RESUMEN

In order to explore the mechanisms underlying the vasoconstriction induced by blockade of inward rectifier Kchannels (K) with BaCl, myogenic tone of isolated rat coronary artery (RCA) was recorded with wire myograph. The dependence of BaCl- induced contraction on intracellular Ca([Ca]) release and extracellular Ca([Ca]) influx was studied by Cadeprivation and restoration. The mechanisms underlying BaCl-induced RCA contraction were investigated with specific inhibitors. BaCl(0.1-1.0 mmol/L) contracted isolated RCA in a concentration-dependent manner and the maximal contraction was (5.69 ± 1.07) mN, nearly equal to contraction induced by 60 mmol/L KCl. The contractions induced by BaClin Ca-free solution and by followed restoration of 2.5 mmol/L Caaccounted for (35.44 ± 6.72)% and (64.56 ± 5.94)%, respectively. Calcium channel blocker nifedipine (0.3 μmol/L), cyclooxygenase inhibitor indomethacin (100 μmol/L), ERK1/2 inhibitor PD98059 (10 μmol/L) and chloride channel blocker niflumic acid (100 μmol/L) pretreatment depressed the BaCl-induced maximal contraction by (87.82 ± 5.43)% (P < 0.01), (73.23 ± 5.47)% (P < 0.01), (75.69 ± 7.94)% (P < 0.01) and (83.24 ± 7.69)% (P < 0.01), respectively. These results demonstrate that BaClinduces vasoconstriction in RCA by enhancing both [Ca]release and [Ca]influx, and suggest that increase of prostanoids synthesis, activation of calcium channels and chloride channels, as well as ERK1/2 pathway may be involved in this process.

Objeetive To investigate the clinical significance of combined detection of lipoprotein associated serum phospholipase A2 (Lp-PLa2),homocysteine (Hcy) and cystatin C (CysC) in the diagnosis of hypertensive disorders complicating pregnancy (HDCP).Methods From January 2013 to May 2016 in Changan Hospital, selected 113 cases of pregnancy induced hypertension patients as the observation group, and were divided into three group A,B and C (group A:55 cases of HDCP patients, group B:32 cases of mild preeclampsia and group C for patients with severe preeclampsia 26 cases). At the same period, selected 50 cases of normal college pregnancy as control group, serum Lp-PLa2 (enzyme-linked immunosorbent assay),Hcy (cyclophorase method) and CysC (particle enhanced turbidimetric method),the test results were analyzed and compared. Results Serum Lp-PLa2,Hcy and CysC test results in the control group, the observation group A,observation group B and observation group C increased significantly,in the observation group C increased most obviously.Compared with the control group, the serum levels of Lp-PLa2,Hcy and CysC in the observation group were significantly higher, the difference was statistically significant (F=8.102,7.231 and 6.926,all P<0.05).Pearson correlation analysis showed that there was a positive correlation between serum Lp-PLa2,Hcy and CysC and blood pressure (r=0.71,0.69,0.63,all P<0.05).The abnormal rate of serum Lp-PLa2,Hcy and CysC for three joint detection was higher than that of single detection, and the difference was statistically significant (x2 =6.725,P<0.001).The abnormal rate of serum Lp-PLa2,Hcy and Cys single test results increased with the exacerbation of HDCP,and the difference was statistically significant (x2=9.351,P<0.000).Conclusion Serum Lp-PLa2,Hey,CysC and pregnancy would be closely related to the occurrence and development of hypertension syndrome,so combined detection of HDCP can improve the detection rate of abnormal results, and it has important clinical significance for early diagnosis and prognosis of HDCP.

RESUMEN

Objective: To prepare foliate-conjugated chitosan nanoparticles loaded with berberine hydrochloride (BH/FA-CTS-NPs) and investigate the optimizing technology, physicochemical characterizations, and inhibitory effect on CNE-1. Methods: Folate-conjugated chitosan (FA-CTS) was prepared by amino reaction of folic acid active ester and chitosan molecules; BH/FA-CTS-NPs were prepared using ion cross-linking technique with BH as a model drug. The morphology, particle size, and physicochemical characteristics such as entrapment efficiency (EE), drug-loading, and release in vitro of nanoparticles were studied. The effect of cell anti-migratory and anti-invasive actions of BH/FA-CTS-NPs was investigated using MTT assays, wound healing assays and Annexin-V-FITC single staining assays, respectively. Results: The prepared BH/FA-CTS NPs were round, and the size uniformity adhesion was not found. The results of mean particle size, EE, and drug-loading amount were (282.4 ± 4.5) nm, (89.82 ± 2.91)%, and (9.16 ± 1.01)%, respectively. (80.32 ± 3.24)% of BH in nanoparticles was released within 5 h and then released slowly, and the accumulative release rate within 24 h was (90.92 ± 5.21)%. These results by MTT assay and wound healing assay indicated that BH/FA-CTS NPs not only inhibited the proliferation of CNE-1 cells in a concentration- and time-dependent manner, but can induced apoptosis. Conclusion: BH/FA-CTS NPs with the sustained release effect could be prepared successfully by the ionic crosslinking method. Considering these properties, block proliferation and impair the migration of the CNE-1 cell line, BH/FA-CTS NPs could be an important compound for consideration in the treatment of nasopharyngeal carcinoma.
