RESUMEN

ObjectiveTo evaluate the advantages and disadvantages of endoscopic thyroidectomy by the breast approach. MethodsFrom August 2002 to April 2003, sixty single thyroid nodule patients were divided into 2 groups randomly. Group 1 (30 patients) received endoscopic thyroidectomy(ET) by the breast approach. The other 30 patients underwent conventional thyroidectomy(CT). Preoperative diagnosis,operative time,operative cost,postoperative pain,complications,and cosmetic result between the 2 groups were compared. RESUMEN

Objective To investigate the relationship between the expression of Fas ligand mRNA and the biological behavior and prognosis of colon cancer.Methods The expression of Fas ligand mRNA was examined in 52 cases of colon cancer by in situ hybridization (ISH) technique.Results Fas ligand mRNA expression was found in 30 out of 52 cases of colon cancer (58%). Fas ligand mRNA expression was positive in cancer tissues in 3 out of 12 cases (25%) with pericancer inflammation and in 27 out of 40 cases (68%) with no pericancer inflammation ( P

RESUMEN

Objective: To investigate the expression and function of Fas ligand (FasL) in human colon carcinoma cells. Methods: A total of 6 human colon cancer cell lines were examined for FasL mRNA and protein expression, Fas-dependent cytotoxicity and its modulation were detected by using RT-PCR, Western blot and 3H-TdR release assay. Results: FasL mRNA and protein was expressed in all of these 6 cancer cell lines RPMI 4788, HCT-116, DLD-1, LoVo, WiDr and COLO 320DM. Fas-dependent cytotoxicity to hFas transfectant was exerted in part of these cancer cells, such as DLD-1, LoVo and WiDr. The cytotoxicity was significantly enhanced by PMA and ionomycin. Conclusion: These data suggest that functional FasL is expressed, at least in part, in human colon cancer cell lines and may help to escape from immune surveillance of the host.

RESUMEN

Objective: To investigate the role of TNF-? and IFN-? in modulation of Fas ligand(FasL) expression in human colon carcinoma cells. Methods: A total of 6 human colon cancer cell lines were examined for FasL protein expression and for TNF-? and IFN-? modulation of FasL expression by Western blot method. Results: The results showed that FasL protein was expressed in all of these 6 cancer cell lines.FasL expression was up-regulated by TNF-? and IFN-? for 10 h in DLD-1 colon cancer cells; FasL expression was also up-regulated by TNF-? and IFN-? as well as PMA plus ionomycin for 10 h in SW620 cells, and further up-regulation was got by TNF-? and PMA plus ionomycin for 48 h in SW620 cells,whereas slight up-regulation was obtained by IFN-?. Conclusion: These data suggest that TNF-? and IFN-? may enhance FasL expression in tumor cells and may facilitate to escape from the immune surveillance of the host.

RESUMEN

Objective To investigate the modulation of FasL protein in colon cancer cells by interleukin 18 (IL 18) and it's effect on tumor cells counterattacking T lymphocytes. Methods By using Western blotting assay, FasL protein expression was examined in human colon cancer cell line SW620. The ratio of apoptosis of lymphocytes induced by tumor cells was studied by flow cytometry analysis. Results 10 ?g/L and 100 ?g/L IL 18 upregulated FasL protein expression in SW620 cell line, reaching the peak in 18 and 36 hours respectively, and returned to initial levels in 72 hours. The ratio of apoptosis of Jurkat cells induced by tumor cells increased in accordance with the upregulation of FasL protein. Conclusions IL 18 upregulates FasL protein expression in colon cancer cell line and enhances the ability of cancer cells to counterattack T lymphocytes.
