RESUMEN

Objective:To investigate the relationship between serum miR-501 and miR-195 levels and sensitivity to concurrent chemoradiotherapy in patients with locally advanced cervical cancer (LACC).Methods:Clinical data of 96 patients with LACC admitted to Nanyang Central Hospital from January 2020 to June 2022 were retrospectively analyzed and compared with those of 96 healthy subjects during physical examination in our hospital during the same period to compare the differences of serum miR-501 and miR-195 levels. Tumor status was reviewed at 6 months after concurrent chemoradiotherapy. Patients were divided into the sensitive and resistant groups according to the evaluation criteria of solid tumor efficacy. The relationship between serum miR-501 and miR-195 levels and the sensitivity to concurrent chemoradiotherapy in LACC patients was analyzed by univariate and multivariate analyses. The receiver operating characteristic (ROC) curve was also drawn to predict differential efficacy of concurrent chemoradiotherapy sensitivity in LACC patients. Multivariate analysis was conducted by binary logistic regression analysis. P<0.05 indicated statistically significant differences. Results:In LACC patients, serum miR-501 level was significantly higher, whereas serum miR-195 level was significantly lower than those in physical examination subjects (both P<0.05). Univariate analysis showed that serum miR-501 level at admission in the resistant group was significantly higher, whereas serum miR-195 level was significantly lower than those in the sensitive group (both P<0.05). Multivariate analysis showed that serum miR-501 and miR-195 levels were significantly correlated with the sensitivity to concurrent chemoradiotherapy in LACC patients. The area under the ROC curve (AUC) was 0.736 and 0.913, respectively. Conclusions:The higher the serum miR-501 level and the lower the serum miR-195 level before treatment, the higher the probability of resistance to concurrent chemoradiotherapy in LACC patients. Serum miR-501 and miR-195 levels of LACC patients before treatment have certain predictive value for the sensitivity to concurrent chemoradiotherapy.

RESUMEN

With the development of the genomic sequencing of human commensal microbiome and bioinformatics, the role of esophageal microbiome in the pathogenesis of gastroesophageal reflux disease (GERD), Barrett's esophagus (BE), esophageal adenocarcinoma (EAC), and esophageal squamous cell carcinoma (ESCC) has been attracted much attention. Esophageal microbiome are relatively stable, but changes such as age, diet, use of drugs, oral hygiene, smoking, etc. can break the balance between esophageal microbiome and host immunity, and lead to persistent dysbacteriosis, increasing the risk of local inflammation and systemic inflammation and ultimately lead to the progression of many diseases. This article reviewed the progress in study on relationship between microbiome dysbiosis and pathogenesis of esophageal diseases.

This study seeks to explore the early signs of cognitive impairment in patients with obstructive sleep apnea hypopnea syndrome (OSAHS). According to polysomnography, twenty patients diagnosed with OSAHS and twenty normal controls underwent event-related potential (ERP) examination including mismatch negativity (MMN) and P300. Compared with normal controls, OSAHS patients showed significantly prolonged latency of MMN and P300 at Cz. After controlling age and body mass index (BMI), MMN latency positively correlated with apnea hypopnea index (AHI), oxygen reduction index, stage N1 sleep and arousal index, while MMN latency negatively correlated with stage N3 sleep and mean blood oxygen saturation; and P300 latency positively related to AHI and oxygen reduction index; no relationships were found among MMN latency, MMN amplitude, P300 latency and P300 amplitude. These results suggest that the brain function of automatic processing and controlled processing aere impaired in OSAHS patients, and these dysfunction are correlated with nocturnal repeatedly hypoxemia and sleep structure disturbance.

RESUMEN

Natural killer T-cell lymphoma (NKTL) is a malignant neoplasm which usually involves the nasal cavity or paranasal sinuses, while an orbit origin is extremely rare. Here we report the clinical, radiological and histopathologic features of a patient with NKTL originating from the orbit. We analyzed the clinical and radiologic records in the whole course of the disease. We also reviewed the morphology and immunohistochemistry of the neoplasm biopsy, including the presence of CD56, CD3 and cytotoxic molecules. This case demonstrated that nasal-type NKTL with a poor prognosis can originate from the orbit.

RESUMEN

The present paper is to investigate the variation characteristics and clinical application of event related potentials (ERPs) in patients with bipolar disorder. A total of 70 patients with bipolar disorder, with 39 patients in manic phase and 31 patients in depressive phase, and 50 health volunteers were recruited in the case-control study. The ERPs, such as P300, MMN, CNV, P50, were recorded by the Nihon Kohden evoked potential instrument. Compared with the healthy volunteers' latency in the control group, the latency of MMN, CNV, N2, P3a, P3b was longer in the manic phase group; the latency of MMN, CNV, P3a and P3b were longer and the amplitude of CNV was lower in the depressive phase group. The S2/S1 value of P50 obviously increased among the manic phase patients. With manic phase or depression phase of bipolar disorder, the variation characteristics of ERPs were different. The features of ERP reflect the cognitive dysfunction and sensory gating impairment of patient's brain. The results provide neural electrophysiological basis for clinical diagnosis and therapy of bipolar disorder.

RESUMEN

<p><b>OBJECTIVE</b>To compare the efficacy differences between integrative therapy of electroacupuncture and repetitive transcranial magnetic stimulation (rTMS) and simple rTMS only in the treatment of comorbid anxiety and depression (CAD).</p><p><b>METHODS</b>Eighty-five cases of CAD were randomized into two groups. The observation group (40 cases) was treated with electroacupuncture at Baihui (GV 20), Yintang (EX-HN 3), Shenting (GV 24), Shenmen (HT 7) and Neiguan (PC 6) mainly, combined with rTMS. The control group (45 cases) was treated with simple rTMS. Separately, before treatment, after 5 days and 10 days treatment, Hemilton Depression Scale (HAMD) and Hemilton Anxiety Scale (HAMA) were used for the scoring of depression and anxiety. Additionally, the efficacies were compared between two groups.</p><p><b>RESULTS</b>After 5 days, 10 days treatment, the scores of HAMA, HAMD were reduced remarkably as compared with those before treatment in two groups (all P < 0.001), HAMA score in observation group was lower than that in control group after 10 days treatment (P < 0.05). The remarkable effective rates of depression and anxiety in observation group those 10 days that treatment were 87. 1% (27/31) and 90.3% (28/31) respectively, which were higher apparently than 61.8% (21/34) and 61.8% (21/34) in control group separately (both P < 0.05).</p><p><b>CONCLUSION</b>The integrative therapy of electroacupuncture and rTMS is definitely effective on CAD. This method is better than simple rTMS.</p>

OBJECTIVE To compare the detection rates of Ureaplasma urealyticum(Uu),by 4 kinds rits and find out the best one.METHODS Using four kinds of mycoplasma detection kits,namely are French mycoplasmaIST2,Zhuhai Livzon′s ICS,Dark horse′s IES and Shanghai Its Prachanda,under the same conditions,the mycoplasma of the Clinical specimens for 260 cases were tested.RESULTS The detection rate of mycoplasma(Uu) for 48 hours by four kinds of brands kits about mycoplasma liquid culture was as follows 45.4% for French MycoplasmaIST2,36.2% for Zhuhai Livzon′s ICS,37.7% for Dark horse′s IES and 36.8% for Shanghai Its prachanda.The results of susceptibility test: the reagent of IST2 had.the lowest resistance to josamycin,doxycycline,clarithromycin,ofloxacin and azithromycin,and had the better results than the other three kinds of reagents.Mycoplasma reagents form different companies had significant differences in resistance rates to five kinds of antibiotics.CONCLUSIONS The results suggest that when we cultivate clinical mycoplasma should be careful to select of the mycoplasma liquid culture identification kit.

Objective To investigate the brain perfusion changes in patients with refractory depressive disorder (RDD) and non-refractory depressive disorder (NDD) using arterial spin labeling (ASL).Methods Present study included 12 patients with RDD, 37 patients with NDD and 42 controls,and their age, sex and handedness were matched among the three groups.FAIR sequences were performed using a 3 T MR imaging system with an 8 channel phase array head coiL The labelled and controled images were subtracted and then averaged to obtain perfusion-weighted images.The first 2 images were excluded to avoid T, equilibrium effects and then voxel based analysis was performed using SPM2. One way ANOVA analysis using age as covariance ( thresholded at P < 0.01 uncorrected) was performed.Results Patients with NDD showed regional alteration of the brain perfusion, mainly involved in two frontal subcortical circuits, i.e. significantly decreased perfusion in the left frontal and thalamus (the limbic thalamo-cortical circuit) whereas significantly increased perfusion in bilateral hippocampi, fight lentiform and left anterior cingulated gyros (the limbic-striatal-pallidal-thalamic circuit).In contrast, patients with RDD presented significantly decreased perfusion involving bilateral frontal areas (the limbic-thalamo-cortieal circuit) whereby no significantly increased perfusion areas were observed.Conclusion In addition to the reported experimental evidences, our results suggest that the RDD is associated with inactivity of bilateral frontal areas, while the NDD is associated with inactivity of left frontal areas and overactivity of bilateral limbic system.

RESUMEN

Objective Explore the conditions of the cloning,expression and purification of FMRP.Methods The plasmid pET22b(+)-FMR1,constructed by molecular cloning,was transformed into E.coli BL21(DE3) competent cells and induced to express FMRP by IPTG.Recombinant FMRP was purified by affinity chromatography,verified by Western-blot,and tested for its RNA binding ability.Results FMR1 cDNA was successfully cloned into pET22b(+) vector and expressed in E.coli BL21(DE3).A protein with Mr 79 000 was purified and confirmed to be FMRP.This protein retained the RNA binding ability of FMRP.Conclusion We successfully expressed recombinant hFMRP with high purity and activity in E.coli,which provided a reliable material to study the function of FMRP.

RESUMEN

<p><b>OBJECTIVE</b>To clone, express and purify nucleocapsid protein from SARS coronavirus PUMC2 strain.</p><p><b>METHODS</b>According to the published SARS coronavirus genome sequences, the full length cDNA of N protein from SARS coronavirus PUMC2 strain was cloned by RT-PCR and the cDNA was cloned into the pET32a expression vector. The recombinant N protein was expressed in E. coli BL21 (DE3), and purified by Ni(2+)-NTA.</p><p><b>RESULTS</b>Prokaryoticly expressed and purified N protein of SARS coronavirus PUMC2 strain was obtained.</p><p><b>CONCLUSIONS</b>The SARS coronavirus recombinant N protein obtained by genetic engineering methods can be used for further functional study of SARS coronavirus N protein.</p>

RESUMEN

<p><b>OBJECTIVE</b>To get the cDNA clones which cover the whole genome of SARS-CoV PUMC2 strain.</p><p><b>METHODS</b>Using the SARS-CoV PUMC2 strain genomic RNA as the template, the cDNA fragments were amplified by RT-PCR, the PCR products were further purified and ligated into the pGEM-T vector, and all the clones obtained were sequenced.</p><p><b>RESULTS</b>The cDNA clones which cover the whole genome of SARS-CoV PUMC2 strain were obtained.</p><p><b>CONCLUSIONS</b>These cDNAs can be provided for the function study of SARS-CoV proteins and the construction of full-length infectious cDNA clone of SARS-CoV.</p>

<p><b>OBJECTIVE</b>To perform variation and phylogenetics analysis on the SARS-CoV genome sequence (PUMC01) isolated in the Peking Union Medical College Hospital.</p><p><b>METHODS</b>The cDNA library of SARS-CoV (PUMC01 isolate) was constructed by means of random-priming strategy. Random selected plasmid was sequenced and the genome sequence of SARS-CoV-PUMC01 was assembled by conventional methods (The Genebank Accession No. of SARS-CoV-PUMC01 is AY350750). The variation and phylogenetics analysis were performed by comparing the PUMC01 sequence with other SARS-CoV isolates.</p><p><b>RESULTS</b>Ten variation sites were found by comparing PUMC01 isolate with Tor2 and Urbani isolates. In phylogenetic analysis of 18 SARS-CoV isolates, two classes were observed and there is different differential time between these two classes and the different isolates in each class.</p><p><b>CONCLUSIONS</b>The evidence of phylogenetic analysis of different SARS-CoV isolates from different region is instructive for understanding the clinical relations between the different isolates and the transmission chain of SARS-CoV.</p>
