Objective: To assess the effect of application of Biodentine (BD), Photobiomodulation (PBM) using 810 nm diode laser and both on the proliferation and odontogenic differentiation of human dental pulp stem cells (HDPSCs). Material and Methods: HDPSCs were collected, isolated, and characterized and then divided into six groups: groups 1, control; groups 2, biodentine (BD); group 3, irradiation at 1 J/cm 2 of 810-nm diode laser; group 4, irradiation at 1 J/cm 2 and culture with BD; group 5, irradiation at 2 J/cm 2, and group 6, irradiation at 2 J/cm 2 and culture with BD. Viability assay was measured through MTT assay and Alkaline phosphatase (ALP) enzyme activity and mRNA levels of RUNX2, collagen 1 (Col-1) and BMP2 were also assessed. Results: Photobiomodulation at 1 and 2 J/cm 2 combined with biodentine significantly promoted HDPSCs proliferation (in MTT assay results) and odontogenic differentiation (through the gene expression of RUNX2, Col-1 and BMP2 levels (p < 0.05). Conclusion: Photobiomodulation at 2 J/cm 2 combined with biodentine enhanced proliferation and odontogenic differentiation of cultured HDPSCs and thus could further be beneficial for dentin regeneration (AU)


Objetivo: Avaliar o efeito da aplicação de Biodentina (BD), Fotobiomodulação (PBM) usando diodo de laser de 810 nm e ambos na proliferação e diferenciação odontogênica de células tronco cultivadas da polpa dental (HDPSCs). Material e Métodos: HDPSCs foram coletadas, isoladas, caracterizadas e então divididas em seis grupos: grupo 1, controle; grupo 2, biodentina (BD); grupo 3, irradiação com diodo de laser a 1 J/cm2 de 810- nm; grupo 4, irradiação a 1 J/cm 2 e cultivo com BD; grupo 5, irradiação a 2 J/cm2, e grupo 6, irradiação a 2 J/cm2 e cultivo com BD. A viabilidade foi mensurada através do teste MTT e a atividade da enzima Fosfatase alcalina (ALP), e níveis de RNAm de RUNX2, de colágeno 1 (Col-1) e de BMP2 foram também mensurados. Resultados: Fotobiomodulação a 1 e 2 J/cm 2 combinada com biodentina promoveu significativa proliferação de HDPSCs (nos resultados do teste MTT) e diferenciação odontogênica (através da expressão genética dos níveis de RUNX2, Col-1 e BMP2 (p < 0.05)). Conclusão: Fotobiomodulação a 2 J/cm2 combinada com biodentina aumentou a proliferação e diferenciação odontogênica de HDPSCs cultivadas e dessa forma poderia ser benéfica para a regeneração dentinária. Objective: Endodontic perforation is a challenging mishap that should be repaired with a biocompatible material, Mineral trioxide aggregate (MTA) and Biodentine are the most commonly used repair materials. However, these materials are expensive, (MTA) has prolonged setting time and difficult manipulation. The purpose of this study is to prepare the experimental nano calcium-aluminate/tri-calcium-silicate (CA/C3S) material and comparing its physical properties with biodentine and MTA, to evaluate the experimental material eligibility to compete the commercial repair materials. And to perform part two (animal study) that will evaluate the cytotoxicity, the biocompatibility and the efficacy of (CA/C3S) in furcal perforation repair compared to diode laser. Material and Methods: A mixture of calcium carbonate and aluminum oxide was used to formulate calcium aluminate phase (CA), tri-calcium-Silicate phase (C3S) was formulated by firing of calcium carbonate and quartz. The produced powders were investigated by X-ray diffraction, then (CA) and (C3S) mixed with water.(CA/ C3S) compared with MTA and biodentine for setting-time, micro-hardness, dimensional-stability and solubility. Results: Mean setting time of (CA/C3S) was (32.70±0.75min) which is significantly higher than MTA and Biodentine. The Mean microhardness of (CA/C3S) was (56.50±7.41VHN) which has no statical difference with MTA and Biodentine. Solubility results showed weight increase for (CA/C3S) as following (6.29±3.05)and loss of weight for MTA and Biodentine. The percentage of change in dimensions for(CA/C3S) increased as following (0.64±0.78) while decreased for MTA and Biodentine. Conclusion: The experimental (CA/C3S) material showed good microhardness, dimensional stability and acceptable setting time that could be improved in further work (AU)


Objetivo: A perfuração endodôntica é um percalço desafiador que deve ser reparado com um material biocompatível, Agregado de trióxido mineral (MTA) e Biodentina são os materiais de reparo mais comumente usados. No entanto, esses materiais são caros, (MTA) tem tempo de presa prolongado e difícil manipulação. O objetivo deste estudo é preparar o material experimental de nano aluminato de cálcio/silicato tricálcico (CA/C3S) e comparar suas propriedades físicas com biodentina e MTA, para avaliar a elegibilidade do material experimental para competir com os materiais de reparo comerciais. E realizar a segunda parte (estudo animal) que avaliará a citotoxicidade, a biocompatibilidade e a eficácia do (CA/C3S) no reparo de perfuração de furca em comparação ao laser de diodo.Material e Métodos: Uma mistura de carbonato de cálcio e óxido de alumínio foi usada para formular a fase de aluminato de cálcio (CA), a fase tri-cálcio-silicato (C3S) foi formulada por queima de carbonato de cálcio e quartzo. Os pós produzidos foram investigados por difração de raios X, em seguida (CA) e (C3S) misturados com água. (CA/ C3S) comparados com MTA e biodentina para tempo de presa, microdureza, estabilidade dimensional e solubilidade. Resultados: O tempo médio de presa de (CA/C3S) foi (32,70±0,75min) que é significativamente maior que MTA e Biodentine. A microdureza média de (CA/C3S) foi (56,50±7,41VHN) que não tem diferença estática com MTA e Biodentine. Os resultados de solubilidade mostraram aumento de peso para (CA/C3S) conforme a seguir (6,29±3,05) e perda de peso para MTA e Biodentine. A porcentagem de mudança nas dimensões para (CA/C3S) aumentou como segue (0,64±0,78), enquanto diminuiu para MTA e Biodentine. Conclusão: O material experimental (CA/C3S) apresentou boa microdureza, estabilidade dimensional e aceitável tempo de presa, que pode ser melhorado em trabalhos futuros (AU)

Objective: Glucocorticoids induced osteoporosis and its related fragility fractures represent a costly human and socioeconomic load worldwide. All the current pharmacological therapies possess multiple adverse effects and high cost. Thus, the pesent study aimed to evaluate the bone healing ability of Moringa oleifera (MO) on glucocorticoids induced osteoporosis in the jawbone of Albino rats. Material and Methods: Osteoporosis was prompted by a daily intraperitoneal injection of 200 µg/ 100 g dexamethasone for 30 days. Next,the animals were randomly divided into 2 groups; osteoporotic and MO treated group. The treated group receivd a daily oral dose of 200mg/kg of MO. Rats from the MO group were sacrificed after 4 weeks from the beginning of treatment, and the same sacrifice date was used for the osteoporotic group. Bone regeneration was evaluated by dual energy x-ray absorptiometry (DEXA), real time polymerase chain reaction (RT-PCR), histopathological and histomorphometric examination. Results: After the sacrifice, the DEXA analysis revealed a significant upregulation in the BMD in the MO treated group (p <0.001). The RT-PCR test showed a significant decline in RANKL gene expression and a significant rise in OPG gene expression in the MO group (p < 0.001, p = 0.002, respectively). The histopathological examination of the MO group displayed a marked healing of the jawbone micro-anatomy. The histomorphometric analysis also showed that the bone area percentage increased significantly in the MO group (p <0.05). Conclusion: A cheap, easy to get, yet a powerful plant like MO leaves, can be cosidered an effective treatment for osteoporosis (AU).


Objetivos: A osteoporose induzida por glicocorticóides e suas fraturas por fragilidade relacionadas representam um custo humano caro e carga socioeconômica em todo o mundo. Todas as terapias farmacológicas atuais possuem múltiplos efeitos adversos e alto custo. Assim, o presente estudo teve como objetivo avaliar a capacidade de cicatrização óssea de Moringa oleifera (MO) em osteoporose induzida na mandíbula de ratos albinos. Material e Métodos: A osteoporose foi induzida por uma injeção intraperitoneal diária de 200 µg / 100 g de dexametasona por 30 dias. A seguir, os animais foram divididos aleatoriamente em 2 grupos; grupo tratado com osteoporose e MO. O grupo tratado recebeu uma diária dose oral de 200 mg / kg de MO. Os ratos do grupo MO foram eutanasiados após 4 semanas do início do tratamento, e a mesma data de eutanásia foi usada para o grupo osteoporótico. A regeneração óssea foi avaliada por espectrometria de raio-x de energia dupla (DEXA), reação em cadeia da polimerase em tempo real (RT-PCR), análise histopatológica e histomorfométrica. Resultados: Após a eutanásia, a análise DEXA revelou uma regulação positiva significativa na DMO no grupo tratado com MO (p <0,001). O teste RT-PCR mostrou um declínio significativo na expressão do gene RANKL e um aumento significativo na expressão do gene OPG no grupo MO (p <0,001, p = 0,002, respectivamente). O exame histopatológico do grupo MO revelou uma cicatrização acentuada da microanatomia do maxilar. A análise histomorfométrica também mostrou aumento significativo na porcentagem de área óssea no grupo MO (p <0,05). Conclusão: A MO é uma planta barata, de fácil obtenção, e suas folhas ainda podem ser consideradas poderosas como tratamento eficaz para a osteoporose. (AU)

Salivary gland tumors show complex histopathology and the treatment depends mainly on the stage of cancer. Induced pluripotent stem cells (iPS) have a great role in regenerative medicine as they can generate pluripotent stem cells from any available cell types as fibroblast. Thus, the aim of this work is to investigate the possible therapeutic effect of (iPS) on induced salivary gland cancer through evaluation of the silent information regulators of sirtuin-1 (Sirt-1), Tgf-β genes and their protein expressions in addition to LncRNA MALAT-1 expression. Thirty male albino rats were employed and divided into three groups (ten rats for each group), group 1 (control): Rats were injected with phosphate buffered saline (PBS), group II induced squamous cell carcinoma (SCC): rats were injected with squamous carcinoma cells (SCC), group III (induced SCC/iPS): SCC treated rats treated with 5 × 106 iPS cells. Submandibular specimens were taken and prepared for histological, histochemical and immunohistochemical studies for Bax and TGF-β3 protein. Also, Real time PCR was performed for Sirt-1, Tgf-β, and MALAT-1 LncRNA genes expression. SIRT-1 and TGF-β protein level expression was assessed by western blot technique. Group III (iPS treated group) revealed more or less normal acinar structure with normal rearrangement of acini and normal intralobular ducts with an increase in their number. In the iPS treated group there was increasing in the amount of mucopoly saccharide in the acinar cells and intensity of BAX immunostaining while, TGF-β3 was decreased in its intensity in comparison to that of the cancer treated group. In addition to Sirt-1, Tgf-β, and MALAT-1 LncRNA expressions were increased in cancer group compared to iPS treated and control groups. Induced pluripotent stem cells play a potential therapeutic role in treatment of induced submandibular gland carcinoma. Aims: To assess the occupational risk factors that contribute to the occurrence of work- related musculoskeletal disorders among Operation Room (OR) nurses and MSDs relation to biomarker c-telopeptide of type II collagen (CTX-II). Study Design: A cross sectional study. Place and Duration of Study: Cairo University hospitals from January to April 2014. Methodology: The study was conducted upon 184 OR nurses from different specialties. They include 155 females and 29 males with age ranged 20-50 years and work duration ranged 2-35 years. They represent 52.6% of the OR nurses work power all over Cairo University hospitals. The individuals were subjected to questionnaires including Standardized Nordic Questionnaire (SNQ), Job Content Questionnaire (JCQ) and Quick Exposure Check (QEC) for ergonomic risk assessment. Urinary CTX-II was measured. Results: Out of 184 OR nurses, 180 have musculoskeletal symptoms with a prevalence of 97.8%. CTX-II in females was non-statistically significant higher than in males. Higher non-statistically significant levels of CTX-II were found in those aged ≥ 40 years than in those < 40. Non- statistically significant differences were in correlation between CTX-II and various occupational risk factors as BMI, age, duration of employment, physical risk factors except for outreached arm (r= 0.16, p=0.03) and psychosocial factors except for deficient rest break (r=-0.15, p=0.038) which showed statistically significant differences. No statistically significant correlation was found between CTX-II and any of musculoskeletal symptoms during the past twelve months by applying SNQ. A statistically significant difference (Z=2.26, p=0.02) was found in OR nurses group with knee crepitus in comparison with nurses group without knee crepitus among those with high level of urinary CTX-II. Other MSDs showed no statistically significant differences between symptomatizing and non-symptomatizing groups. Conclusion: Operating room nurses are exposed to multiple occupational risk factors leading to MSDs. CTX-II may be a valuable biomarker in early osteoarthritis.
