RESUMEN

Objective:To investigate the effect and potential mechanism of serum exosome-derived cirC_0009362 on the osteogenic differentiation of human bone marrow mesenchymalstem cells (hBMSCs) .Methods:Serum samples from patients with osteoporosis (OP) were collected and exosomes were isolated. The expression level of circ_0009362 in exosomes was detected by qRT-PCR. hBMSCs osteogenesis was induced and the expression of circ_0009362 and miR-29b-3p was detected. The interaction between circ_0009362 and miR-29b-3p was detected by dual luciferase reporter assay. Alkaline phosphatase (ALP) kit was used to detect ALP activity and alizarin red (ARS) staining was used to detect calcium deposition.Results:Compared with healthy control group, the expression of circ_0009362 in serum exosomes of OP patients was increased, and the expression of circ_0009362 was decreased after inducing hBMSCs osteogenesis (all P<0.05) . The ALP activity and the percentage of calcium deposition in hBMSCs were decreased by exosomes, and this effect was achieved by secreting circ_0009362. The effect of exosomes was partially offset by circ_0009362 expression in knockdown exosomes (all P<0.05) . The expression of miR-29b-3p was increased after inducing hBMSCs osteogenesis ( P<0.05) . Circ_0009362 had a targeting relationship with miR-29b-3p, and exosomes inhibited the expression of miR-29b-3p by secreting circ_0009362. The ALP activity and the percentage of calcium deposition in hBMSCs were promoted by overexpression of miR-29b-3p, which was partially offset by exosomes (all P<0.05) . Conclusion:Serum exosomes of OP patients inhibit the osteogenic differentiation of hBMSCs by secreting circ_0009362 to down-regulate the expression of miR-29b-3p.

Objective To investigate the correlation of serum(SF)and hepcidin(Hepc)level with cardi-ac function and chronic heart failure(CHF).Methods A total of 80 elderly patients with CHF who had received treatment in our hospital from October 2015 to February 2017 were assigned to CHF group and 80 patients without heart failure were assigned to a control group. Serum SF and hepidin levels in both the groups were measured and compared.Results Serum hepidin and hs-C-reactive protein levels were all significantly higher in CHF group than in the control group[(69.58 ± 27.16)ng/ml vs.(128.46 ± 33.28)ng/mL;(97.16 ± 16.81)ng/mL vs.(54.12 ± 15.76)ng/mL;(3.3 ± 1.64)mg/L vs.(2.78 ± 1.36)mg/L].Multiple logistic regression analysis showed that SF was an independent protective factor for CHF(P < 0.05);Hepc and hs-CRP were independent risk factors for CHF (P<0.05).Conclusions Serum hepcidin and hs-C-reactive protein may be risk factors for chronic heart failure. Measurement of serum hepidin and hs-CRP levels is helpful for early prevention and treatment of heart failure.

RESUMEN

Objective To explore the association of osteoporosis with blood pressure variability in elderly patients with hypertension complicated by osteoporosis, as well as its mechanisms. Methods A total of 189 elderly patients with hypertension were selected. According to the diagnostic criteria for osteoporosis, the patients were divided into three groups. 24-hour ambulatory blood pressure and IL-6 were detected in all the patients, and then compared among different groups. Results For the three group, there were statistical differences in levels of IL-6 and blood pressure variability (P < 0.05). Blood pressure variability was negatively correlated with BMD (P < 0.05). 3. IL-6 was positively related with blood pressure variability (P < 0.05), but had a negative correlation with BMD (P < 0.01). IL-6 had significant statistical differences among different sexes (P < 0.01). Conclusions Osteoporosis is closely associated with blood pressure variability in the elderly patients. IL-6 and blood pressure variability may play a synergistic role in the process of osteoporosis; and IL-6 may be a risk factor for postmenopausal osteoporosis in women.

RESUMEN

Data were gathered from epidemiological survey of Xinjiang Hetian area in 2008 and 2 228 subjects were included.Waist-height ratio (WHtR),body mass index (BMI),Waist-hip ratio (WHpR) were calculated.Kappa test was applied to determine the concordance among different methods.The results showed that according to International Diabetes Federation 2005 diagnostic criteria for metabolic syndrome,it was better to apply WHtR in predicting metabolic syndrome by the receiver operating characteristic (ROC) curve than BMI,abdominal circumference,and WHpR.When the cutoff of WHtR was 0.53,the sensitivity and specificity in diagnosing MS were 91.2% and 71.8 %,with its area under ROC curve 0.878.Kappa test showed an intensive concordance between WHtR and waist circumference (WC).The specificity and sensitivity of diagnosing MS would be significantly raised by using both WHtR and WC.

RESUMEN

<p><b>OBJECTIVE</b>To investigate the association of the 1891-1892del TC polymorphism of regulator of G-protein signalling 2 (RGS2) gene with hypertension in Xinjiang Kazakh population.</p><p><b>METHODS</b>The case-control study was performed in 444 cases and 489 controls. The genotypes of the individuals in the 1891-1892del TC locus were identified by TaqMan method.</p><p><b>RESULTS</b>A significant association was observed between the1891-1892TC insertion/deletion polymorphism with hypertension in men (OR=1.698, P=0.03) and in the total population (OR=1.32, P=0.044). The mean systolic blood pressure and serum uric acid levels of the ID+DD carriers were significantly higher than that of the II carriers (adjusted, P=0.04 and P<0.01).</p><p><b>CONCLUSION</b>The results suggest that the D allele of the 1891-1892TC insertion/deletion locus of the RGS2 gene might be an independent risk factor for hypertension in Xinjiang Kazakhs; and the polymorphism may have some influence on serum uric acid level in this population.</p>
