RESUMEN

Objective To explore the application of nerve ultrasound score (DCEC score) in the diagnosis of diabetic peripheral neuropathy. Methods Seventy-three patients with type 2 diabetes were divided into subclinical diabetic peripheral neuropathy group (n=16) and diabetic peripheral neuropathy group (n=58). DCEC score was obtained from all participants. The cut-off value of DCEC score was defined by the receiver operating characteristic (ROC) curve, and its sensitivity, specificity, accuracy, positive likelihood ratio and negative likelihood ratio were analyzed. Results When the DCEC score was ≥14.5, 50 cases of diabetic peripheral neuropathy were diagnosed. The sensitivity was 81.0% , the specificity was 80.0% , the accuracy was 80.8% , the positive likelihood ratio was 4.05, and the negative likelihood ratio was 0.24. Conclusion DCEC score can effectively diagnose diabetic peripheral neuropathy which can be used as a new method to diagnose diabetic peripheral neuropathy.

RESUMEN

PURPOSE: This study was done to identify the status of smartphone addiction among nursing students and to examine the mediating effects of Internet ethics on the relationship between smartphone addiction, physical symptoms and psychological well-being. METHODS: Data were collected between May 20 and June 23, 2014. Participants were a convenience sample of 214 nursing students of one university in Seoul. Structured questionnaires were used to measure degree of smartphone addiction, physical symptoms, psychological well-being and Internet ethics. Data was analyzed using Independent t-test, One-way ANOVA, Pearson correlation coefficient, multiple hierarchical regression. RESULTS: Of the nursing students, 14.5% had high level of risk of smartphone addiction and 16.8% had potential risk. Higher frequency of physical symptoms and lower level of Internet ethics were related to the higher level of smartphone addiction. Voluntary-control in Internet ethics had mediating effects on the relationship between smartphone addiction and physical symptoms. CONCLUSION: The results suggest that the level of smartphone addiction may lowered in nursing students by intensifying Internet ethics especially voluntary-control. Based on these findings, Internet ethics program for nursing students should be developed to facilitate the effective use of smartphones.

RESUMEN

Objective To discuss the diagnosis and treatment of the solid pseudopapillary tumor of the pancreas(SPTP). Methods Six consecutive cases operated in our hospital from Aug. 2008 to Apr. 2010were reviewed. Results Those cases aged ranging from 17 to 31 years, with an average age of 21.8 years. The maximal dimension of tumor ranged from 6.6 to 13.0 cm, averaging 8.2 cm. Those tumors originated from pancreatic head (2), and body or tail (4). Abdominal B-ultrasound or CT scan examination disclosed that pancreas had solid-cystic mass in 4 cases, and the other two had solidmass. All tumors were resected completely and defined pathologically. We performed 5 open operations, including pancreaticoduodenectomy (2 cases), spleen-preserving distal pancreatectomy (2 cases) and distal pancreatectomy plus splenectomy (1 case). One case underwent laparoscopic distal pancreatectomy plus splenectomy. Conclusions The young females are more likely suffered from the SPTP than others, which usually has a clear boundary and large volume. The abdominal B-ultrasound and CT scan are important diagnostic methods. The surgical modality of pancreatectomy is dependent on the tumor's location and extent, furthermore the resectability of SPTP is high. It is possible to perform a spleen-preserving distal pancreatectomy for tumors located in pancreatic body or tail.

RESUMEN

Objective To observe the enhanced inhibitory effect of adenovirus (Ad)-mediated HCCS1 combined with 5-FU on the growth of LoVo cells, and to explore the molecular mechanisms. Methods RT-PCR and Western blot were used to detect the expression of HCCS1 in LoVo cells infected with Ad HCCS1. CCK-8 assay was applied to observe different inhibitory effects of different treatments on growth of LoVo cells. The apoptotic rates were detected by using flow cytometry. The apoptotic proteins were detected by using Western blot. Results ① The recombinant adenovirus, Ad HCCS1, could trigger the expression of HCCS1 in LoVo cell. ② In comparison with controls (92.23%±3.77%), the cell viability rate of LoVo was only (11.23±4.61 )% on 96 h after the combination treatment of 5-FU and Ad-HCCS1 (P<0. 01). ③ The apoptotic rate was (27.57±1.78)% on 72 h after the combination treatment, which was higher than that in 5-FU treated cells (8.64±0.94)%, Ad-HCCS1 treated cells (13.19±1.32)% and 5-FU Ad treated cells (12.16±1.28)%, (P<0. 01). ④ Cathepsin D was only detected in Ad HCCS1-infected cells. When treated with 5-FU, the procaspase-8 was decreased and the cleaved Bid was increased in cytosol. The lowest level of Bax and the highest level of cytoso C and cleaved caspase-3 were detected in cytosols of 5-FU+Ad HCCS1 treated cells. Conclusion The inhibitory and proapoptotic effects are significantly enhanced in LoVo cells when treated with Ad-HCCS1+5-FU. The key protein of the cross-talk is Bax and these data provided a new strategy to treat colorectal carcinomas.

RESUMEN

Objective To investigate the protective effects of α-1 antitrypsin on human islets injured by protease released from pancreas exocrine cells. Methods ( 1 ) in vivo experiment. Parts of the cadaveric pancreas was digested with collagenase, islets were selected artificially, and pancreatic exocrine cells were collected. 8-9 weeks olds male BALB/c-Nu nude mice were induced into diabetic mice with STZ (240 mg/kg body weight, i. p) and randomly divided into two groups: the control group (n = 6), 250 islets were transplanted into left kidney subcapsule of diabetic nude mice; cotransplant group (n = 7), 250 islets and the equal volume of pancreatic exocrine cells were transplanted into different regions of left kidney subcapsule. Blood glucose level was monitored. Nephrectomies were performed after 28 days. The expression of anti-amylase antibodies in subcapsule was detected by using immunohistochemical staining. (2) Islets culture: Three groups were randomly set up. Group 1: purified islet group, 250 islets were incubated into a 6-well culture plate; Group 2: non-purified islet group, 250 purified islets and equal volume of exocrine cells were incubated; Group 3: nonpurified islet + Al AT group, 250 purified islets and equal volume of exocrine cells were incubated with α-1 antitrypsin added (0. 5 mg/ml). After 48 h, insulin content of islets in each well and trypsin concentration in the supernatant of each well were measured. Results 10000 islets were collected. After islets transplantation, the blood glucose levels in control and co-transplant groups were normal, but a delayed islet function in reversing diabetes was in the co-transplant group, and ehe mice in both groups became hyperglycemic after nephrectomy. A large number of anti-amylase antibody-positive cells were found in renal subcapsule in the co-transplant group while little seen in the control group.Insulin levels in the non-purified islet group were decreased as compared with purified islet group, those in the non-purified islet group + A1AT group were higher than in the non-purified islet group, but lower than in the purified islet groups. Trypsin concentration in the non-purified islet group was increased as compared with purified group, that in the non-purified islet group + A1AT group was lower than the non-purified islet group, but higher than in the purified islets group (all P<0. 01).Conclusion Protease released from acinar cells during pancreatic digestion has detrimental effect on islet function after transplantation. Co-cultivation of islets and pancreatic exocrine cells with A1AT added can prevent islet cell damage caused by trypsin.

RESUMEN

To investigate the effect of NVP-DPP728, a DPP-Ⅳ inhibitor on new-onset diabetes and the autoimmune response in non-obese diabetic ( NOD ) mice. Diabetes could be reversed in 75% of NVP-DPP728 treated 20 NOD mice. In these 15 mice with remission, insulitis scores were significantly lower than those of the control group. The percentage of Tregs was increased in the thymus and celiac lymph nodes, plasma TGF-β1 and GLP-1 were also significantly increased ( P<0. 01 ). NVP-DPP728 treatment may reverse new-onset diabetes in NOD mice by reducing insulitis and increasing Tregs.

RESUMEN

Objective To screen and identify photosynthetic bacteria that could be used to treat bean wastewater. Methods Photosynthetic bacteria (PSB) were screened by enrichment culture from the sludge. The morphological and biochemical characteristics of the bacteria were determined using transmission electron microscope, live cells'scanning and testing of utilization of different carbon sources. In addition, the sequence of 16S rRNA of the PSB was analysed and the homology was compared with the standard strain according to the sequence in the GenBank. Results A strain NS-04 was screened from the sludge of the pond in Nanjing. The bacteria was Gram-negative in shape of ovoid or rod, with size of 0.6-0.9 ?m wide and 1.2-2.0 ?m long. The multiplication manner of the bacteria was binary fission. The photosynthetic membrane was lamellar type, which was under and parallel to intro-cytoplasmic membrane. The clone was brown with major pigments riched in carotenoids and bacteriochlorophylla. The bacteria could grow both under anaerobic condition in the light and aerobic condition in the dark. The bacteria could utilize many kinds of carbon sources and the growth of the cells could be prompted by yeast extract. The analysis of phylogenetic tree of 16S rRNA sequence indicated that the similarity of strain NS-04 and the standard strain of Rhodopseudomonas palustris in GenBank was 97%. Conclusion According to Bergey's Manual of Systematic Bacteriology, strain NS-04 was identified to be Rhodopseudomonas palustris.
