RESUMEN

Abstract The sexing of species is an important tool for population management. In tortoises and turtles, sexing is usually done according to secondary characteristics, such as plastron morphology or phallus eversion techniques. In this paper, we present the phallus eversion method and compare its efficiency to other existing techniques. This new method aims at shortening the management time and reducing the physical aftereffects on the animals, fostering the clinical and reproductive management of Geoffroys side-necked turtle (Phrynops geoffroanus) and related species. A sample of fifty individuals was taken in the central region of Piauí state, Northeastern Brazil. The proposed method consisted of moving the head from its resting position. The methods achieving a success rate higher than 80% were compared to each other as for the stimulation time required for phallus eversion. The method as here proposed achieved 100% efficiency in the sex determination of specimens with a linear carapace length between 10 and 34 cm. Also, when compared to the second most efficient method, it substantially reduced the time needed for managing the specimens (91%), avoiding possible sequelae in the individuals handled.

RESUMEN

Abstract Several species of Cichla successfully colonized lakes and reservoirs of Brazil, since the 1960s, causing serious damage to local wildlife. In this study, 135 peacock bass were collected in a reservoir complex in order to identify if they represented a single dominant species or multiple ones, as several Cichla species have been reported in the basin. Specimens were identified by color pattern, morphometric and meristic data, and using mitochondrial markers COI, 16S rDNA and Control Region (CR). Overlapping morphological data and similar coloration patterns prevented their identification using the taxonomic keys to species identification available in the literature. However, Bayesian and maximum likelihood from sequencing data demonstrated the occurrence of a single species, Cichla kelberi. A single haplotype was observed for the 16S and CR, while three were detected for COI, with a dominant haplotype present in 98.5% of the samples. The extreme low diversity of the transplanted C. kelberi evidenced a limited number of founding maternal lineages. The success of this colonization seems to rely mainly on abiotic factors, such as increased water transparency of lentic environments that favor visual predators that along with the absence of predators, have made C. kelberi a successful invader of these reservoirs.

Abstract This study evaluated the effect of the volatile oil of Alpinia zerumbet (VOAz) on caveolin-1 gene expression and muscular fibrosis. The rats were immobilized to induce fibrosis of the gastrocnemius muscle, and they were treated with VOAz. Collagen quality was assessed by histology and the expression of the caveolin-1 (CAV-1) gene was evaluated using qPCR. Histomorphological analysis indicated a significant reduction in the perimeter, width, and intensity of collagen in the treated groups, thus showing that the oil was effective in regulating the quality of collagen at the three concentrations. The results of expression levels suggested a decrease in the lesioned group and in two treatment groups (0.0115 µg/g and 0.009 µg/g). However, with the lowest concentration (0.0065 µg/g), no significant difference was observed, with levels similar to those found in healthy tissue. Therefore, the results showed that VOAz has the potential to be a non-invasive and low-cost alternative to aid in the treatment of muscular fibrosis.

Abstract Objective Existing scales that seek to measure alterations in self-experience were developed based on studies conducted in developed countries. Therefore, the aim of this study was to evaluate the psychometric properties of the Ego Dissolution Inventory (EDI) after translating and adapting it for the Brazilian context. Methods The measure was translated by two translators fluent in both English and Portuguese, followed by back-translation into English to ensure there was no loss of meaning. The scale was used in an online survey exploring substance use. A total of 528 participants answered the full scale. We calculated the Kaiser-Meyer-Olkin (KMO) measure to evaluate sampling adequacy, then ran exploratory factor analysis (EFAs) to investigate the factor structure of the EDI. Results The scale showed acceptable psychometric properties, with excellent internal consistency and sampling adequacy for factor analysis. Kaiser-Guttman's criteria and Hull's method indicated a three-factor solution, while parallel analysis suggested a two-factor solution. All items showed salient loadings, with two items exhibiting cross-loading. Positive but weak correlations were found between EDI factors 1 and 2 and nature relatedness. Conclusions The validated scale showed solid psychometric properties, with potential differences in factor structure in relation to the English version. Considering validation is an ongoing process, it is recommended that studies be conducted comparing ego dissolution scores across distinct substances and different regions of the country.

RESUMEN

ABSTRACT Purpose: To analyze the histology and distribution of abdominal testicular vessels in human fetuses Patients and Methods: We studied 19 fetuses (34 testes) ranging in age from 12 to 19 weeks post-conception. The fetuses were evaluated regarding crown-rump length (CRL), total length (TL) and body weight immediately before dissection. Each testis was dissected and embedded in paraffin, from which 5 µm thick sections were obtained and stained with Masson's trichrome and Anti-CD31 antibody to quantify the vessels. The stereological analysis was carried out with the Image Pro and Image J programs, using a grid to determine volumetric densities (Vv). Means were statistically compared using the unpaired T-test (p<0.05). Results: The fetuses presented mean weight of 222.5g, mean CRL of 15.3 cm and mean TL of 23.2 cm. All testes were in the abdominal position. The mean percentage of vessels (Vv) in the upper portion of the testis was 7.6% (4.6 to 15%) and in the lower portion the mean was 5.11% (2.3 to 9.8%), with a significant difference (p=0.0001). In the analysis between the upper portion of the right and left testes (p=0.99) and in the analysis of the lower portion of the right and left testes (p=0.83), we did not observe significant differences. Conclusion: The upper portion of the abdominal testis in human fetuses had a higher concentration of vessels than the lower portion. These results suggest that manipulation of the lower end of the testis during Fowler-Stephens surgery should be avoided in order to preserve the collateral circulation.

RESUMEN

The effect of brine on bacteria isolated from leafy vegetables was evaluated. Fresh waterleaf (Talinum triangulare), Pumpkin leaves (Telfairia occidentalis) and Bitter leaves (Vernonia amygdalina) including cooking salt were bought from vendors in Mile 3 market, Port Harcourt, Rivers State. Brine concentrations of 1, 2, 3, 4 and 5% were prepared by dissolving appropriate grams of salt in distilled water. Nine millilitres of the respective concentrations were transferred into clean test tubes, labelled, stoppered with foil and autoclaved at 121? for 15 minutes at 15psi. Sterile distilled water served as a control. The test isolates were standardized based on 0.5McFarland and 1mL each was introduced into different brine concentrations. The standard plate count was used to monitor brine effects on isolates and this was done hourly for six hours. Inoculated plates were incubated at 37? for 24 hours. After incubation, enumerated colonies were used to deduce isolate mortality. The total heterotrophic bacterial (THB) load of bitter, Pumpkin and water leaves were 3.49±0.4×106, 3.25±0.4×106 and 1.99±0.2×106 CFU/g, respectively. The staphylococcal counts for bitter, Pumpkin and water leaves were 1.65±0.3×104, 3.13±0.5×104 and 1.55±0.4×104 CFU/g, respectively. Total coliform counts for bitter, Pumpkin and water leaves were 1.52±0.8×105, 2.85±0.1×105 and 1.75±0.6×105 CFU/g, respectively. Staphylococcal counts of pumpkin leaves were significantly (P?0.05) higher than those obtained for bitter leaf and water leaf. There was no significant difference(P>0.05) in the THB and Coliform counts of all samples. E. coli was predominant in Pumpkin and water leaves while Staphylococcus sp was predominant in bitter leaves. The LC50 values for E. coli, Klebsiella, Staphylococcus, and Bacillus sp were; 5.39, 3.88, 1.62, and -0.41mg/ml, respectively. The LC50 showed that the brine was very lethal on Bacillus sp and Staphylococcus sp. High brine concentration is recommended to achieve reduced bacterial load.

RESUMEN

Infectious diseases such as typhoid fever lead to the formation of free radicals which can have a detrimental effect on the body. Typhoid fever is caused by poor sanitation, lack of clean water and resistance of germs to antibiotics and reactive oxygen species (ROS). Hence an urgent needs to find alternative treatments with little or no toxicity for the treatment of this disease. Objective: This work aims to evaluate the in vivo antisalmonellal and antioxidant activity of the 95% hydroethanolic extract of the leaves of Bauhinia rufescens (Fabaceae) in rats experimentally infected with Salmonella Typhi ATCC6539, as an alternative therapy. Methods: The rats were randomly divided into twelve groups (six per sex) of animals. Thus 3 control groups: (T0) uninfected and untreated; (T-) infected and untreated; (T+) infected and treated with ciprofloxacin (14 mg/kg) and 3 test groups: T1, T2 and T3 infected and treated with different doses of the extract (40, 80 and 117.71 mg/kg respectively). The evolution of the infection and the effectiveness of the treatment were monitored by blood culture, food consumption and weight growth were assessed during the trial; at the end of which the animals were sacrificed and the different parameters were evaluated. Results: Infected animals treated with different doses of the extract showed zero bacterial loads from the twelfth day post infection in both sexes. Regardless of sex, animals treated with the extract at the dose of 117.71mg/Kg were cured by the seventh day after the start of treatment while those treated with the doses of 40 mg/Kg and 80 mg/kg were cured by the ninth day after the start of treatment. Infection induced a significant (p < 0.05) decrease in food consumption and weight growth, while treatment induced, at all doses, an increase in food consumption and weight growth. Infection also caused a significant (p < 0.05) increase in NO and MDA levels, as well as a significant decrease in catalase and peroxidase activities in animal tissue homogenates. However, treatment resulted in a significant decrease in NO and MDA levels, and a significant (p < 0.05) increase in catalase and peroxidase activities. Conclusion: These results showed that the 95% hydroethanolic extract of Bauhinia rufescens leaves has mixed antisalmonellal and antioxidant activity in vivo and could be developed for the treatment of typhoid fever.

RESUMEN

Background: Despite control measures to curtailed salmonella fruit contamination over the years, pathogenic disease outbreaks caused by the ingestion of Salmonella contaminated fresh-cut-fruits pose a significant problem to human health by the consumption of fresh and minimally processed fruits. This study aimed to enumerate and determine the prevalence of Salmonella species isolated in ready-to-eat fruits vended in Bukuru Market Jos South, Plateau state. Methodology: A total of seventy-eight ready-to-eat vended fruit samples were purchased and cultured for the enumeration of bacterial isolates according to National Food Safety Standard for Microbiological Examination. Pulp pH value of each fruit was obtained by immersing litmus paper into the pulp and results recorded. Results: Of the 78 fruits specimen, 22 (28.2%) were Salmonella positive. The prevalence rates of salmonella isolated were found to be higher (22.7%) in both coconut and avocado followed by watermelon (18.2%) and sweetmelon (13.6%) as compared with other fruits in the study area. Lower rates of 4.5% were found in banana, pawpaw, and dates with a rise in apple with 9.1% respectively. Conclusion: The study showed a high p<0.05 (7.811) prevalence of pathogenic Salmonella species isolated in ready-to-eat fruits in the study area revealing that the spread of salmonella is not independent of fruits thereby suggestive of contamination made available by fruit vendors in this part of the world. Thus, epidemiological traceability and significant measures must be taken to check the safety of these vended products before consumption.

RESUMEN

Diabetes mellitus is associated with impaired wound healing. The topical use of insulin is a promising therapy because it may favor all phases of the wound healing process. This study aimed to investigate the therapeutic outcomes of insulin gel in wounds of hyperglycemic mice. After diabetes induction, a 1-cm2 full-thickness wound was created on each animal's dorsum. The lesions were treated daily for 14 days with insulin gel (insulin group) or vehicle gel without insulin (vehicle group). Tissue samples were extracted on days 4, 7, 10, and 14 after the creation of the lesion. The samples were analyzed with hematoxylin/eosin and Sirius red staining, immunohistochemistry, Bio-Plex immunoassays, and western blotting. Insulin gel favored re-epithelialization at day 10 and increased the organization and deposition of collagen. Additionally, it modulated the expression of cytokines (interleukin (IL)-4 and IL-10) and increased the expression of arginase I, VEGF receptor 1, and VEGF on day 10. Activation of the insulin signaling pathway occurred via IRβ, IRS1, and IKK on day 10 and activation of Akt and IRS1 on day 14. These results suggested that insulin gel improved wound healing in hyperglycemic mice by modulating the expression of inflammatory factors, growth factors, and proteins of the insulin signaling pathway.

Antimicrobial resistance remains a threat to patient safety and healthcare outcomes and largely arises from inappropriate antimicrobial prescriptions. This study aimed to determine the pattern of antibiotic prescriptions in the Paediatrics department of Rivers State University Teaching Hospital, Port Harcourt.Method:A point prevalence survey was conducted in the Paediatric wards and Special Care Baby Unit (SCBU) on 13 November 2021. Records of all children admitted before or at 8:00a.m. on the day of the survey were descriptively analysed using the protocol and web-based management system of the Global Point Prevalence Survey of Antimicrobial Consumption and Resistance, University of Antwerp. Results: The antibiotic prevalence in this study was 77.4%. The most common indication(s) for antibiotic use in SCBU was infection prophylaxis (81.3%) and in paediatric wards: Pneumonia, Ear Nose Throat and Soft tissue infections accounted for (23.1%) each. Third-generation cephalosporins and aminoglycosides were predominantly used in all wards and were empirical-based prescriptions. Regarding antibiotic quality indicators of prescriptions: In SCBU: 19 (90.5%) had indication(s) for antibiotics documented, 10 (46.7%) were guideline compliant, and 1 (4.8%) had documented review/stop date. In the paediatric medical and surgical wards, 17(85.0%) vs. 4(100%) had indication(s) for antibiotics documented, 6(30.0%) vs. 0(0%) were guideline compliant, and 1(5.0%) vs. 4(100.0%) had a review/ stop date. Conclusion:High prevalence of antibiotic use, suboptimal antibiotic quality indicators and absence of laboratory evidence for antibiotic prescriptions were observed in the paediatric units. There is a need to reorientate prescribers and institute strategic measures to improve antimicrobial stewardship

RESUMEN

Abstract The inheritance of the seedless fruit characteristic of Annona squamosa has not yet been explained. Molecular techniques may aid breeding programs, mainly in the assisted selection of the target gene. The INO gene may be related to seed development in these fruits. The objective of the present paper was to investigate the inheritance of seedlessness in the 'Brazilian seedless' sugar apple and INO gene conservation in Annona squamosa and Annona cherimola x Annona squamosa genotypes by assessing their homology with the INO database genes. The F1 generation was obtained by crossing the mutant 'Brazilian seedless' (male genitor) (P1) with the wild-type A. squamosa with seeds (M1 and M2, female genitors). The INO gene was studied in mutant and wild-type A. squamosa (P1, M1, M2 and M3) and in the Gefner atemoya (A. cherimola x A. squamosa) (M4) cultivar. The DNA was extracted from young leaves, and four sets of specific primers flanking the INO gene were amplified. The seedless characteristic was identified as stenospermatic in the fruits of parental P1, suggesting monogenic inheritance with complete dominance. High sequence similarity of the INO gene amplifications in the sugar apple accessions (M1, M2, M3) and the atemoya cultivar Gefner (M4) reinforces the hypothesis of their conservation.

Abstract The work aims were to describe the histological and histochemical structure of the gastroesophageal tube of Iguana iguana and verify the occurrence and distribution of immunoreactive serotonin (5-HT) and somatostatin (SS) cells. Fragments of the gastrointestinal tract (GIT) of five iguanas were which underwent standard histological and immunohistochemistry technique. Immunoreactive cells for 5-HT and SS were quantified using the STEPanizer. The oesophagus has ciliated columnar pseudostratified epithelium with staining Alcian blue (AB) + and goblet cells highly reactive to periodic acid Schiff (PAS). In the cervical oesophagus, the numerical density of 5-HT cells per unit area (QA [5-HT cells]/µm2) was 4.6x10-2 ± 2.0 and celomatic oesophagus presented QA = 4.0x10-2 ± 1.0. The epithelium of the stomach is simple columnar, PAS and AB +. The cranial and middle regions of the stomach presented (QA [5-HT cells]/µm2) = 6.18x10-2 ± 3.2 and the caudal region, QA = 0.6x10-2 ± 0.2. The SS cells were only observed in the caudal stomach, with numerical density (QA [SS cells]/µm2) = 1.4x10-2 ± 0.9 In I. iguana, variation was observed in terms of the distribution of mucus secretions and the pattern of occurrence of serotonin and somatostatin-secreting enteroendocrine cells in the TGI, which possibly will result in an interspecific adaptive response.

O epitélio do estômago é colunar simples, PAS e AB positivo. As regiões cranial e média do estômago apresentaram (QA [células 5-HT] / µm2) = 6.18x10-2 ± 3.2 e a região caudal, QA = 0.6x10-2 ± 0.2.

The inheritance of the seedless fruit characteristic of Annona squamosa has not yet been explained.
