OBJECTIVES@#To establish the ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) method to detect ethanol metabolites phosphatidylethanol (PEth) in whole blood.@*METHODS@#An appropriate amount of aqueous solution including 1% formic acid was added to 100 μL whole blood, the protein was precipitated with acetone, centrifuged and the supernatant was purified and enriched by using Bond Elut Certify column. The eluent was redissolved with 1/1 isopropanol/acetonitrile (v/v) solution after nitrogen blowing and then tested by UPLC-MS/MS. Selective reaction monitoring scanning was carried out in negative ionization mode, and quantitative analysis was performed by external standard method.@*RESULTS@#PEth showed a linear relationship over the concentration range of 1-160 ng/mL in whole blood (r=0.999 9) with peak area. The detection limit was 0.2 ng/mL, the quantification limit was 1 ng/mL, the recovery rate was 97.43%-103.61%, the accuracy was 0.99%-1.77%, the intra-day precision was 0.4%-2.4%, and the inter-day precision was 1.1%-3.3%, and the matrix effect was 91.00%-99.55%. PEth was not detected in the in vitro blood samples supplemented with ethanol. PEth was detected positive in three drunk driving cases, and the concentration were 195.49, 83.67 and 876.12 ng/mL, respectively.@*CONCLUSIONS@#The established method has high sensitivity and specificity and the analysis results are accurate. It is suitable for the qualitative and quantitative analysis of PEth in whole blood.

Abstract@#The metabolic level of children and adolescents is of great significance in the healthy growth and development of children and adolescents. Irisin, as a myokine, was first discovered and named in 2012. Early studies showed that irisin can participate in body metabolism and inhibit the occurrence of obesity. Later studies found that irisin can improve the symptoms of type 2 diabetes mellitus, metabolic syndrome, osteoporosis and other diseases. At the same time, irisin can promote glucose, lipid metabolism and bone metabolism in children and adolescents, and its mechanism is still controversial. In this review, research update regarding the effect of irisin/FNDC5 on metabolism (lipid metabolism, glucose metabolism, bone metabolism) in children and adolescents were summarized in order to provide theoretical reference for researchers in the field of the metabolism of children and adolescents.

Multi-factor research design is widely applied in scientific research. It can simultaneously explore the effects of multiple factors on outcome indicators. The consideration of the interactive effects of different factors is a critical issue when analyzing this type of data. The analytic strategy for main effects or simple effects depends on the significance of the interactive effect. However, many researchers tend to skip the analysis on interactive effects, or wrongly select statistical analysis method because of ignoring the test result. In this study, SPSS 20.0 and R 3.6.1 statistical software were used to simulate and illustrate how to analyze data from two most popular multi-factor design data——factorial design and repeated measurement design. The significance of evaluating interactive effect and corresponding key point analysis was explained. The possible consequences of ignoring the statistical significance of interactive effects were indicated, that include leading to low inspection efficiency, prone to draw wrong conclusions, loss of valuable information in the original data, or loss of practical significance of the analytic results. It is suggested that in the analysis of research data, we should first judge whether there are interactive effects, and then correctly choose main effect analysis or single effect analysis to avoid one-sided and wrong conclusions.

AIM:To investigate the effects of Auricularia auricular-judae polysaccharide ( AAP) on pulmonary tissues of rats with LPS-induced acute lung injury (ALI) and its mechanisms.METHODS: Adult Sprague-Dawley rats were randomly divided into control group , LPS group, low-dose AAP group, middle-dose AAP group, high-dose APP group, and dexamethasone group.The rats were injected with LPS (8 mg/kg, ip) to induce ALI.The rats in the AAP groups were treated with AAP for 7 d before the induction of ALI .The protein concentration in the bronchoalveolar lavage fluid (BALF) was measured.The lung edema degree was measured by detecting the wet /dry weight ratio.The myeloper-oxidase ( MPO) , total antioxidant capacity ( T-AOC) , total superoxide dismutase ( T-SOD) , nitric oxide synthase ( NOS) and malondialdehyde ( MDA) levels were determined .The pathological changes of the lung tissues were evaluated by HE staining.RESULTS:Treatment with AAP significantly improved LPS-induced lung pathological changes , attenuated the protein concentration in the BALF and wet/dry weight ratio , inhibited the activities of MPO and NOS , reduced MDA level and increased the activities of T-AOC and T-SOD.CONCLUSION: AAP protects against LPS-induced acute lung injury in rats.

Objective:To observe the short-term therapeutic effect and safety of amlodipine combined terazosin on middle and old aged male patients with essential hypertension (EH).Methods:A total of 64 middle and old aged male EH patients treated in our hospital from Jan 2011 to Feb 2012 were enrolled.They were randomly and equally divided into amlodipine group and combined therapy group (received amlodipine combined terazosin treatment).Re-sults:Total effective rate of combined therapy group (93.75%)was significantly higher than that of amlodipine group (65.63%),P =0.028;there were significant reductions in levels of systolic blood pressure (SBP),diastolic blood pressure (DBP),total cholesterol (TC),triglyceride (TG)and fasting plasma glucose (FPG)in both groups after treatment (P <0.05~<0.01),compared with amlodipine group,there were more significant reductions in these indexes [SBP:(138.55±7.96)mmHg vs.(110.65±7.28)mmHg,DBP:(93.35±5.86)mmHg vs.(80.11 ±5.93)mmHg,TC:(5.67±0.76)mmol/L vs.(4.22±0.63)mmol/L,TG:(2.67±0.86)mmol/L vs.(2.01± 0.75)mmol/L,FBG: (5.69±0.86)mmol/L vs.(4.31±0.58)mmol/L]in combined therapy group,P <0.05 all;incidence rate of adverse reactions in combined therapy group (6.25%)was significantly lower than that of am-lodipine group (18.75%),P <0.05. Conclusion:Amlodipine combined terazosin possesses more therapeutic effect in middle and old aged male patients with essential hypertension.It can effectively reduce systolic and diastolic blood pressure;the adverse reactions are fewer,so it's worth extending in clinic.

<p><b>OBJECTIVE</b>To probe into a new therapy with less pain, good therapeutic effect and convenience for facial palsy in acute stage.</p><p><b>METHODS</b>Sixty cases were randomly divided into 2 groups, an observation group and a control group, 30 cases in each group. The observation group was treated with routine acupuncture plus moxibustion on temperature-sensitive points, and the control group with the routine acupuncture. Their therapeutic effects were assessed by the criteria for assessment of facial nerve function stipulated by Japanese Institute for Researching the Facial Nerve in 1997.</p><p><b>RESULTS</b>There were significant differences in cumulative scores of symptoms before and after treatment in the two groups (both P < 0.01) and there was a significant difference in the cumulative score of symptoms after treatment between the two groups (P < 0.01). The cured rate was 76.67% in the observation group and 50.00% in the control group with a significant difference between the two groups (P < 0.05). The total effective rates were respectively 93.33% and 76.67% in the two groups with no significant difference between the two groups (P > 0.05).</p><p><b>CONCLUSION</b>Both acupuncture combined with moxibustion on temperature-sensitive points and simple acupuncture have good therapeutic effects on idiopathic facial palsy, but the observation group is better than the control group.</p>

<p><b>OBJECTIVE</b>Observe therapeutic effect of suspended moxibustion on temperature-sensitive acupoints for chronic prostatitis, to explore a new therapy.</p><p><b>METHODS</b>Sixty cases were randomly divided into an observation group and a control group (n = 30 in each group). The suspended moxibustion on temperature-sensitive acupoints plus herbalenema were practiced in the observation group. The moxibustion at Guanyuan (CV 4), Zhongji (CV 3), Shenshu (BL 23) combined with herbalenema were practiced in the control group. The score of syndromes, the score of symptom indexes (NIH-CPSI), and WBC in prostatic secretion (EPS-WBC) were used as observation index for chronic prostatitis. The therapeutic effect was compared between the two groups.</p><p><b>RESULTS</b>The markedly cured rate was 50.0% in the observation group, better than that of 26.7% in the control group (P < 0.05). The score of syndromes, NIH-CPSI and EPS-WBC in the observation group were lower than the control group (P < 0.05, P < 0.01).</p><p><b>CONCLUSION</b>The therapeutic effect of suspended moxibustion on temperature-sensitive acupoints plus medication is better than moxibustion plus medication for chronic prostatitis in improvement of symptoms and EPS-WBC.</p>
