RESUMEN

OBJECTIVE@#To present on a prenatally diagnosed case with complex structural rearrangements of chromosome 8.@*METHODS@#Chromosome karyotyping, chromosomal microarray analysis (CMA) and fluorescence in situ hybridization (FISH) were carried out for a fetus with increased nuchal thickness.@*RESULTS@#The karyotype of the amniotic fluid sample showed extra materials on 8p. FISH revealed a centromeric signal at the terminal of 8p with absence of telomeric signal. CMA revealed partial deletion of 8p23.3 [(208049_2256732)×1], partial duplication of 8p23.3p23.2 [(2259519_3016818)×3], and partial duplication of 8q [8q11.1q12.2(45951900_60989083)×3].@*CONCLUSION@#The complex structural rearrangements of chromosome 8 in this case has differed from the commonly seen inv dup del(8p).

RESUMEN

Objective@#Anesthesia-induced cognitive impairments are common for elder patients after surgery. Oxidative stress is the predominant factor contributing to the impairments. This study was to assess the therapeutic potential of an anti-oxidative naturally occurring flavonoid, chrysin, in attenuating sevoflurane-induced cognitive impairments in rat models. @*Methods@#Rat models of cognitive impairments were constructed by exposing aged rats (18 months old) to sevoflurane for 2 h. Chrysin was administered via oral gavage at the dose of 25, 50, and 100 mg/kg/day for seven days. The elevated plus maze test was used to assess anxiety and explorative behaviors. Spatial memory tests were performed using novel object recognition test, object location memory task, and water maze experiments. Oxidative stress was evaluated by measuring levels of malondialdehyde, nicotinamide adenine dinucleotide phosphate, 4-hydroxynonenal, and glutathione using colorimetric assays. Quantitative real-time polymerase chain reaction and Western blot were used to analyze how chrysin affects nuclear factor E2-related factor (Nrf) signaling. @*Results@#While sevoflurane anesthesia led to significant decline in cognitive performance in object recognition test, object location memory task, and water maze test, chrysin exerted significant effects in alleviating the impairments. Oxidative stress was also reduced in the hippocampus tissue of rats after chrysin intake. Nrf signaling was activated by chrysin treatment in sevoflurane-induced cognitive impairment models. @*Conclusion@#Chrysin was effective in alleviating cognitive impairments induced by sevoflurane anesthesia, which was at least in part facilitated by its effects in reducing oxidative stress via activating Nrf signaling.

Objective:To investigate the correlation between CD133 expression and clinicopathological features in triple negative breast cancer (TNBC) patients, and the impact of CD133 on prognosis in these patients.Methods:Data of 70 patients who received surgical treatment in our center from Jan. 2008 to Dec. 2012 were collected. Immunohistochemistry was used to examine the expression of CD133. Patients were divided into two groups according to CD133 expression. Univariate analysis, Cox and Logistic regression multivariate analysis were used in order to investigate the correlation between CD133 expression and clinicopathological features. Kaplan-Meier curve and Log-rank analysis were used to evaluate DFS (disease-free survival) and OS (overall survival) .Results:CD133 was expressed in cytomembrane and cytoplasm with expression rate of 95.71% (67/70) . Of which, 64.29% (45/70) of patients were low CD133 expression and 35.71% (25/70) were high expression. High CD133 expression was significantly correlated with younger age (≤50) ( P=0.007) and larger tumor size (>2 cm) ( P=0.020) . Tumor size ( P=0.035) , axillary status ( P=0.001) , Ki67 ( P=0.005) and CD133 expression ( P=0.014) were independent predictors of recurrence and metastasis in TNBC patients. Axillary status was independent predictor of death event ( P=0.008) . Increased CD133 was associated with poor prognosis. Compared with high expression, patients with low CD133 expression had better DFS ( P=0.002) and OS ( P=0.088) , while OS did not reach significant difference. Conclusion:CD133 expression was correlated with age and tumor size in TNBC patients. High expression was associated with recurrence, metastasis and poor prognosis. Thus, CD133 may be a potential biomarker in predicting prognosis in TNBC.

OBJECTIVE:To realize refined management of tablets i n the inpatient pharmacy ,and to ensure the medication safety of patients. METHODS :Based on intelligent pharmacy ,the dispensing and packaging process under the automated drug dispensing and packaging system (ADDPS)mode was optimized and modified ;PDA barcode scanning technology was applied in all links of taking ,dismantling and adding ,so as to realize the real-time tracking of batch number and inventory ,and improve the drug closed-loop management of tablets. The error rate ,staff consumption time and pharmacist/nurse satisfaction were compared before and after the process improvement. RESULTS :After the process improvement ,the dispensing error rate was decreased from 1.637‰ before improvement to 0.082‰(P<0.01);the staff consumption time decreased from (7.52±0.33)h to (5.11±0.24)h (P<0.01);the pharmacist/nurse satisfaction increased from 66.5% to 93.4%(P<0.01). CONCLUSIONS :Based on ADDPS ,the application of PDA barcode scanning technology standardizes the tablets management of inpatient pharmacy ,supplements and improves the drug closed-loop information ,realizes batch number tracking and inventory management ,reduces the occurrence of tablet dispensing errors ,improves the work efficiency and satisfaction of pharmacists ,and ensures the safety of clinical medication.

RESUMEN

Tuberculous pleurisy (TP) is a common disease, especially in areas with high prevalence of tuberculosis. However, because of the lack of bacterial characteristics in pleural effusion, the detection sensitivity of traditional smear and culture method is low, and the pleural biopsy has certain risks; therefore, the diagnosis of tuberculous pleurisy is challenging. Due to the limitations of traditional detection methods, to find a fast, accurate and effective method for TP diagnosis is important. This article reviews the progress of molecular, biochemical and immunological methods for diagnosis of TP.

Objective:To investigate the clinical significance of gonadotropin-releasing hormone (GnRH) and its receptor (GnRHR) expression in breast cancer.Methods:GnRH and GnRHR expression data in The Cancer Genomes Atlas (TCGA) database and its clinical information were downloaded. Statistically assessed was performed for relationship with clinicopathological factors and prognosis.Results:The expression of GnRH in breast cancer tissues was lower than that in normal breast tissue (0.42 vs 1.27, P=0.000) , and it was correlated with age ( P=0.046) , race ( P=0.000) , lymphnode status ( P=0.003) , ER ( P=0.000) , PR ( P=0.000) , and HER2 ( P=0.000) . The GnRH expression was higher in patients whose age ≤55 years, black or African American, lymphnode negative, ER negative, PR negative, and HER2 negative. Survival analysis suggested that the Overall Survival (OS) in GnRH high expression group was better than in low expression group ( P=0.018) . The expression of GnRHR in breast cancer tissues was similar to normal breast tissue (0.08 vs 0.07, P=0.778) , and it was correlated with age ( P=0.031) , race ( P=0.000) , ER ( P=0.000) , PR ( P=0.000) , and HER2 ( P=0.030) . The GnRHR expression was higher in patients whose age >55 years, white, ER positive, PR positive, and HER2 negative. There was no significant difference in OS between GnRHR high expression group and low expression group ( P=0.719) . Subgroup analysis showed that OS in GnRHR high expression group was better than in low expression group ( P=0.028) in the triple negative breast cancer subgroup, while GnRHR was not associated with OS in the non-triple negative subgroup ( P=0.976) . Conclusion:The expressions of GnRH and GnRHR are correlated with some clinicopathological parameters of breast cancer, and the prognosis of oreast cancer (especially triple negative breast cancer) . The GnRH and GnRHR signaling pathways maybe have tumor suppressor activity.

Objective To investigate the mechanism of AR/let-7 signaling pathway in inhibiting the proliferation of TNBC and its significance for survival.Methods Human breast cancer MDA-MB-453 cells were cultured in vitro and divided into experimental group and control group.The experimental group was added with androgen dihydrotestosterone(DHT),and the control group was added nothing.The cell proliferation was detected by CCK-8,cell cycle was detected by flow cytometry,AR expression was detected by Western blot,and let-7 expression was detected by real-time fluorescent quantitative PCR.The AR,let-7 expression data and survival data of TNBC patients were downloaded from the Cancer Genomes Atlas (TCGA).The expression of AR and let-7 between cancer tissues and normal breast tissues and their relationship with survival was analyzed.Results Cellular experiments showed that the proliferation rate of cancer cells in the experimental group was significantly lower than that in the control group(1.22±0.11 vs 2.26±0.23,t=7.065,P<0.05),and the ratio of G1/S in the experimental group was greater than in the control group (1.08±0.03 vs 0.68±0.03,t=17.321,P=0.000).The AR and let-7a,b,c,and d were overexpressed in the experimental group.The TCGA data showed that AR,let-7a-1,let-7a-2,let7a-3,and let-7c were lower in breast cancer tissues than in normal tissues (P<0.05),while let-7d was higher in breast cancer tissues (P<0.05).The AR,let-7a-1,let-7a-2,let-7a-3,and let-7c were used to cluster the patients into high-expression group and low-expression group,and the overall survival in the high-expression group appeared to be higher,while the difference was not statistically significant (P=0.163).Conclusions The AR/let-7 signaling pathway is up-regulated by DHT activation,which blocks cells in the G1 phase and inhibits cell proliferation.Patients with high expression of AR,let-7a-1,let-7a-2,let-7a-3,and let-7c may have better overall survival.It is suggests that the AR/let-7 signaling pathway may become a new target for TNBC.

Objective To investigate the expression of microRNA (miRNA)-3620 in the plasma of patients with anti-tuberculosis drug-induced hepatotoxicity (ATDH).Methods A total of 35 patients with ATDH and 35 non-ATDH paired individuals were included in this study.Plasma miRNA-3620 levels were detected using real-time Polymerase chain reaction.Comparison between two groups was done with t test.Receiver operation characteristic (ROC) curve analysis was performed to determine the diagnostic value of miRNA-3620 in ATDH.Results The relative expression of plasma miRNA-3620 of patients with ATDH and non-ATDH were 1.65±1.43 and 0.71±0.45, respectively, with significantly statistical difference (t=3.703, P<0.01).The cut off value of miRNA-3620 expression was 1.15 and the area under ROC curve were 0.71(95% CI: 0.43-1.45).Based on this cutoff value, the sensitivity and specificity of miRNA-3620 in diagnosing ATDH were 60.0% and 82.9%, respectively;the positive predictive value was 77.8% and the negative predictive value was 67.4%.Twenty-one ATDH cases and 29 non-ATDH cases was correctly diagnosed, with the accuracy of 71.4%.Conclusion The expression of miRNA-3620 in plasma is significantly increased in ATDH patients.

Objective:To observe the effect of combining Chinese medicine and tuina along the meridians on motor function and activities of daily living (ADL) in patient with post-stroke upper limb spasticity. Methods:A total of 220 patients with post-stroke upper limb spasticity were randomly allocated into a treatment group (n=110) and a control group (n=110). Patients in the treatment group received tuina along the meridians combined with spasticity-alleviating and collateral-unblocking Chinese medicine, whereas patients in the control group received routine rehabilitation therapy. Patients in both groups were treated for 3 weeks. Then the patients’ motor function, ADL and muscle tone were evaluated before and after treatment using the Fugl-Meyer assessment scale (FMA), modified Barthel index (MBI) and modified Ashworth scale (MAS). Results:After treatment, the FMA scores, MBI scores, and muscle (shoulder intortor, elbow flexors and wrist flexors) tones were significantly improved (P<0.05), but the improvement was more significant in the treatment group than that in the control group (P<0.05). Conclusion:Tuina along the meridians combined with spasticity-alleviating and collateral-unblocking Chinese medicine can substantially alleviate muscle tone on the affected side and remarkably improve the patients’ motor function and ADL.

Objective To explore the effect of family rehabilitation on growth and development of children with hypoxic ischemic en-cephalopathy (HIE). Methods 80 children with moderate HIE were divided into control group (n=40) and observation group (n=40). The control group received routine intervention and nursing, and the observation group accepted family rehabilitation in addition. Mental devel-opment index (MDI) and psychomoter developmental index (PDI) were compared when they were 3 and 6 months old with intellectual de-velopment testing, and their development quotients (DQ) were compared at the age of 1 year. Results The MDI and PDI scores were higher (P<0.01) in the observation group than in the control group when they were 3 and 6 months old. The DQ were higher (P<0.05) in the obser-vation group than in the control group when they were 1 year old. Conclusion Family rehabilitation can promote the development of intelli-gence in children with HIE.

The gene encoding thermostable lactate dehydrogenase (Tm-LDH) was cloned into the plasmid pHsh from Thermotoga maritima, and expressed in Escherichia coli JM 109. The recombinant protein was purified to homogeneity by a simple step, heat treatment. The recombinant enzyme had a molecular mass of 33 kDa. The optimal temperature and pH of Tm-LDH were observed 95 degrees C and 7.0. The purified enzyme had a half-life of 2 h at 90 degrees C, and exhibited better stability over a pH range from 5.5 to 8.0. The K(m) and V(max) values were 1.7 mmol/L, 3.8 x 10(4) U/mg of protein for pyruvate, and 7.2 mmol/L and 1.1 x 10(5) U/mg for NADH, respectively. The expression of Tm-LDH in T7 system could not obtain high efficiency, but it has been soluble over-expression in pHsh system and reached 340 mg/L. The superior stability and productivity of Tm-LDH will lay the foundation of its industrial-scale fermentation and application in the NAD regeneration.

Objective To study the expression of insulin-like growth factor-Ⅱ receptor(IGF-Ⅱ R) and its relationship with the prognosis of human epidermalgrowth factor receptor 2 (HER2) positive breast cancer.Methods The expression of IGF-Ⅱ R in 104 specimens of HER2 positive breast cancer was evaluated by immunohistochemistry.The expression was rated on 2 scales:negative and positive.The age,pathological type,tumor size,lymph node involvement,status of estrogen receptor(ER),and progesterone receptor(PR) of all the cases were reviewed.The relationship between the expression of IGF-Ⅱ R and the above pathological parameters were analyzed by statistical methods.The relationship between the expression of IGF-Ⅱ R and the disease-free survival and overall survival was studied.Results The positive expression rate of IGF-Ⅱ R was 43.3% (45/104).The positive rate of IGF-Ⅱ R was significantly higher in the lymph node positive group than in the lymph node negative group (61.5 % vs 25.0%,P =0.000).No significant correlation was observed between the expression of IGF-Ⅱ R and age,pathological type,tumor size,status of ER,or PR.The disease-free survival and overall survival was significantly lower in IGF-Ⅱ R positive group than in IGF-Ⅱ R negative group.(The 5-year disease-free survival:62.6% vs 84.7%,P =0.022; The 5-year overall survival:71.5% vs 89.6%,P =0.024).Cox regressive analysis showed that the lymph node status was an independent risk factor of disease-free survival and overall survival,while the IGF-Ⅱ R was not.Conclusion The positive expression of IGF-Ⅱ R in HER2 positive breast cancer indicates poor prognosis.The expression of IGF-Ⅱ R may be regulated by insulin-like growth factor-Ⅱ (IGF-Ⅱ).

Objective To study the clinicopathologic characteristics of triple-negative breast cancer (TNBC) and its value in the prediction of prognosis. Method In this study,500 cases of female breast cancers were examined immunohistochcmically for the TNBC. The clinicopathologic characteristics of the 243 TNBC cases were inspected. Results TNBC accounted for 17.6% (88/500) of the 500 breast cancers. The histological types of the TNBC included mainly infihrative ductal carcinoma, metaplastic carcinoma and medullar carcinoma. Among those, histological grade Ⅲ accounted for 72.7% (64/88) of all the TNBC and was more common than that in hormone receptor positive breast cancers (HR+ group ) and Her-2 overexpression breast cancers (Her-2 group)(P=0.000). The positive rates of CK5/6 and EGFR in the TNBC were 30.7% (27/88) and 34.1% (30/88), respectively. The positive rates of ERCC1 and KIT in the TNBC were 28.4% (25/88) and 34.1% (30/88), respectively, Both of which were higher than those in the HR + group and Her-2 group, respectively (P=0.032 and P=0.026). 3-year survival rate of the TNBC was 71.5% and it was lower than that of HR group (P=0.021) and not significantly different from that of Her-2 group (P=0.474). Conclusions TNBC is the breast cancer with high aggressive pathologic futures and poor prognosis. EGFR and ERCC1 expression were positive in a portion of TNBC cases.

Objective To establish a cell line and a convenient and well-duplicated minimal residual leukemia (MRL) model. Methods We chose the international standard domestic inbred DBA/2 mouse to inoculate the different numbers of L1210 leukemic cells to investigate the relationship between inoculating cell number and mouse survival time. On the third day after 1×106 leukemic cells per mouse were inoculated, different doses of CTX were used for the chemotherapy. Results The inoculating number of L1210 leukemic cells increased while the mouse survival time decreased;The survival time increased following the increase of the doses of the CTX;The survival time of the inoculated mouse treated with CTX at 125 mg/kg was equal to that of the mouse inoculated with 500 L1210 leukemic cells without treatment. Conclusion The observation results are that the dose of 125 mg/kg for creating the model of MRL mouse.

Objective To compare the clinical effects of 3i nternal fixation devices,130?comp res-sion-plate,Richards screw-plate system and Gamma locking nail,in tre atment of intertrochanteric fractu res of femur in order to provide evidence fo r clinical choice.Methods The 3different internal fixation devices were statistically compared in terms of t heir surgery time,volume of operative bleeding,hospitalization time,postop-erative complications,as well as excellent and good rate of function in t reatment of intertrochanteric frac tures of femur.Results Compared with the other two treatmen t groups,the Gamma locking nail grou p presented the best therapeutic effects.Its posto perative complications were significantly lower and its excellent and f ine rate of function was significantly higher t han those of the other two groups.Conclusion Gamma locking nails are a kind of effective internal fixation device for the treatment of intertro chanteric fractures in elderly patients,because it has better biomechanical properties which facilitate fracture healing and recovery of hip joint function.[

Objective To investigate the effect of pancreatic transplantion(PTA) using diverse operative (methods) in rats. Method Inbred SD rats were used as donor and recipient, and were randomly assigned (into) 4 groups:Normal control group( Group NC), consisted of 10 rats;diabetes group(Group DC) consisted of 10 rats;PTA with enteric drainage group(Group E-D), consisted of 22 diabetic rats;PTA using bladder reconstruction group(group B-D), consisted of 22 rats.Arterial supply of transplanted pancreas was (performed) by using end-to-side anastomosis of the donors' abdominal aorta(with splenic artery) and (recipients)′ abdominal aorta; and venous drainage was performed by using end-to-end anastomosis of the (donors)' portal vein segment(with splenic vein) and (recipients)′ left renal vein(cuff method). The fasting blood glucose, body weight, food intake, water intake and urine volume of the recipient were monitored before transplantation and on the 1st, 3rd, 7th, 14th, 30th day after operation, and also the reasons of failures were analyzed. Objective To investigate the effect of a new cloned full length gene, C2 gene which encodes a translation initiation factor, on cell cycle and the relationship between C2gene and apoptosis. Methods Construct a eukaryotic vector carrying the full length of C2 gene, then transfected it into a gastric cell line-SGC7901 cells and gained cells expressing C2 protein transiently or stably. Test the expression of C2 protein and the change of cell cycle of the transfected cells using the flurescence activatied cell sorting(FACS) and the changes of their ultra structure electromicroscopically. Results The C2 gene eukaryotic expression vector was successfully constructed. FACS showed the C2 protein expression ratio of transfected cells was 32.3%, while the empty control group was 0.9%. The test of cell cycle showed that there was appeared apoptosis peak in transiently or stably transfected cells. The apoptosis cells can be seen electromicroscopically among the C2 transfected cells. Conclusions C2 gene transfection can lead SGC7901 cells to apoptosis, but the mechanism has to be further study.

Objective To investigate the effect of a new cloned full length gene, C2 gene, which encodes human eukaryotic translation initiation factor, on tumorigenesis of human gastric cancer cells in vivo and in vitro. Methods A constructed eukaryotic vector carrying the full length of C2 gene was amplified, purified, and transfected into a gastric cell line SGC7901 cell. The expression of C2 protein was examined with fluorescence activated cell sorting (FACS) and Western Blot. The effect of C2 gene on tumorigenesis of human gastric cancer cells was investigated in vitro and in vivo with methods of clone formation in plate and oncogenesis in nude mouse. Results FACS and Western Blot showed that C2 protein was expressed highly and stably in transfected cells. The average clone formation rate of C2 gene transducted cells is 43.8%, the rate which is lower than that of vector transducted cells (76.9%). In vivo, the time of tumorigenesis of C2 gene transducted cells in nude mouse is prolonged from 1 week to 2 weeks, and the volume of tumor node is smaller than that of vector transducted cells, that is, 1.20 and 5.58 cm 3 respectively ( P

AIM: To determine if aquaporin-1 (AQP-1) expression and function is influenced after lipopolysaccharide (LPS) stimulation in rat lung microvessel endothelial cells and to investigate the mechanisms of lung fluid abnormal metabolism in acute lung injury. METHODS: LPS at different concentrations (100 ?g/L, 1 mg/L or 10 mg/L) was used to stimulate cultivated rat lung microvessel endothelial cells in vitro at different stimulatory times (4 h, 12 h or 24 h), respectively. Tritium water permeation was conducted for determining the intracellular signal intensity in rat lung microvessel endothelial cells.
