RESUMEN

Objective To observe the quantitative relation between new subarachnoid hemorrhage (SAH) grading and scores of Garcia neurological scale in mouse models of SAH.Methods Mouse models of SAH were induced by filament perforation; neurological scale was performed 24 h after SAH; and then,these mouse models were sacrificed and performed SAH grading;the relation between scores of neurological scale and SAH grading was analyzed by SPSS 13.0 software.Results Fourteen of 21 mice survived after SAH. The higher the SAH grading, the worsen the neurological scale results; these 2 were negatively correlated (r=-0.841,P=0.000). Conclusion The scores of Garcia neurological scale and SAH grading has a quantitative negative relation and can be used to evaluate the hemorrhage in mouse models induced by filament perforation.

RESUMEN

Objective To investigate the protective effect of erythropoietin(EPO)on the mitochondria in the brain neurons against cerebral ischemia/reperfusion(IR)injury in rats.Methods Thirty SD rats are randomly allocated into 3 groups,namely the EPO group(n=10),IR group(n=10),and shamoperation group(n=10).In EPO group and IR group,the rats were subjected to middle cerebral artery occlusion(MCAO)to induce cerebral IR injury,followed by treatment with intraperitonal EPO injection at 3000 U/kg and the same volume of saline,respectively.In the sham operation group,the carofid artery was only isolated without MCAO or subsequent treatment.Seventy-two hours after the IR injury,the mitochondrial membrane potential in the neurons was detected,and the changes in superoxide kismutase(SOD)activity,malondialdehyde(MDA)and nitric oxide(NO)concentrations,and Na+-K+-ATPase activity in the neuronal mitochondria were determined.The number of caspase-3-positive neurons in the hippocampus Was observed immunohistochemically. Results The mitochondrial membrane potential and activities of SOD and Na+-K+-ATPase Were significantly higher.whereas the MDA and NO concentrations and the number of caspase-3-postive neurons significantly lower in EPO group than in IR group after the treatment.Conclusion Protecting the neuronal mitochondrial function is one of the important mechanisms of EPO for brain protection against IR injury,and this mitochondrial protection effect is mediated essentially by stabilizing the mitochondrial membrane potential.

RESUMEN

<p><b>OBJECTIVE</b>To assess the value of (1)H-magnetic resonance spectroscopy ((1)H-MRS) in evaluating cerebral vasospasm resulting from subarachnoid hemorrhage (SAH).</p><p><b>METHODS</b>Six dogs were subjected to autologous non-heparinized blood injection via cisternal puncture twice at one-day interval to establish models of SAH, and another 6 received injections with normal saline in an identical manner. (1)H-MRS scan was performed on the 3rd, 7th and 14th days after the injections to measure the changes of N-acetylaspartate (NAA), creatine (Cr) and choline (Cho). After the (1)H-MRS scan, all the dogs underwent brain digital subtraction angiography (DSA) for determining the basilar artery diameter.</p><p><b>RESULTS</b>DSA results on day 3 presented development of obvious vasospasm of the basilar artery, which was most evident on day 7 and recovered obviously on day 14. (1)H-MRS results demonstrated obvious changes of NAA, Cho and Cr on days 3 and 7 in SAH model group, and NAA declined to the lowest level on day 3 followed by gradual ascending till reaching the normal level on day 14. Cho decreased slightly on day 3, then increased and reached the peak level on day 7 and then decreased. Cr rose steadily from day 3 to 14, but since day 7, the rise slowed down obviously and Cr maintain a level not significantly different from that on day 14 (P>0.05). The functional results of (1)H-MRS were consistent with the DSA results.</p><p><b>CONCLUSION</b>(1)H-MRS can be used to monitor the development of cerebral vasospasm resulting from SAH as a good evaluation method for functional imaging.</p>
