RESUMEN

Objective To investigate the effect and mechanism of Feixin Decoction(Astragali Radix,Pericae Semen,Carthami Flos,Descurainiae Semen Lepidii Semen,Paeoniae Radix Rubra, etc.)on monocrotaline-induced pulmonary arterial hypertension(PAH)rats based on peroxisome proliferator-activated receptor-γ/nuclear factor-κB(PPAR-γ/NF-κB)signaling pathway. Methods Forty-eight male SD rats were randomly divided into normal group, model group,Sildenafil group(0.025 g·kg-1)and low-,medium-and high-dose of Feixin Decoction groups(11.7,23.4,46.8 g·kg-1).PAH rat model was established by single intraperitoneal injection of monocrotaline solution(60 mg·kg-1).After 1 hour of modeling, the rats were given intragastric administration once a day for 28 days.Hemodynamic and echocardiographic parameters including right ventricular systolic pressure(RVSP),mean pulmonary artery pressure(mPAP),right ventricular hypertrophy index(RVHI),pulmonary artery acceleration time(PAAT),pulmonary artery ejection time(PET),tricuspid annular plane systolic excursion(TAPSE),right ventricular internal diameter(RVIDd)and right ventricular anterior wall thickness(RVAWT)were measured in each group. The pathological changes of pulmonary arterioles were observed by HE staining. The expression level of α-smooth muscle actin(α-SMA)in rat pulmonary artery was detected by immunofluorescence. The levels of plasma interleukin-1β(IL-1β),IL-6 and tumor necrosis factor-α(TNF-α)were detected by ELISA.The expression levels of PPAR-γ/NF-κB signaling pathway-related proteins were detected by immunohistochemistry and Western Blot. Results Compared with the normal group, the RVSP,mPAP,RVHI,RVIDd and RVAWT of the model group were significantly increased(P<0.01).PAAT,PAAT/PET and TAPSE were significantly decreased(P<0.01).The wall of pulmonary arterioles was significantly thickened, and the percentage of wall thickness of pulmonary arterioles to vascular diameter and the percentage of vascular wall area to total cross-sectional area of pulmonary arterioles were significantly increased(P<0.01).The positive expression rate of α-SMA protein in pulmonary artery was significantly increased(P<0.01).The levels of plasma IL-1β,IL-6 and TNF-α were significantly increased(P<0.01).The positive expression rate of PPAR-γ protein in lung tissue was significantly decreased(P<0.01),and the positive expression rate of NF-κB protein was significantly increased(P<0.01).The protein expressions of PPAR-γ and IκB-α in lung tissue were significantly down-regulated(P<0.01).The protein expression ratio of p-NF-κB/NF-κB was significantly increased(P<0.01).Compared with the model group,RVSP,mPAP,RVHI,RVIDd and RVAWT in each administration group were significantly decreased(P<0.05,P<0.01),while PAAT,PAAT/PET and TAPSE were significantly increased(P<0.05,P<0.01).The thickness of the vascular wall was significantly reduced, and the percentage of the wall thickness of the pulmonary arterioles to the diameter of the blood vessels and the percentage of the vascular wall area to the total cross-sectional area of the small arteries were significantly reduced(P<0.05,P<0.01).The positive expression rate of α-SMA protein in pulmonary artery was significantly decreased(P<0.05,P<0.01).The plasma levels of IL-1β,IL-6 and TNF-α were significantly decreased(P<0.05,P<0.01).The positive expression rate of PPAR-γ protein in lung tissue was significantly increased(P<0.05,P<0.01),and the positive expression rate of NF-κB protein was significantly decreased(P<0.05,P<0.01).The protein expression of PPAR-γ in lung tissue was significantly up-regulated(P<0.05,P<0.01),and the protein expression ratio of p-NF-κB/NF-κB was significantly decreased(P<0.01).The protein expression of IκB-α in the lung tissue of rats in the high-dose group of Feixin Decoction was significantly up-regulated(P<0.01).Conclusion Feixin Decoction can improve pulmonary artery pressure, right ventricular dysfunction and pulmonary vascular remodeling in PAH rats induced by monocrotaline. The mechanism may be related to the regulation of PPAR-γ/NF-κB signaling pathway to inhibit inflammatory response.

RESUMEN

【Objective】 To observe the effects of mirror neuron system theory(MNST) on hand dexterity and fine motor function in preschool children with developmental coordination disorder(DCD), so as to provide reference for the rehabilitation of children with DCD. 【Methods】 A total of 51 children with DCD treated at Nantong Maternal and Child Health Care Hospital from June 2021 to April 2023 were enrolled in this study, and were randomly assigned to treatment group (n=26) and control group (n=25) except for 5 cases who missed in the follow-up. Both groups received conventional rehabilitation training, while the treatment group received MNST additionally. The hand dexterity and fine motor function of both groups were assessed using the hand dexterity subscale of the Movement Assessment Battery for Children-Second Edition (MABC-2), Peabody Developmental Motor Scale-Fine Motor (PDMS-FM), and Function Independence Measure for Children (WeeFIM) before and after 12 weeks of treatment. 【Results】 Before treatment, there were no significant differences in hand dexterity subscale of MABC-2, PDMS-FM, and WeeFIM scores between the two groups (P>0.05). After treatment, both groups showed improvements in hand dexterity subscale of MABC-2, PDMS-FM, and WeeFIM scores (treatment group: t=35.620, 42.084, 40.072; control group: t=14.000, 12.017, 14.054, P<0.001), with the treatment group showing significantly greater improvements compared to the control group (t=2.611, 3.120, 2.331, P<0.05). 【Conclusion】 MNST combined with conventional rehabilitation training can enhance hand dexterity and fine motor function in children with DCD, thereby improving children′s activities of daily living.

RESUMEN

Objective:To understand the sensory characteristics of children with autism spectrum disorder (ASD), and to explore its relationship with the clinical symptoms and to provide a reference basis for early diagnosis and intervention treatment of ASD.Methods:A cross-sectional survey was conducted to collect 216 children diagnosed with ASD according to DSM-5 from September 2021 to December 2021.All children were investigated with short sensory profile (SSP), autism behavior checklist (ABC) and social response scale (SRS). SPSS 21.0 was used for statistical analysis and the correlation between sensory characteristics and clinical symptoms of ASD children was analyzed by Pearson correlation. Results:The results showed that 120 of 216 with ASD children had abnormal SSP total score, with an abnormal rate of 55.6%.Among the seven dimensions, the abnormal rate of auditory filtering was the highest, reaching 83.8%, followed by low strength/weakness(60.2%), motor sensitivity(43.5%), low response/sensation seeking(39.8%), taste/smell sensitivity(37.5%), vision / hearing sensitivity(27.8%) and tactile sensitivity(17.6%), and the total incidence of paresthesia was 92.1%.There were significant differences in the total score of ABC and the five dimensions of sensation, communication, body movement, language and life self-care of ASD children with different sensory characteristics ( F=39.15, 39.99, 46.02, 29.18, 50.98, 51.76 respectively, all P<0.01). There were significant differences in the total score of SRS and social perception, social cognition, social communication, social motivation among ASD children with different sensory characteristics ( F=19.33, 29.47, 24.82, 29.68, 31.48, 39.86 respectively, all P<0.01). With the aggravation of sensory abnormalities, the total scores of ABC and SRS and the scores of each dimension increased. The total score of SSP was negatively correlated with the scores of sensation, communication, body movement, language, self-care and the total score of ABC ( r=-0.613, -0.631, -0.643, -0.558, -0.657, -0.679 respectively, all P<0.01). The total score of SSP was negatively correlated with social perception, social cognition, social communication, social motivation, autism behavior and the total score of SRS ( r=-0.440, -0.527, -0.504, -0.522, -0.542, -0.595 respectively, all P<0.01). Conclusion:Sensory abnormalities are common in ASD children, and with the aggravation of sensory abnormalities, the core symptoms and behavioral performance of ASD children are more obvious and positively correlated.
