RESUMEN

Objective:To study the expression of 11 cancer-testis (CT) genes and autologous humoral immune response to NY-ESO-1 in gastric carcinoma in China to provide some data for developing a vaccine against this cancer. Methods: Pathology tests were conducted in 101 gastric carcinoma samples,including tumor tissue and paired adjacent normal non-tumor tissue. Reverse transcription-polymerase chain reaction (RT-PCR) was used to test for 11 CT antigen gene expression and ELISA for autologous humoral immune response to NY-ESO-1.The antigen of NY-ESO-1 was also detected in tumor tissues by mAb E978 in IHC.Results: About 74.3% of the samples expressed at least one CT antigen,and the most frequently expressed one was MAGE-3 (41.6%). Twelve out of the 101 samples (11.9%) were found to be NY-ESO-1 mRNA positive. Simultaneously, autologous humoral immune response to NY-ESO-1 was detected in 6 of 12 NY-ESO-1 mRNA positive patients. Conclusion: Multiple CT genes were expressed in gastric cancer samples. MAGE-3,SSX-1,SSX-4, NY-ESO-1/LAGE-1, and MAGE-1 can be chosen as candidate antigens for cancer vaccines due to their higher expression rate. The potential of a multivalent CT antigen vaccine, including NY-ESO-1 and MAGE-3 along with other CT antigens, should be evaluated in patients with gastric cancer.

RESUMEN

Objective:To investigate the expression of metabolic enzymes of fluoropyrimidines in primary colorectal cancer. Methods: Sixty-one cases of pathological confirmed primary colorectal cancer were collected in Beijing Cancer Hospital from August 2003 to February 2004. The expressions of thymidylate synthase (TS), dihydropyrimidine dehydrogenase (DPD) and orotate phosphoribosyl transferase (OPRT) were detected in paired tumor tissues and non-cancerous tissues by RT-PCR. Results: Expression of each enzyme in tumor tissues was positively correlated with that in paired non-cancerous tissues respectively. Compared with the expression of each enzyme in cancer with that in non-cancer, OPRT was higher in cancer while that of DPD and TS was comparable. The expression of OPRT was approximately 4.38-folds higher in colorectal cancer tissues than that in non-cancerous tissues (P0.000). The lowest expression of OPRT was found in mucinous carcinoma while the highest levels in poorly-differentiated adenocarcinoma. There were correlations between the ratio of tumor tissues / non-cancerous tissues OPRT expression (T/N) and metastasis to the lymph nodes (r0.36; P0.005), small vessel invasion (r0.26; P0.041) and differentiation (r-0.33; P0.009). Conclusion: The detection of three enzymes by means of RT-PCR is practical and can be used in clinical application. Our results also suggest that OPRT is a potential biomarker in predicating prognosis of colorectal cancer.

RESUMEN

Objective To evaluate the prognosis of laryngeal cancer postoperative recurrence, and to investigate the relationship of Cyclin D1, PCNA, and Ki 67 protein expression in patients with recurrent laryngeal carcinoma. Methods 574 cases of laryngeal carcinoma from 1958 to 1999 were retrospectively reviewed. All patients were follow up surveyed for 3 years postoperatively. The whole set of tissue microarrays from 574 patients contained tumor tissue, including recurrent tumor tissue from 56 patients, and 39 normal laryngeal tissue specimens from normal laryngeal were collected as control. The tissue chips were analyzed by the SABC immunohistochemical method. Results Cyclin D1, PCNA, and Ki 67 expressed in most of the laryngeal carcinoma and normal laryngeal tissues in a different positive ratio. There was higher expression of Cyclin D1, PCNA, and Ki 67 in recurrent laryngeal carcinoma compared to that in laryngeal carcinoma without recurrence and normal laryngeal tissue ( P
