@#Abstract: Objective To investigate the relationship between plasma exosomal microRNA (miRNA)-346 and treatment outcomes in patients with multidrug-resistant tuberculosis (MDR-TB), to provide more reference basis for the treatment of MDR-TB patients. Methods A total of 406 patients with MDR-TB admitted to Tuberculosis Control and Prevention Institute of Shaanxi Provincial between January 2018 and May 2021 were selected as the study subjects. General clinical data of the patients were collected and analyzed. The expression level of plasma exosomal miR-346 was detected by real-time fluorescence quantitative polymerase chain reaction. The predictive value of plasma exosomal miR-346 for treatment outcome was analyzed using receiver operating characteristic (ROC) curve analysis. Furthermore, the relationship between the expression of plasma exosomal miR-346 and treatment outcome was analyzed using a multivariable logistic regression model. After standard treatment, patients were divided into good treatment outcome group (n=226) and poor treatment outcome group (n=180) according to the treatment outcome. Results Typical exosomes were identified by transmission electron microscopy, particle size analysis and Western blot, that is, plasma exosomes were successfully extracted. In the poor treatment outcome group, more patients were complicated with diabetes or HIV infection, and the proportion of patients with pulmonary cavity, acid-fast bacilli smear positive rate >1+, previous treatment history and fluoroquinolone resistance was also significantly increased, and the levels of white blood cells, neutrophils, and monocytes were significantly increased, while the level of albumin was significantly decreased, with statistical significance (P>0.05). Compared with the good treatment outcome group [0.61 (0.46, 0.74)], the expression level of plasma exosomal miR-346 in the poor treatment outcome group [1.23 (0.60, 2.02)] was significantly higher (Z=-13.185, P<0.001). ROC curve analysis showed that the area under the curve of plasma exosomal miR-346 to predict adverse outcomes of MDR-TB treatment was 0.881 (95%CI: 0.846-0.915), with a sensitivity of 78.3% and specificity of 86.7%. The corresponding cut-off value was 0.81. Multivariate logistic regression analysis showed that diabetes, AIDS virus infection, pulmonary cavity, AFB smear positive degree>1+, previous treatment history, fluoroquinolones resistance and high expression of plasma exosomal miR-346 were independent influencing factors for poor treatment results of MDR-TB (P<0.05). Conclusions High expression of plasma exosomal miR-346 is associated with the high risk of adverse outcome of MDR-TB treatment, and it is promising to be a useful marker for predicting the outcome of MDR-TB treatment.

ABSTRACT Introduction: Volleyball is a team sport where winning the confrontation at the net largely determines characteristics. Volleyball players must constantly strive to improve team cooperation, defensive ability, and speed skills to improve their ability to perform over the net. Objective: Establish a fixed intensity load model for speed training on the athlete's visual response in volleyball. Methods: This paper selected several volleyball players as research subjects, randomly divided into two groups (regular training and experimental groups). The experimental group was subjected to a visual response speed training protocol based on routine training. Mathematical statistics were employed to analyze the athletes' reaction speed and defensive ability against the net. Results: The qualities analyzed, mainly movement speed, swing speed, agility, and endurance for fast swing and fast movement, showed a significant differential after the intervention. The speed response was concentrated in the center of body balance. The lower limbs increased the endurance for speed. After visual reaction speed training, the experimental group's ability to cope with attacks on the net was statistically superior (P<0.05). Conclusion: The proposed protocol can improve the reaction speed training of volleyball players. Volleyball players should reinforce reaction speed training daily to perform better close to the net. RESUMO Introdução: O voleibol é um esporte coletivo e suas características de vitória são determinadas em grande parte pelo confronto em rede. Os jogadores de vôlei devem se esforçar constantemente para melhorar a cooperação da equipe, a capacidade defensiva e habilidades de velocidade para melhorar a capacidade de atuarem pela a rede. Objetivo: Estabelecer um modelo de carga de intensidade fixa para o treinamento de velocidade na resposta visual do atleta em voleibol. Métodos: Este trabalho seleciona vários jogadores de vôlei como objetos de pesquisa, divididos aleatoriamente em dois grupos (grupos de treinamento regular e grupos experimentais). O grupo experimental teve o diferencial de ser submetido a um protocolo treinamento de velocidade de resposta visual baseado no treinamento de rotina. Estatísticas matemáticas foram empregadas para analisar a velocidade de reação dos atletas e a capacidade defensiva contra a rede. Resultados: As qualidades analisadas, principalmente velocidade de movimento, velocidade de balanço, agilidade e resistência para balanço rápido e movimento rápido mostraram um diferencial significativo após a intervenção. A velocidade de resposta concentrou-se no centro de equilíbrio corporal. A resistência para a velocidade foi incrementada pelos membros inferiores. Após o treinamento da velocidade de reação visual, a capacidade do grupo experimental de enfrentar os ataques sobre a rede foi estatisticamente superior (P<0,05). Conclusão: O treinamento da velocidade de reação dos jogadores de vôlei pode ser aprimorado com o protocolo proposto. Os jogadores de vôlei devem reforçar o treinamento da velocidade de reação em seu treinamento diário afim de uma melhor capacidade de atuação próxima à rede. RESUMEN Introducción: El voleibol es un deporte de equipo y sus características ganadoras están determinadas en gran medida por el enfrentamiento en la red. Los jugadores de voleibol deben esforzarse constantemente por mejorar la cooperación en equipo, la capacidad defensiva y las habilidades de velocidad para mejorar su capacidad de actuación sobre la red. Objetivo: Establecer un modelo de carga de intensidad fija para el entrenamiento de velocidad sobre la respuesta visual del deportista en el voleibol. Métodos: Este trabajo selecciona a varios jugadores de voleibol como sujetos de investigación, divididos aleatoriamente en dos grupos (grupos de entrenamiento regular y grupos experimentales). El grupo experimental tuvo el diferencial de ser sometido a un protocolo de entrenamiento de la velocidad de respuesta visual basado en el entrenamiento rutinario. Se emplearon estadísticas matemáticas para analizar la velocidad de reacción y la capacidad defensiva de los atletas contra la red. Resultados: Las cualidades analizadas, principalmente la velocidad de movimiento, la velocidad de balanceo, la agilidad y la resistencia para el balanceo rápido y el movimiento rápido mostraron un diferencial significativo después de la intervención. La velocidad de respuesta se concentró en el centro de equilibrio del cuerpo. La resistencia para la velocidad se incrementó en los miembros inferiores. Tras el entrenamiento de la velocidad de reacción visual, la capacidad del grupo experimental para afrontar los ataques sobre la red fue estadísticamente superior (P<0,05). Conclusión: El entrenamiento de la velocidad de reacción de los jugadores de voleibol puede mejorar con el protocolo propuesto. Los jugadores de voleibol deben reforzar el entrenamiento de la velocidad de reacción en su formación diaria para mejorar su capacidad de actuación cerca de la red. RESUMEN

In the present study, the interaction between Perfluorohexadecanoic acid (PFHxDA) and human serum albumin (HAS) was studied by fluorescence spectroscopy, molecular docking, dynamic simulation and circular dichroism (CD). The interaction character and the effect on human serum albumin conformation were measured by simulating the physiological condition (pH= 7.4). Experiments and simulation results revealed that PFHxDA molecules and HSA have regular fluorescence quenching, and the quenching mechanism is static quenching and non-radiative energy transfer. Thermodynamic analysis revealed the binding behavior was mainly governed by hydrophobic forces. Specific binding site experiments showed that the binding site of PFHxDA was a site I of HSA. The results from the CD spectrum demonstrated that PFHxDA changed the molecular conformation of HSA, which is consistent with the results obtained by molecular docking and dynamic simulation.

@#With the gradual maturity of laser technology, it has become widely considered a new method for disease treatment. Nd:YAG laser and Er:YAG laser are two representative solid-state lasers. These lasers are easy to use, comfortable and safe, and thus, they have recently become a research hotspot in dental treatment. Nd:YAG laser and Er:YAG laser have been used for the treatment of dentin hypersensitivity and dental caries, root canal therapy, pulp preservation and apical surgery. They are effective adjuvant methods for the treatment of dental pulp diseases and provide new avenues for clinical treatment. In this paper, the application of Nd:YAG laser and Er:YAG laser in the treatment of dental pulp disease is described to provide a reference for clinical treatment options.

Semaphorin 4A plays a regulatory role in immune function and angiogenesis. However, its specific involvement in controlling lung fibrosis, a process that is closely related to angiogenesis and inflammation is still poorly understood. In the present study, we show that treatment of Sema4A on normal lung fibroblasts induces expression of proteins that contribute to a contractile phenotype, including α-smooth muscle actin (α-SMA), ezrin, moesin, and paxillin. We confirm that Sema4A enhances the ability of lung fibroblasts to contract collagen gel. Sema4A treatment led to resistance to apoptosis in normal lung fibroblasts. Relative to normal lung fibroblasts, fibroblasts cultured from scars of patients with the fibrotic disease Systemic Sclerosis (SSc) showed elevated Sema4A secretion, enhanced α-SMA, ezrin, moesin, and paxillin expression, and high ability to induce collagen gel contraction. Using neutralizing antibody against Sema4A receptor, PlexinD1, we found that endogenous Sema4A signalling in SSc fibroblast was through PlexinD1 receptor. We then identified the signalling mechanism through which Sema4A-PlexinD1 promotes the ability of normal fibroblasts to contract a collagen gel matrix. Western blot analysis showed that Sema4A activated the Akt pathway in lung fibroblasts, and the specific inhibitor of Akt pathway, Akt inhibitor III, blocked the ability of Sema4A to promote the ability of lung fibroblasts to contract a collagen gel matrix. Thus, blocking Sema4APlexinD1- Akt cascades might be beneficial in reducing pulmonary fibrosis.

This study examined endogenous cannabinoid (ECB)-anandamide (AEA) and its can-nabinoid receptors (CBR) in mice liver with the development of schistosomajaponicum.Mice were infected with schistosoma by means of pasting the cercaria onto their abdomens.Liver fibrosis was pathologically confirmed nine weeks after the infection.High performance liquid chromatography (HPLC) was employed to determine the concentration of AEA in the plasma of mice.Immunofluorescence was used to detect the expression of CBR 1 and CBR2 in liver tissue.Morphological examination showed typical pathological changes,with worm tubercles of schistosoma deposited in the liver tissue,fibrosis around the worm tubercles and infiltration or soakage ofinfiammatory cells.Also,CBRI and CBR2 were present in hepatocytes and hepatic sinusoids of the two groups,but they were obviously enhanced in the schistosoma-infected mice.However,the average optical density of CBR1 in the negative control and fibrosis group was 13.28±7.32 and 30.55±7.78,and CBR2 were 28.13±6.42 and 52.29±4.24 (P<0.05).The levels of AEA in the fibrosis group were significantly increased as compared with those of the control group.The concentrations of AEA were (0.37±0.07) and (5.67±1.34) ng/mL (P<0.05).It is concluded that the expression of endocannabinoids AEA and its cannabinoid receptor CBR were significantly increased in schistosoma-infected mice.Endogenous endocannabinoids may be involved in the development of schistosoma-induced liver fibrosis.
