RESUMEN

【Objective】 To establish a method for detecting human immunoglobulin Fc function based on surface plasmon resonance technology. 【Methods】 Based on the characteristic that FcγRI can be binded to the Fc segment of IgG, the affinity constant of the sample was detected by surface plasmon resonance, and its Fc function was the KD ratio of the sample to the standard. The method was validated for specificity/specificity, precision and robustness. The method and the pharmacopoeia method were used to detect the Fc function of 30 human immunoglobulins, and the correlation and consistency of the detection results were analyzed. 【Results】 The method validation results showed that this method has strong specificity/specificity (t values were 0.15, 0.22, both P>0.05), good precision (CV value 5.37%~10.69%) and good robustness (CV value 10.06%). The detection results of this method and the pharmacopoeia method have high correlation (r=0.96, P<0.05) and high consistency (Bias-2.060, 95% Limits of Agreement-5.628~1.508). 【Conclusion】 A method for detecting human immunoglobulin Fc function based on surface plasmon resonance has been successfully established.

RESUMEN

Objective@#To understand physical fitness and physiological function level of children and adolescents with different nutritional status in Beijing Tianjin Hebei Region, and to provide reference for physical health promotion and intervention of children and adolescents.@*Methods@#From September to December 2019, 4 424 children and adolescents aged 7-18 years old in Beijing Tianjin Hebei Region were tested for different nutritional status, physical fitness and physiological function by using stratified cluster sampling method.@*Results@#The detection rates of malnutrition, normal, overweight and obesity were 7.93%, 67.36% and 24.71%, respectively.There were differences in heart rate, systolic blood pressure, diastolic blood pressure, vital capacity body mass index, grip strength body mass index, oblique pull up/pull up, standing long jump, 50 m running, 1 000 m running, and BMI in boys with different nutritional status statistical significance ( Z =9.6, 55.2, 118.6, 332.5, 122.8, 15.6, 49.5, 47.5, 12.6, 113.5, P <0.01); There were differences in heart rate, systolic blood pressure, diastolic blood pressure, vital capacity body mass index, grip strength body mass index, sit up for 1 minute, standing long jump, 50 m running, 800 m running and physical fitness index in girls with different nutritional status statistical significance ( Z =33.9, 24.5, 46.5, 262.5, 102.6, 32.5, 27.5, 33.6, 27.6, 51.6, P <0.01). Correlation analysis showed that the nutritional status of boys was positively correlated with systolic blood pressure, diastolic blood pressure, 50 m running, and 1 000 m running ( r =0.27, 0.31, 0.14, 0.16, P <0.05), and was correlated with heart rate, vital capacity, body mass index, grip strength body mass index, oblique pull up/pull up, standing long jump, and BMI were negatively correlated ( r =-0.07, -0.62, -0.41, -0.21, -0.35, -0.29, P <0.05); nutritional status of girls it was positively correlated with systolic blood pressure, diastolic blood pressure, and 800 m running ( r =0.21, 0.27, 0.22, P <0.05), and negatively correlated with heart rate, vital capacity BMI, grip strength BMI, and BMI ( r =-0.12, -0.49, -0.32, -0.18, P <0.05).@*Conclusion@#The nutritional status of children and adolescents in Beijing Tianjin Hebei Region is related to physical fitness and physiological function. Overweight and obese child have lower physical fitness and physiological function. In the future, childhood overweight and obesity should be effectively controlled to better promote the development of physical fitness and physiological function.

Objective:To study the application of systemic meta-competence and its difficulties in the clinical practice of initial family therapy by qualitative research, with the aim of providing an empirical basis for developing local family therapists' clinical practice competence in systemic thinking.Methods:A purposive sample of 4 senior and 4 novice family therapists were gained, and a total of 12 hours of initial interviews were recorded.The materials were coded by thematic analysis with reference to the theoretical models of National Institute for Health and Care Excellence(NICE) and the American Association for Marriage and Family Therapy (AAMFT), and one-on-one interviews (totally 4 hours)were conducted with the therapists based on the coding results.Results:The application of systemic meta-competence in the initial family therapy was mainly manifested in four superordinate themes: adapting a non-pathological perspective, maintaining a relational orientation, focusing on interpersonal interactions and applying systemic intervention strategies, under which a total of 16 subordinate themes were included.The application difficulties of therapists with different qualification had both commonalities and differences.Conclusion:The systemic meta-competency is a key competence in the practice of family therapy.The application of systemic meta-competence in initial interview will benefit the therapeutic relationship, family information collections and formulation of therapy goals, and the application difficulties may reflect the culturally practical needs of professional trainings and supervisions.

Objective To investigate the effect and mechanism of electroacupuncture at acupoints on radiation-induced hippocampal neuronal apoptosis in C57 mice. Methods Forty one-month-old C 57/6J mice were equally and randomly divided into control group, model group(8 Gy), acupoint-electroacupuncture group, and non-acupoint-electroacupuncture group. In the acupoint-electroacupuncture group,mice received electroacupuncture intervention at Baihui, Fengfu, and Shenshu points. The novel object recognition test was used to evaluate the cognitive, learning, and memory abilities in mice. Immunohistochemistry was used to determine the levels of caspase-3 and apoptosis-inducing factor(AIF). TUNEL assay was used to detect DNA breaks. The data were analyzed by one-way analysis of variance and the Tukey's multiple comparison. Results Compared with the control group, the model group had a significantly reduced recognition index(P= 0.040);compared with the model group, the acupoint-electroacupuncture group had a significantly increased recognition index(P=0.043),shown by significantly longer time spent exploring novel objects(P=0.035).According to the results of immunohistochemistry,the model group had significantly higher apoptosis rate and levels of caspase-3 and AIF in hippocampal neurons than the control group(P= 0.002, 0.003, 0.023), while the acupoint-electroacupuncture group had significantly lower apoptosis rate and levels of caspase-3 and AIF in hippocampal neurons than the model group(P=0.027, 0.005, 0.004). Conclusions Radiation can induce hippocampal neuronal apoptosis and damage cognitive function in mice. Electroacupuncture intervention can, to some degree, reduce above damages.

Objective To explore the effects of different courses of electro-acupuncture at Baihui,Fengfu,and Shenshu of both sides on the behavior of mice exposed to radiation.Methods Fifty C57BL/6Jmice 30 days old were randomly and equally divided into control group,model group (exposure to radiation of 8 Gy),electro-acupuncture group 1 (1 week of treatment),electro-acupuncture group 2 (2 weeks of treatment),and electro-acupuncture group 3 (3 weeks of treatment).The mouse model of radiation-induced brain injury was established by exposure to radiation of 8 Gy and treated with electro-acupuncture.The open field test and Morris water maze test were used to evaluate the cognitive function and spatial learning and memory,respectively.Group t-test difference.Results In the open field test,the model group had significantly fewer vertical or horizontal movements than the control group (P=0.032,0.029,0.028),while the electro-acupuncture groups 2 and 3 had significantly more vertical or horizontal movements than the model group (P=0.001,0.017,0.000).In the Morris water maze test,compared with the control group,the model group had significantly longer latency,a significantly lower frequency of entry into the goal box,and significantly shortened residence time,indicating substantially worse performance of spatial learning and memory (P=0.023,0.001,0.001).Compared with the model group,the electro-acupuncture groups 2 and 3 had significantly shortened latency,a significantly higher frequency of entry into the goal box,and significantly longer residence time (P=0.046,0.017,0.049,0.007,0.031,0.009).Conclusions Different courses of electro-acupuncture treatment at Baihui,Fengfu,and Shenshu of both sides can substantially improve the cognitive function and spatial learning and memory in mice exposed to radiation.Particularly,2 and 3 weeks of electro-acupuncture treatment give the best outcomes.

RESUMEN

Objective To investigate the effects of electroacupunctures at Baihui,Fengfu,and Shenshu on the cognitive function and learning memory of mice exposure with ionizing radiation.Methods C57BL/6J (30-day old) mice were randomly divided into l0 groups consisting of the control group,X-ray exposure groups (4,8 and 16 Gy),electroacupuncture-nonacupiont (E-nonA) groups (4,8 and 16 Gy) and electroacupuncture-acupiont (EA) groups (4,8 Gy and 16 Gy),with 10 mice assigned to each group.The mice were irradiated (IR) on the left cerebral hemisphere with doses of 4,8 and 16 Gy.Some mice were electroacupunctured for 3 courses of treatment.Using weight as a measurable parameter,the open-field test and Morris water maze test were used to evaluate the curative effects.Results Compared with the control group,the weights of mice in the exposure groups (8 and 16 Gy) significantly decreased (t =2.917,9.650,3.043,2.882,P < 0.05) at 7 and 14 d,while the weights of the E-nonA (8 and 16 Gy) and EA groups (8 and 16 Gy) were not significantly altered.Compared to the model groups (8 and 16 Gy),the weights of EA groups (8 and 16 Gy) significantly increased (t =6.602,5.409,19.660,2.975,P < 0.05).In the open-field test,the crossing times and the rearing time of exposure model groups (8 and 16 Gy) significantly decreased compared to the control group (t =2.183,2.119,2.369,2.231,P < 0.05).Compared with model groups (8 and 16 Gy) and E-nonA groups (8 and 16 Gy),the crossing times and rearing time of the EA groups were significantly increased (compared with model:t =8.914,2.727,4.325,4.178,P < 0.05;compared with E-nonA:t =4.030,3.014,2.150,4.771,P <0.05).In the Morris water maze,compared with the control,the latent period of model group (4 Gy) was significantly increased and the crossing times significantly decreased (t =4.544,3.422,P < 0.05);compared to model and E-nonA groups (4 Gy),the latent period of EA group significantly increased (compared with model:t =2.877,P < 0.05;compared with E-nonA:t =3.001,P < 0.05).At day 2 and 3,the latent period of model groups (8 and 16 Gy) significantly increased (t =2.544,3.706,3.796,7.934,P < 0.05) and the crossing times (t =2.120,2.393,P < 0.05) significantly decreased,resident time in target area of model group (8 Gy) was significantly decreased (t =2.543,P < 0.05) compared to the control group.Compared with model and E-nonA groups (8 and 16 Gy),the latent period of EA gourps significantly decreased (compared with model:t =5.404,7.869,4.104,15.590,P < 0.05;compared with E-nonA:t =2.938,7.955,6.566,10.350,P < 0.05) while the through times and resident time in target area significantly increased in the EA group (8 Gy) (compared with model:t =2.673,2.613,P < 0.05;compared with E-nonA:t =3.345,2.179,P < 0.05).Conclusions Irradiation with a dose higher than 8 Gy could induce mice brain injury.Electroacupuncture can alleviate the radiation damage of cognitive function as well as learning and memory functions in mice.

RESUMEN

For rapid screening, we constructed two levels pools (primary and secondary pools) of the bacterial artificial chromosome (BAC) library of Chinese fine wool merino sheep. The primary pools were based on the individual 384-well microtiter plate and were prepared with a three-dimensional pooling scheme. Three dimension (plate, row and column) pools were made for each. The secondary pools were based on the entire BAC library. We developed a PCR based strategy to identify positive BACs from sheep BAC library. First, we analyzed secondary pools DNAs, according to the result, we analyzed correlative primary pools. It was one-step screening (66 PCR reactions) that we could screen a single positive clone from 74 000 BACs by our method, or three-step screening (less than 100 PCR reactions) could screen more clones. By one-step screening (66 PCR reactions), we screened successfully a positive clone 373D13 with polymorphism marker BF94-1.

OBJECTIVE: To study the effects of Naomaitong Recipe, a compound Chinese herbal medicine, on the expressions of nuclear factor-kappaB (NF-kappaB) and nitric-oxide synthases (NOSs) in brain tissue after focal cerebral ischemia reperfusion in aged rats and the dosage-effect relationship. METHODS: We adopted the middle cerebral artery occlusion (MCAO) in aged rats as the focal cerebral ischemia model, with 3-hour ischemia and 12-day reperfusion, and observed the changes of the nerve dysfunction score, the water content of cerebral constitution and the expressions of NF-kappaB, heat-shock protein 70 (HSP70) and NOSs in cerebral cortex. RESULTS: The nerve dysfunction score, the water content of cerebral constitution, the expressions of NF-kappaB, HSP70, neuronal nitric-oxide synthase (nNOS), endothelial nitric-oxide synthase (eNOS) and inducible nitric-oxide synthase (iNOS) in the untreated group were higher than those of the sham-operated group. The nerve dysfunction score, the water content of cerebral constitution, the expressions of NF-kappaB, iNOS and nNOS were lower, while the expressions of HSP70 and eNOS were higher in the high-, medium- and low-dose Naomaitong-treated groups and nimodipine-treated group than those of the untreated-group. The nerve dysfunction score and the expression of nNOS were lower, while the expression of eNOS was higher in the medium-dose Naomaitong-treated group than those of the nimodipine-treated group. The expressions of HSP70 and eNOS were higher, while the other indices were lower in the medium-dose Naomaitong-treated group than those of the low-dose Naomaitong-treated group. CONCLUSION: Naomaitong Recipe can protect the brain cells from damage after focal cerebral ischemia reperfusion injury by adjusting the expressions of NF-kappaB, HSP70 and NOSs, and the medium-dose Naomaitong Recipe is more effective.

Objective To study the protective mechanism of Buyang Huanwu decoction(补阳还五汤) on brain damage after focal cerebral ischemia/reperfusion(I/R)in aged rats from the expression changes of nuclear factor-?B(NF-?B),heat shock protein 70(HSP70)and nitric oxide synthase(NOS).Methods A focal cerebral ischemia model was established by middle cerebral artery occlusion(MCAO),and the models were duplicated.Ninety-six aged rats were randomly divided into four groups:sham-operated group,model group,nimodipine group(6 mg?kg~(-1)?d~(-1)),Buyang Huanwu decoction group(0.9g?kg~(-1)?d~(-1)).The ischemia(I)was performed for 3 hours and reperfusion(R),for 1,3,6 and 12 days.The effects of Buyang Huanwu decoction on the nervous dysfunction score,the water content of cerebral constitution and the expressions of NF-?B,HSP70 and NOS were observed at different time points of reperfusion.Results The nervous dysfunction score,the water content of cerebral constitution,the expressions of NF-?B,HSP70, endothelial nitric oxide synthase(eNOS),neuronal nitric oxide synthase(nNOS),and inducible nitric oxide synthase(iNOS)in the model group were higher than those of the sham-operated group(P

Objective:To study the protective effect of electroacupuncture on stem cell proliferation and differentiation in dentate gyrus (DG) in aged rats with focal cerebral ischemia reperfusion injury. Methods:We duplicated focal cerebral ischemia model with MCAO,with ischemia 3h,I/R1,3,7,14,21d points. The effect of electroacupuncture on the nervous dysfunction score,the water content of cerebral constitution and the stem cell proliferation and differentiation in dentate gyrus were observed,and the non-acupoint group was as control. Objective:To study the protective effect of electro-acupuncture on glutamate(GLU),glycine(GLY),aspartate(ASP)and NF-?B expression in rat with cerebral ischemia reperfusion.Methods:The middle artery occlusion(MCAO)model was established and high pressure liquid chromatography(HPLC)fluorescence assay was used to detect contents of glutamate(GLU),glycine(GLY)and aspartate(ASP)in rat brain.The effect of electro-acupuncture on the nervous dysfunction score,the water content of cerebral constitution and the expression of NF-?B were observed.The group with non-acupoint group was as control.Results:The nervous dysfunction score,the water content of cerebral constitution,NF-?B,GLUand ASP in model group were higher than those of the sham-operated group.GLY levels had no obvious changes,there was no significant diference between the model group and the non-acupoint group.The nervous dysfunction score,the water content of cerebral constitution,the expression of NF-?B,GLU and ASP in electro-acupuncture group were lighter than that of the model group,GLY levels had no obvious changes.Conclusion:Electro-acupuncture could protect brain cell from damage after focal cerebral ischemia reperfusion injury by inhibiting the cerebral expression of NF-?B and excitatory amino acid.

Objective: To study the protective e ects and dosage-e ect relationship of Naomaitong on focal cerebral ischemia reperfusion in aged rats.Methods : Focal cerebral ischemia model ( ischemia 3h and I/R12d)was duplicated with MCAO.The changes of the nervous dysfunction score ,the water content of cerebral constitution and the expression of TNF-?,VCAM-1,ICAM-1and its mRNA were observed. Results :The nervous dysfunction score ,the water content of cerebral constitution, the expression of TNF-?,VCAM-1,ICAM-1and its mRNA in aged model group were higher than those of the sham-operated group;but all of these were decreased in three Naomaitong groups and Nimodipine group compared with that of aged model group;the nervous dysfunction score and the expression of VCAM-1,ICAM-1 mRNA in Naomaitong group(0.9g?kg-1) were lighter than that of the Nimodipine group;the nervous dysfunction score,the water content of cerebral constitution, the expression of TNF-?,VCAM-1,ICAM-1and its mRNA in Naomaitong group(0.9g.g-1.d-1) were higher than that of Naomaitong group(0.45g?kg-1?d-1).Conclusion :Naomaitong could protect brain cell from damage after focal cerebral ischemia reperfusion injury by inhibiting the expression of TNF-?,VCAM-1,ICAM-1, with the middle dose of Naomaitong being more e ective.

RESUMEN

Objective To establish the quality standard of Fengsui Pills. Methods The qualitative analysis of Fructus Amomi and Radix Glycyrrhizae in Fengsui Pills was carried out by TLC,and the content of berberine hydrochloride,an effective component of Cortex Phellodendri in Fengsui Pills was determined by HPLC. Results The chromatographic sports of Fructus Amomi and Radix Glycyrrhizae were identified without the interference of negative control. A linearity of berberine hydrochloride was obtained from 0.272?g to 1.360 ?g with a good correlation (r=0.999 9,n=5).The average recovery was 99.01 %and the RSD was 0.98%. Conclusion The qualitative and quantitative methods established in this study are simple and feasible,accurate and reproducible,and can be used as the quality control standard for Fengsui Pills.
