RESUMEN

Objective In May 2013,an abnormal increase of surgical-site infection among post cesarean section patients was reported at one hospital in Zhongshan. An investigation was conducted to identify the risk factors and related control measures. Methods All the reported surgical-site infection records among post cesarean section patients were checked. A review of cesarean section schedules of health workers was also performed. An 1∶2 case-control study was conducted among surgical-site infection cases in May 2013. Microbiologic cultures were performed on 2 surgical site secretion samples and 12 samples from the environment. All the positive isolates were molecular typed by pulsed field gel electrophoresis(PFGE). Results In May 2013,4 post cesarean section patients who developed surgical-site infection symptom at one hospital in Zhongshan were reported, with an attack rate as 10.3%(4/39). The emergence time of symptom was 2-3 days after operation. All of the 4 cases underwent an emergency operation. The median time interval for cases from admission to operation was 7.2 hours(ranged from 2 to 9 hours),lower than that seen in the controls, with a median time of 20.8 hours (Z=5.50,P=0.03). Two of the 4 cases took type-B ultrasonic inspection 1.4 h and 8.4 h before the operation, and the other two cases took continuous fetal heart monitoring 2 hours before the operation. Skin of the operation area on the 4 cases had been exposed to ultrasonic couplant, without a thorough clean. The proportion of type-B ultrasonic inspection or continuous fetal heart monitoring was much higher in cases than in controls(χ2=5.19, P=0.01). Burkholderia cepacia (BC) isolates were discovered from:one surgical site secretion,2 type-B ultrasonic probe samples, one ultrasonic couplant in use and one ultrasonic couplant unopened. All the isolates were identified as 100%identical by PFGE. Conclusion The skin of operation area of cesarean section patients had been exposed to BC contaminated ultrasonic couplant without thorough cleaning, which seemed to be related to the outbreak of surgical-site infection,in our case.

RESUMEN

Objective: The chemical composition, extraction and biological activity of antioxidant substance of sweet potatoes were studied. Method: The antioxidant substance was extracted with alcohol and purified with AB-8 big-hole resin and its elimination rate of _?O 2 and OH and SOD activity were determined. Results: The content of antioxidant substance stockpiled respectively in the root tubers, stems and leaves of sweet potatoes and was different in varieties. The highest content in the root tuber was 0.324%, which accounted 70% of total content in sweet potatoes. Its extraction ratio was 85%. The major composition of the antioxidant substance was phenol acid, including chlorogenic acid, isochlorogenic acid, neochlorogenic acid and 4-0-caffeonyl quinic butyl ester. They had intense antioxidant function and their elimination rate to _?O 2 and?OH at concentration 90 mg/ml was 88.02% and 82.21% respectively. The antioxidantsubstance restrained oxidation of lipid and hemolysis of red cell induced by H2O2,and increased SOD activity in serum and skin of mice. Conclusion: Several kinds of phenol acid as antioxidant substance stockpiled in sweet potatoes were proved to have strong antioxidant activity.

RESUMEN

Objective:To determine the difference of miRNA expression profiles in 6 types of human cancer cells by microarray technique. Methods:The microarray was prepared, with contained 210 oligonucleotides, including 206 probes complementary with human and mouse miRNA sequences and 4 positive control oligos.MiRNAs were extracted from HeLa(human cervical cancer epithelial cells),MCF-7(human breast cancer cells),A549(human lung adenocarcinoma cells),HT-29(human colonic cancer cells),ES-2(ovarian carcinoma cells),and K562(chronic myelogenous leukemia cells) cells and were labeled with Cy3 for hybridization to the miRNA microarray. The slides were scanned by ScanArrayTMExpress1.0 and images were analyzed by ScanArray3.0 and Cluster3.0;the results were confirmed by Northern blotting and RT-PCR.Results:Totally 115 miRNAs were found to be differentially expressed in the 6 cancer cell lines, with miR-21 expression up-regulated and miR-125b,let-7 expression down-regulated.The expression of miR-17-5p and miR-20a was in cluster and was more higher in ES-2 cells than in other cells.HeLa and MCF-7 cells were located on a single branch of the dendrogram in cluster analysis.Northern blotting showed that both pre-and miR-17-5p expressed in K562,pre-miR-17-5p was weakly expressed in A549 and ES-2 cells, and obvious pre-miRNA expression and weak miRNA expression were noticed in MCF-7,HeLa,and HT-29 cells.RT-PCR showed that expression of pre-miR-17-5p in K562 cells was higher than those in other cells.Expression of miR-21was high in all 6 cell lines and the highest expression was seen in A549 cells. Conclusion:Microarray can be used to detect miRNA expression files in cancer cells, which contributes to the study of the relation between miRNA and tumor.
