RESUMEN

Non-coding RNA (ncRNA) used to be considered as a class of gene transcripts without protein-coding capacity. Whereas emerging evidence has demonstrated that a small fraction of ncRNAs are still capable of producing functional peptides. Such ncRNAs and corresponding peptides are highly conserved and homologous, and can be detected by sequencing or mass spectrometry analysis. In this paper, we searched several databases with the keywords of "non-coding RNA" and "peptide", and briefly reviewed the characteristics of ncRNA that can be translated into functional small peptides, detection methods of peptides, biological functions of peptides and clinical application value of peptides. The results show that these functional peptides are often involved in disease processes, such as regulation of tumor progression, muscle activity and immune disorders. NcRNA-encoded peptides can be used as novel and efficacious disease diagnostic, therapeutic, and prognostic tools, and further develop as anticancer therapeutic targets to provide new ideas for individual precise treatment of tumors.

Objective@#The purpose of this study is to explore the biological function of long non-coding RNA (lncRNA) HMMR-AS1 in proliferation and metastasis of lung adenocarcinoma (LUAD). @*Methods@#Real-time fluorescence quantitative polymerase chain reaction (RT-qPCR) was used to detect the expression of HMMR-AS1 and its sense strand HMMR in LUAD cell lines. Then we knock down the HMMR-AS1 expression through small interfering RNA and evaluate the transfection efficiency and its effect on the expression of HMMR. CCK-8 (cell counting kit), clone formation, flow cytometric analysis, wound scratch assay and transwell assay were used to assess the biological function of A549 and H1299 cells. Western blot was used to detect the protein expression of HMMR in the two cell lines after transfection with si-HMMR-AS1. @*Results@#The expression of HMMR-AS1 in A549 and H1299 cells of LUAD cell line was markedly higher than that in normal lung epithelial cell BEAS-2A by upregulating approximately 3.06 and 5.02 folds (P<0.05), respectively. After transfection with si-HMMR-AS1, the expression of HMMR-AS1 markedly reduced in both levels of transcription and protein (P<0.05). Furthermore, knocking down of HMMR-AS1 significantly inhibited the proliferation, migration and invasion abilities, and increased the apoptosis rates of A549 and H1299 cells. @*Conclusion@#LncRNA HMMR-AS1 could promote malignant progression of LUAD cells through enhancing the growth, migration and invasion ability of LUAD cells.

Objective To evaluate the efficacy of entecavir treatment in chronic hepatitis B virus ( HBV ) infected patients with mild hepatic dysfunction and marked pathological injury.Methods One hundred and fifty five chronic hepatitis B ( CHB) patients with HBV DNA>1.0 ×104 U/mL admitted in Lishui People’ s Hospital during January 2008 to October 2011 were enrolled in the study.Patients were divided into three groups: those with serum ALT 5 ×ULN were in control group 2 (n=42).All patients were given entecavir (0.5 mg, 1/d, p.o) treatment.ALT normalization rates, HBV DNA negative rates, HBeAg negative conversion rates and seroconversion rates at 12-, 24-,48-, 96-and 144-week were observed and compared among groups.Variance analysis andχ2 test were performed for measurement data and numeration data, respectively.Results ALT normalization rates in observation group were 86.7%, 90.7%, 90.7%, 92.0%and 96.0%at 12-, 24-, 48-, 96-and 144-week, which were higher than those in control group 1 (χ2 =2.04, 2.15, 2.78, 2.69 and 2.47, P 0.05) . HBV DNA negative rates in observation group were 70.7%, 78.7%and 82.7%at 12-, 24-and 48-week, which were higher than those in control group 1 (χ2 =4.56, 4.23 and 4.28, P0.05).HBeAg negative conversion rates in observation group were 6.6%, 21.3%and 25.3%at 48-, 96-, and 144-week, which were higher than those in control group 1 (χ2 =4.68, 4.78 and 5.01, P0.05).HBeAg seroconversion rate in observation group was 4.0%at 24-week, which were higher than that in control group 1 (χ2 =2.87, P <0.05), but the seroconversion rates at 96-and 144-week were lower than those in control group 2 (χ2 =2.92 and 3.14, P<0.05).Conclusion The efficacy of entecavir treatment for HBV infected patients with mild hepatic dysfunction and marked pathological injury is satisfactory.

RESUMEN

Objective:To evaluate the clinical efficacy and safety of repaglinide and gliclazide in the treatment of elderly type 2 di-abetes mellitus (T2DM). Methods:A total of 225 T2DM patients with the age over 60 were enrolled and randomly divided into repa-glinide group (n=112) and gliclazide group (n=113). The repaglinide group was given repaglinide 0. 5~2 mg, po, tid, while the gliclazide group was given gliclazide modified release tablets 30~120 mg, po, qd,both with the treatment course of 12 weeks. The level of FPG, 2h PG, HbAlC, TC and TG and the side effects were observed and the clinical efficacy was compared between the two groups. Results:The level of FPG, 2h PG, HbAlc and TC in the two groups showed significant decrease after the treatment (P0. 05). Conclusion:Both repaglinide and gliclazide show safe and reliable efficacy in the treatment of elderly T2DM.

<p><b>OBJECTIVE</b>To investigate the role of N-Methyl-D-aspartic acid (NMDA)-type glutamate receptors in the central nucleus of the amygdale (CeA) in food and water intake.</p><p><b>METHODS</b>Male Sprague-Dawley rats with stainless steel cannulae implanted unilaterally into the CeA were used. The prototypic NMDA receptor agonist NMDA, or the selective NMDA receptor antagonist D(-)-2-amino-5-phosphonopentanoic acid (D-AP-5) was microinjected into the CeA of satiated and euhydrated rats.</p><p><b>RESULTS</b>Intra-CeA injection of 8.50, 17.00, or 34.00 nmol NMDA did not alter food intake but significantly increased water intake 0-1 h after the injection (F(3,32)=3.191, P=0.037) independent of food intake. Without affecting the food intake, injection of 6.34, 12.70, or 25.40 nmol D-AP-5 into the CeA significantly decreased water intake 0-1 h after the injection (F(3,28)=3.118, P=0.042) independent of food intake.</p><p><b>CONCLUSION</b>NMDA receptors in the CeA may participate in the control of water intake rather than food intake.</p>

RESUMEN

Objective To investigate the points of perioperative nursing of patients with diabetic foot ulcers (DFUs)treated with continuous thrombolysis.Methods A total of 78 cases of DFUs patients treated with continuous infusion of urokinase through femoral artery from February 2006 to August 2011 were recruited into the study.Besides,psychological care,education guidance,and materials preparation were given to them before the operation.Meanwhile,a module of “Nursing observation and assessment indicators to DFUs treated with interventional thrombolysis” was set up and used to give a dynamic evaluation.Results Foot ulcers of all the subjects were healed,and no case occurred serious complications such as bleeding or infection and so on.Conclusions Our results indicate that professional psychological care,education guidance,and fully materials preparation in perioperative period lay the foundation of clinical efficacy.The establishment and application of “Nursing observation and assessment indicators to DFUs treated with interventional thrombolysis” is the key to the success of thrombolytic therapy because it can effectively reduce postoperative adverse events and promote ulcer healing.

To observe the relationship between positive rate of β1-adrenergic and AT1 receptors autoantibodies with serum concentration of cystatin C in 371 patients with diabetic nephropathy patients,107 patients with type 2 diabetes,and 47 subjects as healthy control.In patients with diabetic nephropathy,the positive rates of the β1 and AT1 receptors autoantibodies were significantly higher than those in patients with type 2 diabetes and normal controls.The titers of β1 and AT1 receptors autoantibodies in diabetic nephropathy patients with abnormal cystatin C were significantly higher than those with normal cystatin C concentration.These findings suggested that β1 and AT1 receptors autoantibodie may play important roles in the pathogenesis of diabetic nephropathy.

Objective To investigate the circadian change of autonomic nervous system in hyperthyroidism (HT) patients. Methods Recording 24 h dynamic electrocardiography from 33 HT patients( HT group) and 35 controls (control group). The cardiac autonomic nervous function was evaluated by the time domain and frequency domain analysis of heart rate variability( HRV). Results Comparing with control group, the SDNN, SDANN, ASDNN and rMSSD were significantly lower in HT group[ (82.3 ± 29.0)ms vs. (139.4±40.2 ) ms, ( 75.0 ± 27.4) ms vs. ( 130.3 ± 43.9) ms, (29.9 ± 14.9 ) ms vs. (57.3 ± 14.4) ms,( 19.8 ± 10.9 ) ms vs. (29.5 ± 9.4) ms ] (P < 0.01 or < 0.05 ), the long term total power (TP), high frequency (HF), low frequency (LF) and very low frequency (VLF) were significantly lower in HT group [ (566.1±573.2) ms2/Hz vs. ( 1894.2 ± 984.3)ms2/Hz, (68.1 ± 88.9 ) ms2/Hz vs. (232.7 ± 155.5 ) ms2/Hz, ( 127.4 ±163.0) ms2/Hz vs. (551.3 ± 390.6) ms2/Hz, (330.3 ± 300.6) ms2/Hz vs. (1073.2 ± 570.2) ms2/Hz] (P <0.01 ). Comparing with control group, short term VLF was higher in HT group during most time in 24 hours (P< 0.05 ). Short term LF was higher in HT group mainly in day time (P< 0.05 ). Short term HF was higher in HT group occasionally in the whole day (P<0.05). Short term LF/HF didn't show significant difference between HT group and control group at most time points (P>0.05).Conclusion In HT patients, cardiac chronotropic property is impaired, sympathetic activity increases in whole day, vagal activity increases correspondently but the increase in day is more marked than that in night.

Objective To evaluate the clinical effect of intervertional devascularization and surgical devascularization in treatment of patients with upper gastrointestinal hemorrhage .Methods Ninety-nine cases treated with intervertional devaacularization or surgical devascularization were retrospectively studied including 48 cases in intervertional group and 51 cases in surgical group.The postoperative resection, complications and rebleeding time were compared by X2 test and t test between two methods.Results The 12 months, 24 months and 36 months cumulative rebleeding rates after intervertional devascularization were 12.5% (6/47) ,24.5% (11/45), 27.9% (12/43) respectively.The rate after surgical devaacularization were 29.2% (14/48) ,44.7% (21/47), 48.9% (22/45) respectively.There were statistically significant differences between the intervertional and surgical groups (X2=3.843,4.150,4.083, P < 0.05).The complications of intervertional devascularization included fever 85.4% (41/48), bellyache 81.3% (39/48), portalvein thrombosis 4.2% (2/48), intraabdominal hemorrhage 2.1% (1/48), infection 2.1% (1/48) and death 2.1% (1/48).The complications of surgical devaacularization included fever 68.6% (35/51), bellyache 62.7% (32/51), generous hydroperitoneum 25.5% (13/51), portalvein thrombosis 37.3% (19/51), splenic vein thrombosis 11.8% (6/51), hepatic encephalopathy 3.9% (2/51), hepatorenal syndrome 2.0% (1/51), intraabdominal hemorrhage 2.0% (1/51), death 3.9% (2/51) and infection 15.7% (8/51).There were statistically significant differences of fever, bellyache, generous hydroperitoneum, portalvein thrombosis, splenic vein thrombosis between the intervertional and surgical groups(X2=4.174,3.098,16.199,6.011,5.536,14.085,P <0.05).Conclusions The intervertional devascularization procedure is simple, safe and effective method for treating upper gastrointestinal hemorrhage for portal hypertensive.The clinical effect is better than that of surgical devascularization.

Objective To assess whether the T~ -786 →C mutation of endothelial nitric oxide synthase (eNOS) gene is associated with endothelial dysfunction in type 2 diabetes. Methods A total of 162 type 2 diabetic men without angiopathy were studied. PCR/alleles-specific oligonucleotide (ASO) probes were used to analyse the T~ -786 →C mutation of eNOS gene, and high resolution ultrasound was used to measure endothelium-dependent arterial dilation (EDD). Objective: To observe the effect of urapidil (agonist of 5-HT 1A receptor ) on morphine withdrawal syndrome Method: A physical dependent model in mice was established by subcutaneous injection of quantitative morphine The intensity of withdrawal syndrome was evaluated by observing jumping latency, jumping times and lose of body weight after intraperitoneal injection of naloxone (5 mg/kg) The effect of urapidil was observed after intraperitoneal injection it with 3 different dosages (100, 200, 400 mg/kg) Contents of NO in plasma and brain tissue were measured by nitrate reduction method Results: Morphine withdrawal syndrome was inhibited by urapidil at any dosage Content of NO in brain tissue reduced by urapidil 400 mg/kg, while plasma NO increased Conclusion: Urapidil inhibits morphine withdrawal syndrome in mice, increase the content of NO in their brain tissue

Objective To investigate molecular changes in growing rats masticatory muscle after functional mandibular protrusion.Methods SD rats were randomly divided into eight groups as control 3d,7d,14d,21d and experimental 3d,7d,14d,21d.The RTPCR and qPCR methods were used to find expression and any differences of mRNA of MMPs in growing rats masticatory muscle after functional mandibular protrusion. (Results)(① Expression) of MMP-2,MMP-9 mRNA expressed in all growing rats masticatory muscle.② Expression differences of MMP-2 and MMP-9 mRNA in 3d,21d were found between control and experimental groups after functional mandibular protrusion.Conclusion ①MMP-2 and MMP-9 mRNA participate in masticatory muscle normal development.(② MMPs) mRNA involve changes in growing rats masticatory muscle after functional mandibular protrusion.
