@#【Objective】 To investigate the endometrial pathological composition in abnormal uterine bleeding(AUB),and whether it related to age,body mass index,waist circumference or waist-hip ratio.【Methods】From January 2017 to June 2017,206 patients with AUB underwent diagnostic curettage or hysteroscopy and endometrial biosy in the daytime operating room of the Third Affiliated Hospital of Sun Yat- sen University were included ,and age,height, weight ,waist circumference ,hip circumference and endometrial pathology were collected and analyzed statistically.【Results】① The proportions of Normal endometrium(NE),Disorder Proliferative Endometrium(DPE),Endometrial Polyp(EP),Endometrial Hyperplasia without atypia (EH without atypia),Endometrial Hyperplasia with atypia (EH with atypia),and Endometrial Cancer(EC,type I)were 32.52%,8.25%,6.31%,40.77%,9.22% and 2.91% respectively in endometrial pathological grouping. ②The age of EC group was 50.0(S=7.3)years old,which was older than that of NE group 42.0(S=13.0,P<0.01). ③The abdominal circumference of EC group(89.0±4.9)cm were higher than that of NE group(81.4±5.4)cm(P<0.01). WHR of EH with atypia group and EC group were(0.877±0.035)and(0.912±0.042) respectively,which were higher than that of NE group[(0.847±0.036);P<0.01,P<0.01].【Conclusions】About half of patients with abnormal uterine bleeding have endometrial hyperplasia,and the proportion of endometrial cancer is nearly 3%,both of which are associated with central obesity(such as increased waist circumference and/or waist-to-hip ratio).

RESUMEN

Idiopathic macular hole ( IMH ) refers to full thickness defect of retinal neurepithelium layer in macular area with unclear reasons. Early IMH can be observed. Vitrectomy combined with internal limiting membrane ( ILM ) peeling is a conventional method for treating IMH. The continuous improvement and innovation of ILM surgery contribute greatly to the diversity and maturation of IMH treatment. At present, the pharmacological vitreolysis and pneumatic vitreplysis have gradual progress in the prevention and treatment of IMH. In this article we review the current management of IMH.

Objective To investigate the effect of Dahuang Ling Xian Capsules(DLXC) on miRNAs in bile duct cells at inflammatory state. Methods The cultured well-grown bile duct cells at logarithmic phase were chosen for the experiment. Lipopolysaccharide (LPS , 5 μg/mL) was used to induce the model of bile duct cells at inflammatory state, and then DLXC (1 mg/mL) was added. After co-culturing for 72 h, the total RNA of bile duct cells was collected for miRNA chip detection and bioinformatics analysis. Results DLXC showed regulatory effect on the expression of 30 miRNAs in bile duct cells at inflammatory state, through which to affect biological processes involving with bile duct cell signal transduction, organic compound reaction and hypoxia stress, cell components such as cytoplasm , endoplasmic reticulum and membrane rafts , molecular biological functions of signal transduction activity , DNA binding in transcription regulation region and voltage-gated potassium channel activity , and multiple pathways of hypoxia-inducible factor-1(HIF-1) signaling pathway , uanosine 3 ' , 5 '-cyclic monophosphate-protein kinase C (cGMP-PKG) signaling pathway , tumor necrosis factor alpha (TNF-α) signaling pathway, chemokine signaling pathway, apoptosis signaling pathway, phosphatidylinositol-3-kinase (PI3K)-Akt signaling pathway and Toll-like signaling pathway. Conclusion DLXC exert the multidimensional regulation of bile duct cells at inflammatory state with miRNAs as targets.

Objective To investigate the bystander effect injury to lung-heart-liver-spleen caused by 12C6+ beam radiation,and explore the prevention and treatment effect of Guiqiyiyuan ointment on the injury and its mechanism.Methods Ninety healthy male Wistar rats were randomly divided into 3 groups:NC group (normal control,normal saline 2ml/kg,n=30),SR group (simple radiation,8Gy,2ml/kg,n=30),GO group (Guiqiyiyuan ointment 11.83g/kg,radiation,8Gy,n=30).All the rats received intragastric administration for 7 days.The right side of the lung was modeled by 12C6+ beam radiation.After modeling,the rats were killed at 48h.The heart,liver and spleen were taken.The malonaldehyde (MDA),glutathione (GSH),glutathione peroxidase (GSH-Px),superoxide dismutase (SOD) contents were measured by colorimetry,DNA methylation rate was assayed by ELISA,and the expressions of Dnmt1,Dnmt3a and Dnmt3b were detected by immunohistochemistry.Results Compared with NC group,the contents of SOD,GSH and GSH-Px decreased (P<0.01),MDA increased (P<0.01),the level of DNA methylation decreased (P<0.01),and the expressions of Dnmt1,Dnmt3a and Dnmt3b increased in SR group (P<0.01).Compared with SR group,the contents of SOD,GSH and GSH-Px increased (P<0.01),MDA decreased (P<0.01),the level of DNA methylation increased (P<0.01),and the expressions of Dnmt1,Dnmt3a and Dnmt3b decreased in GO group (P<0.01).Dnmtl,Dnmt3a and Dnmt3b proteins were expressed in the cytoplasm of myocardial cells,hepatocytes and peripheral B cells of the white pulp in spleen,in all the groups.The color of NC group was light brown-brown,showing a weak positive expression.The color of SR group was brown-brown,showing a strong positive expression.The color of GO group was light brown-tan,showing a moderate positive expression.Conclusion The Guiqiyiyuan ointment can reduce the bystander effect caused by the 12C6+ beam radiation,and its mechanism is related to improving the oxidative stress reaction and the level of DNA methylation.

RESUMEN

Objective To compare the efficacy of atorvastatin,simvastatin and rosuvastatin in the treatment of hypercholesterolemia.Methods Selected 120 cases of patients with hypercholesterolemia who were treated in our hospital from January 2014 to December 2016,divided into atorvastatin group,simvastatin group and rosuvastatin group.The blood lipid levels and the incidence of adverse reactions were compared between the three groups before treatment,after treatment for fourth weeks and after treatment for eighth weeks.Results The serum lipid level were significantly improved in three groups after treatment (P < 0.05),and rosuvastatin group was significantly better than atorvastatin group and simvastatin group (P < 0.05);the incidence of adverse reactions of the three groups showed no significant difference.Conclusion Atorvastatin,simvastatin and rosuvastatin can effectively improve the blood lipid levels in patients with hypercholesterolemia,and the effect ofrosuvastatin is the best.

Eleven flavonol glycosides were isolated from the ethanol extract of Lysimachia clethroides by a combination of various chromatographic techniques including column chromatography over silica gel, Sephadex LH-20, and reversed-phase HPLC. Their structures were identified as astragalin (1), isoquercitrin (2), isorhamnetin-3-O-β-D-glucopyranoside (3), quercetin-3-O-β-D-6"-acetylglucopyranoside (4), quercetin-7-O-β-D-glucopyranoside (5), prunin (6), 2-hydroxynaringin-5-O-β-D-glucopyranoside (7), kaempferol-3-O-rutinonoside (8), kaempferol-3-O-robinobioside (9), rutin (10) and kaempferol-3,7-di-O-β-D-glucopyranoside (11). Among them, compounds 4, 7 and 11 were obtained from the Lysimachia genus for the first time, while compounds 3, 5 and 9 were firstly reported from this plant. In the preliminary assays, compounds 2, 6 and 8 possessed significant inhibition against aldose reduc- tase, with IC50 values of 2.69, 1.00, 1.80 μmol · L(-1), respectively; none of compounds 1-11 exhibited obvious cytotoxic activity (IC50 > 10 μmol · L(-1)).

RESUMEN

To study the chemical constituents of the 95% ethanol extract of Psidium guajava. Compounds were separated by using a combination of various chromatographic methods including silica gel, D101 macroporous resin, ODS, Sephadex LH-20 and preparative HPLC. Their structures were elucidated by physicochemical properties and spectral data Eighteen compounds were isolated and identified as (+) -globulol (1), clovane-2beta, 9alpha-diol (2), 2beta-acetoxyclovan-9alpha-ol (3), (+) -caryolane-1 ,9beta-diol (4), ent-T-muurolol (5), clov-2-ene-9alpha-ol (6), isophytol (7), tamarixetin (8), gossypetin (9), quercetin (10), kaempferol (11), guajaverin (12), avicularin (13), chrysin 6-C-glucoside (14), 3'-O-methyl-3, 4-methylenedioxyellagic acid 4'-O-beta-D-glucopyranoside (15), p-hydroxy-benzoic acid (16), guavinoside A (17) and guavinoside B (18). Compounds 2-9 and 14-16 were isolated from this plant for the first time. The ethanol extract showed 61.3% inhibition against the proliferation of colon cancer cell line SW480.

<p><b>OBJECTIVE</b>To explore the associated biomarkers influencing recurrence, metastasis and prognosis in patients with gastrointestinal stromal tumors (GIST) after complete resection.</p><p><b>METHODS</b>Tumor tissue samples of 148 patients with GIST undergoing complete resection from January 1990 to December 2008 in Sun Yat-sen University Cancer Center were collected. The expressions of Ki-67, E-cadherin, MMP7, CD44, nm23, P53, survivin, Cyclin D1, COX-2, and VEGF in tumor tissue samples were detected by tissue microarray and immunohistochemistry (IHC). The association of above factors expressions with recurrence, metastasis and prognosis was examined.</p><p><b>RESULTS</b>Log-rank test showed that Ki-67, E-cadherin, MMP7, CD44, P53 and survivin were associated to disease-free duration after complete GIST resection (all P<0.05), and the Ki-67, E-cadherin, P53 and survivin were associated to overall survival (all P<0.05). Cox multivariate analysis revealed that disease-free survival was associated with Ki-67, CD44 and P53 (all P<0.05), and the overall survival was only associated with Ki-67 (P<0.05).</p><p><b>CONCLUSION</b>Ki-67, CD44 and P53 are closely associated with recurrence and metastasis after complete GIST resection, and Ki-67 can predict the prognosis of GIST.</p>

RESUMEN

The Portable dynamic state electrocardiogram collecting system is introduced by using TMS302VC5402, TLC320AD50C, liquid crystal display model, and so on. This dissertation describes the work principle of the system and uses the united algorithm based on wavelet to identify and locate the ECG characteristic waves. This system has as follows of advantages: big memory, low noise,high common mode rejection ratio, the low power consume,the long record time etc.

<p><b>OBJECTIVE</b>To explore factors influencing acetabular development after closed reduction in developmental dislocation of the hip (DDH), to provide theoretical basis for improving the therapeutic effects of closed reduction treatment of DDH.</p><p><b>METHODS</b>A total of 100 children with single side DDH who were treated by closed reduction from January 2002 to December 2005 were followed up, and the AI and AI (D/W) of dislocation hip were measured at the 12th month after reduction. Ten factors such as gender, side, age, AI and AI (D/W) of dislocation hip before reduction, degree of dislocation, the rate of width of femoral head, femoral neck anteversion of dislocation hip before reduction, h/b rate,cut off adductor and skeletal traction et al were taken as independent variable, AI and AI (D/W) at the 12th month after reduction were taken as dependent variable,then multiple linear stepwise regression analysis was used to screen the major influencing factors.</p><p><b>RESULTS</b>Age, gender, degree of dislocation, h/b rate, the rate of width of femoral head and femoral neck anteversion exerted obviously influence on AI and AI (D/W),and then age,degree of dislocation and FNA had positive correlation with AI and negative correlation with AI (D/W); the rate of width of femoral head and h/b rate had negative correlation with AI and positive correlation with AI(D/W). The development of acetabular of female patients was faster than that of male patients.</p><p><b>CONCLUSION</b>Age, gender,degree of dislocation, concentric reduction or not, the development degree of femoral head before reduction and proximal femoral shape are the major factors influencing acetabular development.</p>

Through PCR amplification, 1.3 kb of 5'-proximal promoter (TA, 1.3 kb) of the beta-actin gene of white cloud mountain minnow Tanichthys albonubes was obtained. Using Genome Walker, a 1.7 kb 5'-upstream sequence from the proximal promoter of the beta-actin gene was isolated, and a further promoter (3.0 kb in size) was amplified according to the isolated 5'-proximal and upstream sequences (TLA, 3.0 kb). Both the 1.3 kb and 3.0 kb promoters contain elements that were critical to the transcription activity of other species, including the CCAAT Box (-89 approximately -85), CArG Box (-59 approximately -49), TATA Box (-26 approximately -20). Results of putative transcription binding sites analysis of the promoters by software TRANSFAC 6.0 revealed the presence of E-box, several transcript binding sites NF-Y, SP1 (Stimulating Protein 1), AP1 (Activator Protein 1), and some more transcription binding sites existing in the further promoter. The two promoter sequences were inserted into the expression vector to construct the recombinant expression vector, pTA-DsRed and pTLA-DsRed, respectively. The vectors were microinjected into the fertilized eggs of Tanichthys albonubes and higher positive rate was obtained and stronger red fluorescence was observed in pTLA-DsRed transgenic fish. RT-PCR analysis showed that RFP (Red fluorescent protein) mRNA level in pTLA-DsRed transgenic fish was 35.7% higher than that of the pTA-DsRed transgenic fish of 15-days-post-hatched. The present study showed that both the proximal and further promoter sequences have effective transcription activities and the 3.0 kb promoter possesses higher potent activity than that of the 1.3 kb promoter.

RESUMEN

Objective: To observe the short-term curative effect of radiofrequency ablation with active electrode cooling in the treatment of hepatocarcinoma. Methods: Radiofrequency ablation therapy was targeted to 117 tumor bodies of 43 patients with hepatocarcinoma using cooled-tip radiofrequency energy delivery machine (HG-3000). The change of tumor size was observed using CT and type B ultrasonic examination. The level of alpha-fetoprotein (AFP), carcinoembryonic antigen (CEA), the liver function were recorded and compared with pretreatment. Results: Radiofrequency ablation therapy significantly inhibited the growth of 117 tumor bodies. The AFP value obviously decreased compared with pretreatment (P<0.05); the CEA value had the tendency to continually increase. The CEA value had significant difference after 3-, 4-, and 5-month therapy compared with pretreatment (P<0.05). The liver function had obvious changes after 1- or 2-week therapy and recovered to the primary value before treatment after 4 to 5 weeks. Conclusion: The radiofrequency ablation with active electrode cooling may achieve satisfactory efficacy and has little complications.

<p><b>OBJECTIVE</b>To evaluate the effect of dynein inhibitor on mouse oocyte in vitro maturation and its cyclin B1 transcription level.</p><p><b>METHODS</b>Immature mouse oocytes were cultured in vitro with a known dynein ATPase activity inhibitor-sodium orthovanadate (SOV) to detect the changes of maturation rate, and semi-quantitative RT-PCR and single cell RT-PCR were performed to detect the changes of cyclin B1 mRNA level.</p><p><b>RESULTS</b>In dose-dependent experiments, the maturation rates of oocytes were significantly different between 5 micromol/L SOV and control groups (P < 0.05), and decreased with SOV increasing doses. However, the elevation of cyclin B1 mRNA level of immatured oocytes cultured for 12 h depended on SOV concentrations ranging from 50 to 500 micromol/L. In incontinuity exposed SOV experiments, the maturation rates of oocytes markedly reduced after the first incubation with 400 micromol/L SOV at least for 1 h and were first cultured in SOV-free medium for 4 h or 8 h before exposure to SOV (P < 0.05). In time-course experiment, the opposite changes of cyclin B1 mRNA level in oocytes between SOV and control groups were observed.</p><p><b>CONCLUSION</b>Dynein inhibitor might delay oocytes meiosis process, and cause ectopic expression of cyclin B1 in oocytes. Most Oocytes incubated with SOV blocked at germinal vesicles (GV) stage or M I to anaphase transition due to dynein dysfunction and ectopic transcription level of cyclin B1.</p>

RESUMEN

Cystatin, which widely distributed in both tissues and body fluids of animal and plant, was a superfamily of cysteine proteinase inhibitors. It could form activity-inhibitor complexes with cysteine proteinases to inhibit the hydrolytic activity of proteinases. Cystatin played important roles not only in the inhibition of the proteolytic degradation of fish muscle, but also in biological defense systems against invaders. To explore the functions of fish cystatin and the potential values in fish disease prevention and cure, as well as seafood processing, the recombinant yeast strains which could express Chinese sturgeon cystatin were constructed. First, the cystatin cDNA of Chinese sturgeon, which had been PCR modified, was subcloned into yeast integrated vector pPICZaA. After extracted and purified, the recombinant plasmids were linearized by Sac I. The yeast Pichia pastoris GS115 strain was transformed by use of the Lithium Chloride transformation method, and the recombinant cystatin yeast strains got. After 0.5% methanol induction, SDS-PAGE analysis of the culture supernatant indicated that the yield of recombinant cystatin was about 215mg x L(-1) with the percentage about 73.6%. The recombinant cystatin was purified through Q-Sepharose anion-exchange chromatography, and the purity reached about 94.2%. The inhibitory activity of recombinant cystatin was measured by inhibiting the proteinase activity of papain. The results showed that about 1 microg recombinant cystatin could inhibit the activity of 15 microg papain. Heat stability assay results showed that there was a decrease in inhibitory activity of cystatin with the increasing of temperature. When solution of recombinant cystatin was kept at 70 degrees C for 5min, the inhibitory activity reduced fast. While the recombinant cystatin was heated to 90 degrees C for 5min, the inhibitory activity of recombinant cystatin was undetected. The inhibitory activity for recombinant Chinese sturgeon cystatin was higher than that of CPI (cysteine proteinase inhibitor) from seeds of corn, that about 1 microg purified CIP could inhibited the activity of 0.278 microg papain. But the heat stability of recombinant cystatin is lower than that of the corn CPI. The expression level and the activity of recombinant cystatin from yeast Pichia pastoris were higher than those from E. coli. Moreover, recombinant cystatin from Pichia pastoris was easier to separate and purify. This paper reported that recombinant fish cystatin was produced in a highly efficient expression system based on the methylotrophic yeast, further work will focus on the function of recombinant Chinese sturgeon cystatin to resist fish disease and explore the value of cystatin as a food additive to inhibit cysteine proteinases during surimi processing.
