RESUMEN

Objective@#To investigate the specifity of amyloid precursor protein(APP), brain derived neurotrophic factor (BDNF)and gamma aminobutyric acid(GABA) in peripheral blood in children with autism spectrum disorder, so as to explore the biomarkers for early screening of ASD and its relationship with the severity of ASD.@*Methods@#A total of 41 children diagnosed with autism from January to December 2019 were enrolled in the ASD group. Meanwhile, 41 healthy children with normal growth and development who were examined in the same period were selected as control group. And the sera total sAPP, sAPP α, sAPP β, BDNF and GABA of all participants were tested by sensitive enzyme linked immunosorbent assay method, and were compared between the two groups.@*Results@#The serum sAPP level in ASD group(2 132.98±333.28 ng/mL) was higher than control group(1 734.76 ±357.97 ng/mL),the serum sAPP α level(335.11±33.87 pg/mL) was higher than control group(274.84±32.12 pg/mL) and the serum GABA level(4.17±0.95 μmol/L)was lower than control group(6.35±0.84 μmol/L). GABA level (4.17±0.95 μmol/L) was lower than that of control group (6.35±0.84 μmol/L), the differences were statistically significant ( t =3.92, 4.25, -7.27, P < 0.05 ). In addition, the serum GABA level in children with severe ASD (3.48±0.77 μmol/L)was lower than children with mild to moderate (4.94±0.98 μmol/L).The difference was significant ( t =-3.31, P <0.05). ROC curve showed that total sAPP( AUC = 0.77 ,95% CI =0.66-0.87), sAPP α( AUC =0.77,95%CI=0.67-0.87), and GABA ( AUC =0.95,95% CI =0.90-0.93)had diagnostic efficacy for ASD( P <0.05), among which the AUC of GABA was the largest (0.95)and its sensitivity(85.65%) and specificity(80.76%) were the highest. The results of binary Logistic regression showed that the abnormal expression of sAPPα ( OR =1.04,95% CI = 1.00- 1.07) and GABA( OR =0.02,95% CI =0.00-0.32) were associated with risk for ASD( P <0.05).@*Conclusion@#Considering the specific change of total sAPP, sAPPα andGABA in peripheral blood in ASD children, total sAPP, sAPP α and GABA can be considered as promising biomarkers in the early diagnosis of ASD, among which GABA has the highest sensitivity and specificity.

RESUMEN

Objective:To explore the clinical significance of three-dimensional pelvic ultrasound parameters in patients with stress urinary incontinence (SUI).Methods:A total of 100 patients with SUI in the First Hospital Affiliated to Hunan Traditional Chinese Medical College from January 2018 to April 2020 were prospectively selected as the observation group, and 100 women who had undergone birth without SUI and other urinary system diseases who came for physical examination during the same period were selected as the control group. Comparing the three-dimensional pelvic floor ultrasound parameters of resting state and Valsalva action in the patients with different degrees of illness, the patients with different efficacy of the two groups and the observation group. Analyzing the correlation between three-dimensional pelvic floor ultrasound parameters and the severity and efficacy of SUI patients.Results:⑴ The posterior urethrovesical angel at rest and after Valsalva action, the pelvic diaphragmatic hiatus area after Valsalva action in the observation group were larger than those in the control group, and the puborectalis muscle thickness at rest and after Valsalva action was smaller than that of the control group, with statistically significant difference( P<0.05); ⑵ The posterior urethrovesical angel at rest and after Valsalva action, the pelvic diaphragmatic hiatus area at rest, and the puborectalis muscle thickness at rest and after Valsalva action were related to SUI ( P<0.05); ⑶ Comparison of patients with stress urinary incontinence of different severity: the posterior urethrovesical angel at rest and after Valsalva action, the pelvic diaphragmatic hiatus area after Valsalva action: mild<moderate<severe; the puborectalis muscle thickness at rest and after Valsalva action: mild>moderate>severe; ⑷ After treatment, the posterior urethrovesical angel and pelvic diaphragmatic hiatus area of patients with poor curative effect at rest and after Valsalva action were larger than those of patients with good curative effect, and the puborectalis muscle thickness of patients with poor curative effect at rest and after Valsalva action was smaller than that of patients with good curative effect, with statistically significant difference ( P<0.05); ⑸ The posterior urethrovesical angel at rest and after Valsalva action, the pelvic diaphragmatic hiatus area after Valsalva action were positively correlated with the degree of disease and clinical efficacy, while the puborectalis muscle thickness was negatively correlated with the degree of disease and clinical efficacy at rest and after Valsalva action ( P<0.05). Conclusions:Three-dimensional pelvic floor ultrasound examination can effectively reflect the changes in pelvic floor structure of SUI patients under different action states, and provides important anatomical and functional basis for the judgment of SUI disease degree and the evaluation of curative effect.

RESUMEN

After binding to the specific insulin-like growth factor 1 receptor (IGF1R) on the surface of target tissue cells, IGF can regulate physiological processes such as apoptosis, proliferation and senescence, which are closely related to growth and development of the body, and occurrence and development of diseases. The binding between IGF1 and IGF1Rα can cause conformational changes of the beta subunit of IGF1R, lead to activation of receptor tyrosine kinase, initiation of the downstream phosphatidylinositol 3-kinase pathway and mitogen-activated protein kinase pathway, and finally participate in the occurrence of acne, psoriasis and other skin diseases. This review summarizes research advances in the role of the IGF1R signaling pathway in the pathogenesis of related skin diseases.

RESUMEN

ObjectiveTo investigate the fusion sequence complexity of EML4-ALK in non-small cell lung cancer (NSCLC) patients, and the potential mutation in tyrosine kinase ( TK ) domain of ALK gene.MethodsIn routine practice,a novel echinoderm microtubule-associated protein-like4 and anaplastic lymphoma kinase (EML4-ALK) V3c variant was detected by rapid amplification of cDNA ends-polymerase chain reaction ( RACE-PCR )-sequencing technology in a patient with NSCLC.The further consecutive 39 cases( total of 40 cases)were screened by use of reverse transcription (RT)-PCR for EML4-ALK fusion.Positive PCR products were purified and cloned into T vectors, transformed into DH5a germ cells and colony picked up and sequenced for sequence complexity analysis.Tyrosine kinase domain of ALK was amplified by RT-PCR and sequenced.ResultsThree out of 40 cases had EML4-ALK fusion. One case had six novel variants of EML4-ALK co-existing,termed as V3c ( 64.6% ),V3d ( 25.0% ),V3e ( 2.1% ),V3f (4.2% ),V3g(2.1% )and V3h(2.1% ) variants, whereas without common V3a and V3b variants. In other two positive cases, one was V1 variant, another was concurrent V2,V3a and V3b variants. No mutations were detected in the TK domain of EML4-ALK in any case.ConclusionsSeveral EML-ALK variants could co-exist in a given lung cancer tissue, which suggest that the diversity and sequence complexity of EML4-ALK fusion are exist.Attentions should be paid to screen all the variants in clinic to improve the pick-up rate.
