RESUMEN

Endometriosis is a female health disorder that occurs when cells from the lining of the uterus grow in other areas of the body. The cause of endometriosis is unknown. The purpose of this study was to investigate TP53 gene codon 72 polymorphism in women with endometriosis and compared it with healthy samples in Isfahan. We undertook a case-control study to examine the possible association of the TP53 gene codon 72 polymorphism with the risk of endometriosis in Isfahan. Ninety whole blood specimens from normal people as controls and ninety endometriosis specimens were analyzed. p53 codon 72 genotypes were identified using allele-specific polymerase chain reaction. Frequency of genotype Arg/Arg [Arginine/Arginine] in the samples of endometriosis was 28.9% and in healthy samples 42.2%. Frequency of genotype Pro/Pro [Proline/Proline] in the samples of endometriosis was 15.6% and in healthy ones. Frequency of heterozygote's Arg/Pro was 55.6% in endometriosis samples and 54.45% in healthy ones 3.3%. By comparing statistical genotype Pro/Pro with two other genotypes in both groups there was a statistical meaningful difference between control group and endometriosis group. [p=0.009, CI=95%, OR=5.34 [1047-19.29]]. Recent research shows that genotype Pro/Pro codon72 exon4 TP53 gene may be one predisposing genetic factor for endometriosis in Isfahan

Chromomycin A3 [CMA3] staining has been used to assess protamine deficiency. The aim of this study was to determine credibility of CMA3 along with semen parameters for assessment of fertility potential. Semen analysis and CMA3 staining were carried out on 234 fertile and 178 subfertile individuals. Semen analysis was assessed according to WHO criteria. Protamine deficiency was assessed by CMA3 staining. Means, range of variables, coefficients of correlation and receiver operating characteristic [ROC] analyses of semen parameters and protamine deficiency were determined. Mean values of three main sperm parameters and the percentage of sperm with negative CM A3 were significantly different between fertile and sub fertile groups. The results of CMA3 assessment showed significant correlation with sperm density, percentage of motility and normal morphology in the total population, while in the subfertile group the results of CMA3 showed significant correlation with sperm density and normal morphology. However in fertile men, the only significant correlation was observed between sperm with negative CMA3 and normal morphology. ROC analyses revealed that CMA3 staining has a higher potential to predict fertility status, compared to semen parameters. Assessment of protamine deficiency could be considered as one of the complementary tests along with semen analysis for assessment of fertility

RESUMEN

The aim of this study was to evaluate the effect of human sperm MTT viability assay on outcome of intracytoplasmic sperm injection. MTT is a tetrazolium salt, routinely used for cell proliferation and cytotoxicity assays.Material and 50microl of processed semen was treated with MTT solution, while the remaining used as the control. Meanwhile, 109 donated human oocytes [metaphase II] obtained from 12 patients were divided into two groups. Fifty five oocytes were injected using MTT positive sperms, while 54 oocytes were injected with sperms from the control group. Then the injected oocytes were cultured and observed at 18, 42, 66, 90, and 114 hours pos- ICSI. Finally, the fertilization and embryo development rates were compared in both groups.No significant differences were observed between fertilization and embryo development rates in the MTT and control groups.In future studies after approving that the MTT has not cytotoxic or teratogenic effects, then sperm MTT viability assay might be useful for ICSI in patients with absolute or severe asthenospermia or in patients with highly deformed sperm tails
