RESUMEN

Breast cancer is a malignant tumor with high mortality, and multidrug resistance (MDR) mediated by ABCG2 (ATP-Binding cassette G2) is an important cause of chemotherapy failure. It is an urgent problem to explore the mechanism of ABCG2-mediated drug resistance and its key molecules. Epithelial cell adhesion molecule (EpCAM) is involved in multiple tumor drug resistance and is closely related to breast cancer MDR. However, its role in ABCG2-mediated breast cancer drug resistance has not been clarified. The purpose of this study was to explore the regulation of EpCAM on ABCG2-mediated MDR in breast cancer cells and its mechanism. CCK8 cytotoxicity assays confirmed that the drug resistance of MCF-7/MX cell line to mitoxantrone (MX) was significantly increased compared with MCF-7 drug-sensitive strain of human breast cancer. Western blotting results showed that ABCG2 was highly expressed and EpCAM was up-regulated in MCF-7/MX cells compared with MCF-7. SiRNA knockdown of EpCAM in MCF-7/MX cells down-regulated ABCG2 expression and restored sensitivity to MX. Cell morphology was observed under an inverted microscope, and it was found that knocking down EpCAM reduced cell-cell connections between MCF-7/MX cells. The co-localization of EpCAM and claudin 1 in MCF-7/MX cells was observed by immunofluorescence. Furthermore, Western blotting results showed that EpCAM knockdown reduced claudin 1 expression in MCF-7/MX cells. In conclusion, EpCAM may promote ABCG2-mediated mMDR in breast cancers by enhancing intercellular tight junctions through interaction with claudin 1.

RESUMEN

BACKGROUND:Cycloserine with low hepatotoxicity exhibits no cross-resistance with the existing anti-tuberculosis drugs,and has been commonly used for the treatment of drug-resistant tuberculosis.However,its oral administration or injection leads to a certain degree of neurotoxicity.OBJECTIVE:To prepare poly(lactic-co-glycolic acid) (PLGA)-cycloserine sustained-release microspheres which are expected to reduce the neurotoxicity and adverse reactions,and maintain the drug concentration in the bone tuberculosis region for a long time,and to observe the in vitro drug release of the microspheresMETHODS:Double emulsion solvent evaporation method was used to prepare PLGA-cycloserine microspheres that were bonded into sponge implant by Bletilla striata polysaccharide extract.Then,morphology,particle size,encapsulation efficiency and in vitro performance of the microspheres were observed.The drug loading,burst release,appearance and dispersion of the microspheres were observed at 0,1,2 months after the microspheres were placed in room temperature (25 ℃),high temperature (60 ℃) and high humidity (93%),respectively.RESULTS AND CONCLUSION:The PLGA-cycloserine microspheres that were round and spherical presented with the mean particle size of (143±38) μm,the drug loading of 38.38% and the encapsulation efficiency of 67.54%.No burst release occurred,and the cumulative release of drug within 50 days was 65.62% After being stored at room temperature,high temperature and high humidity for 1 and 2 months,the microspheres were intact in the appearance and morphology,and showed insignificant changes in drug loading and burst release.To conclude,the time of degradation and the release of drug accord with the biological requirements of bone restoration.

RESUMEN

BACKGROUND: The technology of biodegradable materials covering growth factors can be used to make sustained-release microspheres, which provides the feasibility for the efficient utilization of growth factors. OBJECTIVE: To prepare nano/micron-sized spheres using recombinant human bone morphogenetic protein 2/poly(lactic-co-glycolic acid) (rhBMP-2/PLGA)copolymer and to compare their release behaviors by in vivo and in vitro release experiments. METHODS: The rhBMP-2/PLGA nano/micron-sized spheres were prepared by emulsion solvent evaporation method to control the rate of pulp mixing.(1)In vitro release experiment:Prepared nano/micron-sized spheres were dissolved in PBS for 70 days,and then ELISA method was used to detect the rhBMP-2 concentration in the supernatant at different time.(2)In vivo release experiment:Forty-four New Zealand rabbits were divided into two groups, and rhBMP-2/PLGA nano/micron-sized spheres were respectively implanted into trochanteric defects. The concentration of rhBMP-2 in the defect site was detected by ELISA within 70 days after implantation. RESULTS AND CONCLUSION:In vitro sustained release experiment:There was a sudden release of nanospheres in the former 3 days,and the cumulative release nearly reached 41%, followed by a steady and slow release, and then the cumulative release was up to approximately 83% at 70 days. The initial release of micron-sized spheres was less than that of nanospheres, and the cumulative release was about 20% within the former 3 days and reached to 70% at 70 days.In vivo sustained release test:There was a sudden release of the nanospheres,the cumulative release was nearly 35%, followed by a steady and slow release, and then the cumulative release was up to approximately 72% at 70 days. The initial release of micron-sized spheres was less than that of nanospheres, and the cumulative release was about 21% in the former 3 days and increased to about 63% at 70 days.In both in vivo and in vitro release experiments,the release duration of micron-sized spheres was longer than that of nanospheres in the former 3 days. To conclude, the release time of rhBMP-2/PLGA micron-sized spheres fulfills the need of bone growth cycle, therefore, rhBMP-2/PLGA micron-sized spheres are more favorable than rhBMP-2/PLGA nanospheres for bone defect repair in clinical practice.

RESUMEN

Objective To study the effects and mechanism of Glyphosate induced cell cycle arrest and apoptosis in mouse spermatogenic cells(GC-2).Methods Morphology of the GC-2 cells was observed by optical microscope and Transmission Electron Microscopy.Flow cytometry were used to detect cell cycle arrest and apoptosis.Cell cycle arrcst and apoptosis related proteins were detected by Western blot.Results Glyphosate could inhibit cell proliferation and impair endoplasmic reticulum and mitochondria significantly,which was associated with G1 phase arrest and apoptosis in GC-2 cells.Glyphosate lead to cell cycle arrest and apoptosis by regulating the expressions of G1/S phase checkpoint regulatory factor proteins such as p21,CyclinD1,CyclinE and activating caspase3.Conclusion Glyphosate has adverse effects on male germ cells by inducing cell cycle arrest and apoptosis.

OBJECTIVE@#To explore effect of different anesthesia methods and different anesthetics on erythrocyte immune function in mice.@*METHODS@#The mice were anesthetized by isoflurane and ether inhalation, and also under intraperitoneal anesthesia with sodium pentobarbital and chloral hydrate. Blood was collected from the ventro-cardinal vein. Automatic blood cell analyzer was used for routine blood examination, and the canthine oxidase method was used to measure the superoxide dismutase (SOD) activity. Lipid peroxidation product malondialdehyde (MDA) was measured with TBA, and glutathione peroxidase (GSH-Px) was measured with DTNB, and then the effect of different anesthesia methods and different anesthetics on erythrocyte immune function in mice was observed.@*RESULTS@#Hct level of chloral hydrate intraperitoneal injection group was significantly higher than the other three groups (P<0.05). And the MDA levels in the pentobarbital sodium group were significantly higher than the other three groups (P<0.05). SOD and GSH-Px of the chloral hydrate and sodium pentobarbital intraperitoneal injection group were significantly lower than the other two groups; RBC-C 3bRR and RBC-ICR of the chloral hydrate and sodium pentobarbital intraperitoneal injection group were significantly lower than the other two groups.@*CONCLUSIONS@#Different drugs can induce changes in immune function of mice at different levels. Isoflurane and ether have less damage to animal body, while chloral hydrate and sodium pentobarbital intraperitoneal injection have a certain inhibitory effect on the animal body respiratory system and can cause greater damage to the body. Therefore, the reasonable selection and control of anesthetics are very important in order to avoid the experimental errors caused by anesthesia.

RESUMEN

<p><b>OBJECTIVE</b>To assess the association between single nucleotide polymorphism in M-type phospholipase A2 receptor (PLA2R) gene and membranous nephropathy (MN) in a Chinese Han population.</p><p><b>METHODS</b>A total of 430 non-related Chinese Hans were enrolled, which included 145 patients with idiopathic membranous nephropathy (IMN), 53 patients with secondary MN and 232 normal controls (NC). The polymorphism of rs35771982 in PLA2R gene was determined with polymerase chain reaction-restriction fragment length polymorphism assay. Serum anti-PLA2R antibodies were detected by Western blotting.</p><p><b>RESULTS</b>The genotypic and allelic frequencies for rs35771982 was significantly different among the three groups (P=0.004; P<0.001). CC genotype and C allele were significantly more common in IMN group compared with NC group (P=0.002; P<0.001) or secondary MN group (P=0.011; P=0.001). In the IMN group, the CC genotype was correlated with serum albumin (Alb), 24-hour urine protein (24h UP) and positive rate of serum anti-PLA2R antibody (P<0.001, P<0.001, P=0.010), and was a risk factor for IMN (OR=8.927, 95%CI:2.107-37.821, P=0.003).</p><p><b>CONCLUSION</b>The CC genotype and C allele at rs35771982 in PLA2R gene are associated with susceptibility to IMN in Chinese Hans. The associations between CC genotype and severity of IMN as well as serum anti-PLA2R antibody have indicated that production of anti-PLA2R autoantibody in IMN patients is associated with mutation at the rs35771982 locus of PLA2R gene.</p>

RESUMEN

Objective To study the self-adaptive anti-control of a single H-R neuronal system transformed from periodic dynamics pattern to chaos. Methods Numerical analysis was performed by adding sine function self-delay feedback while coupling intensity and time-delay, respectively, as the control parameters. Results By numerical simulation and analysis, it was found that in a certain range of the combination of coupling intensity and time-delay, the time-interval sequences of the dynamical pattern of a single H-R neuronal system could be controlled from a periodical pattern of 3 spikes onto chaos and other periodical patterns. Conclusions The method of self-adaptive feedback of sine function is effective for the anti-control of H-R neuron, and the coupling intensity and time-delay are both important parameters. The particular self-adaptive dynamics of information identification to neuron is reflected in the control process.

<p><b>OBJECTIVE</b>To investigate the therapeutic effect of digital dorsal flap based on vascular network for skin defects at fingers.</p><p><b>METHODS</b>From August 2005 to August 2010, 49 cases with 61 skin defects at fingers were treated with the digital dorsal flaps, including 19 at the index fingers, 19 at the middle fingers, 16 at the ring fingers and 3 at the little fingers. The defects were 42 at the distal end, 12 at the middle segment and 7 at the proximal segment. The flaps size ranged from 27 mm x 20 mm to 10 mm x 8 mm.</p><p><b>RESULTS</b>61 flaps in 49 cases survived completely. 52 fingers were followed up for 6-22 months. The aesthetic and functional results were both satisfactory. The flaps sensation was graded as S3, with a discrimination distance of 6-9 mm (median, 7.6 mm). No complication happened in donor sites.</p><p><b>CONCLUSIONS</b>The digital dorsal flap based on vascular network has reliable blood supply with less morbidity to the main artery. It is safe and easily performed with good effect for skin defect at finger.</p>

<p><b>OBJECTIVE</b>To explore a new method for repair of concurrent skin and nerve defect at palm and carpal on ulnar side.</p><p><b>METHODS</b>From April 2000 to August 2009, five cases with concurrent skin and nerve defect at palm and carpal on ulnar side were reconstructed with free medial plantar flaps. Palmar nervous proprii defect at ulnar side of little finger was repaired by the first toe tibia nervous proprii in one case. The superficial branch of radial nerve was applied to repair the defect of ulnar nerve, as well as its deep or superficial branch in two cases. The superficial branch of radial nerve was also used to repair the defect of superficial branch of ulnar nerve, common palmar digital nerve of the fourth finger, Little finger ulnar palmar nervous proprii in one case. The dorsal branch of ulnar nerve was applied to repair the defect of superficial branch of ulnar nerve, common palmar digital nerve of the fourth finger, little finger ulnar palmar nervous proprii in one case. The vascular bundle of medial plantar flap was anastomosed with ulnar vascular bundle. The wounds at donor sites were covered with free skin grafts which were obtained from upper leg.</p><p><b>RESULTS</b>All the flaps and skin grafts were survived completely. The five patients were followed up for six months to four years with no muscular atrophy or claw hand deformity. The esthetic result was satisfied. The Sensory of flaps and fingers recovered to S3 to S3+. The two-point discrimination distance on flaps was range from 7 mm to 10 mm. The postoperative comprehensive evaluation was excellent in the cases whose superficial and deep branches of ulnar nerve were repaired.</p><p><b>CONCLUSIONS</b>Free medial plantar flap is an effective method to repair concurrent skin and nerve defect at palm and carpal on the ulnar side.</p>

RESUMEN

<p><b>OBJECTIVE</b>To investigate the therapeutic effect of free tissue flap anastomosed with reverse descendant branch of lateral femoral circumflex artery for severe soft tissue defect at leg.</p><p><b>METHODS</b>The severe soft tissue defect at leg, without any vessels for anastomosis of free tissue flap, was reconstructed with free tissue flap, which was anastomosed with proximal end of descendant branch of lateral femoral circumflex artery and great saphenous vein. From Oct. 2004 to Dec. 2009, 36 cases were treated with 15 cases of latissimus dorsi musculocutaneous flaps, 12 cases of anterolateral femoral flaps, and 9 cases of thoracoumbilicus flaps.</p><p><b>RESULTS</b>All the 36 free flaps survived completely. The patients were followed up for 6 months to 2.5 years with good cosmetic results.</p><p><b>CONCLUSIONS</b>It is effective and practical to repair the severe soft tissue defects at legs with the reverse descendant branch of lateral femoral circumflex artery to carry the free flaps.</p>

RESUMEN

<p><b>OBJECTIVE</b>Development of new methods, ELISA and immunostrip test, for the diagnosis of nasopharyngeal carcinoma.</p><p><b>METHODS</b>The engineering purified antigens coat plate or absorb on nitrocellulose filter. The plate and diagnostic strips carrying antigens were used for detection of IgG antibody in the sera from nasopharyngeal carcinoma patients and outpatients patients.</p><p><b>RESULTS</b>127 cases sera from nasopharyngeal carcinoma patients were parallel detected TK/IgG antibody by ELISA and immunostrips. The TK/IgG antibody are all positive in the 127 cases of nasopharyngeal carcinoma patients. 55 cases show positive by ELISA, 58 cases positive by immunostrips in 247cases sera from outpatient. The antibody positive rate to early antigen p54 lower then to TK. Conclusion ELISA and imuunostrips are sensitive and specific means for detection of the IgG antibody to TK of EBV and the diagnosis of nasopharyngeal carcinoma.</p>

RESUMEN

<p><b>OBJECTIVE</b>To discuss the application of reverse flap based on two dorsal metacarpal artery for reconstruction of degloved fingertip avulsion.</p><p><b>METHODS</b>From Jan. 2005 to Mar. 2008, 28 cases with degloved fingertip avulsion were treated with reverse flaps based on two dorsal metacarpal artery. The defects were located distal to the distant interphalangeal joints and were 0.8-2.2 cm in length. 10 defects was in the index fingers, 13 in the middle fingers and 5 in the ring fingers. 24 fingers were treated in an emergency surgery. 4 fingers were treated due to skin necrosis.</p><p><b>RESULTS</b>Epidermal necrosis happened at the distal end of flaps in 3 cases. All the other flaps survived completely. 25 cases were followed up for 4-27 months with satisfactory cosmetic and functional results. The 2-points discrimination distance was 6.0-9.0 mm (average, 7.6 mm).</p><p><b>CONCLUSIONS</b>The reverse flap based on two dorsal metacarpal artery is easily performed and reliable for degloved fingertip avulsion with satisfactory results.</p>

<p><b>OBJECTIVE</b>To investigate the therapeutic effect of island flaps pedicled with digital artery for the defects at the end of fingers.</p><p><b>METHODS</b>63 cases with 72 soft tissue defects at the end of fingers were treated with the island flaps. The flap size ranged from 8 mm x 11 mm to 21 mm x 27 mm.</p><p><b>RESULTS</b>All the flaps survived completely. The follow-up period ranged from 4 to 19 months. The functional and cosmetic results were satisfactory. The two-point discrimination was 7 to approximately 9 mm. The TAM functional examination showed excellent in 59 fingers, good in 11 fingers and common in 2 fingers.</p><p><b>CONCLUSIONS</b>The technique with island flap is simple and reliable for the defects at the end of fingers.</p>

RESUMEN

To investigates the condition and the mechanism of the formation of mummy in Shanghai region. Three cases of mummy were normaly examinated. It showed that the formation condition of mummy were influenced by season, scene, and the corpse keeping methods etc. In warm and damp southern region in Shanghai, the mummy formation depended on lower temperature, dry environment season, dehydrated and hunger before death, and interfered factors like ventilating and turning over corpse frequently.
