RESUMEN

OBJECTIVE:To study the effects of Mahuang xixin fuzi decoction on Toll-like receptors (TLRs)response and cytochrome C oxidase (Cyt-CO)-mediated apoptosis regulation in mice with influenza disease of kidney-yang deficiency. METHODS:Totally 48 male Balb/c mice were randomly divided into normal group (n=12)and modeling group (n=36). The modeling group was intraperitoneally injected with estradiol benzoate solution (8 mg/kg)and intranasally injected with influenza virus H 1N1(20 μL/mice)to establish the influenza disease compound model of kidney-yang deficiency. After modeling ,the mice were randomly divided into model group ,positive drug group (Oseltamivir phosphate capsules ,0.195 g/kg),Mahuang xixin fuzi decoction group (1.802 g/kg,by crude dru g),with 12 mice in each group. Each group was given relevant medicine intragastrically, normal group and model group were given, corresponding volume of normal saline intragastrically 20 mL/kg,once a day ,for consecutive 6 days. During admi-nistration,body weight and anal temperature of mice were mail:xsy407861520@163.com measured daily ;the percentage of initial body weight was calculated. After last medication ,the organ (spleen, thymus and lung )indexes were calculated ;the pathological changes of lung tissue were observed. The viral load of influenza A virus H 1N1 in lung tissue was detected (reflected by M gene mRNA expression);mRNA expressions of TLR3,TLR7,myeloid differentiation factor (MyD88)and Caspase- 3 in cardiac tissue as well as the activity of Cyt-CO and the content of cytochrome C (Cyt-C)were also determined. RESULTS :Compared with normal group, initial body weight percentage and anal temperature of the model group continued to decrease (P<0.05);the spleen and thymus indexes were decreased significantly (P<0.05),while lung index was increased significantly (P<0.05);the lung tissue lesions were serious. Viral load in lung tissue ,mRNA expressions of TLR 3,TLR7,MyD88 and Caspase- 3 in cardiac tissue as well as the content of Cyt-C were increased significantly (P<0.05 or P<0.01),while the activity of Cyt-CO in cardiac tissue was significantly decreased (P<0.01). Compared with model group ,initial body weight percentage and anal temperature of mice in Mahuang xixin fuzi decoction group showed an increasing trend from the fourth day of administration (P<0.05 or P<0.01). The spleen and thymus indexes were increased significantly (P<0.05),while the lung index was significantly decreased (P<0.05);the pathological injury of lung tissue was significantly improved ;viral load in lung tissue ,mRNA expressions of TLR 3 and Caspase- 3 as well as the content of Cyt-C in cardiac tissue were decreased significantly (P<0.05 or P<0.01),while the activity of Cyt-CO was increased significantly in cardiac tissue (P<0.01). CONCLUSIONS :Mahuang xixin fuzi decoction can improve influenza disease of kidney-yang deficiency in mice ,the effect may related to inhibit TLRs response and apoptosis regulation pathway mediated by Cyt-CO.

RESUMEN

OBJECTIVETo observe the clinical efficacy of transcutaneous electrical acupoint stimulation (TEAS) at Hegu (LI 4) and Neiguan (PC 6) on treatment and prevention of postoperative sore throat (POST) after tracheal intubation under general anesthesia.

METHODSOne hundred patients who received elective thyroid gland lobectomy with gradeⅠand Ⅱ of American Society of Anesthesiologists criteria were randomly assigned into a TEAS group and an anesthesia group according to random number table method, 50 cases in each group. All the patients were treated with tracheal intubation under general anesthesia. Patients in the TEAS group were treated with TEAS (2 Hz/100 Hz, 8 to 12 mA) at Hegu (LI 4) and Neiguan (PC 6) from 30 min before anesthesia induction to the end of operation. Patients in the anesthesia group were treated with TEAS at the same acupoints but no electrical stimulation was given. The incidence rate, severity and visual analogue scale (VAS) of POST were recorded 1h, 6h, 12h and 24h after tracheal extubation, respectively.

RESULTSThe incidence rate of POST was 12.0% (6/50), 22.0% (11/50) and 18.0% (9/50) 1h, 6h, 12h after tracheal extubation respectively in the TEAS group, which was significantly lower than 30.0% (15/50), 42.0% (21/50) and 36.0% (18/50) in the anesthesia group (all<0.05). However, the incidence rate was not significantly different between the two groups 24h after extubation[14.0% (7/50) vs 28.0% (14/50),>0.05]. Moreover, the VAS scores of the patients with POST in the TEAS group at each time point were lower than those in the anesthesia group (all<0.05).

CONCLUSIONSTEAS at Hegu (LI 4) and Neiguan (PC 6) can effectively reduce the incidence rate and severity of POST induced by tracheal intubation under general anesthesia.

RESUMEN

The latest study shows that the expression abnormalities of programmed death 1/programmed death ligand 1 (PD-1/PD-L1) signaling pathway in a variety of solid tumor tissues and cells are related to disease progression and poor prognosis, and directly involve in tumor immune escape, becoming the recent focus. With the further research, PD-1/PD-L1 signaling pathway in blood diseases, for example, in the development of lymphoma, is catching more attention gradually. The signaling pathway not only is associated significantly with malignancy, progression, recurrence of lymphoma and prognosis of patients, but probably becomes a new target for cancer immunotherapy. This paper summarizes the expression and research status of the biological characteristics and mechanism of PD-1/PD-L1 signaling pathway in lymphoma, in order to provide a new way for lymphoma treatment.

RESUMEN

Objective:To analyze clinical characteristics, treatment, and prognosis of B-cell lymphoma, unclassifiable, with features in-termediate between diffuse large B-cell lymphoma and Burkitt lymphoma (DLBCL/BL). Methods:The clinical and pathological data of 13 DLBCL/BL patients, who were treated in the First Affiliated Hospital of Zhengzhou University between January 2013 and December 2014, were collected. Overall survival (OS) and progression-free survival (PFS) were estimated by the Kaplan-Meier method. Through the log-rank test, survival curves were compared among groups classified by clinical stage, age, serum lactate dehydrogenase (LDH) lev-el, international prognostic index (IPI) score, or first chemotherapy regimen. Results:Among the 13 patients with DLBCL/BL, 12 pa-tients showed extra-nodal involvement. The median OS and PFS were only 10 and 6 months, respectively. Univariate analysis showed that the LDH levels and IPI scores exerted statistically significant effects on prognosis. Some borderline differences in survival were not-ed among the CHOP, CHOP-like, and intensive chemotherapy groups. Conclusion:DLBCL/BL is an aggressive B-cell lymphoma with a short survival time. The majority of patients presented extra-nodal involvement. DLBCL/BL did not respond well to CHOP or CHOP-like regimen, and more intensive chemotherapy may improve survival. Elevated LDH levels and high IPI scores were predictors of poor sur-vival.

RESUMEN

Background Retinal ichemic-reperfusion injury (RIRI) affects vision and life quality and has no ideal treatment outcome up to now.The mechanism of RIRI is associated with apoptosis.Researches showed that subretinal transplantation of bone mesenchymal stem cells (BMSCs) can relieve RIRI,but its mechanism is unclear. Objective This study was to observe the influence of BMSCs subretinal transplantation on the expressions of apoptosis protein Fas/FasL,caspases-3 after RIRI,and to explore the neuroprotecitve mechanism of BMSCs transplantation to treat RIRI.Methods Bone marrow was isolated and extract form femur and tibia of SD rat by the density gradient centrifugation method and then the BMSCs were collected.BMSCs suspension was prepared using DMEM containing low-glucose with the cell density 1 x 106-2× 106/ml.RIRI models were established by ligation of optical nerve (Daniel M method).Sixty-four SD rats were randomly divided into the normal control group, model control group,BMSCs transplantation group and PBS control group.Twenty-four hours after RIRI,BMSCs suspension with the cells density 5× 104 cells was subretinally injected in the rats of the BMSCs transplantation group, and equal volume of PBS was injected in the same way in the rats of the PBS control group. The expressions of Fas/FasL and caspase-3 were dynamically detected using immunohistochemistry at 6,24,48 and 72 hours after BMSCs transplantation. Results The positive response cells for Hoechst33324 were seen in subretina 72 hours after BMSCs transplantation.A few of positive cells for Fas,FasL,caspase-3 proteins were expressed in the rats of the normal control group at 6,24,48 and 72 hours after BMSCs transplantation. However, the positive cells for Fas,FasL,caspase-3 protein were significantly increased in the model control group and the BMSCs transplantation group compared with the normal control group at various time points after injection (all at P<0.01),and the number of positive cells was significantly less in the BMSCs group than those of the RIRI group and the BMSCs transplantation group (P<0.05,P<0.01).No significant differences were found in the numbers of positive cells of Fas,FasL and caspase-3 proteins at various time points between the model control group and the PBS control group (all at P>0.05).Conclusions The subretinal transplantation of BMSCs down-regulate the expression of Fas/FasL and caspases-3 proteins in RIR1 rats, and thus alleviate the apoptosis of retinal neural cells.
