RESUMEN

To explore the effects of serum insulin on the expression of ChREBP, ACC and FAS in vivo, KKAy mice which were characterized with high levels of both serum insulin and glucose and DIO mice which were characterized with high serum insulin level alone were utilized, separately. The age-matched C57BL/6J mice fed with standard chow were used as normal control (Con). Expressions of hepatic ChREBP, ACC and FAS were detected by Western blotting. As the results, in KKAy mice, a positive correlation between the levels of serum insulin and glucose (r = 0.902, P < 0.000), as well as between the levels of serum insulin and TG (r = 0.732, P < 0.000), was observed. Meanwhile, the expressions of hepatic ChREBP, ACC and FAS increased significantly and accompanied with its hyperinsulinemia and hyperglycemia, separately. In DIO mice, correlation between the levels of serum insulin and TG (r = 0.722, P < 0.001) also showed positive, and the expressions of hepatic ChREBP, ACC and FAS increased significantly and also accompanied with its hyperinsulinemia. However, their blood glucose values were almost normal. These demonstrated that hyperinsulinemia may cause glycolipid metabolic disorders by up-regulating the expression of ChREBP in vivo.

RESUMEN

This study is to investigate the effects of a Chinese prescription (FF), compatibility of Rhodiola crenulata, Cordyceps militaris and Rheum palmatum, on nephropathy in type 1 diabetic rats induced by streptozocin. According to fasting blood glucose level, diabetic rats were divided into three groups: model group, insulin-treated group and FF-treated group. Parameters for evaluating the glucose & lipid metabolism and the renal function were monitored dynamically. Levels of oxidative stress were detected ten weeks later. The results show that FF could significantly decrease the level of serum glucose and lipid profiles, improve the renal functions by decreasing blood urea nitrogen, urine albumin excretion and urease activity; FF could also affect on oxidative stress. In conclusion, Chinese prescription FF could ameliorate hyperglycemia-mediated renal damage in type 1 diabetic rats. These effects may be related to its regulation on the metabolism of glucose and lipid, the microcirculation disturbance and the oxidative stress.

RESUMEN

<p><b>OBJECTIVE</b>To study digitized virtual hepatic three-dimensional reconstruction and virtual hepatic surgery.</p><p><b>METHODS</b>The whole series of hepatic images taken from the database of digitized Virtual Chinese Human Female Number 1 (VCH-F1) was employed to reconstruct a three-dimensional (3D) liver. First, studied some algorithms for registration of human liver tissue images, and then, segmented the regions of liver, vein, bile duct, and gallbladder from the images. Based on them, the 3D visualization human liver model was reconstructed. Finally, a 3D visualization demo system of liver was developed based on personal computer and Windows operation system.</p><p><b>RESULTS</b>This demo system of liver provided a graphics user interface to rotate, scale the 3D liver to observe the 3D hepatic structure, and a virtual liver simulation system of resection with primary function.</p><p><b>CONCLUSIONS</b>The study may be beneficial to the future research on digitized virtual hepatic and virtual hepatic surgery, and the 3D visualization demo system of liver may be beneficial to the research on the hepatic structure.</p>

<p><b>OBJECTIVE</b>To study the role of plasminogen activator inhibitor-1 (PAI-1) and tissue plasminogen activator (tPA) in the accumulation of extracellular matrix (ECM) in the kidney of KKAy mice with type 2 diabetes.</p><p><b>METHODS</b>KKAy mice, a type 2 diabetic animal model, and C57BL-J mice were sacrificed at 16, 20, and 24 weeks of age, respectively. The local expression of renal laminin was analyzed with immunohistochemistry. Chromogenic substance was used to show the activity of PAI-1. The mRNA expression of tPA was determined by RT-PCR. The mRNA expression of PAI-1 was measured by reverse transcription-polymerase chain reaction (RT-PCR).</p><p><b>RESULTS</b>Laminnin expression was significantly increased in all age groups of KKAy mice. The tPA mRNA was significantly lower than that in C57BL mice, especially at the age of 16w (only 47%). Otherwise the PAI-1 mRNA expression was remarkably up-regulated than that in C57BL mice.</p><p><b>CONCLUSION</b>In type 2 diabetes KKAy mice, the accumulation of ECM may be associated with the abnormal expression of PAI-1/tPA mRNA.</p>

<p><b>OBJECTIVE</b>To study the three-dimensional (3D) reconstruction and 3D visualization of the pancreas and create anatomy of the digitalized visual pancreas so as to construct a concrete basis for virtual operation and surgical operation on pancreas.</p><p><b>METHODS</b>The digital imaging data of pancreas, duodenum, common bile duct, arteries and veins were obtained from the Virtual Chinese Human-Female 1 (VCH-F1) and processed using ACDSee and Photoshop so as to reconstruct 3D pancreas digitally and realize 3D visualization of pancreas.</p><p><b>RESULTS</b>We successfully 3D reconstructed and visualized the pancreas and the peri-pancreatic structures: the duodenum, the common bile duct, the inferior vena cava, the portal vein vessels, the aorta, the ceoliac trunk vessels. The 3D and visualized pancreas manifested itself with its complete structure as well as its adjacency to other tissues.</p><p><b>CONCLUSIONS</b>The 3D reconstruction and 3D visualization of the pancreas based on the digital data of VCH-F1 produces a digitally visualized pancreas, which promises us a novel method for virtual operation on pancreas, clinical operation on pancreas and anatomy of 3D visualized pancreas.</p>

RESUMEN

<p><b>OBJECTIVE</b>To investigate the early pathological changes of KKAy mouse model of type 2 diabetes.</p><p><b>METHODS</b>Five male KKAy mice and six C57BL mice each were studied at the age of 8, 16, 20 and 24 weeks. With each mouse a 24-hour urine collection was made for the tests of proteinuria. Plasma insulin, lipids, serum creatinine and urea were also measured. Renal tissues were observed to assess glomerular and tubulointerstial pathology.</p><p><b>RESULTS</b>The KKAy mice developed hyperglycemia, hyperinsulinemia and obesity by 16 weeks of age (P < 0.01). The proteinuria increased with the increasing of age (P < 0.005), but there were no changes in control. The glomerular hypertrophy was observed in KKAy mice at the age of 16 weeks. Computer map analysis system (CMIAS) indicated the expansion of mesangial matrix in KKAy mice with ageing. There was significant tubular dilation, accompanied with focal tubular atrophy and interstitial fibrosis. On electron microscopy, GBM undergo progressive thickening (P < 0.01), accompanied with podocytes fusion and increasing of proteinuria.</p><p><b>CONCLUSIONS</b>KKAy mice developed hyperglycemia, hyperinsulinemia and obesity after 16 weeks, with proteinuria, mesangial matrix accumulation, GBM thickening and tubular dilation. It was considered a good animal model for the early pathology changes of DN.</p>
