RESUMEN

OBJECTIVE@#To summarize the research progress on the role of macrophage-mediated osteoimmune in osteonecrosis of the femoral head (ONFH) and its mechanisms.@*METHODS@#Recent studies on the role and mechanism of macrophage-mediated osteoimmune in ONFH at home and abroad were extensively reviewed. The classification and function of macrophages were summarized, the osteoimmune regulation of macrophages on chronic inflammation in ONFH was summarized, and the pathophysiological mechanism of osteonecrosis was expounded from the perspective of osteoimmune, which provided new ideas for the treatment of ONFH.@*RESULTS@#Macrophages are important immune cells involved in inflammatory response, which can differentiate into classically activated type (M1) and alternatively activated type (M2), and play specific functions to participate in and regulate the physiological and pathological processes of the body. Studies have shown that bone immune imbalance mediated by macrophages can cause local chronic inflammation and lead to the occurrence and development of ONFH. Therefore, regulating macrophage polarization is a potential ONFH treatment strategy. In chronic inflammatory microenvironment, inhibiting macrophage polarization to M1 can promote local inflammatory dissipation and effectively delay the progression of ONFH; regulating macrophage polarization to M2 can build a local osteoimmune microenvironment conducive to bone repair, which is helpful to necrotic tissue regeneration and repair to a certain extent.@*CONCLUSION@#At present, it has been confirmed that macrophage-mediated chronic inflammatory immune microenvironment is an important mechanism for the occurrence and development of ONFH. It is necessary to study the subtypes of immune cells in ONFH, the interaction between immune cells and macrophages, and the interaction between various immune cells and macrophages, which is beneficial to the development of potential therapeutic methods for ONFH.

RESUMEN

Objective:To investigate the therapeutic effects of small peptide,alkaloid,total flavonoids and polysaccharides extracted from sunflower plate powder on the hyperuricemia in the mice,and to clarify that the influence of sunflower powder in the uric acid(UA)level and joint swelling and its protective effect on the liver and kidney tissues.Methods:A total of 96 male Kunming rats were randomly divided into 8 groups,each group of 12 rats.One group was used as blank control group,the others were used to set up hyperuricemia models.The successful mouse models of hyperuricemia were randomly divided into model group and positive control group (allopurinol group),small molecule peptide group,alkaloi group,total flavone group,polysaccharide group and sunflower disk group.The serum UA levels were measured 7 d after administration and the liver and kidney tissues of the mice in various groups were taken.The morphological changes of liver and kidney tissues were observed by HE staining.Acute gouty arthritis model was established by injecting 3 mg sodium urate into the knee joint of the male SD rats.The experiment was divided into blank group,model group,positive control colchicine group, sunflower disc extracts(SDE)group with different concentrations(0%,20%,40%,60%,80% and 100%), 4 rats in each group.The joint circumference was measured at 0,12,24 and 48 h,respectively.The serum levels of interleukin-10(IL-10)and macrophage inflammatory protein-1α(MIP-1α)were detected by ELISA kit. Results:Compared with model group,the serum UA levels of the mice in allopurinol group,small molecule peptide group,alkaloid group,total flavonoids group and sunflower group were significantly decreased after administration (P<0.05);and the serum UA level in allopurinol group was decreased the most,followed by small molecule peptide group.The HE staining results showed that the hepatocytes in small molecule peptide group were clear, the hepatocytes had full cytoplasm,and no abnormality was observed in the kidney cells compared with blank control group.After 48 h injection of sodium urate,the degree of joint swelling was reduced by up to 27.1% in SDE group compared with model group.Compared with model group,the serum IL-10 level in 20% SDE group was significantly increased(P<0.01),and the serum MIP-1α level was also significantly increased(P<0.01). Conclusion:Sunflower powder can reduce the UA level and has protective effect on the liver and kidney tissues and it can improve the body's anti-inflammatory activity and reduce the body's inflammatory response to achieve anti-gout arthritis activity.

Intervertebral disc degeneration (IVDD) takes great affection on people health all over the world.With the extension of human life and aging,the number of sick people will grow rapidly.At present,both conservative and surgical treatment can only alleviate the symptoms,rather than the fundamental treatment for the process of degeneration.Therefore,the application of cell therapy in the treatment of degenerative disc regeneration is an innovative treatment.However,it's a great bottleneck for exogenous transplanted stem cells to survive in degenerated intervertebral disc with harsh environment containing strong force,acidic pH,hypoxia,hyperosmolarity and limited nutrition.In recent years,tissue endogenous repair has been demonstrated to be present in almost all human tissues,and endogenous stem cell play an important role in the process.Similarly,nucleus pulposus was also confirmed the existence of such a kind of tissue specific stem cells-nucleus pulposus derived stem cells (NPSCs)-a new promising candidate for the treatment of IVDD.NPSCs,as a new kind of stem cells,hold either the characteristic of stem cell and the special potential of themselves,such as the nice adaption of the environment in intervertebral disc and the superior potential of cartilage differentiation.Nonetheless,the isolations of NPSCs were different;cell phenotypes were lack of wildly acceptable standard and the biological characteristics were various among different NP sources.In addition,whether the application of NPSCs could be used in nucleus pulposus tissue engineering still calls for further research.Due to the unclear in degeneration mechanism of IVDD,the study on degeneration mechanism of NPSCs may play a great important role in clarifying this issue.

Objective To study the protective effects of choline on myocardial ischemia rat heart and its potential mecha -nisms .Methods Ischemia hypoxia environment was simulated with low value of pH (pH 6 .8) and lack of oxygen .Calcium currents were recorded by whole cell patch under the voltage clamp configuration .The alternations in[Ca2 + ]induced by KCl was detected by laser scanning confocal microscope in ventricular myocytes ,then disccuss the effects of choline on calcium and calcium store in cells . Results The normalized peak currents of ICa-L in ventricular myocytes were larger in pH 6 .8 group than those in pH 7 .4 group , which can be attenuated by choline .The(Ca2 + )i induced by KCl in ventricular myocytes were significantly increased in pH 6 .8 Ty-rode solution and its increasing can be suppressed by choline .4-DAMP can inhibit the suppressing effect of choline .Conclusion The possible mechanism of M 3 receptor involved in the protection of ischemic myocardium by inhibiting myocardial cells in ICa-L ,in-hibiting intracellular calcium overload .

Objective To observe the vasodilation effect of extract of Jasminum samba (EJs), a kind of traditional Chinese medicine, on ex vivo rat thoracic aortic rings, and to investigate its mechanism. Methods On ex vivo aortic ring perfusion device, influence of EJs on contraction of the aorta induced by phenylephrine (PE) or potassium chloride (KCl) was observed. Influence of N-nitro-L-arginine-methylester ( L-NAME ), barium chloride ( BaCl2 ), glibenclamide ( Gli ) on vasodilating effect of EJs (0. 5, 1, 2, 4, 8 g·L-1 ) was detected. Effect of EJs on the contraction of calcium chloride (CaCl2 ) and PE in Ca2+-free medium was detected. [ Ca2+ ] i in vascular smooth muscle cells was determined by using laser scanning confocal microscope (LSCM). Results In blood vessels with intact endothelium, EJs concentration-dependently decreased PE- or KCl-induced vasoconstriction, the maximum dilating effect being (105. 0±3. 2)% and (78. 0±6. 5)% , respectively; L-NAME affected the vasodilatory effect of EJs on thoracic aorta rings ( P<0. 01), the maximum dilatory effect being (58. 0 ± 6. 9)% . BaCl2 and Gli had significant influence on vasodilation of EJs, and the contraction was obviously attenuated (P<0. 01), the maximum dilatory effect being (37. 0±5. 2)% and (78. 0±10. 0)% , respectively. EJs significantly inhibited contracting effect of PE on thoracic aorta rings in Ca2+-free medium (P<0. 01). The maximum contraction effect was (70. 0±6. 3)% . EJs inhibited CaCl2-induced vasoconstriction (0. 5-8 mmol·L-1 ), and vasoconstriction was decreased by (65. 0±3. 2)% . LSCM recorded that Fmax / F0 of 4 and 8 g·L-1 EJs was (2. 0±0. 2) and (1. 5±0. 2), respectively. Conclusion EJs exerted a dose-dependent vasodilating effect on rat isolated aorta rings. The mechanism might be related to promoting NO release, activating K+channels and decreasing the concentration of cytoplasmic Ca2+.

RESUMEN

Objective To observe the efficacy of bloodletting therapy plus narrow band ultraviolet B (NB-UVB) in treating prurigo nodularis.Method According to the randomized controlled principle, the enrolled patients were divided into a treatment group and a control group. The treatment group was intervened by bloodletting cupping at the selected acupoints and the topical areas plus NB-UVB once every other day; the control group was by orally taking Mizolastine sustained release tablets and external application of Halometasone cream.Result The total effective rate was 85.7% in the treatment group versus 61.9% in the control group, and the difference was statistically significant (P<0.01).Conclusion Bloodletting therapy plus NB-UVB can produce a content efficacy in treating prurigo nodularis, with few adverse reactions.

RESUMEN

Based on the characteristics of salt.alkalinized soil in northeastem China,twenty-five kinds of salt alkaline conditions with different salinities and pH were simulated by mixing NaCl,NaHCO3,Na2SO4,and Na2CO3,in various proportions and applied to sunflower (Helianthus annuus L.) seedlings to investigate the coordinative effects of salt and alkali stresses on its antioxidant enzyme system.The soil was conditioned with a salt concentration range between 50 to 250 mmol/L and pH values from 7.12 to 10.46.Several physiological indexes of stressed seedlings were measured,including the activities of superoxide dismutase (SOD),catalase (CAT),and pemxidase (POD),as well as the content of malondialdehyde (MDA).The results showed that the responses of the antioxidant enzyme system in sunflowers were influenced by salinity and alkalinity,which all three antioxidant enzymes exhibited a rise-drop pattern as salinity increased,whereas their responses to alkalinity appeared to be diverse:decreased for SOD and CAT,and increase for POD along when increasing alkalinity.A two-way analysis of variance (ANOVA) showed that the effects of salinity and alkalinity on the activities of the three enzymes were significant (P<0.001).The effect of salinity on POD and SOD was greater than that of alkalinity,whereas the effect of alkalinity on CAT was greater than that of salinity.The interrelation of salinity and alkalinity on each antioxidant enzyme was significant (P<0.001) except for SOD.The correlation and stepwise regression analyses between the activity of the antioxidant enzyme system and the MDA content were significant to difierent extend,SOD was a dominant factor,and POD was neglectable.

RESUMEN

[Objective]To compare the clinical results,advantages and disadvantages of the mono-segment lateral lumbar disc herniated patients with the treatment of micro-endoscopic discectomy(MED) and fenestration discectomy(FD).[Methods]Sixty-nine cases operated with mono-segment discectomy were analyzed from July 2003 to July 2005,among them,32 cases operated with microendoscopic discectomy and 37 cases operated with fenestration discectomy.The operating time,blood loss,time stay-in-bed,duration of hospitalization after operation were compared.[Results]The rate of excellent and good outcomes was 93.7% in MED group,and 91.9% in FD group.No significant difference was found between them,and the same with the postoperative sciatica relief significant differences ould be observed in the operation time,blood loss,duration of hospitalization after operation and postoperative visual analog scales.[Conclusion]The symptoms of the lumbar disc herniation can be reliefed effectively by both methods,but micro-endoscopic discectomy has more advantages of less trauma,less blood loss,earlier rehabilitation,less postoperative low back pain.

BACKGROUND: At present, genetic recombinant technique has been utilized to express recombinant human bone morphogenetic protein-2 (rhBMP-2)and to induce either orthotopic or ectopic regenerated bone successfully. But,because that osteoblastic activity induced by rhBMP-2 is lower than that by natural BMP-2, the perfect vectors have not been discovered yet.OBJECTIVE: BMP recombinant adenovirus (rAdV) was constructed so as to provide feasible vector for the basic treatment of bone defect.DESIGN: Single sample was designed in the experiment.SETTING: Experimental Center of Molecular Biology in First Hospital of Xi'an Jiaotong University.MATERIALS: PACCMV-PLPA plasmid, PJM17 plasmid, 293-cell line.METHODS: The experiment was performed in Experimental Center of Molecular Biology in First Hospital of Xi'an Jiaotong University from September 2001 to June 2002. Reverse Transcription-Polymerase Chain Reaction (RT-PCR) was used to clone the whole-length gene of BMP-2 and construct AdV vector. DNA-calcium phosphate coprecipitation was used to transfect accessory plasmid PJM17 into 293cell and homologous recombination was used to construct rAdV. The titer was assayed and CsC1 density gradient centrifugation and purification were applied.construction of rAdV.binant plasmid is named as pGEM-T/hBMP2, (3 015+1 213)bp in size.It was indicated in agar gel electrophoresis that the practical value was in BMP2 shuttle plasmid: The plasmid was about 10 Kbp in size. Since PACCMV-PLPA plasmid multiclone sites belonged to PUC plasmid series,EcoR Ⅰ enzyme digestion was used to linerarizate recombinant plasmid,10 Kbp in size. 8.8 Kb and 1.2 Kbp visible fragments were obtained durtion of rAdV: PCR technique was used to identify rAdV. The target fragment 1.2Kbp that was amplified by BMP2 specific primer of the whole length was in conformity completely with the theoretic value.CONCLUSION: Successful construction of BMP2 rAdV lays a foundation for the feasible genetic treatment with vector of bone defect.

RESUMEN

Objective To investigate the protective mechanisms of nerve growth factor (NGF) on spinal cord injury.Methods The spinal cord injury (SCI) of Wistar rats was performed by a 10g×2.5cm impact on the posterior T12 spinal cord.The experimental animals received NGF liquid by subarachnoid space tube.The radioimmunological techniques were applied to examine the level of endothelin.Results The level of endothelin was significantly increased after the injury as compared with that in control group(P<0.01).The level of endothelin in NGF group as obviously lowered as compared with that in normal saline group 4 h after injury (P<0.01).Conclusion NGF can protect spinal cord against injury in vivo.One of the mechanisms is that NGF could inhibit endothelin-induced vicious circle.

RESUMEN

Objective Acute spinal injury(ASI) is a kind of disease commonly seen in the orthopedic department, with secondary pathological injury causing the delayed damage of tissue structure. This study is focused on finding the injury mechanism of endothelin and the relation with calcium in SCI, and developing an effective treatment of SCI through animal experiment for clinic application in the future. Methods ASI animal model with radioimmuolo gical techniques are applied to examine the level of endothelin, and to find the pathological changes under microscope and electron-microscope. Results The quantity of endothelin and calcium in the cell with 1, 4-dihydropyridine calci um channel inhibitor is decreased, as a result, depolarization was lightened. The mechanism delays the development of secondary injury significantly. Conclusion This type of treatment may be used in emergency for spinal cord injury in order to protect the function and to gain much precious opportunity for spinal recovery and other treatment.
