticales en el ensamblaje de larvas de peces en ambientes costeros con importancia ecológica y económica, será útil para diseñar programas de muestreo más eficientes.(AU)

BACKGROUND: Soil salinity causes huge economic losses to agriculture productivity in arid and semiarid areas world-wide. The affected plants face disturbances in osmotic adjustment, nutrient transport, ionic toxicity and reduced photosynthesis. Conventional breeding approaches produce little success in combating various stresses in plants. However, non-conventional approaches, such as in vitro tissue culturing, produce genetic variability in the development of salt-tolerant plants, particularly in woody trees. RESULTS: Embryogenic callus cultures of the date palm cultivar Khalas were subjected to various salt levels ranging from 0 to 300 mM in eight subcultures. The regenerants obtained from the salt-treated cultures were regenerated and evaluated using the same concentration of NaCl with which the calli were treated. All the salt-adapted (SA) regenerants showed improved growth characteristics, physiological performance, ion concentrations and K+/Na+ ratios than the salt non-adapted (SNA) regenerants and the control. Regression between the leaf Na+ concentration and net photosynthesis revealed an inverse nonlinear correlation in the SNA regenerants. Leaf K+ contents and stomatal conductance showed a strong linear relationship in SA regenerants compared with the inverse linear correlation, and a very poor coefficient of determination in SNA regenerants. The genetic fidelity of the selected SA regenerants was also tested using 36 random amplified polymorphic DNA (RAPD) primers, of which 26 produced scorable bands. The primers generated 1-10 bands, with an average of 5.4 bands per RAPD primer; there was no variation between SA regenerants and the negative control. CONCLUSION: This is the first report of the variants generated from salt-stressed cultures and their potential adaptation to salinity in date palm cv. Khalas. The massive production of salt stress-adapted date palm plants may be much easier using the salt adaptation approach. Such plants can perform better during exposure to salt stress compared to the non-treated date palm plants.

Abstract The response of two local maize (Zea mays L.) genotypes designated as Sahwal-2002 (salt-tolerant) and Sadaf (salt-sensitive) to salt stress was investigated under controlled growth conditions. The role of phenylalanine and seed priming under salt stress in maize with different morphological parameters were studied. The genotype Sadaf, being salt-tolerant, experienced more oxidative damage than the Sahiwall-2002 genotype under salt stress. The salinity affected both growth and physiological attributes of the maize species whereas the phenylalanine successfully increased the salinity tolerance in maize species at the seedling stage.

Abstract Silicon accumulation is known to improve tolerance of plants under both biotic and abiotic stress. Salinity stress is an inevitable crisis causing wide spread damage to rice leading to food insecurity. The influence of Si (1mM) on two rice cultivars cv. Ghanteswari (high accumulator) and cv.Badami (low accumulator) which differs in Si uptake potential under saline (10ds/m EC) and non- saline conditions were studied in nutrient culture. The Si transporter genes were isolated and characterized to determine their function in salinity tolerance. Under stress, there was an increase in Si accumulation, Na+/K+ ratio, electrolyte leakage, lipid peroxidation and antioxidant activities. On addition of silicon, the K+ uptake increased, membrane damage reduced and osmolytes balance improve under salinity. But, the level of resurgence was varied in both cultivars, due to their differential Si-accumulation. Molecular characterizations of Lsi1 protein revealed its involvement in the movement of ion and water and therefore prevent osmotic stress. The Lsi2 is responsible for removal of Na+, reducing salt toxicity. Silicon accumulation is responsible for maintenance of cell water status, osmotic balance and Na+ ion exclusion during high salinity. The variable relative expression of Lsi2 provides a possible explanation for differential genotypic uptake of silicon.

OBJECTIVES: This study examined the salinity of soups provided at childcare centers by measuring the salinity for three years and providing basic data for sodium reduction.METHODS: The soup salinity was measured using a Bluetooth salinity meter from January 2015 to December 2017 at 80 childcare foodservice establishments enrolled in the Suseong Center for Children's Foodservice Management in Daegu.RESULTS: An analysis of the soup salinity each year showed that the salinity decreased significantly from 0.48% in 2015 to 0.41% in 2017, particularly in clear soups and soybean soups compared to other soups (P < 0.05). The salinity and sodium content in seafood soups (0.45% and 179.1 mg/100 g, respectively) were highest, followed by soybean soups (0.44%, 175.2 mg/100 g), with perilla seed soups containing the lowest (0.42%, 167.2 mg/100 g) (P < 0.05). The salinity was significantly higher in institutional foodservice establishments than small foodservice establishments (P < 0.001). The salinity and sodium content were the highest in foodservice establishments with a small number of measurements, and the salinity was the lowest in foodservice establishments with salinity measurements performed an average of 151 times each year (three times a week) or more (P < 0.05). The soup salinity was low in the order of winter, spring, summer, and autumn, and the salinity decreased significantly year by year in all seasons. (P < 0.05).CONCLUSIONS: The soup salinity was significantly lower in foodservice establishments where the salinity was measured more than three times a week, indicating that continuous salinity management is effective.
