RESUMEN

@#ObjectiveTo explore the protective effect of Sox11 gene on cerebral ischemia reperfusion injury(CIRI)in mice and its mechanism,so as to provide a new target for the treatment of CIRI.MethodsThe mouse middle cerebral artery occlusion(MCAO)model and Neuro2A cell oxygen glucose deprivation reperfusion(OGDR)model were established and detected for the temporal and spatial distribution of Sox11 in the models by real-time quantitative PCR,Western blot,immunohistochemistry(IHC)and immunohistofluorescence(IHF).The altered expression of some crucial genes in the pathway of apoptosis and inflammation in OGDR model after the disruption of Sox11 expression was detected by Western blot.ResultsThe expression level of Sox11 mRNA and protein increased significantly in both MCAO and Neuro2A OGDR models(P = 0.000 1 ~ 0.038 8);After CIRI,Sox11 expression was elevated in the hippocampal dentate gyrus(DG)region of mice;After interfering with the expression of Sox11 in OGDR model,the expression of apoptosis-related protein Cleaved Caspase 3 significantly increased,while the expression of anti-apoptosis protein Bcl-2 significantly decreased,and the expression of phosphorylated NF-κB(p-NF-κB)protein related to inflammatory reaction also up-regulated significantly.Conclusion Sox11gene had a protective effect against CIRI in mice,and was involved in the regulation of apoptosis and inflammation pathways after CIRI.

RESUMEN

To establish the online two-dimensional liquid chromatography by using double gradient liquid chromatography system and UV detector, in order to simultaneously determine the content of paeoniflorin, paenol, amygdaloside and cinnamic acid. A pump of the two-dimensional liquid chromatography was adopted as the one-dimensional separation pump. C18 (3.0 mm x 150 mm, 3 microm) was used as the analytical column, with acetonitrile as the organic phase and 0.08% phosphoric acid + 0.08% triethylamine as the aqueous phase for gradient elution at the flow rate of 0.5 mL x min(-1). Another pump of the two-dimensional liquid chromatography was adopted as the two-dimensional separation pump. PAII C18 was used as the analytical column, with acetonitrile as the organic phase and 20 mmol, pH 3.0 monopotassium phosphate as the aqueous phase for gradient elution at the flow rate of 0.8 mL x min(-1). The detection wavelengths were set at 218, 230, 275 nm by using wavelength time-switching program. The linearity range of paeoniflorin, amygdaloside, paeonol and cinnamic acid were 5.55-222 (r = 0.999 7), 6.6-264 (r = 0.999 8), 3.3-132 (r = 0.999 5) and 0.315-12.6 mg x L(-1) (r = 0.999 7), respectively. The average recoveries of the four components were between 96.12% and 103.9%. The experiment proved that this method was so rapid and accurate in determination results that it could be used for evaluating drug quality.

<p><b>OBJECTIVE</b>To analyze the meiotic segregation results of male reciprocal chromosome translocation by fluorescence in situ hybridization (FISH).</p><p><b>METHODS</b>Multi-color FISH using 3 combined probes located in any 3 chromosome segments on both sides of two breakpoints was performed on the de-condensed sperm head to analyze the sperm chromosomal contents and segregation patterns.</p><p><b>RESULTS</b>Four male reciprocal translocation carriers were included in the study, with the karyotypes of 46, XY, t(2;18) (p16; q23); 46, XY, t(4;6) (q34;q21); 46, XY, t(8;13) (q23;q21) and 46, XY, t(4;5) (4q31;5q13), respectively. The results showed that 4 carriers had different proportions of various segregated spermatozoa. The spermatozoa of alternate, adjacent-1, adjacent-2, 3:1, non-disjunction in meiosis II, and 4:0 or diploidy accounted for 27.1%-49.4%, 26.9%-37.6%, 2.7%-15.7%, 8.6%-32.7%, 0.2%-1.9%, and 0.1%-0.4%, respectively.</p><p><b>CONCLUSION</b>For each-reciprocal translocation carrier seems to have a particular meiotic segregation results, FISH analysis on sperm head should be done for each carrier in order to provide an accurate genetic counseling.</p>

RESUMEN

Objective To study the changes of anticoagulation and fibrinolysis in neonatal respiratory distress syndrome(NRDS).Methods The levels of plasma protein C(PC),total protein S(TPS),antithrombin Ⅲ(AT-Ⅲ), D-Dimer(D-D) and von Willebrand factor(vWF) were measured with ELSIA assay and immunoturbidimetry in 27 cases NRDS, 20 cases prematures and 15 cases full-term newborns as normal controls.Results The levels of PC, TPS and AT-Ⅲ were lower, but the levels of D-D and vWF were significantly higher than those of prematures and normal controls.The PC,TPS of prematures were lower than normal controls otherwise there is no difference of AT-Ⅲ,D-D,vWF between prematures and normal controls. Conclusions There exists the activation of anticoagulation and fibrinolysis and the injury of vascular endothelium in critical NRDS, and they are early sensitive indexes for the diagnosis of disseminated intravascular coagulation(DIC).

RESUMEN

Objective To investigate the levels of interleukin-15(IL-15) in children with acute leukemia and contribute to understand their function on acute leukemia about the process.Methods Every group of acute lymphocytic leukemia(ALL),acute nonlymphocytic leukemia(ANLL) or non-leukemia group had 20 children who did not receive any treatment. Peripheral blood of every group and 20 other healthy children were aspirated .The levels of IL-15 in serum was analyzed by enzyme linked immunosorbent assay (ELISA).Results The levels of IL-15 in ALL group,ANLL group, non-leukemia group and healthy children were (34.37?2.8) ng/L, (29.61?3.2 )ng/L, (117.54?3.9) ng/L, (122.23?4.2) ng/L. Compared with the levels of the healthy group or the non-leukemia group, the difference of ALL or ANLL was signicant in statistics(q=3.95,4.03 Pa0.05).Conclusion Detection of serum IL-15 levels may have clinical significance in judging the anti-tumor immune condition in children with leukemia.

RESUMEN

Objective To investigate children′s myelodysplastic hematopoietic cells reaction to interleukin (IL)-15.Methods CD 34 + cells in bone marrow from 18 myelodysplast syndrome(MDS) patients were purified by an immunomagnetic beads sorting system. Apoptosis of hematopoietic precursors was assayed by propidium iodine staining and flow cytometric analysis.Results On 8th cultured day,when IL-15 concentration was between 0-100 ng/ml,it could suppress apoptosis of hematopoietic cells in MDS patients in a dose-and- time dependent manner. IL-15 in study group significanthy lower than that of control group.Conclusion IL-15 may partly suppress apoptosis of hematopoietic cells in MDS patients.
